ABSTRACT

OBJECTIVE:To study the effects of Purple frute scens leaves polysaccharides (PPLPs)on oxidative stress and PI3K/AKT/GLUT4 signaling pathway of pancreatic tissues in diabetes mellitus (DM)model mice. METHODS :Totally 60 mice were given intraperitoneal injection of STZ (60 mg/kg)to induce DM model. The 40 successful modeling mice were randomly divided into model group ,metformin group (positive control ,200 mg/kg),PPLPs high-dose and low-dose groups (400,200 mg/kg),with 10 mice in each group. Another 10 healthy mice were selected as the normal group (normal saline ). They were given relevant medicine intragastrically ,once a day ,for consecutive 28 days. During the experiment ,general information and body weight of mice were observed ;oral glucose tolerance (OGTT)test(determining FBG at 0,30,60,120 min after giving 40% glucose solution )was conducted. After last medication ,the changes of related blood glucose indexes (FBG,FINS,ISI,IRI), blood lipid indexes (HDL-C,LDL-C,TC,TG)and oxidant stress indexes (MDA content and the activities of SOD ,CAT, GSH-Px)as well as the protein expressions of PI 3K,p-AKT and GLUT 4 in pancreatic tissue were determined. RESULTS :During the experiment ,compared with normal group ,the mice were slow in action ,the feed consumption and water consumption increased,and body weight significantly increased in model group (P<0.05). 0,30,60,120 min after giving glucose ,the FBG content of mice were all increased significantly (P<0.05). After last medication ,the contents of FINS and HDL-C in serum as well as ISI ,the activities of SOD ,CAT and GSH-Px as well as the protein expressions of PI 3K,p-AKT and GLUT 4 in pancreatic tissue were all decreased significantly (P<0.05);the contents of FBG and LDL-C ,TC and TG in serum as well as IRI , 疗。E-mail:sunguangping83@163.com MDA content in pancreatic tissue were all increased significantly(P<0.05). Compared with model group ,the general condition and OGTT of mice in each administration group was improved;the contents of FINS and HDL-C in serum as well as ISI ,the activities of SOD ,CAT and GSH-Px as well as the protein expressions of PI 3K,p-AKT and GLUT 4 in pancreatic tissue were all increased significantly (P<0.05);the contents of FBG,LDL-C,TC and TG in serum as well as IRI ,MDA content of pancreatic tissue were decreased significantly (P<0.05). CONCLUSIONS:PPLPs has anti-diabetic effects ,which are related to reducting oxidative stress level and promoting the activation of PI 3K/AKT/GLUT4 signaling pathway.

ABSTRACT

Objective Idiopathic membranous nephropathy(IMN)is a common cause of nephrotic syndrome in adults,and the prognosis of IMN varies among individuals. The aim of this study was to investigate the prognosis and risk factors,especially the prognostic indicators in kidney patholo?gies for IMN. Methods Patients diagnosed as IMN by renal biopsy from January 2006 to December 2011 were collected and those who finished at least 12 months of follow?up were enrolled in this study. The clinical and renal pathological indicators were collected and the correlation between the pathological indicators and the prognosis was analyzed. Results Totally 137 patients were enrolled in this study,18 patients(13.14%)reached the follow up ending point(defined as eGFR<30 mL/min). The prognosis of kidney function was poor in IMN patients with interstitial fibrosis,tubular atrophy and severe vascular sclerosis and those who were older at disease onset and demonstrated to have higher MAP,poorer renal function,intersti?tial fibrosis,tubular atrophy and worse vascular sclerosis. Conclusion Renal pathological characteristics during renal biopsy were correlated with the renal prognosis of IMN. Severe interstitial fibrosis,tubular atrophy and vascular sclerosis were identified as risk factors for the poor prognosis of re?nal function in IMN patients.

ABSTRACT

Objective To investigate whether sodium ferulate(SF)can prevent contrast-induced renal injury in chronic renal insufficiency patients and its possible mechanism.Methods Eighty-six patients with chronic renal insufficiency,whose serum creatinine(Scr)levels ranged from 132 μmol/L to 358 μmol/L were randomly divided into control group(saline 100 ml iv drip)and SF treatment group(treatment group,saline 100 ml + 0.3 g SF iv drop)based on sufficient hydration.Patients were treated with the above-mentioned protocols at 3 h be-fore the administration of contrast medium and in the following 3 days,Scr,plasma malondialdehyde(MDA)and urine nitric oxide(NO)were measured before angiography and on the third day after angiography.Results The incidence rate of contrast-induced nephropathy(CIN)in control group was significantly higher than that in treatment group(P 〈 0.05).The contrast medium increased Scr levels and plasma MDA,and decreased urine NO excretion,while SF treatment attenuated the above changes(P 〈 0.05).Conclusion The preventive effect of SF on con-trast-induced renal injury might be associated with its suppressive effect on endothelin,increasing renal production of NO and anti-oxydation.

ABSTRACT

Objective To understand the effect of anti-cysticercus therapy for patients with cerebral cysticercosis and the changes of cysticercus on CT image after treatment. Methods The patients with cerebral cysticercosis were classified by the presentation of their brain CT image before treatment, then the effect of anti-cysticercus therapy on them after treatment was analyzed and the presentations of their brain CT images between before and after treatment were compared. Results There were different changes on CT image of cysticercus in brain tissues after anti-cysticercus therapy for different types of patients with cerebral cysticercosis. Type Ⅰ: the focus was absorbed completely after treatment in the majority of patients and calcificated in the minority. Almost all the patients were cured clinically after anticysticercus therapy. Type Ⅱ: the focus was absorbed completely in the minority, and one to two or more calcification dots were observed in the majority of patients. Anti-cysticercus therapy was effective. Type Ⅲ and Ⅳ: the absorption of focus was not very good and the effect of anti-cysticercus therapy was lower relatively. Conclusion The changes of CT image such as absorption, calcification, has important significance in forecasting prognosis and instructing clinical usage.
