ABSTRACT

Objective@#To investigate the impact of overweight and obesity on knee joint biomechanics during running in children and to provide theoretical support for scientific exercise prescription and reduced risk of exercise injury in overweight and obese children.@*Methods@#Fifteen children aged 7-11 years old with normal weight (healthy group) and overweight/obesity (overweight/obesity group) were selected from June to August 2020. Participants ran through the force measuring platform at a speed of [3.5×(1-5%)~3.5×(1+5%)]m/s. The kinematic and dynamic data of the knee joint were collected simultaneously by the Simi Motion motion capture system and the Kistler three dimensional force measuring platform, and the surface area and density of the tibial plateau were measured by peripheral quantitative computed tomography. The difference of knee joint angle, impact mechanics and knee joint torque based on tibial plateau dimension were compared between the two groups.@*Results@#The knee abduction peak angle of the overweight/obesity group[(6.14±4.16)°] was higher than that of the healthy group[(2.57±1.36)°] ( t=-3.16, P < 0.05 ). Based on the standardization of tibial plateau dimension, the impact mechanics (peak vertical ground reaction force, peak impact force, maximum load rate and average load rate) and knee joint torque (knee flexion, extension, adduction peak torque) in the overweight/ obesity group were higher than those in the healthy group ( t=-4.26, -4.52, -2.97, -2.74, -2.17, -4.27, -3.70, P <0.05).@*Conclusion@#Overweight/obese children show abnormal running mechanics and knee joint load. Higher joint load may indicate increased risk of anterior cruciate ligament injury among overweight/obese children.

ABSTRACT

OBJECTIVETo study the promoter methylation status of SFRP genes and the effect of 5- aza- 2'- deoxycytidine (5- Aza- CdR)induced apoptosis via Wnt/β- catenin pathway by demethylation in Jurkat cells.

METHODSJurkat cells were treated with different concentrations of 5- Aza- CdR. The cell proliferation level of Jurkat cells was detected by MTT assay. Apoptosis was evaluated by flow cytometry. Methylation- spcific PCR (MSP) was used to determine the methylation status of SFRP genes. The expressions of SFRP genes were detected by real time fluorescence quantitative PCR. The mRNA expression levels of survivin, c- myc and cyclin- D1 were analyzed by RT- PCR. Western blot was used to detect the levels of β-catenin protein.

RESULTSCompared with control group, the different concentrations of 5-Aza-CdR could significantly inhibit the proliferation of Jurkat cells in a time-dose dependent manner (P<0.05). After being treated by 5- Aza- CdR for 48 hours, the cell early apoptosis rate in experiment group was significantly higher than that in control group (P<0.05). The promoters of SFRP1, SFRP2, SFRP4, SFRP5 genes were hypermethylation state in the control group, after being treated by 5-Aza-CdR for 72 hours, the brightness of SFRP1, SFRP2, SFRP4, SFRP5 genes' methylation strips weakened in a dose- dependent manner. SFRP mRNA expression increased (P<0.05) when 5- Aza- CdR concentration increased, and the level of β- catenin protein was dampened in a dose- dependent manner (P<0.05). As compared to the control group, the mRNA expressions of associated apoptosis genes survivin, c-myc and cyclin- D1, respectively were obviously down- regulated in a dose- dependent manner (P<0.05).

CONCLUSIONThe effect of demethylation could up- regulate SFRP genes expressions by reversing its hypermethylation and induced apoptosis by down-regulation of β-catenin and associated apoptosis genes.

ABSTRACT

Graphene/Room temperature ionic liquids ( GN/IL ) nanocomposite was prepared by grinding graphene and ionic liquid 1-butyl-3-methylimidazolium bromide ( BMIMPF6 ) which mixed together in appropriate proportion. Atomic force microscopy ( AFM ) was utilized to characterize the formation of the GN-ILs. Due to the synergistic effect between ionic liquids and graphene, the nanocomposite exhibited excellent performance toward H2 O2 reduction. A novel uric acid ( UA) electrochemical sensor was fabricated based on uricase-GN/IL modified glassy carbon electrode. The experimental results showed that the response displayed a good linear response toward UA in the concentration range from 0. 002-4. 5 mmol/L. The corre1ation coefficient was 0. 995 and the detection limit was 0. 85 μmol/L. The easily prepared electrochemical sensor had favorable stability and selectivity and could be applied to the quick determination of UA in human serum, thus providing a new UA detection method for clinical trial.

ABSTRACT

Objective To investigate the relationship between promoter hypermethylation of secreted frizzled-related protein (SFRP) gene and acute leukemia (AL). Methods We examined the promoter methylation starus of SFRP1, 2, 4 and 5 in primary or relapsed AL patients, cell lines ( HL60,NB4, Molt-4 and Jurkat) and peripheral blood mononuclear cells from healthy people with methylationspecific PCR (MSP). Results None of the normal mononuclear cells showed methylation of any SFRP genes. The frequencies of aberrant methylation among the samples were 33.9% (20/59) for SFRP1,23.7%(14/59) for SFRP2, 6. 8% (4/59) for SFRP4 and 10. 2% (6/59) for SFRP5 in acute myelocytic leukemia (AML), and 39.3% (11/28) for SFRP1, 28.6% (8/28) for SFRP2, 25.0% (7/28) for SFRP4 and 32. 1% (9/28) for SFRP5 in acute lymphoblastic leukemia (ALL). Hypermethylation of SFRP1, 2, 5genes were present in the 4 AL cell lines. SFRP4 was methylated in NB4, Molt-4 and Jurkat cell lines. However, methylation and unmethylation of SFRP4 were both detected in HL60. Conclusions Hypermethylation of SFRP genes is a common event in the evolution of AL. Methylation of SFRP genes might serve as potential independent biomarkers for early detection of AL.

ABSTRACT

Objective To observe the effects of triptolide(TPL) on the anti-oncogene-APC gene of acute lymphoblastic leukemia cell line Jurkat in vitro. Methods The effects of TPL on proliferation Jurkat cells were assayed by using cell culture, MTT. The effects of TPL on APC gene of Jurkat cells were analyzed by nested methylation specific PCR and RT-PCR. The effects of TPL on the proteinum expression of APC gene were detected by Western blotting analysis. Results Following the treatment of TPL, the cell proliferation rate was degraded as the treatment concentration increased and the culture time extended. The effects were dose and time-dependent. The 48 hour IC50 was 19.7 ng/ml. TPL can reverse hypermethylation of APC gene, and induce the expression of the mRNA and the proteinum. Conclusion Low dose TPL could depress the proliferation rate of Jurkat. The possible mechanism might be its reversing the hypermethylation of APC gene and activiting the expression of APC gene.
