ABSTRACT
Valproic acid is used in the epilepsy, bipolar and migraine therapy. As a histone deacetylase inhibitor, Valproic acid has been recently under investigation in cancer treatment, either alone or in combination with either chemotherapy or radiotherapy. This study was done to determine the effect of Valproic acid and radiotherapy on viability of MCF-7 breast cancer cell line. In this descriptive - analytic study, MCF-7 cell line was obtained from the Iranian Pasteur Institute. The cells were treated and incubated by different concentrations of Valproic acid [1, 2, 4, 8, 16, 32, 64 and 128 mM] either alone or in combination with various dosages [0 .5, 2, 4, 6 and 8 Gray] of radiotherapy. After cell viability assay, using the Neutral red staining, the most nearest results to LD50 were selected. Cell viability was evaluated with trypan blue staining. The most nearest concentrations of LD50 was doses of 2, 8 and 16 mM of valproic acid and dosage of Gray 4 of radiation. There was a significant dose-dependent correlation between reduction of cell viability with valproic acid concentration [P<0.05]. Valproic acid, either alone or combination with radiotherapy caused a significant decline in the cell viability of MCF-7 breast cancer cell line

Carbohydrates are considered as the major source of energy in physical activity. Studies show that consumption of carbohydrate foods before exercise can balance blood glucose and free fatty acids and increase athletes' performance. In this study, we compared the effect of three kinds of foods with different glycemic indices on blood glucose [BG] and serum free fatty acids [FFA] in cycling ,male athletes. In this clinical trial, 21 members of national cycling team randomly allocated to three equal groups of glucose [low glycemic index]' lentil [low glycemic index] and potato [high glycemic index]. First, Fasting blood samples [5ml] were obtained to measure BG and FFA. Then the subjects were asked to eat their foods. After 45 mins of rest, they pedaled with maximal oxygen consumption VO2max for two hours and again their blood samples were taken to compare with the levels of before interventions. Glucose consumption resulted in a significant decrease in I FFA level after 2 hours of pedaling [P = 0.01] but no significant change j in BG level. Plasma glucose was higher after eating lentil than that of | potato [P<0.05], but it was not true for FFA level of both groups. Based on the results, the preexercise use of low glycemic index [lentil] compared to high glycemic index [potato] can better lead to increased blood glucose during exercise

The present study was performed to investigate the effects of trace elements particularity Se, Zn and Cu on tumor genesis in breast cancer. The inhibitory effect of Se, Zn and Cu, on telomerase activity was analyzed in human breast tumor tissues and breast cancer [T47D] cells. Tissue specimens from 24 women with benign breast disease and 32 women with breast cancer specimens [ductal carcinoma, lobular carcinoma] were collected during surgery. In addition venous blood samples were obtained for assessing the trace elements. T47D cell line was cultured and treated with trace elements. Telomerase activity then was measured with TRAP assay in cell line and tissue extracts. There was a significant difference between tissue and serum levels of Cu, Se and the ratio of Cu/Zn in patients and controls [P<0.001]. After treating with 100 microm/L Zn So4, 10 um /L Cu So4 for 6 hours, telomerase activity of T47D cells was markedly increased. But after treating with 10, and 30 um /L selenium-L- methionin, telomerase activity was markedly inhibited. Telomerase activity of T47D cells for 24 hours were 0.93, 0.60 and for 48 hours were 0.76, 0.12 respectively [control 49.2%]. There were variations in serum level of Zn and Cu in breast cancer patients. Association between trace elements level and telomerase activity level can be exploited as prognostic and diagnostic marker for breast cancer
