ABSTRACT

Objective:To analyze the current situation of major projects undertaken by medical colleges and universities in China, and the medical research competitiveness and its changing trend.Methods:Based on the data of National Key R & D Program, the research competitiveness and trend of medical subject in ten medical colleges and universities with strong competitiveness in medical scientific research during 2016-2020 were analyzed by range ratio, coefficient of variation and competitiveness index.Results:Ten medical colleges and universities have undertaken 459 projects of National Key R & D Program, and have been funded 5 788 million yuan of central financial funds. From 2016 to 2020, the coefficient of variation of National Key R & D Program on major medical colleges and universities increased from 56.25% to 62.91%. The change trend of competitiveness index shows that the medical research competitiveness in universities with strong initial competitiveness has increased, while the weaker ones decreased.Conclusions:Projects of National Key R & D Program undertaken by medical colleges and universities have a large volume and high support intensity. National Key R & D Program on the whole presents the Matthew effect, and the competitiveness of medical scientific research has been differentiated. There is an obvious trend of interdisciplinary integration between medical discipline and other disciplines in the process of undertaking projects of National Key R & D Program.

ABSTRACT

Objective:This article aims to analyze and explore practical and effective operation and management mechanisms of the independent interdisciplinary institutes in universities.Methods:Adopting case analysis and literature review as research methodologies to explore the operation and management mechanisms of mature interdisciplinary research institutions in foreign universities, and providing constructive suggestions for domestic academic institutions.Results:Case analysis found that flexible talent structure, sufficient scientific research platform sharing mechanism, and close partnership with industry operation mechanism can effectively promote the virtuous circle of interdisciplinary knowledge generation, dissemination and transformation, thereby driving the efficient operation of interdisciplinary institutes. At the same time, scientific and appropriate management scheme and evaluation system provide a strong systematic administrative support.Conclusions:Interdisciplinary institutes in domestic universities should adopt a matrix-style team formation method to assure the flow of talents with diverse backgrounds, thereby enhancing the internal driving-force of interdisciplinary development. Besides, a substantial resource sharing mechanism should be created to promote the establishment of interdisciplinary research communities. Furthermore, exploring " extensional" knowledge transfer mechanisms to promote collaborative innovation with enterprises. At the same time, establishing scientific and clear institutional management which take into full consideration of the key factors of interdisciplinary research methods and evaluation system.

ABSTRACT

Objective To explore ways to promote international research cooperation ,enhance research innovation and interna-tional impact of Peking University Health Science Center (PKUHSC) ,we analyzed the international (regional) cooperation and ex-change program of National Natural Science Foundation of China (NSFC) .Methods Taking the major international (regional) joint research projects as an example ,we analyzed the current situation of international research collaboration of PKUHSC and proposed pos-sible suggestions .Results PKU HSC is active in international research cooperation and gained advantage in undertaking major interna-tional (regional) joint research projects .To further enhance our international competitiveness ,the international research cooperation projects need to be further optimized .Conclusions It is necessary to further explore the international cooperation mode of "project-tal-ent-base",and enhance international level of scientific and technological innovation .

ABSTRACT

Objective To understand the current problems of medicine related scientific and technical innovation bases in China,provide suggestions for further policy-making and reforming.Methods Questionnaires tailored for the director,principal investigator and administrators of the bases were designed to collect problems found during the operation and management process of different bases.Results 66.7% and 43.4% respondents respectively of national bases and provincial/ministerial level bases believed that bases had the ability of science and technology innovation.However,organization form needed to be optimized,team needed to be strengthen,achievement transformation ability needed to be improved,equipment management needed to be rationalization,assessment needed to be adapted and usage rule of fund should be more flexible.Conclusions By the joint efforts of government,support units and bases,the management of science and technology innovation bases can be further optimized.

ABSTRACT

Objective To promote the "first-class universities and first-class disciplines" program in Peking University Health Science Center (PKUHSC),this article analyzed the research projects and explored novel methods to improve project organization.Methods Taking the National Natural Science Foundation of China (NSFC) as an example,we analyzed the advantages and challenges of research projects,proposed suggestions for improvement.Results PKUHSC did a great job in leading scientists training and research team building.However,the overall number of NSFC approved projects,especially the number of youth projects,needs to be increased.Conclusions Lessons should be constantly summarized to optimize management systems,improve service capabilities,and to build world-class Peking University medicine with "Chinese characteristics".

ABSTRACT

Objective To understand the current situation of scientific and technical innovation bases about medcine in China,provide evidence for further policy-making and reforming.Methods Based on the information from websites and the lists of laboratory evaluation,analyzed the types,research fields and the supporting units of these bases.Results Among the 347 bases,there were 160 national level bases and 187 ministerial-level bases.In terms of the research areas,clinical medicine accounted for 34.9 %,pharmaceutical sciences accounted for 26.5 %,basic medicine / biology accoun ted for 20.5%,public health accounted for 8.1%,translational medicine accounted for 1.4 %,medical equipment accounted for 1.4% and medical materials accounted for 0.9%.About 95.4% of bases were supported by single unit.Conclusions The research areas of medicine related scientific and technical innovation bases are unbalanced,and the number of industry-university-research cooperation bases is quite small.The government should strengthen the top-level design and optimize the layout.

ABSTRACT

Objective:To determine the effects of silencing glucose regulated protein ( GRP94 ) on the proliferation and apoptosis of breast carcinoma MCF7 cells. Methods:Chemically synthesized siRNA targeting GRP94 gene and transfected into MCF7 cells used by Liopfectamine RNAIMAX. The mRNA and protein expression levels of GRP94,cyclinD1,Bax and Bcl-2 were detected by Real-time PCR and Western blot. CCK8 assay was used to detect the effect of specific GRP94 siRNA on cell proliferation and the effect on cell cycle and apoptosis were analyzed by flow cytometry and Hoechst 33258 staining. Results:Compared with the siRNA-NC cells, the expression of GRP94 was significantly down-regulated in MCF7 cells. Knockdown of GRP94 in MCF7 cells decreased cell proliferation and promoted cell apoptosis. The expression of cyclinD1and Bcl-2levels were significantly reduced, and Bax level was increased in siRNA-GRP94 MCF7 cells. Conclusion: The siRNA-mediated GRP94 silence significantly inhibits MCF7 cell proliferation,promoted cell apoptosis by down-regulating cyclin D1,Bcl-2 expression and up-regulating the Bax expression in MCF7 cells.

ABSTRACT

Objective:To investigate the effects of ALEX1 overexpression on cell proliferation and apoptosis of human breast cancer cell line MCF-7.Methods: MCF-7 cells were infected recombinant lentivirus LV5-ALEX1 and the negative control lentivirus LV5-NC,respectively.After 72 h, the expression of ALEX1 was detected by Real-Time PCR and Western blot.CCK8 assay were performed to observe the proliferation ability after 24 h, 48 h, 72 h, 96 h.The effect of overexpression ALEX1 on apoptosis was determined by flow cytometry.The level of Bax,BCL-2 and active caspase3 was detected by Western blot.Results:The mRNA level of ALEX1 markedly increased after 72 h(165.81±12.14 vs 52.29±2.32,P<0.01).In CCK8 assay,the results revealed that the cell pro-liferation was inhibited compared with control group in 48 h,72 h,96 h( P<0.05).The results revealed that overexpression of ALEX1 enhanced MCF-7 apoptosis(20.55%±2.50 % vs 3.60%±1.614%,P<0.05).The results by Western blot showed that the protein levels of Bax and active caspase were increased in LV5-ALEX1 group compared with LV5-NC group.However,the protein levels of BCL-2 was decreased in LV5-ALEX1 group compared with LV5-NC group.Conclusion:Overexpression of ALEX1 significantly reduced MCF-7 cancer cells proliferation and induced MCF-7 cells apoptosis.

ABSTRACT

[Abstrct] Objective The deformity of severe varus knee (varus angulation > 15°) can be treated by many ways in primary total knee arthroplasty , but the efficacy and cost are different. The topic of this study provides a new surgery: autograft (wedge grafts) implant and substitute medial tibial bone defects. Methods From Jan. 2006 to Jan. 2014 , a retrospective study was conducted in the First Affiliated Hospital of BaoTou Mendical College. Out of 687 patients, 721 primary knees were operated. Bone defects in proximal tibia (varus angulation >15° ) were encounted in 16 patients in 21 knees in the pattern of non-inclusive slope bone defects in the mean varus degree of 27°. Their IBM was 31 kg/m2. The average age of patients was (63 ± 5) years. The score of clinical knee society score (KSS) before operation was 18 on average. Among of them, the pain score were 12 points, the average range of motion (ROM) were 12 points. the average funtional score were 20 points. During operation,the slope bone defect of the medial tibial plateau was tailored into wedge-shaped ladder bone defect by osteotomy,and then the defect was filled with the resected tibial plateau autograft in which the thickness and shape were matched completely. The high-intensity cortical part of the autograft was placed in the rim to sustain the tibial prosthesis. Three months after operation ,patients started to walk without the help of crutch. Results The average followup period was 5.3 y , We observed that an average postoperative scores of clinical KSS from 42 points to 88 points with the score of functional KSS from 57 points to 92 points. Fusion was observed by trabeculae crossing from the graft to the tibial metaphysis on plain films at least 6 mouths without collapse, stress fracture, untight and bone ununion. Confirmation of vascularization of the graft was observed in bone scans of nuclide. showing increased uptake in the operated area. Inflammatory makers were normal. Conclusion Autograft (wedge grafts) reconstruction of medial tibial bone defects, is of high rate of healing in implant bones, good restoration of the function and low cost.
