ABSTRACT
OBJECTIVE@#To explore the effect of dichloromethane extraction phase of ethanol extract from stem of Patrinia scabiosaefolia Fisch.(DPSS) on proliferation and differentiation of K562 cells and its related mechanism.@*METHODS@#MTT assay was used to detect the effects of DPSS at 0, 25, 50, 100 and 200 μg/ml on the proliferation of K562 cells at 24, 48 and 72 hours. Flow cytometry was used to analyze the changes of cell cycle and apoptosis at 24 and 48 hours. Wright-Giemsa staining was used to observe the morphological changes of K562 cells. The cell surface antigens CD33 and CD11b were detected by flow cytometry.@*RESULTS@#The proliferation of K562 cells treated with different concentrations of DPSS was inhibited in a time-dose dependent manner (r=-0.96). Cell cycle analysis showed that with the increase of DPSS concentration, cells in G2/M phase increased (r=0.88), and cells were blocked in G2/M phase. Flow cytometry results showed that with the apoptosis rate of K562 cells was the highest when treated with 200 μg/ml DPSS for 48 h. Morphological observation showed that the K562 cell body increased, the amount of cytoplasm increased, the ratio of nucleus to cytoplasm decreased, and the nuclear chromatin was rough after DPSS treatment. Cell differentiation antigen, CD33 and CD11b, were positively expressed after treated with DPSS.@*CONCLUSION@#DPSS can induce apoptosis through cell cycle arrest, inhibit the proliferation of K562 cells, and induce K562 cells to differentiate into monocytes, which has a potential anti-leukemia effect.

This study aims to analyze the effect of berberine on serum inflammatory factors and carotid atherosclerotic plaques in ppatients with acute cerebral ischemic stroke(AIS). In the study, 120 patients with AIS were randomly divided into berberine group(n=60) and general group (n=60). The 60 cases in the general group were provided with general therapy according to the latest guidelines of diagnosis and treatment of AIS. The berberine group received berberine 300 mg(tid) in addition to the therapy of the general group. The levels of serum inflammatory factors, the nerve function defect grades and the indexes of carotid atherosclerosis plaques [including the total plaque area(TPA), intima-media thickness(IMT) and the number of unstable carotid atherosclerotic plaques] were measured and compared. The results indicated that the levels of serum inflammatory factors, the NIHSS(national institute of health stroke scales) cores and the indexes of carotid atherosclerosis plaques were not significantly different between the berberine groups of general group, with positive correlation between serum inflammatory factors and NIHSS scores(P<0.05). The levels of serum inflammatory factors and NIHSS scores of the berberine groups on 14 d were significantly lower than those on 1 d(P<0.05). The levels of serum inflammatory factors and NIHSS scores of the berberine group on 14 d were significantly lower than those of the general group(P<0.05). The TPA and the number of unstable carotid atherosclerotic plaques of the berberine groups on 90 d were significantly lower than those of general group, with significant differences(P<0.05). The IMT showed a downward trend, but with significant difference.The mRS(modified rankin scale) scores of the berberine group on 90 d were significantly lower, with a higher rate of short-term favorable prognosis (P<0.05). There was no significant difference in the incidence of adverse reactions between the two groups. This study showed that berberine in addition to the general therapy can significantly lower the levels of serum MIF and IL-6, reduce the degree of carotid atherosclerosis to some extent and improve neurological impairment and the prognosis of patients with AIS.

Objective:To examine the effects of chronic intermittent hypoxia(CIH)on the NF-κB,IL-6 and PGE2 level in rats with periodontitis.Methods:32 male SD rats(6 weeks old)were randomly divided into 4 groups(n =8),group A(normoxic control),B (normoxic periodontitis),C(CIH)and D(periodontitis +CIH).Periodontitis model was established in the upper second molars by liga-tion technique and high-glucose diet in the rats of group B and D.The rats in the group C and D were subjected to CIH in a cycle of al-ternative nitrogen and oxygen in a closed chamber.The chamber was filled with nadir and zenith ambient oxygen every 1 20 seconds per cycle for 8 hours per day.The rats were sacrificed and the gingival tissues were examined for the detection of IL-6 and PGE2 expression by ELISA,and NF-κB expression by immunohistochemistry.Results:Histology revealed apical migration of junctional epithetlium and crestal alveolar bone resorption in group B and D,and in the above phenomena of group D was the severest.The content of NF-κB,IL-6 and PGE2 in group B,C,D was higher than that in group A(P <0.05),and in group D was the highest(P <0.05).Conclusion:Chro-nic intermittent hypoxia can aggravate the inflammation of periodontitis.

Objective:To investigate the expression of tumor necrosis factor-α(TNF-α)and soluble intercellular adhesion molecule -1(sICAM-1)in blood serum and gingival tissue in rat periodontitis model in normoxia and chronic intermittent hypoxia environment. Methods:32 male SD rats were randomly divided into 4 groups(n =8):normoxia control group(group A),normoxia periodontitis group(group B),hypoxic control group(group C)and hypoxia periodontitis group(group D).The periodontitis models were estab-lished by ligating the bilateral maxillary second molar and raised by periodontitis diet.The rats in hypoxia groups were raised under chronic intermittent hypoxia environment,while those in normoxia groups were raised under normoxia environment.After 8 weeks, plaque index(PLI),bleeding index(BI)and attachment lost(AL)were measured,TNF-αand sICAM-1 concentrations in blood serum and gingival tissue were measured by ELISA.Results:TNF-αand sICAM-1 concentrations in blood serum and gingival tissue in group A were higher than those in group B,C(P <0.05);in group D were lower than in group B and C(P <0.05).TNF-αand sICAM-1 levels in blood serum and gingival tissue were positively correlated with AL(P <0.05).Conclusion:In chronic intermittent hypoxic environment TNF-αand sICAM-1 may aggravate periodontitis,and promote the inflammatory response to peripheral vascular system.
ABSTRACT
<p><b>OBJECTIVE</b>To investigate the antagonizing effect of Hirsutella sinensis (HS) on renal tubular epithelial-myofibroblast transdifferentiation (TEMT) and its possible pathogenic mechanism in rats with chronic aristolochic acid nephropathy (CAAN).</p><p><b>METHODS</b>Eighteen male Sprague-Dawley rats were equally divided into 3 groups, the model (M) group, the intervention (I) group and the control (C) group. The 24 h urinary protein (UP) in rats was measured before intervention and at the end of the 1st, 4th, 8th, and 12th week, and creatinine clearance rate (CCr) was measured before intervention and at the end of the 12th week respectively. All rats were sacrificed at the end of the 12th week, their kidney was taken for examining the degree of fibrosis in renal interstitial with Masson's stain and determining mRNA and protein expressions of transforming growth factor-beta1 (TGF-beta1), Snail, alpha-smooth muscle actin (alpha-SMA) and cytokeratin in renal tissue by Real-time RT-PCR and immunohistochemistry staining, respectively.</p><p><b>RESULTS</b>Compared with the C group, CCr was significantly lower, while 24 h UP was higher; the relative area of interstitial fibrosis was significantly larger in the M group; besides, the mRNA and protein expressions of TGF-beta1, Snail and alpha-SMA were significantly up-regulated (P < 0.01 or P < 0.05), and those of cytokeratin were significantly down-regulated (P < 0.01) in renal tissue of the M group. While in the I group, all the above-mentioned abnormalities were restored to some extent (P < 0.05) and showed significant difference (all P < 0.05) as compared with those in the M group.</p><p><b>CONCLUSION</b>HS can downregulate TGF-beta1 and Snail expressions in renal tissue, antagonize TEMT and renal interstitial fibrosis, and improve renal function in CAAN rats.</p>

Objective This study aimed at soliciting preliminary data about the feasibility and effectiveness of a computer-aids for HIV/AIDS voluntary counseling and test (VCT) developed by ourselves and inform further refinement and evaluation of the software.Methods Informant qualitative interviews,structured questionnaires and audio-recorded counseling cases were used in gathering data.Pre-and post-intervention statistical comparisons of quantitative data and expert reviews of qualitative materials were performed in deriving evaluation outcomes.Results Application of the software increased per episode counseling time from 5.55 minutes to 10.73 minutes;coverage of critical counseling components by over 1.5 folds;and successful memory of core knowledge items from 40.51% to 86.34%.Most participating counselors and clients perceive the software as feasible and effective.Conclusions The software put forth merits further researches.
ABSTRACT
Objective This study aims at developing a computer aid for performing HIV/AIDS counseling and test,so as to standardize VCT operating procedures and help frontline counselors provide high quality VCT service.Methods Rapid prototyping was employed in which primitive VCT standard operation procedure (SOP) and prototype software were generated through literature reviews,counseling case studies and expert panel activities first and then refined through consecutive rounds of piloting at various VCT settings.Results A pragmatic SOP and computer-aid for performing routine VCT services were put forward.It consists of 5 main application modules including performing counseling,making appointment,managing data,developing counseling skill and setting system parameters.Conclusions Our study suggests that standardization and computerization of complex VCT service is feasible and the system we had produced provides a new means for improving the service.
ABSTRACT
<p><b>OBJECTIVE</b>To study the variations of protein concentration in saliva stimulated and its effect on clinical diagnosis.</p><p><b>METHODS</b>The saliva from 33 normal controls and 73 patients with Sjögren syndrome (SS) who were stimulated with acid and not were collected. The concentration of beta 2 microglobulin (beta 2-mG), secretory immunoglobulin A (SIgA), and pH were measured by Radioimmunoassay, Rate Nephelometry and pH Detection Paper, respectively. SPSS 10.0 was used to determine the mean statistical differences among these groups.</p><p><b>RESULTS</b>In patients with SS, the concentration of beta 2-mG in saliva stimulated with Vc was significantly lower compared with that in saliva not stimulated (P < 0.01); In saliva stimulated with Vc, the concentration of beta 2-mG in patients with SS was higher than that in normal controls (P < 0.05). In normal controls, compared with that in saliva not stimulated, flow rate in saliva stimulated with 3% acetic acid and Vc was significantly higher (P < 0.01) and pH, concentration of beta 2-mG and SIgA were significantly lower (P < 0.01 and P < 0.05 respectively); there was a significant difference of flow rate, beta 2-mG, SIgA and pH in saliva between the subjects stimulated with 3% acetic acid and with Vc (P < 0.01).</p><p><b>CONCLUSIONS</b>The reason for the decrease of protein concentration in saliva stimulated may be the increase of flow rate caused by the decrease of pH or the decrease of pH itself. Protein detection of saliva stimulated in patients with SS is helpful in diagnosis, but the criterion is different between the saliva stimulated and not stimulated.</p>
