ABSTRACT

OBJECTIVE@#To investigate the effects of genipin on promoting brown adipose tissue activation and white adipose tissue browning.@*METHODS@#The male C57BL/6J mice were divided into three groups: normal control group, genipin group and cold-stimulus group.Genipin group were treated consecutively with genipin at a dose of 15 mg/kg once a day for 9 days, normal control group were treated with the saline.The mice with cold-stimulus were exposed to 4℃ environment for 5 days.Daily food amount and body weight were measured.Morphological changes were observed in the subscapular region, inguinal region and epididymis around the adipose tissue.The expression of uncoupling protein 1 (UCP1) was determined by real-time PCR and Western blot respectively.@*RESULTS@#The wet weight of white fat in genipin-treated mice was decreased by 16% , and 28% in that of cold-stimulus mice, compared with the normal control group (P<0.05).After treatments of genipin and cold-stimulus, the color of white adipose tissues was darker, and the size of lipid droplets in adipocytes was smaller, whereas the number was increased.Compared with the normal control group, UCP1 expression was increased obviously in fat tissues, including the subcutaneous and visceral white adipose tissues, and brown adipose tissue after treated with genipin and cold-stimulus (P<0.05).@*CONCLUSION@#Genipin promoted activation of brown adipose tissue and browning of white adipose tissue by upregulating UCP1 expression, which could contribute to the loss of body weight against obesity.

ABSTRACT

OBJECTIVE@#To investigate the expression of miR-101 and EZH2 in patients with mantle cell lymphoma(MCL) and to analyze its correlation with clinical prognosis of MCL patients.@*METHODS@#RQ-PCR and S-P immunohistochemistry were used to detect the expressions of miR-101 and EZH2 in tissue of MCL patients. CCK-8 was used to assay the effect of miR-100 minics on the proliferation of Jeko-1 and Mino cells; the flow cytometry with Annexin V/PI double staining was used to assay the apoptosis; Western blot was used to assay the effect of miR-101 minics on the expression of EZH2 protein in Jeko-1 and Mino cells.@*RESULTS@#Compared with control group, miR-101 lowly expressed, and EZH2 protein highly expressed in MCL group, with very statistically significant difference(P<0.01).There was negative correlation between miR-101 and EZH2 expression(r=-0.638,P<0.05). The expression of miR-101 and EZH2 significantly correlated with B symptoms, International Prognostic Index(IPI) and Ann Arbor stage, respectively. Survival analysis showed that the overall survival(OS) rate of patients with low expression of miR-101 were significantly lower than that of patients with high miR-101 expression (P=0.0014), the OS rate of patients with EZH2 high expression were significantly lower than that of patients with EZH2 low expression (P=0.0093). The miR-100 minics could inhibit the proliferation of Jeko-1 and Mino cells, and increase the apoptotic rate. The expression of EZH2 protein was markedly suppressed by the miR-100 minics.@*CONCLUSION@#The expression of miR-101 and EZH2 is different in MCL patients with different clinical stage and prognosis. The miR-101 can inhibit the cell proliferation and induce cell apoptosis of mantle cell lymphoma by targeting EZH2.

ABSTRACT

Objective To compare the efficacy and safety of diazepam combined with olanzapine in treatment of agitation symptoms in patients with alcohol withdrawal delirium tremors( DT) . Methods Total-ly 60 inpatients with DT according to CCMD-3 were enrolled in a 7-day,with 30 patients in the diazepam group ( control group) and 30 patients in diazepam combined with olanzapine group ( research group) . The delirium rating scale( DRS-R-98) was used to evaluate the severity of the delirium symptoms. The positive and negative syndrome scale excited component( PANSS-EC) was used to assess the agitation symptoms. The treatment emergent symptom scale ( TESS) was used to assess the safety and the vital signs. Results There was no significant difference in the delirium recovery time between the two groups(M(QR):24 h(24 h)vs 24 h(6 h))(Z=-0. 45,P=0. 65). The PANSS-EC score in research group(17. 00±2. 67) was significant low-er than that in control group(19. 80±2. 43) at the 6 hours after treatment(t=4. 26,P<0. 01),but there were no significant difference between the two groups at the baseline,24 h,48 h and 72 h(P>0. 05) . One case of dizziness,2 cases of lethargy in the control group;1 case of nasal congestion,5 cases of drowsiness,3 cases of constipation, 3 cases of dry mouth, 1 case of abnormal liver function in the research group. Conclusion The combination of olanzapine with diazepam can not reduce the recovery time of DT,but can quickly improve the agitation behavior.

ABSTRACT

Objective:To investigate the effect of Rab7 on cytokine induced by TLR7 (Toll like receptor-7) R848 activated in Raw264.7,and discusses the influence of Rab7 on MAPK signal transduction.Methods: TLR7 downstream cytokines such as TNF-α,IL-6,IFN-α,IFN-β and IP-10 activated by R848 were detected through Q-PCR in Rab7 silenced mouse macrophages,and then analysis of phosphorylation of MAPK determined with Western blot showed the effect of Rab7 on signal transduction of MAPK.Results: Rab7 inhibit production of cytokine activated by TLR7,and also,Rab7 had an inhibitory effect on MAPK signal pathway.Conclusion: The experimental results further illustrate that the Rab7 is the TLR7 signal transduction pathway negative regulatory factor,and to participate in MAPK signaling pathway.

ABSTRACT

Objective To explore the factors of relapse in alcohol-induced psychiatric and behavioral disorders.Methods103 male inpatients met with the diagnostic criteria of alcohol-induced psychiatric and behavioral disorders according to ICD-10 were enrolled.All patients were hospitalized from Wuxi Mental Center from January 2013 to August 2015.As baseline,information was obtained within all patients,and relapse was evaluated one year after discharge.The Kaplan-Meier method was used to statistically analyze the alcohol relapse time of patients with different length of hospitalization.Cox regression was used to explore the risk factors for alcohol relapse,including age,education level,marital status,family history,smoking,fixed income,number of hospitalizations,duration of alcohol intake,average daily alcohol intake,time of psychosis,psychiatric symptoms,length of hospitalization,physical condition and mental condition.Results(1)The analysis (log rank) showed that the length of hospitalization had no significant statistical differences with relapse(χ2=0.069,P=0.966).(2) The number of hospitalizations (RR=1.074,95%CI=1.002-1.151,P=0.042) and average daily alcohol intake(RR=1.035,95%CI=1.012-1.059,P=0.003) were the risk factors for relapse.ConclusionThe number of hospitalizations and average daily alcohol intake are risk factors for relapse within a year in male inpatients with alcohol-induced psychiatric and behavioral disorders.Prolonged hospital stay has no help to reduce relapse in those people.

ABSTRACT

Objective To analyze and detect the whole genome sequence of human mitochondrial DNA (mtDNA) by Ion Torrent PGMTM platform and to study the differences of mtDNA sequence in different tissues.Methods Samples were collected from 6 unrelated individuals by forensic postmortem examination,including chest blood,hair,costicartilage,nail,skeletal muscle and oral epithelium.Amplification of whole genome sequence of mtDNA was performed by 4 pairs of primer.Libraries were constructed with Ion ShearTM Plus Reagents kit and Ion Plus Fragment Library kit.Whole genome sequencing of mtDNA was performed using Ion Torrent PGMTM platform.Sanger sequencing was used to determine the heteroplasmy positions and the mutation positions on HV Ⅰ region.Results The whole genome sequence of mtDNA from all samples were amplified successfully.Six unrelated individuals belonged to 6 different haplotypes.Different tissues in one individual had heteroplasmy difference.The heteroplasmy positions and the mutation positions on HV I region were verified by Sanger sequencing.After a consistency check by the Kappa method,it was found that the results of mtDNA sequence had a high consistency in different tissues.Conclusion The testing method used in present study for sequencing the whole genome sequence of human mtDNA can detect the heteroplasmy difference in different tissues,which have good consistency.The results provide guidance for the further applications of mtDNA in forensic science.

ABSTRACT

Objective To investigate the social cognition and its correlation with social function for attenuated psychosis syndrome (APS). Methods From August, 2014 to December, 2015, 39 patients with APS were recruited as research group. Another 40 normal healthy persons with similar gender, ages, and education levels were selected as control group. The Faux Pas Recognition Test (FPR) and Yoni Task Test were used to evaluate the social cognition, and Social Disability Screening Schedule (SDSS) was used to evaluate the social function. The correlation between FPR, Yoni Task Test and SDSS in the research group was analyzed. Results In FPR test, the faux pas questions score, control questions score and total score of FPR were significantly lower in the research group than in the normal control group (t>2.378, P2.341, P0.05). The SDSS total score was signifi-cantly higher in the research group than in the normal control group (t=13.032, P0.473, P0.448, P<0.01). Conclusion Social cognition func-tion in APS is impaired. It is associated with social dysfunction in APS.

Objective:Using the macrophage cell lines RAW264.7 stably expressing Rab5a and its dominant negative mutant Rab5aN133I as models to analyze the effect and the mechanism of Rab 5a,Rab5aN133I on CpG-induced production of pro-inflammatory cytokines and type Ⅰ IFN.Methods: The eukaryotic expression vectors of Rab5a and Rab5aN133I were transfected into RAW264.7 cells by liposome,and screened with G418.The G418-resistant colonies were obtained and amplified.The transformed cell lines were i-dentified by RT-PCR,Real time-PCR and Western blot.The production of cytokines were measured after transformed cell lines of Rab5a and Rab5aN133I was stimulation with CpG for 8 h.Results: Rab5a expression in transfected cells was significantly higher than the control cell group (P<0.05).Overexpression of Rab5a significantly promoted the production of TNF -α,IL1-β(P<0.01) and IFN-β( P<0.05) in CpG stimulated RAW264.7.The production of cytokines was restored in Rab 5aN133I transfected cell line.Conclusion:Rab 5a promotes CpG-induced pro-inflammatory cytokines and typeⅠIFN in macrophages,it may be act as a positive regulator in TLR9 signaling pathway.

ABSTRACT

Objective:To explore the inhibitory effects of seabuckthorn polysaccharide on hepatic oxidative stress in a mice model of acute liver injury induced by intraperitoneal injection of LPS and D -GalN and detect the expression on hepatic BCL-2/Bax and PPAR-γ.Methods: C57BL/6 male mice were randomly divided into six groups:control group ( CTRL), model group ( L/G), dexamethasone positive control group ( DXM ) , low ( SPL ) , medium ( SPM ) and high dose group ( SPH ) of seabuckthorn polysaccharide.Mice in the SPL,SPM and SPH group were gavaged with 50,100 and 200 mg/kg seabuckthorn polysaccharide for 14 days respectively.Acute liver injury model were established by intraperitoneal injection of LPS (10 μg/kg) and D-GalN (700 mg/kg) .Serum and liver samples were collected 4 h after model establishment .Serum levels of ALT and AST and the content of MDA were de-tected.Hepatic expression of SOD 2 BCL-2 and Bax was determined by Western blot and the expression of PPAR-γwas detected by im-munohistochemistry .Results:ALT and AST levels significantly increased in the model group and decreased dose-dependently after pre-treatment with seabuckthorn polysaccharide .The level of MDA in the model group increased significantly as compared with the control group and decreased in seabuckthorn polysaccharide groups ,while the level of SOD 2 decreased in the model group and recovered in sea-buckthorn polysaccharide groups .The expression of Bax decreased after pretreatment with seabuckthorn polysaccharide .There was no obvious effect on BCL-2 expression after sea buckthorn polysaccharide supplementation .The expression of PPAR-γreduced in the sea-buckthorn polysaccharide group as compared with the model group .Conclusion:Seabuckthorn polysaccharide protects against LPS /D-GalN-induced liver injury.The effect is associated with an upregulation of SOD 2 and downregulation of Bax .

ABSTRACT

To explore the relativity between alexithymia and cognitive function in somatoform disorder patients.A total of 40 patients aged 18-65 years fulfilling the criteria of ICD-10 for somatoform disorders were recruited as research group.And 40 normal healthy subjects were selected as control group.Toronto alexithymia scale-20 (TAS-20) was employed to examine the alexithymia.And cognitive abilities screening instrument (CASI) was used to measure their cognitive ability.The 9-factor scores and total score of CASI test in patients with somatoforill disorder were significantly lower than those in the normal controls (P < 0.05 or < 0.01).The 3-factor scores and total score of TAS-20 test in patients withsomatoforill disorder were significantly higher than those in the normal controls (P < 0.05 or < 0.01).In research group,CASI scores were negatively correlated with the TAS-20 scores (P <0.05 or 0.01).

ABSTRACT

<p><b>OBJECTIVE</b>To study the clinicopathologic features and prognostic factors of Chinese patients with mantle cell lymphoma.</p><p><b>METHODS</b>One hundred and two cases of mantle cell lymphoma occurring in Chinese patients were studied by light microscopy and immunohistochemistry. The follow-up information was also analyzed. The cases were classified as mantle zone, nodular or diffuse patterns and as typical or blastoid variants. Age, Ann-Arbor staging, B symptoms, hematologic parameters, histologic variants, mitotic index and immunophenotype were assessed for possible prognostic implication.</p><p><b>RESULTS</b>The median age of the patients was 59 years (range: 30 to 79 years) and the male-to-female ratio was 2.92:1. Seventy-one patients (87.65%) presented with advanced stage disease (Ann Arbor stage III to IV). B symptoms were present in 45.45% of patients. The commonest site of involvement was lymph node (100%). The other involved sites included bone marrow (64.44%), spleen (63.16%), Waldeyer's ring (31.25%), peripheral blood (29.41%), liver (22.64%) and gastrointestinal tract (14.71%). All cases expressed B-cell markers but were negative for T-cell marker. Majority of cases were positive for cyclin D1 (94.12%) and CD5 (71.43%). Blastoid variant accounted for 24.51% of cases. Amongst the 68 cases with follow-up data available, the median survival was 10 months. Parameters associated with shorter survival included diffuse pattern, blastoid variant, high mitotic index, high proliferative activity and presence of bone marrow involvement.</p><p><b>CONCLUSIONS</b>The clinicopathologic features and prognostic factors of mantle cell lymphoma occurring in Chinese are similar to those in Caucasians. Diffuse pattern, blastoid variant, high mitotic index, high proliferative activity and involvement of bone marrow indicate poor prognosis.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe effect of acupuncture at Sifeng (EX-UE 10) on serum leptin level in the child of anorexia, so as to reveal relation between changes of blood leptin level and anorexia of child.</p><p><b>METHODS</b>Forty-two cases were randomly divided into 3 groups. The treatment group A (n = 15) were treated with acupuncture at Sifeng (EX-UE 10), once each week, 3 consecutive sessions constituting one course; the treatment group B (n = 15) were treated with acupuncture at Sifeng (EX-UE 10), once two weeks, 3 consecutive sessions constituting one course; the medication group (n = 12) were treated with oral administration of Lactein tablets, 1 - 2 tablets each time, 3 times each day, 4 weeks constituting one course.</p><p><b>RESULTS</b>The effective rate was 93.3% in the treatment group A and 93.3% in the treatment group B, which were higher than that in the medication group, with a very significant differences (P < 0.01); after treatment, the serum leptin levels in the 3 groups had very significant changes (P < 0.01); the improvement of serum leptin levels in the treatment group A and B were better than that in the medication group (P < 0.05).</p><p><b>CONCLUSION</b>Acupuncture at Sifeng (EX-UE 10) can promote secretion of serum leptin in the child of anorexia, and improve anorexia.</p>

ABSTRACT

[Objective]To discuss the effects of DHS and compression screw application on femoral intertrochanteric fracture.[Method]Internal fixation was performed with DHS,DHS and compression screw,Ender nail and proximal femoral anatomical plate in 185 patients with femoral intertrochanteric fracture.The main outcome measures were operation duration,coxa adducta rates and postoperative Sanders scoring.[Result]All patients were followed up for 9-48 months.Operation duration was similar in the four groups.There were significant differences in coxa adducta rates and postoperative Sander's scores.According to the Sander's scores,the excellent to good rates in DHS and compression screw,DHS,Ender nail and proximal femoral anatomical plate groups were 94.6%,85.3%,75.4% and 80.6%,respectively.The coax adducta rates were 2.4%,9.5%,22.2% and 19.2%,respectively.[Conclusion]All the four kinds of internal fixation can be used for femoral intertrochanteric fracture,but DHS and compression screw is the best one for its good restoration of joint function and has no or little complication.

ABSTRACT

[Objective]To evaluate the result of operative treatment of tibial plateau fracture and analyze its related factors of influencing the therapeutic effect and provide effective prevntional measures.[Method]Totally 107 cases(male 62 cases;female 45 cases) of tibia plateau fractures were operated with internal fixation from Feb.2000 to Apr.2005 in our hospital.According to Schatzker classification,there were typeⅠfracture 2 cases,typeⅡ 37 cases,typeⅢ 29 cases,type Ⅳ 22 cases,type Ⅴ 10 cases,type Ⅵ 7cases.TypeⅠfractures were treated with cannulated screws,other type fractures were performed with open reduction and internal fixation with buttress plates and autologous and/or allograft bone grafts.Therapeutic effect and its related factors were analyzed concomitantly.[Result]The 107 patients were followed up for 9~64 months(averaged 27 months).The clinical results were assessed according to Hohl scale for knee joint.The excellent and good results was 85.98%.There were obvious complications articular stiffness 1 cases and traumatic arthritis 8 cases including tibia plateau articular depression 4 cases.[Conclusion]Surgical operation is effective methods of treating tibia plateau fracture,The complete fracture reduction suitable internal fixation a good protection and recovery of the soft tissue of the knee joint structure,and correct and effective function rehabilitation are the significant measures for good therapeutic effect in the treatment of tibia plateau fracture.

ABSTRACT

<p><b>OBJECTIVE</b>To make a solid form of Angelica Sinensis essential oil by preparing its inclusion compound with beta-cyclodextrin and evaluate the inclusion rate, dissolution rate and liberating rate of this complex.</p><p><b>METHOD</b>GC-MS, X-ray diffraction and thin-layer chromatography were applied to characterize the essential oil and the inclusion compound.</p><p><b>RESULT</b>The inclusion compound was shown to form a new phase by X-ray diffraction analysis. GC-MS and TLC check-up corroborated the composition of the free oil and the inclusion oil remaining the same.</p><p><b>CONCLUSION</b>The results showed Angelica Sinensis essential oil was more stable with beta-cyclodextrin inclusion. The preparation of this inclusion compound has advantages in limited amount, long time effect, easily preserved and convenient administration as a new dosage form.</p>
