ABSTRACT
The genus Fuchsia is generally used in herbal preparations to treat conditions caused by microorganisms. Based on the popular use of this type of plants, the objective of this study was to obtain sequential extracts of increasing polarity from the branches of Fuchsia lycioides by maceration at room temperature and by the Soxhlet method at 60ºC, to later evaluate the antifungal capacity of the extracts against different clinical isolates of the Candida genus. The ethyl acetate extract exhibited strong anti-fungal activity, selectively inhibiting C. albicans strains with MIC and CMF values of 10 and 15 µg/mL, respectively; comparable with the drug itraconazole®. The analysis of the extract by GC-MS showed a high concentration of terpenoids (mainly phytol) and phenylpropanoids (mainly cinnamic acid), possibly responsible for the antifungal activity of the ethyl acetate extract of F. lycioides.
El género Fuchsia se usa generalmente en preparaciones de hierbas para tratar afecciones provocadas por microorganismos. En base al uso popular de este tipo de plantas, el objetivo de este estudio fue obtener los extractos secuenciales de polaridad creciente de las ramas de Fuchsia lycioides por maceración a temperatura ambiente y por el método Soxhlet a 60ºC, para luego evaluar la capacidad antifúngica de los extractos frente a diferentes aislados clínicos del genero Candida. El extracto de acetato de etilo exhibió una fuerte actividad antifúngica inhibiendo en forma selectiva las cepas de C. albicans con valores de CMI y de CMF de 10 y 15 µg/mL, respectivamente; comparables con el fármaco itraconazol®. El análisis del extracto por CG-EM mostró una alta concentración de terpenoides (principalmente fitol) y fenilpropanoides (principalmente ácido cinámico), posibles responsables de la actividad antifúngica del extracto de acetato de etilo de F. lycioides.

Introduction: Chlamydia trachomatis (CT) is the most prevalent sexually transmitted bacterial infection, affecting mainly young, sexually active women. Untreated infection may lead to reproductive complications due to tubal damage. Infections during pregnancy may cause preterm labor, low birth weight, perinatal death, and neonatal conjunctivitis and pneumonia. There are few data on CT infection in Brazil. The aim of this study was to determine CT prevalence in infertile and pregnant women. Methods: A cross-sectional study included 77 infertile and 60 asymptomatic pregnant women. First-void urine was tested for CT using PCR (Polymerase Chain Reaction). Blood samples were collected for CT IgG antibodies testing using indirect immunofluorescence. A questionnaire about medical, gynecological, and sexual history was completed by all participants. Results: We found statistically similar prevalence of PCR and IgG antibodies between the groups. There was a 61% prevalence of CT IgG antibodies in infertile women and 56.7% in pregnant women. PCR was positive in only one (1.3%) infertile woman and in none pregnant women. Conclusion: There is a high prevalence of CT IgG antibody in Brazilian pregnant and infertile women, but we found a low prevalence of positive PCR in the urine samples. This search is focused on the study of diet compounds that may have any potential chemopreventive effect against cancer. Some compounds that fulfill this requirement are phytoestrogens. Among them we find genistein (1), the most studied, daidzein (2) and equol (3) (figure 1). To compare the sensitivities of different prostate cancer cells to phytoestrogen treatment, sulphorhodamine B dye assay was performed to determine cell viability. DU-145 and PC-3 prostate cancer cell lines treated with various doses of phytoestrogen (0-12.5-25-50 and 100 μM) for different times (24, 48 and 72h). For cell invasion or migration assay cells were seeded in a Transwell chamber with or without coating Matrigel respectively. DU-145 and PC-3 cells were treated previously with phytoestrogen (50 μM) for 24h. The study showed that equol, daidzein and genistein inhibited migration and invasion in prostate cancer cell lines. Moreover, we analyzed the effects of phytoestrogens in MMP-2 and MMP-9 mRNA expression by RT-PCR. The results indicated that equol, daidzein and genistein diminished the expression of MMP-2 and MMP-9 in a cell-dependent manner. Our data suggested that equol, daidzein and genistein inhibited migration and invasion in prostate cancer cell lines. Moreover, the results also suggest that down-regulation of MMP-2 and MMP- 9 might be involved in the inhibition of invasion of PC-3 and DU-145 cells after genistein, daidzein and equol treatment.
Este trabajo se centra en el estudio de los compuestos de dieta que pueden tener potencial efecto quimiopreventivo contra el cáncer. Algunos de estos compuestos son los fitoestrógenos. Entre ellos encontramos la genisteína (1), el más estudiado, la daidzeína (2) y el equol (3) (figura 1). Para comparar el efecto de estos fitoestrogenos sobre las líneas celulares de cáncer de próstata, DU-145 y PC-3, se utilizó el ensayo de sulforodamina B para determinar la viabilidad celular tras los tratamientos con diferentes concentraciones de fitoestrógenos (0-12.5-25-50-100 μM) durante diferentes tiempos (24, 48, 72 h). Para analizar el efecto sobre la migración celular, las células DU-145 y PC-3 fueron tratadas previamente con una concentración de fitoestrógrno (50 μM) durante 24 horas y sembradas en una cámara Transwell sin recubrir. El estudio mostró que el equol, daidzeína y genisteína inhibió en MMP-2 y MMP-9 expresiones de genes en líneas celulares de cáncer de próstata, la PC-3 y DU-145. Los resultados indicaron que la daidzeína disminuyó la expresión de MMP- 2 y MMP-9 en DU-145 células. Nuestros datos sugieren que equol, daidzeína y genisteína inhiben la migración y la invasión de líneas celulares de cáncer de próstata.

Otholobium glandulosum (L.) J.W. Grimes (= Psoralea glandulosa L.) (Fabaceae) is a resinous bush that grows in Chile. The chemical composition of its resinous exudate was determined for the first time. Three meroterpenic: bakuchiol (8), 3-hydroxybakuchiol (11), 12-hydroxyisobakuchiol (12) and a new compound kuchiol (13) were isolated and their structures were determined. The antioxidant activity of the terpenic compounds and resin was evaluated using the bleaching of DPPH radical, Ferric Reducing Antioxidant Power (FRAP) and Total Reactive Antioxidant Properties (TRAP) assays.
Otholobium glandulosum (L.) J.W. Grimes (= Psoralea glandulosa L.) (Fabaceae) es un arbusto resinoso que crece en Chile. La composición química de su exudado resinoso fue determinada por primera vez. Tres meroterpenos: bakuchiol (8), 3-hidroxibakuchiol (11), 12-hidroxiisobakuchiol (12) y un nuevo compuesto kuchiol (13) fueron aislados y sus estructuras fueron determinadas espectroscópicamente. La actividad antioxidante de los compuestos terpénicos y la resina se evaluó mediante tres métodos antioxidantes DPPH, Poder Reductor de Hierro III (FRAP) y Capacidad Antioxidante Total (TRAP).

Context: Laurelia sempervirens (R. et P.) Tul., is an evergreen tree that growths in southern Chile, its leaves and bark are used in folk medicine as an infusion. Objective: The antimicrobial activities of the essential oil and ethyl acetate extract obtained from the bark of Laurelia sempervirens were investigated. Materials and methods: Ethyl acetate extract and essential oil were analyzed by GC- mass and the antimicrobial activity was investigated against gram positive and gram negative bacteria. Results: The extract and essential oil showed a strong antimicrobial activity against bacteria such as Acinetobacter baumanii, a relevant world nosocomial pathogen Discussion and conclusion: These findings demonstrate that the ethyl acetate extract and essential oil of L.sempervirens bark have excellent antimicrobial activities and thus have great potential as a source for natural health products.
Contexto: Laurelia sempervirens (R. et P.) Tul., es un árbol de hoja perenne que crece en el sur de Chile, sus hojas y corteza se utilizan en medicina popular como infusión. Objetivo: Investigar la actividad antimicrobiana del aceite esencial y el extracto de acetato de etilo obtenido de la corteza de Laurelia sempervirens. Materiales y métodos: El extracto de acetato de etilo y el aceite esencial se analizaron por GC-masa y la actividad antimicrobiana se analizó contra bacterias gram positivas y gram negativas. Resultados: El extracto y aceite esencial evidenció una fuerte actividad antimicrobiana frente a la bacteria, Acinetobacter baumanii patógeno que causa infecciones nosocomiales de relevancia mundial. Discusión y conclusiones: Estos hallazgos demuestran que el extracto de acetato de etilo y aceite esencial de corteza de L.sempervirens tienen excelentes actividades antimicrobianas y por lo tanto tienen un gran potencial como fuente de productos naturales para la salud.
