ABSTRACT

ABSTRACT Lippia alba (Mill.) N.E.Br. ex Britton & P. Wilson, Verbenaceae, is considered a great source of a bioactive volatile oil. Due to the wide range of known chemotypes, its chemical analysis is very important. Among the several activities of this volatile oil, a potential larvicidal action against Culicidae species is highlighted. However, the low water miscibility of volatile oils limits their application in aqueous media. Oil in water nano-emulsions are in the spotlight of novelty to solve this main problem. Thus, the aim of the present study was to obtain this nanostructured system with L. alba volatile oil (citral chemotype) and evaluate its larvicidal activity against Aedes aegypti and Culex quinquefasciatus larvae. The major compounds were geranial (30.02%) and neral (25.26%). Low mean droplet size (117.0 ± 1.0 nm) and low polydispersity index (0.231 ± 0.004) were observed and no major changes were observed after seven days of storage. LC50 values against C. quinquefasciatus and A. aegypti third-instar larvae were respectively 38.22 and 31.02 ppm, while LC90 values were, respectively, 59.42 and 47.19 ppm. The present study makes use of a low energy, solvent-free and ecofriendly method with reduced costs. Thus, this paper contributes significantly to phyto-nanobiotechnology of larvicidal agents, opening perspectives for the utilization of L. alba volatile oil in integrated practices of vector control.

ABSTRACT

Studies have suggested that total energy intake and diet composition affect lifespan and ageing. A high-fat diet induces oxidative stress and affects the development of diseases. In contrast, antioxidants are capable of reducing its harmful effects. Yerba mate beverages are an important source of antioxidants, but there is scarce knowledge about their effects on suppressing fat accumulation. Here, we investigated the compounds present in yerba mate extracts and assessed their effects on Drosophila melanogaster given a high cholesterol diet. LS-ESI-MS analysis showed the presence of matesaponins, phenolic compounds and methylxanthines in all of the examined extracts. In Drosophila, under extract treatment conditions, the mean lifespan was significantly extended from 38 to 43 days, there was an increase in the ability to support induced stress and decrease in lipid peroxidation products. Moreover, yerba mate extracts recovered the glutathione S-transferases (GST) activity and reduced the cholesterol level. Taken together, our results support that extracts can extend lifespan by reducing the detrimental effect of a high-fat diet in D. melanogaster, and this outcome can be associated with the compound content in the extracts. This study improves the understanding of natural interventions that reduce stress-induced oxidative damage, which is fundamental in promoting healthy ageing.

ABSTRACT

O Mesoclemmys vanderhaegei (Testudines: Chelidae) é um quelônio de água doce cuja ocorrência é descrita nas bacias dos rios Amazonas, Tocantins, Paraguai, Paraná e Uruguai. Consta na lista vermelha de espécies ameaçadas da International Union for Conservation of Nature, como espécie de baixo risco, mas que poderá se tornar ameaçada, sendo necessário a atualização de seus dados ecológicos e biológicos. Considerando que planos de manejo e conservação de espécies dependem também de vasto conhecimento sobre a biologia reprodutiva, apresentamos a primeira descrição macroscópica dos órgãos genitais de fêmeas jovens e adultas de M. vanderhaegei correlacionando esses achados ao tamanho do espécime e ao período do ano. Exemplares de M. vanderhaegei foram coletados no município de Chapada dos Guimarães, área de ampla ocorrência natural da espécie. A descrição dos órgãos genitais foi realizada a partir de 17 fêmeas fixadas em formol a 10% e dissecadas para evidenciação de particularidades relacionadas à anatomia externa e interna. Os órgãos genitais M. vanderhaegei jovens e adultas são constituídos por pares de ovários e ovidutos que desembocam dorsolateralmente na cloaca, formando junto com o ureter a papila urogenital. Os ovários são órgãos alongados, com a extremidade cranial mais larga e caudal afunilada, já os ovidutos são longos e localizados lateralmente aos ovários, sendo em adultas, muito diferenciado na sua forma e tamanho em relação ao das jovens. Em ambas faixas etárias, os órgãos genitais são sustentados por pregas de membrana celomática que emergem do teto da cavidade, constituindo os mesovário e mesoviduto. De acordo com a forma e o padrão de mucosa do oviduto em fêmeas adultas, o segmento cranial corresponde as regiões do infundíbulo e magnum, o segmento médio, ao istmo e no caudal identificam-se as regiões útero e vagina. No assoalho do urodeum aloja-se o clitóris. O comprimento retilíneo de carapaça e massa corpórea entre fêmeas imaturas e adultas variam, respectivamente, entre 134-155,6mm e 134,43-365g. A maioria das fêmeas jovens foram capturadas no período chuvoso e as adultas sem e com ovos no período seco. As características macroscópicas dos órgãos genitais observadas em M. vanderhaegei são compartilhadas com outros Testudines, exceto pela papila urogenital e presença do clitóris.(AU)


Mesoclemmys vanderhaegei (Testudines: Chelidae) is a freshwater turtle with occurrence in Amazon, Tocantins, Paraguai, Paraná and Uruguai rivers basins. Although according to International Union for Conservation of Nature, it has low risk of extinction, there is an uptade necessity of ecological and biological data. Considering that the management and conservation plans are related to a wide knowledge of reproductive biology, a first macroscopic description about the young and adults females of M. vanderhaegei is important. These points were correlated to the specimens size and period of the year. The samples of M. vanderhaegei were collected in Chapada dos Guimarães county, Mato Grosso, Brazil, an area of large natural occurrence of the specimens. Genital organs of seventeen females were fixed in 10% formalin and then dissected to demonstrate the particularities related to external and internal anatomy. The young and adult M. vanderhaegei genitals organs are composed of ovaries and oviducts pairs that dorsolaterally discharge into the cloaca, forming with the ureter, the urogenital papilla. The ovaries are elongated organs with larger cranial and elongated caudal portions. The oviducts, which are in adults very differentiated in its shape and size compared to the young, are long and located laterally to the ovaries. In both age groups, the genital organs are supported by celomatic membrane folds that emerge from the ceiling of the cavity, constituting the mesovary and mesoviduct. In adult females, according to the shape and pattern of the oviduct mucosa, the cranial segment corresponds to the regions of the infundibulum and magnum; the middle segment, the isthmus and the caudal segment identify the uterus and vagina regions. The clitoris is sited on the floor of the urodeum. The carapace linear length and body mass between immature and adult females vary to 134-155.6mm e 134.43-365g respectively. The main part of young females was captured in rainy period and the adults, with and without eggs, at dry period. The macroscopic characteristics of the M. vanderhaegei genital organs are also observed in others Testudines, with the exception of urogenital papilla and by the clitoris presence.(AU)

ABSTRACT

ABSTRACT Detection of virulent strains associated with aquatic environment is a current concern for the management and control of human and animal health. Thus, Vibrio diversity was investigated in four estuaries from state of Ceará (Pacoti, Choró, Pirangi and Jaguaribe) followed by antimicrobial susceptibility to different antimicrobials used in aquaculture and detection of main virulence factors to human health. Isolation and identification were performed on TCBS agar (selective medium) and dichotomous key based on biochemical characteristics, respectively. Nineteen strains of genus Vibrio were catalogued. Vibrio parahaemolyticus (Choró River) and V. alginolyticus (Pacoti River) were the most abundant species in the four estuaries. All strains were submitted to disk diffusion technique (15 antimicrobials were tested). Resistance was found to: penicillin (82%), ampicillin (54%), cephalotin (7%), aztreonan (1%), gentamicin, cefotaxime and ceftriaxone (0.5%). Five pathogenic strains were chosen to verification of virulence factors. Four estuaries showed a high abundance of species. High number of tested positive strains for virulence is concerning, since some of those strains are associated to human diseases, while others are known pathogens of aquatic organisms.

ABSTRACT

Macacos-da-noite são muito susceptíveis ao estresse e por isso a contenção química ou física deve ser cuidadosamente avaliada antes de qualquer procedimento. Protocolos anestésicos podem alterar alguns parâmetros fisiológicos, sendo o eletrocardiograma (ECG) um exame muito utilizado para avaliação do ritmo e da frequência cardíaca. O objetivo deste estudo foi avaliar a influência de quatro diferentes protocolos de contenção sobre o ECG realizado em Aotus azarae infulatus. Para isso foram utilizados 10 animais, machos, adultos, submetidos à contenção com a associação tiletamina/zolazepam (TZ), isoflurano (ISO), associação cetamina e midazolam (CET) e contenção física (CF). Não foram observadas diferenças nos parâmetros de ondas e complexos obtidos no ECG em todos os grupos testados, no entanto, durante a avaliação do traçado os animais do grupo CF apresentaram uma quantidade maior de alterações. Concluiu-se que os protocolos de contenção utilizados não alteraram os valores do ECG e que não foi possível considerar o grupo CF como controle devido causar mais alterações do que todos os protocolos testados.(AU)


Feline night monkeys are very susceptible to stress and therefore the physical or chemical restraint should be carefully evaluated before any procedure. Anesthetic protocols can alter some physiological parameters, and electrocardiogram (ECG) is a commonly exam used for evaluation of the rhythm and heart rate. The objective of this study was to evaluate the influence of four different protocols of restraining on ECG in Aotus azarae infulatus, in order to identify the best method of restraining in these primates. For that, we used 10 adult feline night monkeys, males, submitted to restraining protocols with the association tiletamine and zolazepam - (TZ), isoflurane (ISO), association ketamine and midazolam (CET) and physical restraint (CF). No differences were observed in the parameters wave and ECG complexes obtained in all groups tested; however, during the evaluation of the trace the animals of group CF showed a greater amount of change. It was concluded that the restraint protocols used did not change the values of the ECG, and it was not possible to consider the CF group as control due to more caused alterations than all other tested protocols.(AU)

ABSTRACT

O jacaré-do-pantanal (Caiman yacare) é uma espécie abundante no ecossistema do Pantanal. Sua exploração comercial está regulamentada desde 1990 e se tornou um agronegócio em expansão. Para atender essa demanda, uma unidade processadora instalada em Mato Grosso, vem comercializando carne de jacaré em diferentes cortes, entre os quais o filé de dorso, oriundo do pescoço, nos últimos quatro anos. O objetivo desta pesquisa foi descrever os músculos e correspondentes bases ósseas desse corte. Para a descrição de ossos, utilizaram-se seis carcaças desossadas de exemplares juvenis de jacaré-do-pantanal, além de um exemplar adulto obtido após morte do animal, por doação, do Zoológico da UFMT. Os ossos foram macerados em água corrente, clareados com solução de água oxigenada a 10 volumes, e seus detalhes anatômicos foram descritos. Para descrever o músculo, 24 exemplares juvenis foram obtidos após abate e esfola, conservados em freezer e descongelados quando utilizados, sem qualquer fixação. Após a evisceração, foram dissecados em ambos os antímeros, para verificação de simetria de ocorrência, fixações musculares, relacões de sintopia, forma e arquitetura muscular. Verificou-se que a coluna cervical em C. yacare apresenta nove vértebras cervicais (VC), associadas com as respectivas costelas, que servem de base principal ao filé de dorso, que é constituído pela musculatura cervical, exceto os músculos intertransversais cervicais e intercostais cervicais externos.


The yacare Caiman (Caiman yacare) is an abundant species in the Pantanal ecosystem. Commercial exploitation was regulated in 1990 and has become a thriving business. In order to fulfill this demand, a processing plant settled in Mato Grosso state, Brazil, has been supplying for the last four years different cuts of Pantanal Caiman meat, including the "filé de dorso" (back sirloin) obtained from the neck. The aim of this study was to describe the muscles and corresponding bones related to this cut. To describe the bones, we used six boned carcasses from juvenile yacare Caiman, as well as an adult specimen obtained after animal death, by donation from the Federal University of Mato Grosso Zoo. The bones were macerated in water, bleached with 10 volume-hydrogen-peroxide solution, and their anatomical details were recorded. In order to describe the muscles, 24 juvenile specimens were obtained after slaughter and skinning, preserved in a freezer at -20oC, and thawed at the time of use, without any fixation. After evisceration, the specimens were dissected on both sides to verify symmetry of structures, muscle attachments, sintopy relations, shape, and muscular architecture. The cervical spine of C. yacare features nine cervical vertebrae (CV) associated to their ribs, serving as the main base for the back sirloin cut, which is formed by neck muscles, except for the intertransverse cervical and external intercostal cervical muscles.

Sida tuberculata (Malvaceae) is a medicinal plant traditionally used in Brazil as an antimicrobial and anti-inflammatory agent. Here, we aimed to investigate the different extractive techniques on phytochemical parameters, as well as to evaluate the toxicity and antioxidant capacity of S. tuberculata extracts using in silico and in vitro models. Therefore, in order to determine the dry residue content and the main compound 20-hydroxyecdysone (20E) concentration, extracts from leaves and roots were prepared testing ethanol and water in different proportions. Extracts were then assessed by Artemia salina lethality test, and toxicity prediction of 20E was estimated. Antioxidant activity was performed by DPPH and ABTS radical scavenger assays, ferric reducing power assay, nitrogen derivative scavenger, deoxyribose degradation, and TBARS assays. HPLC evaluation detected 20E as main compound in leaves and roots. Percolation method showed the highest concentrations of 20E (0.134 and 0.096 mg/mL of extract for leaves and roots, respectively). All crude extracts presented low toxic potential on A. salina (LD50 >1000 µg/mL). The computational evaluation of 20E showed a low toxicity prediction. For in vitro antioxidant tests, hydroethanolic extracts of leaves were most effective compared to roots. In addition, hydroethanolic extracts presented a higher IC50 antioxidant than aqueous extracts. TBARS formation was prevented by leaves hydroethanolic extract from 0.015 and 0.03 mg/mL and for roots from 0.03 and 0.3 mg/mL on egg yolk and rat tissue, respectively (P<0.05). These findings suggest that S. tuberculata extracts are a considerable source of ecdysteroids and possesses a significant antioxidant property with low toxic potential.

ABSTRACT

O consumo de carne de jacaré-do-Pantanal tornou-se uma tendência de mercado e uma cadeia produtiva em ascensão no Estado de Mato Grosso, sendo 28,40% da carne comercializada nos últimos quatro anos oriundos do tronco. Estudos evolutivos, morfofisiológicos, ontogenéticos e tecnológicos foram desenvolvidos, mas não há descrição da musculatura e bases ósseas dos cortes comerciais. Objetivou-se descrever os músculos e correspondentes bases ósseas dos cortes filé de lombo, filé mignon e aparas. Na descrição óssea, utilizaram-se seis carcaças desossadas de exemplares juvenis de jacaré-do-Pantanal, além de um exemplar adulto, obtido por doação após óbito, do Zoológico da UFMT. Os ossos foram macerados em água corrente, clareados e descritos. Para a descrição muscular, 24 exemplares juvenis foram abatidos e esfolados, conservados em freezer e descongelados quando utilizados, sem qualquer fixação. Após a evisceração, foram dissecados em ambos os antímeros. Os músculos semiespinhal, longuíssimo e iliocostal, fixados nas vértebras e costelas torácicas, lombares e sacrais, formam o filé de lombo. O corte aparas é constituído pelos músculos grande dorsal, serrátil, peitoral e abdominais (oblíquo externo, oblíquo interno, transverso e reto), cuja base óssea corresponde as costelas torácicas, lombares e sacrais, a gastrália, o esterno e o epipúbis. Por sua vez, o m. puboisquiofemoral interno cranial, localizado na região sublombar e o m. troncocaudal, da superfície ventral da pelve, compreendem o filé mignon...


Yacare Caiman meat consumption has become a marketing trend and a commodity on the rise in Mato Grosso state in Brazil. In the last four years, cuts from the trunk represented 28.40% of total meat sales. Although evolutionary studies, morphophysiological ontogenetic and technology research have been carried out, characterization of muscle and bone bases of cuts from the torso has not been previously reported. The aim of this research is to describe the muscles and corresponding bones related to sirloin, filet mignon and meat trims cuts. To describe the bones, we used six boned carcasses from juvenile Yacare Caiman, as well as an adult specimen, obtained by donation after death from the Federal University of Mato Grosso Zoo. The bones were macerated, bleached and their anatomical details recorded. In order to study the muscle, 24 juvenile specimens were obtained after slaughter and skinning and dissected on both sides. The sirloin cut consists of the semispinal, longissimus and iliocostalis muscles, which are inserted on thoracic vertebrae and ribs, as well as lumbar and sacral ribs. The meat trims cut is formed by latissimus dorsi, serratus, pectoral and abdominal (external oblique, internal oblique, transversus and rectus) muscles, based in various bones: bone ribs are the thoracic, lumbar, and sacral ribs, the gastralia, the sternum and epipúbis. The filet mignon cut is formed by the internal puboischiofemoralis cranial (sublumbar) muscle and by the troncocaudal (ventral surface of the pelvis) muscle...

Ácaros do gênero Otodectes são parasitos encontrados frequentemente no ouvido de cães e gatos, sendo reconhecidos como os principais causadores de otite externa nesses animais. Trezentos e vinte cães, oriundos do município de Cuiabá, Mato Grosso, foram examinados através da otoscopia e do exame do swab parasitológico, com o objetivo de avaliar, através da análise bayesiana para a estimativa da prevalência, sensibilidade e especificidade destes métodos no diagnóstico da infestação causada pelo ácaro utilizando-se de dados a priori informativos e não informativos. Cada animal foi considerado uma unidade experimental. Do total de cães examinados, 142 (44,37%) apresentaram-se positivos para otocariose. Em 100 animais (31,25%) a infestação foi diagnosticada pelos dois métodos, em 31 (9,69%) apenas pelo exame do swab parasitológico, em 11 (3,44%) apenas pela otoscopia e em 178 (55,62%) animais o resultado foi negativo por ambos os métodos. A sensibilidade da otoscopia, quando comparada ao exame do swab parasitológico, foi considerada menor, a especificidade dos testes, porém, não difere estatisticamente. O modelo, a priori informativo, demonstrou ser o mais plausível e reduziu consideravelmente o intervalo de credibilidade das estimativas dos parâmetros. Mites of the genus Otodectes are parasites found frequently in the ear of dogs and cats, being recognized as the main causers of external otitis in these animals. Three hundred and twenty dogs, deriving of the city of Cuiabá, Mato Grosso, had been examined through the otoscopy and of the examination of parasitological swab, with the objective to evaluate through the bayes'analysis for the estimate of the prevalence, sensitivity and especificity of these methods in the diagnosis of the infestation caused for the mite, using dates informative and not informative a priori. Each animal was considered an experimental unit. Of the total of examined dogs, 142 (44.37%) had been presented positive for otocariosis. In 100 animals (31.25%) the infestation was diagnosed by the two methods, in 31 (9.69%) only for the examination of parasitological swab, in 11 (3.44%) only for the otoscopy and in 178 (55.62%) animal the result was negative for both the methods. The sensitivity of the otoscopy, when compared with the examination of parasitological swab, was considered lesser, however the especificity of tests they do not differ. The informative model with priori it demonstrated to be most reasonable and considerably reduced the interval of credibility of the estimates of the parameters. It can be concluded that Bayes'analysis is effective in the estimation of prevalence and characteristics of the diagnostic tests...

ABSTRACT

The occurrence of psychrotrophic bacteria in raw milk is studied worldwide due to the difficulties associated with controlling their growth during cold storage and the consequent negative effects upon fluid milk or dairy products. Among the psychrotrophic bacteria, the genus Pseudomonas (represented primarily by P. fluorescens) has been highlighted as the cause of numerous defects in dairy products. In light of its perceived predominance, this species has frequently been chosen as a model organism to assess the effects of psychrotrophic bacteria on milk or to evaluate the efficacy of control measures. However, recent findings derived from the application of molecular biological techniques have exposed a number of deficiencies in our knowledge of the biology of milk-associated psychrotrophs. Furthermore, it has been revealed that microbe to microbe communication plays a significant role in determining both the identities and the extent to which different groups of microbes develop during cold storage. The application of molecular identification methods has exposed errors in the classification of members of the genus Pseudomonas isolated from cold stored milk and has stimulated a reevaluation of the presumed status of P. fluorescens as the predominant milk-associated psychrotrophic species. This article presents a succinct review of data from studies on psychrotrophic bacteria in milk, some of which contest established theories in relation to the microbiology of cold stored raw milk, and poses the question: how much do we really know?.

The Renaissance was a period of extensive scientific and cultural production, which occurred between the fourteenth and sixteenth centuries. One of the exponents of this artistic period was the poet, architect, sculptor and painter Michelangelo Buonarroti, who was born and lived in Italy between 1475 and 1564. Among his best known artworks are the frescoes painted on the Sistine Chapel ceiling. Currently, there is discussion if the paintings are only representations made from the sacred guidance of the church at the time, or if there are other meanings hidden in the images. From this context, we analyzed studies that associated the frescoes painted on the Sistine Chapel ceiling with anatomical structures hidden in the images, taking into account their significance, importance, and if these structures are not simply an imaginative interpretation of the researchers. This study was performed aiming to complement the work published by Ellwanger, Mohr and Campos (2012) in this journal.

ABSTRACT

A criação de jacaré-do-Pantanal tem se consolidado no estado de Mato Grosso como atividade alternativa e legal para as propriedades rurais na área de ocorrência natural da espécie, coibindo a caça predatória e colaborando na preservação da espécie. A racionalização do processo produtivo na criação de jacaré é uma ação relativamente nova, que permite obter pele de melhor qualidade e de utilização integral, diferentemente daquelas oriundas de animais da natureza. Considerando a importância dos intestinos como sede dos principais eventos relacionados à obtenção de nutrientes para o metabolismo corpóreo, este trabalho teve por objetivo caracterizar qualitativa e quantitativamente a mucosa dos intestinos delgado e grosso de jacaré-do-Pantanal jovens. Para a caracterização ao microscópio óptico da estrutura da parede intestinal e da histometria da mucosa foram coletadas amostras intestinais de 16 animais, sendo três deles também destinados ao estudo histoquímico. As amostras foram obtidas de cinco regiões, sendo quatro do intestino delgado e uma do intestino grosso. Elas foram fixadas em solução de Bouin, processadas de acordo com técnicas preconizadas para inclusão em parafina. Cortes histológicos semi-seriados (quatro micrômetros) foram obtidos e o material foi desparafinizado, hidratado e corado pela hematoxilina-eosina para a descrição geral e estudo histométrico, que consistiu da mensuração da freqüência das vilosidades intestinais; dos comprimentos do epitélio e da lâmina própria da mucosa e da espessura da mucosa. Na análise estatística foi empregada a análise de variância e Teste de Tukey. Para o estudo histoquímico foram utilizadas a reação ao ácido periódico de Schiff, alcian blue pH 1,0 contrastado com hematoxilina e alcian blue pH 2,5 conjugado ao ácido periódico de Schiff. Para a descrição das estruturas histológicas foi empregada a terminologia disponível na Nomina Histologia. A estrutura da parede do intestino delgado e grosso do jacaré-do-Pantanal era constituída pelas túnicas mucosa, muscular e serosa. A mucosa era formada por epitélio de revestimento do tipo cilíndrico simples constituído por epiteliócitos colunares e por exocrinócitos caliciformes; lâmina própria de tecido conjuntivo frouxo e muscular da mucosa única. A túnica muscular era constituída por dois estrados, o circular e o longitudinal, sendo o estrato circular o mais desenvolvido. A serosa era típica. As especializações da mucosa observadas no intestino delgado e grosso, respectivamente, foram vilosidades e pregas intestinais, e ao longo das regiões dos intestinos, apresentaram diminuição de sua complexidade. O estudo histométrico da mucosa demonstrou diferença estatisticamente significante entre o intestino delgado e grosso. Embora a estrutura da parede intestinal de C. yacare seja semelhante à de outros crocodilianos, a da tartaruga verde e avestruz, ainda se faz necessário estudos sobre a histofisiologia para que o manejo nutricional da espécie em cativeiro seja incrementado.


Commercial breeding of Caiman yacare has been consolidated in the state of Mato Grosso, Brazil, as an alternative and legal activity for rural properties in the area of its natural occurrence, to restrain predatory hunting and for preservation of the species. Rationalization of the production process in alligator breading is a relatively new action, which allows a better skin quality and integral use, different from the production of skins from animals in nature. Considering the importance of the intestines as being the main place of the major events related to nutrients obtainment for the body metabolism, this study aimed to characterize qualitative and quantitatively the mucosa of the small and large intestine of young C. yacare. For the characterization of the intestinal wall structure in optical microscopy and histometry of the mucosa were collected intestinal samples from 16 animals, three of which were also used for the histochemical study. The samples were obtained from five regions, four from the small intestine and one from the large intestine. They were fixed in Bouin solution and processed according to the techniques recommended for paraffin inclusion. Semiserial histological sections (about four micrometers) were obtained and the specimen was deparaffinized, hydrated, and stained with hematoxylin-eosin for the general description and histometric study, which consisted of measuring the frequency of intestinal villi, lengths of epithelium and lamina propria of the mucosa, and mucosal thickness. In the statistical analysis was applied the variance analysis and Tukey test. For the histochemistry study were used the reaction to periodic acid-Schiff, alcian blue pH 1.0 counterstained with hematoxylin and alcian blue pH 2.5 conjugated to the periodic acid-Schiff. For description of the histological structures was used the terminology available in the Histological Nomina .The wall structure of the small and large intestine of the C. yacare was formed by mucosa, muscular and serosa tunics. The mucosal consisted of lining epithelium type simple cylindrical formed by columnar epithelial cells and goblet cells; lamina propria of loose connective tissue and a single layer of muscularis mucosa. The muscular tunic consisted of two stratums, the circular and the longitudinal, being the circular stratum more developed. The serosa was typical. The specializations of the mucosa observed in the small and large intestines respectively, were intestinal villi and folds, and along the regions of the intestines, showed a reduction of its complexity. The histometric study of the mucosa showed statistically significant difference between the small and large intestines. Although the structure of the intestinal wall of C. yacareis similar to other crocodilians, the green turtle and the ostrich, it's still necessary studies about the histophysiology that the nutritional management of the species in captivity be incremented.

Para realização do estudo microscópico das glândulas anexas à uretra masculina, foram utilizadas duas capivaras (Hydrochoerus hydrocaeris), adultas, das quais foram coletados fragmentos das glândulas genitais acessórias, imersos em solução fixadora de Bouin e lavados cuidadosamente em álcool de 70 por cento ao absoluto. A seguir foram submetidos aos processos histológicos de rotina e corados pelos métodos de Hematoxilina/Eosina e Tricrômico de Masson. Os resultados morfológicos encontrados foram: o ducto deferente possui um espessamento da parede, onde a luz permanece inalterada e sem presença de epitélio granular. A glândula vesicular possui um epitélio secretor do tipo pseudoestratificado colunar. A glândula prostática possui mucosa com pregueamentos altos e ramificados, revestido por epitélio pseudoestratificado cilíndrico. Machos de capivaras possuem glândulas vesiculares e próstata como glândulas uretrais. No material examinado não foi identificado epitélio secretor correspondente a glândula bulbouretral e morfologicamente assemelha-se aos outros histricomorfos.


To perform a microscopic study of the glands attached to the male urethra, we used two adult capybaras (Hydrochoerus hydrocaeris) from which fragments of the urethral glands were collected, immersed in Bouin's fixative solution, washed thoroughly with 70 percent to absolute alcohol, submitted to routine histological techniques and stained with hematoxylin/eosin and Masson's trichromic method. The morphological findings were: Vas deferens has a thickening of the wall without granular epithelium and with unchanged lumen; the vesicular gland has a secretory epithelium of pseudostratified columnar type; the prostate gland has a high and ramified mucous membrane covered with pseudostratified cylindrical epithelium. Male capybaras have vesicular, prostate as urethral glands. In the material examined, no secretory epithelium corresponding to bulbourethral glands was identified; morphologically it resembles to the other histricomorphs.

Changes in urinary porphyrin excretion may be the result of hereditary causes and/or from environmental or occupational exposure. The objective of this study was to measure the amount of some porphyrins in spot urine samples obtained from volunteers randomly selected from a healthy adult population of São Paulo with a sensitive HPLC method and to estimate normal ranges for a non-exposed population. Spot urine samples were collected from 126 subjects (both genders, 18 to 65 years old) not occupationally exposed to porphyrinogenic agents. Porphyrin fractions were separated on RP-18 HPLC column eluted with a methanol/ammonium acetate buffer gradient, pH 4.0, and measured fluorometrically (excitation 405 nm/emission 620 nm). The amount of porphyrins was corrected for urinary creatinine excretion. Only 8-carboxyl (uro) and 4-carboxyl (copro) porphyrins were quantified as µg/g creatinine. Data regarding age, gender, occupational activities, smoking and drinking habits were analyzed by Mann-Whitney and Kruskal-Wallis tests. Uroporphyrin results did not differ significantly between the subgroups studied. Copro and uro + copro porphyrins were significantly different for smokers (P = 0.008) and occupational activities (P = 0.004). With respect to alcohol consumption, only men drinking >20 g/week showed significant differences in the levels of copro (P = 0.022) and uro + copro porphyrins (P = 0.012). The 2.5-97.5th percentile limit values, excluding those for subjects with an alcohol drinking habit >20 g/week, were 0-20.8, 11.7-93.1, and 15.9-102.9 µg/g creatinine for uro, copro and uro + copro porphyrins, respectively. These percentile limit values can be proposed as a first attempt to provide urinary porphyrin reference values for our population, serving for an early diagnosis of porphyrinopathies or as biomarkers of exposure to porphyrinogenic agents.

ABSTRACT

Los avances científicos en el campo de la química y de la medicina se han apropiado la inquietud popular que desde hace muchos años utiliza los recursos que ofrece la naturaleza. La planta Cannabis sativa ha sido tradicionalmente utilizada con propósitos medicinales, así como con fines recreativos. Sin embargo, sus importantes efectos psicoactivos han constituido una limitante para su uso. Algunos gobiernos han comenzado a permitir su uso de forma controlada, con fines medicinales. El descubrimiento de compuestos endógenos con propiedades similares a los cannabinoides (endocanabinoides), y el previo conocimiento de los receptores CB1 y CB2, sobre los cuales actúan, ha despertado interés en el mundo entero por descubrir nuevas rutas terapéuticas para el tratamiento de diferentes patologías. Las posibilidades de aprovechar la estructura de los endocanabinoides para diseñar nuevas moléculas con actividad antineoplásica están llevando a enfrentar este reto, buscando fármacos tan eficaces como los actualmente utilizados pero con menos efectos tóxicos. Se está avanzando en el conocimiento de los aspectos moleculares de muchos de estos compuestos, así como en las rutas bioquímicas sobre las cuales actúan.

ABSTRACT

Brazil is a country of continental dimension with a population of different ethnic backgrounds. Thus, a wide variation in the frequencies of hepatitis C virus (HCV) genotypes is expected to occur. To address this point, 1,688 sequential samples from chronic HCV patients were analyzed. HCV-RNA was amplified by the RT-PCR from blood samples collected from 1995 to 2000 at different laboratories located in different cities from all Brazilian States. Samples were collected in tubes containing a gel separator, centrifuged in the site of collection and sent by express mail in a refrigerated container to Laboratório Bioquímico Jardim Paulista, São Paulo, SP, Brazil. HCV- RNA was extracted from serum and submitted to RT and nested PCR using standard procedures. Nested PCR products were submitted to cycle sequencing reactions without prior purification. Sequences were analyzed for genotype determination and the following frequencies were found: 64.9 percent (1,095) for genotype 1, 4.6 percent (78) for genotype 2, 30.2 percent (510) for genotype 3, 0.2 percent (3) for genotype 4, and 0.1 percent (2) for genotype 5. The frequencies of HCV genotypes were statistically different among Brazilian regions (P = 0.00017). In all regions, genotype 1 was the most frequent (51.7 to 74.1 percent), reaching the highest value in the North; genotype 2 was more prevalent in the Center-West region (11.4 percent), especially in Mato Grosso State (25.8 percent), while genotype 3 was more common in the South (43.2 percent). Genotypes 4 and 5 were rarely found and only in the Southeast, in São Paulo State. The present data indicate the need for careful epidemiological surveys throughout Brazil since knowing the frequency and distribution of the genotypes would provide key information for understanding the spread of HCV.

ABSTRACT

Objetivo: Estudar o impacto da manometria anorretal na avaliação da incontinência fecal e no diagnóstico da doença de Hirschsprung, na Unidade de Gastroenterologia do Departamento de Pediatria (HCRPUSP). Método: Análise retrospectiva dos prontuários de 130 crianças submetidas à manometria anorretal para investigação de constipação intestinal (grupo I) e de incontinência fecal (grupo II), correlacionando os achados manométricos, avaliação ao enema opaco, biópsia retal e evolução clínica. Resultados: No grupo I, o enema opaco foi normal em 55%, a manometria anorretal evidenciou reflexo retoesfincteriano em 80% e a biópsia anorretal mostrou neurônios em 52%. Mediante esses exames e evolução clínica foi possível subdividir as crianças em: com constipação funcional (87), com doença de Hirschsprung (11) e com pseudo-obstrução intestinal (cinco). As 12 crianças restantes tinham estomias, em oito, decorrentes de enterocolite e em quatro, como tratamento provisório de malformação anorretal. Nessas crianças, a avaliação conjunta com enema opaco, manometria e biópsia anorretais possibilitou descartar Hirschsprung. As crianças do grupo II apresentavam meningomielocele (uma), escape fecal (duas), pós-operatório de malformação anorretal (sete), pós-operatório de doença de Hirschsprung (três) e diarréia crônica (duas). A pressão basal anal média em uma criança no pós-operatório de malformação anorretal foi de 50mmHg e, em duas, menos de 20mmHg. Em uma criança com pós-operatório de Hirschsprung a pressão foi de 52mmHg e nas crianças com escape fecal, de 55mmHg. Conclusões: A manometria anorretal é um excelente método na investigação dos distúrbios intestinais na infância. Deve ser utilizada na tentativa da exclusão da doença de Hirschsprung e na avaliação da incontinência fecal, pois, quando é normal, pode, na maioria dos casos, evitar a realização de exames mais invasivos

ABSTRACT

Hidrolisado de bagaço de cana-de-açúcar contendo uma concentração inicial de ácido acético de 3,5g/L foi utilizado como meio de fermentação para a bioconversão de xilose em xilitol pela levedura Candida guilliermondii FTI 20037. Ácido acético (2,0g/L) foi adicionado ao meio em diferentes tempos de fermentação, com o objetivo de avaliar o efeito deste ácido neste bioprocesso. O maior efeito inibitório deste ácido na bioconversão de xilose em xilitol pela levedura ocorreu quando este foi adicionado ao meio após 12h de fermentação. Nesta condição observou-se uma redução de 23,22 porcento e 11,24 porcento, respectivamente, no consumo de xilose e no crescimento celular em relação à fermentação em que a adição deste ácido ocorreu no tempo inicial de incubação. Como conseqüência do efeito inibitório, observou-se os menores valores de rendimento (0,39g/g) e produtividade (0,22g/L.h) de xilitol, correspondendo a uma redução de 36 e 48 porcento, respectivamente, em relação aos valores obtidos com a adição de ácido acético nos outros tempos de fermentação. Os resultados obtidos permitem concluir que, nas condições experimentais empregadas neste trabalho, o efeito inibitório do ácido acético sobre a bioconversão de xilose em xilitol é dependente do tempo de fermentação em que a adição do ácido foi feita e não apenas de sua concentração no meio.
