ABSTRACT

AIM: To compare the effectiveness and complications of treating rhegmatogenous retinal detachment(RRD)with foldable capsule body(FCB)and scleral buckling(SB).METHODS: The clinical data of 81 patients(82 eyes)with RRD who underwent surgery at our hospital from March 2019 to April 2022 were retrospectively analyzed. The differences in retinal reattachment rate, best-corrected visual acuity, the absorption of subretinal fluid, postoperative discomfort and incidence of complications between the two treatments were compared.RESULTS: The retinal reattachment rate was 96% in the FCB group and 92% in the SB group, with no significant difference between the two groups(P>0.05). The best corrected visual acuity of the affected macular eyes was different in the both groups(P<0.01). Both groups effectively promoted the absorption of subretinal fluid. The operation time of FCB group was 16.50(12.75, 25.00)min, while it was 38.00(36.25, 41.75)min in the SB group(P<0.001). Patients in the FCB group also had significantly lower eyelid swelling and pain symptoms than those in the SB group(P<0.001). The visual analogue scale(VAS)score at 1d after operation was 1.00(0.00, 2.00)in the FCB group and 3.00(2.00, 3.00)in the SB group(P<0.001).CONCLUSION: FCB is a safe and effective surgical method to treat RRD that can alleviate patient's pain. Furthermore, FCB has a significantly shorter operation time and milder postoperative adverse reactions than SB.

ABSTRACT

OBJECTIVE@#To investigate the expression and clinical significance of T helper cell 9 (Th9) and its cytokine interleukin 9(IL-9) in peripheral blood of patients with chronic lymphocytic leukemia(CLL).@*METHODS@#A total of 43 newly diagnosed patients with chronic lymphocytic leukemia in the First Affiliated Hospital of Xinjiang Medical University from June 2021 to June 2022 were selected as the case group. The patients were divided into Binet A group (13 cases), Binet B group (20 cases) and Binet C group (10 cases) by Binet staging system, and 20 healthy volunteers who underwent physical examinationin in our hospital in the same period served as control group. The proportion of Th9 cells in peripheral blood was detected by flow cytometry, the expression level of Th9 specific transcription factors PU.1 and IRF4 was detected by Western blot, and the expression level of serum cytokine IL-9 was detected by ELISA. The proportion of Th9, the expression of PU.1, IRF4 and IL-9 in each group were compared, and the correlation between the proportion of Th9, IL-9 and clinicopathological indexes of CLL patients was analyzed.@*RESULTS@#The proportion of Th9, the expression of PU.1, IRF4 and IL-9 in CLL group were significantly higher than those in control group (P<0.05), the proportion of Th9 and the expression of IL-9 in Binet B and C group were higher than those in Binet A group (P<0.05), but there was no significant difference in the proportion of Th9 cells between Binet B group and C group (P>0.05). The expression of IL-9 in Binet C group was significantly higher than that in Binet B group (P<0.05) . The proportion of Th9 cells and IL-9 were highly expression in patients with β2 microglobulin abnormality, IGHV unmutation, P53 abnormality and hepatosplenic lymph node enlargement(P<0.05), but not related to age and sex (P>0.05). The results of Spearman correlation analysis showed that the proportion of Th9 in patients with CLL was negatively correlated with the lymphocytic account and lymphocyte proportion(rs=-0.32,rs=-0.34). The proportion of Th9 and IL-9 were positively correlated with Binet stage, Rai stage and CLL-IPI Scoring (rs=0.79,rs=0.54,rs=0.58; rs=0.72,rs=0.63,rs=0.45), but not with WBC, CD4+ T cells and CD8+T cells (P>0.05). The proportion of Th9 was positively correlated with IL-9 (rs=0.53).@*CONCLUSION@#Th9 cells and IL-9 are abnormally highly expressed in CLL, which is related to the poor prognosis of CLL.

ABSTRACT

OBJECTIVE: To explore the application value of microflow digital imaging (MDI) in sub-visible particle detection of monoclonal antibodies(mAbs). METHODS: Polystyrene latex particles were adopted as standard particles to evaluate the stability and accuracy of MDI, then MDI was used to detect the quantities and compositions of sub-visible particles with diameters of ≥10 μm and ≥25 μm in two groups of mAbs. One group containing three mAbs from different companies against EGFR was selected to show the compositional difference of sub-visible particles. Another group was set to evaluate the impact of storage conditions on stability of sub-visible particles in mAb against TNF-α. The storage conditions included 4℃, 37℃ and exposure under light of 6 000 lx. RESULTS: Using polystyrene latex particles, MDI method showed acceptable stability and accuracy. The analysis showed that the numbers of sub-visible particles in mAbs from Company 1 and Company 2 were comparable, but their main compositions were non-protein and protein particles separately. The number of sub-visible particles in mAb from Company 3 was much higher than that of Company 1 and Company 2 and the sub-visible particles included both non-protein and protein particles, indicating differed strategies should be adopted to control sub-visible particles. It was also found that the selected antibody against TNF-α was much more unstable when stored at 37℃ or exposed under light of 6 000 lx as the quantities of protein particles were increased dramatically, which suggested the obvious impact of storage conditions on stability of protein in mAbs. CONCLUSION: MDI can provide not only diameters and the quantities of sub-visible particles, but also morphological information and tell the nature of the particles. These functions are powerful tools for sub-visible particle control and quality evaluation of mAbs.

Methods 48 cases of severe acute pancreatitis patients in Zhejiang Traditional Chinese Medical hospital from January 2012 to December 2016 were divided into the experimental group 24 cases and the control group 24 cases, the control group was given routine clinical treatment, the experimental group on the basis of the control group was given psychological intervention and somatostatin, the treatment effect of the patients between two groups were compared and analyzed. Results APACHEⅡ score, decline degree of Serum amylase, mortality, hospitalization time, remission time and complication rate in the experimental group was significantly better than the control group, the difference between groups was statistically significant (P<0.05). Conclusion In the treatment of severe acute pancreatitis, the combination of somatostatin and psychological intervention could effectively alleviate the clinical symptoms and control the development of the disease.

ABSTRACT

MicroRNAs (miRNAs) are small noncoding regulatory RNAs ranging in size from 17 to 25 nucleotides which participate many physiological and pathological processes.MicroRNA could also stably exist in peripheral blood,and the detection of circulating microRNA is of great significance for disease diagnosis and prognosis.As so far,there is no unified method and standard for detection of microRNA,which is the major reason for discrepant results between different studies.However,circulating microRNA,as a new disease detection biomarker,has stable properties,convenient detection and high accuracy features,so it till has an important potential value in clinical applications.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the main proteinases responsible for CD16b shedding under different stimulators.</p><p><b>METHODS</b>HEK293 cell line stably expressing CD16b was constructed by lentivirus system. The cell line was then overexpressed with a disintegrin and metalloproteinase 10 (ADAM10) or ADAM17, suppressed with short hairpin RNA of ADAM10 or ADAM17, and reconstituted with ADAM10 or ADAM17, respectively. After each treatment, the cell line was stimulated with ionomycin or phorbol 12-myristate- 13-acetate (PMA) for 12 hours. The soluble CD16b released from cell membrane was detected by immunoprecipition and immunoblot. Quantitation was then implemented to compare the amount of soluble CD16b in cell supernatant after stimulation.</p><p><b>RESULTS</b>HEK293 cell line stably expressing CD16b was successfully established. When CD16b expressing cell line was overexpressed with ADAM10, shedding of CD16b was increased after stimulation with ionomycin but not PMA; when the cell line overexpressed with ADAM17, shedding of CD16b was increased after stimulation with PMA but not ionomycin. Similarly, when ADAM10 was suppressed by short hairpin RNA, CD16b shedding was decreased after stimulation with ionomycin; when ADAM17 was suppressed by short hairpin RNA, CD16b shedding was decreased after stimulation with PMA. The shedding of CD16b was increased again when CD16b expressing cell line was reconstituted with ADAM10 and stimulated by ionomycin or reconstituted with ADAM17 and stimulated by PMA.</p><p><b>CONCLUSIONS</b>Both ADAM10 and ADAM17 could shed CD16b, but they possess differed preferences. ADAM10 is the main sheddase under stimulation of ionomycin, while ADAM17 is the main sheddase under stimulation of PMA.</p>

Objective To investigate an association between Toll-like receptor(TLR)4 Asp299Gly or CD14 C-260T polymorphisms and colorectal cancer in Chinese patients.Methods By a method of polymerase chain reaction-based restriction fragment length polymorphism(PCR-RFLP), TLR4 Asp299Gly and CD14 C-260T polymorphisms in unrelated 110 patients with colorectal cancer and 160 healthy controls in Chinese Han race population were genotyped.Results Significant differences of CD14 genotypes were found between healthy controls and the patients with colorectal cancer.The fre- quency of the CC genotype in healthy controls was extremely lower than that in the group of colorectal cancer(15.6% vs.31.8%,P=0.0027,OR=0.3968,95%CI:0.2209-0.7129)while the frequency of the CT genotype in healthy controls was significantly higher than that in the group of colorectal cancer (48.1% vs.30.9%,P=0.0056,OR=2.074,95%CI:1.246-3.452).No TLR4 Asp299Gly mutant genotype was detected in any patients and healthy controls in Chinese Han population.Conclusion The polymorphism of CD14 C-260T but not TLR4 Asp299Gly is associated with colorectal cancer in Chinese patients.
