ABSTRACT

BACKGROUND:Exosomes are vesicle-like structures secreted by cells into extracellular compartments in the form of cytosol, which contain a large amount of microRNAs with important intercellular communication roles.MicroRNAs in exosomes rely on exosome transport and are able to enter target cells to exert important biological regulatory effects. In common bone and joint diseases, abnormal or damaged bone metabolism releases a large number of exosomes, while some exosome-derived microRNAs also promote the progression of osteoarthritis.Therefore,exosome-derived microRNAs are closely related to the skeletal system and are important for the development as well as diagnosis and treatment of many osteoarticular diseases. OBJECTIVE:To review the research progress of exosome-derived microRNAs in bone metabolism and bone and joint diseases. METHODS:Using"exosomes, extracellular vesicle, microRNA,miRNA,bone, bone diseases, bone formation, bone regeneration, bone resorption, bone destruction"as Chinese and English search terms, articles were searched on CNKI,Metasys, and PubMed databases.Finally,86 articles were included for summarization. RESULTS AND CONCLUSION:Exosome-derived microRNAs can regulate bone metabolism by affecting bone formation and bone resorption, and are closely related to the development of bone and joint diseases such as fracture healing, osteoporosis, osteoarthritis, rheumatoid arthritis, osteonecrosis of the femoral head, and osteosarcoma.Exosome-derived microRNAs will be an effective means of diagnosis and treatment of certain bone and joint diseases in the future. However, the current research on exosome-derived microRNAs in osteoarthritic diseases is limited, and more explorations and researches are still needed to diagnose and treat osteoarthritic diseases using exosome-derived microRNAs.

ABSTRACT

OBJECTIVE:To optimize the extraction technology of Compound Rhodiolae crenulatae granule. METHODS:The pharmacodynamic index of learning and memory ability was used to screen the water extraction, water extraction and alcohol pre-cipitation and alcohol extraction methods for Compound R. crenulatae granule in mice by Y-type water maze test. Using the compre-hensive scores of salidroside, ferulic acid extraction amount and extraction yield as indexes, orthogonal test was used to optimize the ethanol volume fraction, extraction time, solvent amount and extraction times, and verification test was conducted. RESULTS:Products by alcohol extraction can significantly improve learning and memory ability in the 3 methods;the optimal extraction tech-nology was confirmed to 8-fold 70% ethanol reflux extracting for 3 times,1.0 h each time. In verification test, average extraction amount of salidroside and ferulic acid were 4.884 mg/g and 1.669 mg/g,and average extraction yield was 33.52%(all RSD<2%, n=3). CONCLUSIONS:The optimized extraction technology is stable, reliable, simple and feasible, which can be used for indus-trial extraction of Compound R. crenulatae granule.

ABSTRACT

OBJECTIVE: To study the influence factors of the oxidation of DHA and EPA,and to find effective antioxidants.METHODS: The oxidation of DHA and EPA was optimized by orthogonal test with the quality of DHA and EPA as indicators and temperature, light and time as factors. The optimized antioxidants were selected by adding salidroside,BHA,BHT,vitamin E into samples.RESULTS: If there is lower temperature, weaker light and shorter storage, the oxidation of DHA and EPA will be more slowly.Effect of salidroside on the oxidation of DHA and EPA is the strongest.CONCLUSION: The oxidation of DHA and EPA can be reduced by lowering temperature, storing in dark place, reducing storage duration and adding salidroside.

ABSTRACT

Paeoniflorin in antiaging Preparation was qualitatively identified with chemical and TLC methods and quantitatively determined with HPLC. The results showed both tne chemical and TLC method gave fast and accurate results and HPLC was an excellent for quantitative determination with an average recovery of 100.72% and coefficient of variation 1.22%, which is suitable for the quality control of the preparation.
