ABSTRACT

Objective:To study the effect of baicalein on the expression of glutamate receptor related protein in PC12 cells injured by Aβ 25-35. Methods:PC12 cells were divided into control group, model group, estradiol group and baicalein group with different concentrations. The survival condition of PC12 cells in each group were detected by thiazole blue (MTT). PC12 cells were divided into control group, model group, estradiol group and baicalein group. The control group and model group were cultured with DMEM medium, and the estradiol group was added with 1×10 -3 μmol/L estradiol DMEM medium, baicalein group was added with 1 μmol/L baicalein DMEM medium. After 2 hours of intervention, 20 μmol/L Aβ 25-35 was added to the model group, estradiol group and baicalein group with induced PC12 cell injury. After 22 hours of intervention, flow cytometry was used to detect the apoptosis of PC12 cells. The expression of estrogen receptor β (ER β), phosphorylated c-Jun N-terminal kinase (p-JNK/JNK) and ionic receptor N-methyl-D-aspartate receptor 1 (NMDAR1), glutamate receptor 2 (GluR2) and calcium/calmodulin dependent protein kinase Ⅱ (CaMK Ⅱ) were detected by Western blot. Results:Compared with model group, 1 μmol/L baicalein significantly increased the proliferation rate [(95.80±2.47)% vs. (64.34±3.84)%]. The apoptosis rate of PC12 cells[(7.83±0.67)% vs. (12.84±0.91)%] was significantly decreased in baicalein group ( P<0.01). The expression of NMDAR1 (0.582±0.012 vs. 0.352±0.012), GluR2(0.538±0.017 vs. 0.355±0.006), ER β (0.362±0.015 vs. 0.262±0.018) in baicalein group were significantly increased ( P<0.01), the expression of p-JNK/JNK (0.476±0.013 vs. 0.752±0.014) and CaMK Ⅱ(0.499±0.019 vs. 0.670±0.016) in baicalein group were significantly decreased ( P<0.01). Conclusions:Baicalein has a protective effect on PC12 cells injured by Aβ 25-35. Its mechanism may be related to the inhibition of p-JNK/JNK activity by activating ERβ and regulating the expression of glutamate receptor related protein.

ABSTRACT

This study was aimed to investigate the effects of Bu-Yang Huan-Wu (BYHW) decoction to the learning and memory ability of APP/PS1 double transgenic mice through two behavior tests,in order to observe the Alzheimer's disease (AD) treatment effect of BYHW decoction.A total of 60 APP/PS1 double transgenic mice were randomly divided into five groups,which were the model group,the hydrochloride group,the high-,middle-,and low-dose BYHW decoction group,with ten rats in each group.The C57BL/6J mice were used as the control group.After 30 days of drug administration,all mice were subjected to behavior tests,including new object recognition experiment and step-through task.Western blot was used to detect changes of inflammatory factors,including IL-6 and TNF-α,in hippocampus of mice.The results showed that compared with the model group,the high-and middle-dose BYHW decoction can significantly decrease inflammatory factors of IL-6 and TNF-α expression in hippocampus of APP/PS1 mice.It can also obviously improve the learning and memory ability of APP/PS1 mice.It was concluded that BYHW decoction can significantly decrease the inflammatory factor expression in APP/PS 1 double transgenic AD model mice.It can obviously improve its learning and memory ability,which can be used in the treatment of AD.

ABSTRACT

We isolated the strain MBL13 with high collagenase productivity from the soil of piled up animal bones. It was identified as Bacillus cereus. We purified and characterized Bacillus cereus collagenase (BCC). The molecular weight of BCC was 38.0 kDa and the optimum temperature and pH for the enzyme activity were 40 degrees C and 8.0 respectively. The enzyme was stable when the temperature was below 50 degrees C, but only retained 10% activity when kept at 60 degrees C for 1 h. The enzyme activity was stable between pH 7.0-8.5. Some metal ions such as Ca2+, Zn2+, Mg2+ enhanced the enzyme activity, and Cu2+ brought the obvious inhibition. In addition, EDTA and EGTA could inhibit the enzyme activity. We suggested that the purified enzyme was a member of the metalloproteases. Based on the experiment of substrate specificity, we found that the purified enzyme was bone collagenolytic protease, and had a much stronger capacity of hydrolysis for type I collagen than that for type II collagen and type III collagen. By BCC hydrolyzing bone collagen, we obtained polypeptides with different chain lengths. The comparative test indicated that the hydrolysis capacity of BBC was higher than that of standard type I collagenase. The results introduced a new strain and a novel collagenolytic protease for industrial enzyme.

ABSTRACT

OBJECTIVE To investigate the present situation of nosocomial infection and to provide scientific data for surveillance of nosocomial infection.METHODS The survey form of personal case was filled by adopting the method of combining clinical investigation and consulting inpatients medical records.RESULTS The rate of nosocomial infection was 4.04%;infection sites existed mainly in respiratory tract,urinary tract and skin and soft tissue;the application rate of antibiotics was 49.69% at the day of investigation.CONCLUSIONS The present infection rate survey can mainly help doctors to understand the situation of nosocomial infection.Rational use of antibiotics should be emphasized.
