ABSTRACT
Objective To predict the core targets and action pathways of Hedysari Radix based on UPLC-MS/MS and network pharmacology methods,and to verify the results of network pharmacology by molecular docking and molecular dynamics techniques.This article aims to investigate immune regulation mechanism of effective components absorbed into blood from Hedysari Radix.Methods Qualitative quantification of effective components absorbed into blood from Hedysari Radix were operated by using UPLC-MS/MS technique.The corresponding targets of effective components absorbed into blood from Hedysari Radix were screened by TCMSP and HERB databases.Targets of immune-related disease were obtained through DisGeNET,OMIM,TTD,and MalaCards databases.The network of"components absorbed into blood from Hedysari Radix-immune-related diseases"was then constructed.GO and KEGG enrichment analysis and mapped the PPI network were performed.Molecular docking and molecular dynamics techniques were applied for validation.Results A total of 8 prototype components absorbed into blood,synergistically acting on 101 targets,were identified by UPLC-MS/MS.They mediated 538 biological processes including immune response,positive regulation of gene expression,receptor binding,and cytokine activity.Meanuhile,116 signaling pathways,such as HIF-1,Toll-like receptor,JAK-STAT,T cell receptor,PI3K-Akt,and FoxO etc.were involved.The core targets were MAPK14,PTGS2,MMP9,PPARG,CCND1,etc..The results of molecular docking showed that formononetin and calycosin had strong docking binding activity with MAPK14.And molecular dynamics simulations further demonstrated that the binding between MAPK14 and formononetin or calycosin had good structural stability and binding affinity.Conclusion The results of serum pharmacochemistry,network pharmacology and molecular dynamics were verified to reveal the material basis and mechanism of Hedysari Radix in regulating immunity.The aim of this study is to provide scientific basis for its immunomodulatory mechanism.

OBJECTIVE: To optimize reflux extraction technology of total flavonoids from the roots and stems of Angelica sinensis. METHODS: The reflux extraction technology of total flavonoids from the roots and stems of A. sinensis was optimized by Box-Behnken design-response methodology based on single factor test using volume fraction of extraction solvent ethanol, solide-liquid ration, extraction time, extraction times as investigation factors, the content of total flavonoids in extract as evaluation index. RESULTS: The optimal extraction technology of total flavonoids from the roots and stems of A. sinensis was that volume fractions of ethanol were 70％ and 50％; solid-liquid ratios were 1: 40 and 1: 30; extraction time were 1. 3 h and 1. 7 h; The number of extraction times is two times. In verification test, the contents of total flavonoids were 7. 253 6, 25. 144 1 mg/g (RSD= 1. 57％, 1. 49％, n = 3); relative errors of those to predicted value (6. 942 8, 25. 703 5 mg/g) were 4. 28％, 2. 24％. CONCLUSIONS: Optimized extraction technology for total flavonoids from the roots and stems of A. sinensis is simple, reproducible and predictable.
ABSTRACT
Objective To investigate the sleep quality of inland military officers and soldiers stationed and trained at plateau and its possible influence factors.Methods A total of 459 military officers and soldiers stationed and trained at plateau were performed the on site psychological assessment by adopting the psychological stress self-evaluation test (PSET),Pittsburgh Sleep Quality Index (PSQI),work related fatigue feelings (WRFFQ) and self compiled general data questionnaire.Results (1) The mean value of overall sleep quality in militaries stationed and trained at plateau was 5.61-±-3.48.40.5 ％ of respondents had good sleep quality,25.3％ had poor sleep quality and 34.2％ had general sleep quality.(2)The fatigue and psychological stress scores in the militaries with poor sleep quality were significantly higher than those in the militaries with good sleep quality (t1 =10.70,t2 =-9.68,P＜0.01).(3)The psychological stress,fatigue degree,confidence level of self-assessment health status had significantly positive correlation(r =0.517,0.488,0.259,0.352,P＜ 0.01).(4) The psychological stress,fatigue degree,confidence level of self-assessment health status entered the PSQI total score regression equation,moreover the predictive variation amount was 36.1％.Conclusion The psychological stress status and fatigue level in the militaries at plateau affect the sleep quality,and the confidence degree and health status assessment of officers and soldiers on exercise also produce the active influence on sleep.

Objective To investigate the changes of CD4 + CD25 + Foxp3 + regulatory T cell (Treg) and relative cell factors in peripheral blood of steroid-resistant allergic rhinitis (AR) patients,and to study their functions in occurrence of steroid-resistant AR.Methods The CD4 + CD25 + Foxp3 + Treg cell in 30 cases general physical examination (The control group),30 cases steroid-sensitive AR patients (steroidsensitive group) and 30 cases steroid-resistant AR patients (steroid-resistant group) were detected by flow cytometry.The levels of serum interleukin 10 (IL-10) and transforming growth factor β1 (TGF-β1) in the three groups were detected by enzyme-linked immunoadsorbent assay (ELISA).Results Compare to the control group,proportion of CD4 + CD25 + Foxp3 + Treg cell and the levels of serum IL-10,TGF-β1 in steroid-sensitive group and steroid-resistant group were significantly reduced (P ＜ 0.01).The proportion of CD4 + CD25 + Foxp3 + Treg cell and the levels of serum IL-10,TGF-β1 in steroid-resistant group were decreased significantly than steroid-sensitive group (P ＜ 0.01).The proportion of CD4 + CD25 + Foxp3 + Treg cell were positively correlated with the serum IL-10,and TGF-β1 (P ＜ 0.01).Conclusions The reduced proportion of CD4 + CD25 + Foxp3 + Treg cell and lower serum IL-10,TGF-β1 play an important role in the occurrence of steroid-resistant AR patients.

Objective:To construct lentiviral vector which can overexpression miR-137 and produce lentivirus by lentivirus packaging system, and to explore its infection efficiency and expression in HEK293T cells.Methods: miR-137 sequence was chemically synthesized and cloned into lentiviral vector GV209, and the recombinant plasmid containing human miR-137 was obtained and identified.Then miR-137 recombinant plasmid together with two helper plasmids were transfected into HEK293T cells using Lipofectamine 2000.After the HEK293T cells were infected in multiplicity of infection(MOI) 40 for 48 h, the expression of green fluorescent protein (GFP) was observed by fluorescence microscope and the expression level of miR-137 was detected by fluorescence quantitative PCR.Results:The sequencing results showed that the inserted gene sequence was completely consistent with the published human miR-137 gene sequence in GenBank.The GFP was observed in the HEK293T cells infected with miR-137 overexpression lentivirus under fluorescence microscope.The fluorescence quantitative PCR results showed that the expression level of miR-137 in the cells infected with overexpression lentivirus was 12.74 times higher than that in the control cells.Conclusion:The lentivirus containing miR-137 gene is successful packaged, and it could efficiently infect the HEK293T cells.

OBJECTIVETo study the relationship between allergic rhinitis and fractional exhaled nitric oxide (FeNO).METHODS FeNO was measured in 66 allergic rhinitis patients before and after treatment. Visual analogue scale (VAS) score was used to describe the patient's symptoms. RESULTSFeNO and VAS score were significantly decreased after treatment (t=4.25,P=0.0000;t=3.34, P=0.0008), and the decrease proportion were (14.56±27.23)% and (8.68±53.17)% respectively. Correlation analysis showed FeNO and VAS score was significantly correlated before (r=0.43,P=0.0003) and after (r=0.45,P=0.0002) treatment. After treatment, the decreased proportion of FeNO and VAS score were significantly correlated (r=0.25,P=0.0437). CONCLUSIONFeNO can provide objective evidence for the treatment efficacy of allergic rhinitis.

BACKGROUND:The low oxygen environment after femoral fracture and cerebral trauma wil induce series of related cytokines expression, including hypoxia-inducible factor 1αand core binding factorα1, which play key roles in regulating bone healing. However, whether the accelerated bone healing is correlated with the expression of hypoxia-inducible factor 1αand core binding factorα1 is stil unknown. OBJECTIVE:To construct rat models of brain injury, to compare the expression level of hypoxia-inducible factor 1αand core binding factorα1 in femoral fracture combined with cerebral trauma rats and simple femoral fracture rats, and to assess the influence of cerebral trauma on bone healing. METHODS:Rats were randomly divided into blank group, simple femoral fracture group and femoral fracture combined with cerebral trauma group. At 1, 2, 3 and 5 weeks after modeling, rats were executed. Bone healing was evaluated using femoral fracture end X-ray score and hematoxylin and eosin staining at cal us tissues. Besides, the expression levels of hypoxia-inducible factor 1αand core binding factorα1 of three groups were determined with immunohistochemistry. RESULTS AND CONCLUSION:Bone healing in the femoral fracture combined with cerebral trauma group was better than that of simple femoral fracture group. There was significant difference in the expression level of hypoxia-inducible factor 1αand core binding factorα1 between the simple femoral fracture group and femoral fracture combined with cerebral trauma group (P<0.05). At the same time, the level of simple femoral fracture group and femoral fracture combined with cerebral trauma group was significantly higher than that of blank group, and that in femoral fracture combined with cerebral trauma group was significantly higher than that of simple femoral fracture group (P<0.05). Results verified that the expression levels of hypoxia-inducible factor 1αand core binding factorα1 of rats with femoral fracture combined with cerebral trauma were significantly high, which may be the major reason why the bone healing was accelerated after fracture combined with brain injury.

Objective To investigate the function of activator protein-1(AP-1)and nuclear factor-κB(NF-κB)on the treatment of montelukast sodium tables for patients with steroid-resistant allergic rhinitis (AR). Methods A random-ized study was carried out in 50 patients with steroid-resistant AR. 25 cases were treated with montelukast sodium ta-bles(montelukast sodium group), and the others by ATP tables(placebo group). Score system was used to compare the therapeutic effect of these two drugs on clinical symptoms and signs on 21th day after therapy. The DNA binding activ-ities of Ap-1 and NF-κB were measured by electrophoretic mobility shift assay(EMSA). The expression of IL-4, IL-5 and IL-10 were determined by enzyme-linked immunoadsorbent assay (ELISA). Results The therapeutic effect rate in montelukast sodium group was significantly higher than that in placebo group at 21 days after treatment begining (P<0.01). Compared with placebo group, the DNA binding activities of Ap-1 and NF-κB were significantly reduced in montelukast sodium group (P<0.01). The score reduce index (SRI)of the clinical symptoms and signs was negatively correlated with the DNA binding activities in montelukast sodium group (P<0.05), but there was any correlation be-tween them in placebo group(P>0.05).The expression of IL-4 and IL-5 in montelukast sodium group were significantly lower and IL-10 was higher than those in placebo group (P<0.01). Conclusion The treatment effect of montelukast sodium tables for steroid-resistant AR may be aroused by suppressing the Ap-1 and NF-κB DNA binding activities, and effecting the expression of IL-4, IL-5 and IL-10.

@#Objective To analyze the basic situations and curative effect of psychotics treated in open psychotic ward in general hospital,and explore a new managing model with advanced treatment for psychosis patients.Methods 3 803 psychotics,treated in the open psychotic ward since it's setting,were analyzed as a retrospective study.Results The data including the proportion of diseases,the basic information of the patients,the length of stay and the curative effect,were different from that in traditional psychiatric facilities.Conclusion The open psychotic wards setting in general hospital is an effective treating and managing model for psychotics.

@#ObjectiveTo study the significance of Double Therapy Treatment Based On Differentiation Of Syndromes on obsession with obsessive thoughts but without compulsive behavior and its suitability in open wards.Methods47 cases were divided into experimental group(24 cases) and control group(23 cases) randomly in open wards. Experimental group was treated by Double Therapy Treatment Based on Differentiation of Syndromes while control group by Cognitive Therapy combined with Chlorimpramine and Fluoxetine etc. The period of treatment was 5 months, in which positive therapy cost 2 months and free therapy cost next 3 months. Y-BOCS was used to value the effect.ResultsThe recovery rate in experimental group was much higher than that of the control group 2 months (χ2=12.44,P<0.01) and 5 months (χ2=18.00,P<0.01)later. There was significant difference in Y-BOCS score between these two groups 2 months (t=7.140,P<0.001) and 5 months (t=8.191,P<0.001)later.ConclusionDouble Therapy Treatment Based On Differention Of Syndromes shows a satisfied curative effect and prognosis on obsession with obsessive thoughts but without compulsive behavior and should be advocated.
