ABSTRACT
Objective:To control the quality of the reference sample of Wenjingtang by establishing the specific chromatograms. Method:On the basis of analyzing 15 batches of Wenjingtang freeze-dried powder samples, a high performance liquid chromatography (HPLC) specific chromatogram analysis method of Wenjingtang was established. The system adaptability was investigated and the retention time, relative retention value and deviation caused by different chromatographic columns and instruments were calculated by using the same brand of chromatographic columns, four different brands of chromatographic columns and instruments from three different manufacturers. The precision, repeatability and stability of this method was further completed. The possible chemical components of the freeze-dried powders were speculated and identified by ultra-performance liquid chromatography-quadrupole-time-of-flight mass spectrometry (UPLC-Q-TOF-MSn). Chromatographic separation was performed on ACQUITY UPLC BEH C18 column (2.1 mm×100 mm, 1.7 μm) with acetonitrile (A)-0.1% formic acid aqueous solution (B) as mobile phase for gradient elution (0-2.8 min, 10%A; 2.8-8.0 min, 10%-18%A; 8.0-12.2 min, 18%-25%A; 12.2-15.3 min, 25%-40%A; 15.3-17.4 min, 40%A; 17.4-20.5 min, 40%-90%A), and column temperature was set at 30 ℃ with flow rate of 0.4 mL·min-1. Mass spectrometry was performed on electrospray ionization, data were collected under positive and negative ion modes, and the detection range was m/z 50-1 600. Result:Ten characteristic peaks were selected as the distinguishing features in this specific chromatograms, and eight of them were identified by comparing with the reference standards, including paeoniflorin (peak 1), liquiritin apioside (peak 2), liquiritin (peak 3), ferulic acid (peak 4), iquiritigenin (peak 6), cinnamaldehyde (peak 8), paeonol (peak 9)and glycyrrhizic acid (peak 10). By mass spectrometry analysis, 30 compounds were identified, and the source of medicinal materials were assigned. It mainly contained triterpenoid saponins and flavonoids from Glycyrrhizae Radix et Rhizoma, ginsenosides from Ginseng Radix et Rhizoma, monoterpenoid glycosides and tannins from Paeoniae Radix Alba, steroids in Achyranthis Bidentatae Radix, phenolic acids in Angelicae Sinensis Radix. Conclusion:The established characteristic chromatographic analysis method of Wenjingtang is simple, stable and repeatable. The chemical composition of the freeze-dried powder of Wenjingtang is basically defined by mass spectrometry identification and source attribution, which can provide reference for the development and quality control of Wenjingtang in the future.
ABSTRACT
Based on the textual research on literature, the key information of Wenjing Decoction were tested and identified, and 15 batches of lyophilized powder samples of Wenjing Decoction were prepared. The specific components, including paeoniflorin, glycyrrhizin, ginsenosides(Rg_1, Re and Rb_1), glycyrrhizic acid, and paeonol, were used as indexes to establish the HPLC method for quantitative evaluation, and the content ranges and transfer rates of these components were determined. The results showed that the contents of paeoniflorin, glycyrrhizin, ginsenosides Rg_1 + Re, ginsenoside Rb_1, glycyrrhizic acid, and paeonol in the 15 batches of samples were 0.62%-0.86%, 0.25%-0.76%, 0.14%-0.30%, 0.07%-0.21%, 0.63%-1.16%, and 0.09%-0.25%, respectively, and their transfer rates from the decoction pieces to the reference materials were 14.99%-19.42%, 28.11%-40.93%, 25.92%-61.88%, 25.03%-64.06%, 23.43%-35.53%, and 5.34%-10.44%, respectively. The consistency of the transfer rates between batches indicated that the preparation process was stable. It is suggested that the contents of paeoniflorin, glycyrrhizin, ginsenosides Rg_1 + Re, ginsenoside Rb_1, glycyrrhizic acid, and paeonol in Wenjing Decoction should not be less than 0.52%, 0.35%, 0.15%, 0.10%, 0.63%, and 0.12%, respectively. In this study, we determined the contents and analyzed the quantity transfer process of the index components in Wenjing Decoction, which can provide a basis for the follow-up development of Wenjing Decoction and the quality control of related preparations.

Objective To master the prevention knowledge and behavior of echinococcosis and the influencing factors in res-idents in agricultural and pastoral areas of Maerkang City,so as to provide the reference for formulating the policy and strategy of echinococcosis prevention and control. Methods In April,2017,796 residents in agricultural and pastoral areas of Maerkang City were selected by the multi-stage stratified random sampling method, and surveyed with questionnaires. The main questions included the prevention knowledge and related behaviors of echinococcosis. Results The awareness rate of prevention knowl-edge of echinococcosis was 70.1%(558/796).The awareness rate was the lowest in the group aged 15 to 25 years. The difference of knowledge awareness rate was statistically significant among the different age groups(Χ2=16.408,P<0.01).The knowledge awareness rate of the people with middle school or above educational level was higher than that of the people with primary school or below educational level(Χ2=4.869,P=0.027).The knowledge awareness rate of the dog owners was lower than that of the people without a dog(Χ2=11.384,P=0.001).The knowledge awareness rate in the people who received the knowledge training of three times or more was higher than that in the people who received the knowledge training of two times or less(Χ2=11.439,P=0.003).The logistic regression showed that the age, educational level, training times and dog raising were the main influenc-ing factors. The related behavior survey showed that 32.7%(260/796)of residents did not drink unboiled water,37.0%(61/165)of dog owners tied the dog,43.6%(72/165)of dog owners fed the dogs with anthelmintic regularly, and only 10.3%(17/165)of dog owners buried the dog's dung. Conclusions The awareness rate of echinococcosis prevention knowledge in Maerkang City should be improved. The health education should be focused on the young people, people with low educational level, and people who have dogs. The behaviors of drinking unboiled water, tying dogs, feeding dogs with anthelmintic regularly, and burying dog' s dung should be advocated.
ABSTRACT
Objective To investigate the relationship between angiotensin converting enzyme(ACE) gene polymorphism and carotid plaque composition, vessel wall morphology, and clinical symptoms based on vessel wall magnetic resonance imaging. Methods Totally 75 hypertensive patients(75 internal carotid artery plaques) with maximum plaque thickness≥1.5 mm,according to the ACE insertion(I) or deletion(D) gene polymorphism, were divided into ACE 2 genotype group(n=37) and ACE ID/DD genotype group(n=38). The influences of plaque composition, vessel wall morphology, clinical symptoms, and use of ACE inhibitor or angiotensin receptor blocker(ACEI/ARB) on vessel wall morphology were analyzed. Results Compared with ACE 2 genotype group, the ACE ID/DD genotype group had significantly higher incidence of ischemic stroke(Χ=3.921,P=0.048). The plaque composition and vessel wall morphology showed no significant difference between these two groups. Inside ACE ID/DD genotype group, the carotid remodeling index was significantly lower in users of ACEI/ARB than non-users of ACEI/ARB(1.85±0.60 vs. 2.48±0.40;t=3.854,P=0.001).Conclusion In primary hypertension,ACE ID/DD genotype may be associated with carotid atherosclerotic plaque.

In order to determine the appraisal of ABO blood group accurately and ensure the safety of blood transfusion, an investigation was made on the influence of complement activated by antibody in appraising ABO blood group by using hemoagglutination test and direct antiglobulin test. Two samples of ABO blood group were collected from two donors. The results showed that the obverse and reverse patterns did not accord each other in two samples of ABO blood group, and their blood samples were both positive for anti-C(3) and were both negative for anti-IgG. It is concluded that the inconsistency of the obverse and reverse patterns in two samples of ABO blood group were proved to be caused by the complements activated by the antibody against IgM. Blood group O was finally determined for the two samples, and the influences of IgM antibody and complement on ABO blood group were excluded when the test proceeded. This method will be useful to determine ABO blood group accurately.
