ABSTRACT

Abstract Understanding morphological and physiological changes under different light conditions in native fruit species in juveniles stage is important, as it indicate the appropriate environment to achieve vigorous saplings. We aimed to verify growth and morphophysiological changes under shade gradient in feijoa (Acca sellowiana (O. Berg) Burret) to achieve good quality saplings adequate to improve cultivation in orchards. The saplings were grown for twenty-one-month under four shading treatments (0%, 30%, 50%, and 80%). Growth, photosynthetic pigments, gas exchanges, chlorophyll fluorescence, and leaf anatomy parameters were evaluated. Saplings under full sun and 30% shade had higher height and diameter growth and dry mass accumulation due to higher photosynthesis rate. As main acclimatization mechanisms in feijoa saplings under 80% shade were developed larger leaf area, reduced leaf blade thickness, and enhanced quantum yield of photosystem II. Even so, the net CO2 assimilation and the electron transport rate was lower and, consequently, there was a restriction on the growth and dry mass in saplings under deep shade. Therefore, to obtain higher quality feijoa saplings, we recommend that it be carried out in full sun or up to 30% shade, to maximize the sapling vigor in nurseries and, later, this light environment can also be used in orchards for favor growth and fruit production.


Resumo A verificação de mudanças morfológicas e fisiológicas sob diferentes condições luminosas em espécies frutíferas nativas em estágio juvenil é importante, uma vez que indicam o ambiente adequado para a formação de mudas com alto vigor. Objetivou-se verificar o crescimento e as alterações morfofisiológicas sob gradiente de sombreamento em mudas de feijoa (Acca sellowiana (O. Berg) Burret) para obter mudas de boa qualidade, adequadas para fomentar os plantios da espécie em pomares. As mudas foram cultivadas por vinte e um meses sob quatro tratamentos de sombreamento (0%, 30%, 50% e 80%). Foram avaliados parâmetros de crescimento, pigmentos fotossintéticos, trocas gasosas, fluorescência da clorofila e anatomia foliar. Mudas a pleno sol e 30% de sombra apresentaram maior crescimento em altura, diâmetro e acúmulo de massa seca, devido à maior taxa de fotossíntese. Como principais mecanismos de aclimatação sob 80% de sombra, as mudas desenvolveram maior área foliar, redução da espessura do limbo foliar e aumento do rendimento quântico do fotossistema II. Mesmo assim, a assimilação líquida de CO2 e a taxa de transporte de elétrons foram menores e, consequentemente, houve restrição ao crescimento e acúmulo de massa seca das mudas no maior nível de sombreamento. Portanto, para a obtenção de mudas de feijoa de maior qualidade, recomendamos que seja realizada a pleno sol ou até 30% de sombra, para maximizar o vigor das mudas em viveiros e, posteriormente, este ambiente de luz também pode ser utilizado em pomares para favorecer o crescimento e a produção de frutos.

ABSTRACT

Introducción: El infarto esplénico es una de las enfermedades que se presentan secundarias al fallo de aclimatación. En la literatura revisada se relaciona con la presencia de un rasgo sicklémico y desde la década del 50 se tienen reportes de casos a nivel mundial. Objetivo: Describir un caso de infarto esplénico por exposición a la altura. Presentación del caso: Se expuso un paciente que presentó un infarto esplénico después de su exposición a la altura, el cual fue intervenido quirúrgicamente y se presentaron un grupo de complicaciones en el posoperatorio. En la literatura internacional esta enfermedad se relaciona con la presencia de la hemoglobina S, lo cual no concuerda con nuestro paciente, a pesar de no ser el estudio de elección. Sus antecedentes pudieron contribuir a la ocurrencia de esta complicación. Las complicaciones que aparecieron en el posoperatorio coincidieron con las comentadas por otros autores. Conclusiones: El infarto esplénico por exposición a la altura es una complicación que debe tenerse en cuenta cuando se presenta un paciente a su llegada a este ecosistema, aparece con dolor abdominal, donde la inmediatez en la conducta es fundamental para evitar complicaciones de mayor gravedad.


Introduction: Splenic infarction is one of the diseases that occur secondary to failure of acclimatization. In the reviewed literature, it is related to the presence of a sicklemic trait and since the 1950s there have been case reports worldwide. Objective: To describe a case of splenic infarction due to exposure to high altitude. Case report: We report the case of a patient who had a splenic infarct after exposure to high altitude. This patient, who underwent surgery, had a group of complications during the postoperative period. The international literature associates this disease with the presence of hemoglobin S, which does not coincide with the case of our patient, despite not being the study of choice. His history could have contributed to the occurrence of this complication. Those that appeared in the postoperative period did agree with those commented by other authors. Conclusions: Splenic infarction due to exposure to altitude is a complication that must be taken into account when a patient has abdominal pain. Immediacy in behavior is essential to avoid more serious complications.

ABSTRACT

Acute mountain sickness (AMS) is a clinical syndrome of multi-system physiological disorder after acute exposure to low pressure and low oxygen at high altitude. Quantitative proteomics can systematically quantify and describe protein composition and dynamic changes. In recent years, quantitative proteomics has been widely used in the prevention, diagnosis, treatment and pathogenesis of many diseases. This review summarizes the progress of quantitative proteomics techniques and its application in the prevention, diagnosis, treatment of AMS and mechanisms of rapidly acclimatizing to plateau, in order to provide a reference for the pathogenesis, early intervention, clinical treatment and proteomic research of AMS.

ABSTRACT

Abstract Under the directorship of Clemente Onelli (1904-1924), the Jardín Zoológico de Buenos Aires became a major public attraction and gained an international reputation for its innovations in animal keeping and as a supplier of Latin American fauna. It was a hybrid institution that combined the tasks of public instruction, zoological research, and acclimatization of useful animals, and also served as a symbol of national pride. Despite its seemingly peripheral geographical location, the institution was firmly integrated in the global network of zoological gardens. This paper utilizes a transnational perspective to tease out the numerous, multidirectional exchanges of animals and knowledge between the Jardín Zoológico de Buenos Aires and Northern metropolises.


Resumo Sob a direção de Clemente Onelli (1904-1924), o Jardín Zoológico de Buenos Aires se tornou uma importante atração pública e ganhou reputação internacional por suas inovações no abrigo e fornecimento de fauna latinoamericana. Era uma instituição híbrida que combinava instrução pública, pesquisa zoológica e aclimatação de animais úteis, sendo também símbolo de orgulho nacional. Apesar da localização geográfica aparentemente periférica, a instituição estava fortemente integrada na rede global de jardins zoológicos. O artigo utiliza a perspectiva transnacional para lançar luz sobre as muitas trocas multidirecionais de animais e conhecimento entre o Jardín Zoológico de Buenos Aires e as metrópoles do Norte.

Abstract In this study, in vitro propagation and acclimatization of Helianthemum germanicopolitanum Bornm. plant, a local endemic in Çankırı Province (Turkey) with arid and semi-arid lands, and an endangered species taking part among medicinal and aromatic plants were accomplished, which is under-researched. In this study, three basal media [a) Murashige and Skoog b) Gamborg's B5, and c) Nitsch & Nitsch], two gelling agents (agar 7 g/L, and gelrite 2.1 g/L), eight cytokinins and eight auxin doses of plant growth regulators [a) 6-benzyladenin, b) Kinetin-(0, 0.5, 1, and 2 mg/L), c) Indole-3-butyric acid, d) α-napthaleneacetic acid-(0, 0.25, 0.5, and 1 mg/L)] prepared in 64 different combinations with 30 g/L sucrose was added to the basal media and adjusted to pH 5.7 for in vitro propagation of H. germanicopolitanum. During in vitro propagation of the plant, external and internal infections were frequently encountered and this was solved by the developed protocol. The best shoot growth (1.141 cm) and shoot length (0.572 cm) were obtained in the Gamborg's B5 medium in combination with Kinetin (0.5 mg/L)+Indole-3-butyric acid (0.5 mg/L)+gelrite. The maximum number of shoots (19.50) and the best multiplication rate (94%) were obtained in the media containing benzyladenin (1 mg/L)+Indole-3-butyric acid (0.5 mg/L) plant growth regulator in Murashige and Skoog medium solidified with agar. At the rooting stage, the maximum number of roots (30) was reached in the Murashige and Skoog medium containing gelrite and the best rooting rate (92%) with agar. A hundred plants representing the best shoot and root growth were taken to acclimatization stage, and 32 of these plants adapted to external conditions.

ABSTRACT: This study evaluated the occupational exposure of workers to heat during manual harvesting of coffee. The case study was conducted between May to August. Occupational heat in coffee plantations was evaluated using the Wet Bulb Globe Temperature (WBGT) method and the metabolic rate. The WBGT was obtained using the TGD 400 thermal stress meter. The determined WBGT values ​​were compared to two standards: the Occupational Hygiene Standard (NHO) 06 and Regulatory Norm (NR) 09. For acclimatized workers, the exposure limit was not exceeded at any time during the evaluated period. Fornon-acclimatized workers, the exposure limit was not exceeded only in July.


RESUMO: O presente trabalhado teve como objetivo avaliar a exposição ocupacional de trabalhadores ao calor durante a colheita manual do cafeeiro. O estudo de caso foi desenvolvido entre os meses de maio a agosto. Avaliou-se o calor ocupacional em lavoura cafeeira através do método do Índice Bulbo Úmido Termômetro de Globo (IBUTG) e taxa metabólica. O IBUTG foi obtido utilizando o medidor de estresse térmico TGD 400. Os valores do IBUTG determinados foram comparados com duas normativas: Norma de Higiene Ocupacional (NHO) 06 e Norma Regulamentadora (NR) 09. Para trabalhadores aclimatizados o limite de exposição não foi superado em nenhum momento do período avaliado. Em relação aos trabalhadores não aclimatizados o limite de exposição não foi superado apenas em julho.

During the application of phytosanitary products in coffee trees, workers may be exposed to temperatures that could compromise their health. Exposure to occupational heat can lead to progressive dehydration, cramps, exhaustion and the possibility of thermal shock. Thus, knowing the levels of occupational heat that workers are exposed becomes important. This study aimed to evaluate workers' occupational exposure to heat during the application of phytosanitary products with manual costal pump in the coffee crops. The case study was developed at the IFSULDEMINAS school farm - Campus Inconfidentes in the months of September and October 2017. The occupational heat in the coffee crop was evaluated using the WBGTAVERAGE method, using TGD 400 thermal stress meter. The WBGT values found were compared to the exposure limits of NR 15 for the purposes of insalubrity classification and with NHO 06 for acclimatized and non-acclimatized workers. The results showed that the WBGT found is below the tolerance limit of NR 15, for the month of September. In October, the tolerance limit was exceeded in the period from 11:00 a.m. to 2:59 p.m., considering a continuous heavy activity and rest in the workplace, and the activity considered unhealthy. Considering the criteria of the NHO 06 it was observed that the limits of occupational exposure were exceeded in the month of October, for acclimatized and non-acclimatized workers. In September the occupational exposure limit of NHO 06 was exceeded only for non-acclimated workers. Therefore, the workers should be submitted to acclimatization during the phytosanitary application activity in the month of October.


Durante a aplicação de produtos fitossanitários no cafeeiro os trabalhadores podem estar expostos a temperaturas capazes de comprometer sua saúde. Exposição ao calor ocupacional pode levar a desidratação progressiva, câimbras, exaustão e possibilidade de choque térmico. Desta forma, conhecer os níveis de calor ocupacional que os trabalhadores estão expostos torna-se importante. O presente estudo teve como objetivo avaliar a exposição ocupacional ao calor de trabalhadores durante a aplicação de produtos fitossanitários com bomba costal manual na cultura do cafeeiro. O estudo de caso foi desenvolvido na fazenda-escola do IFSULDEMINAS ­ Campus Inconfidentes nos meses de setembro e outubro de 2017. Avaliou-se o calor ocupacional em lavoura cafeeira através do método do IBUTGMÉDIO, utilizando medidor de estresse térmico TGD 400. Os valores do IBUTG encontrados foram comparados aos limites de exposição da NR 15 para fins de classificação de insalubridade e com a NHO 06 para trabalhadores aclimatizados e não aclimatizados. Os resultados demostraram que o IBUTG encontrado está abaixo do limite de tolerância da NR15, para o mês de setembro. No mês de outubro o limite de tolerância foi ultrapassado no período das 11:00 às 14:59h, considerando uma atividade pesada contínua e descanso no próprio local de trabalho, sendo a atividade considerada insalubre. Ao considerarmos os critérios da NHO 06 observou-se que os limites de exposição ocupacional foram ultrapassados no mês de outubro, para trabalhadores aclimatizados e não aclimatizados. Em setembro o limite de exposição ocupacional da NHO 06 foi superado somente para trabalhadores não aclimatizados. Portanto os trabalhadores devem ser submetidos à aclimatização durante a atividade de aplicação de fitossanitários no mês de outubro.

ABSTRACT

RESUMEN Prosopis pallida, conocido como algarrobo, es una especie emblemática de los bosques secos del norte del Perú. Es de gran importancia económica por su uso en la producción de leña y carbón, así como en la producción de algarrobina proveniente de sus frutos. Actualmente las actividades humanas han deforestado grandes poblaciones de algarrobo en el bosque seco, por lo que es muy importante una propagación masiva para planes de reforestación a gran escala en esos ecosistemas, con la finalidad de conservar la especie y también las características genéticas de individuos élite. El objetivo de la investigación fue establecer un protocolo para la propagación in vitro de algarrobo. Previo a la siembra in vitro, se evaluaron tres tratamientos pregerminativos para poder acelerar la germinación de las semillas con la finalidad de obtener mayor material de propagación. Luego se evaluó el efecto del medio de plantas leñosas con la adición de cuatro concentraciones (0; 0,5; 1,0 y 1,5 mg/L) de citoquininas (BAP y ZEA) sobre la propagación in vitro de Prosopis pallida, habiendo realizado tres ensayos debido a la poca efectividad de brotación de las yemas apicales, resultando mejor la ausencia de citoquininas en yemas apicales con sus dos cotiledones, usando tapas de algodón en los tubos de ensayo. En esta etapa se evaluó el número de nudos, altura de plántula y número de brotes. Para el enraizamiento se ensayó con tres concentraciones (0; 0,5 y 1,0 mg/L) de auxinas (NAA, IBA y IAA), y se evaluó el porcentaje de enraizamiento, longitud de la raíz y número de raíces; obteniéndose mejores resultados con 0,5 mg/L IBA. En la fase de aclimatación se evaluó el porcentaje de aclimatación en dos tipos de sustratos, obteniéndose mejores resultados con sustrato comercial de turba y vermiculita.


ABSTRACT Prosopis pallida, known as algarrobo, is an emblematic species of the dry forests of northern Peru. It is of great economic importance for its use in the production of firewood and coal, as well as in the production of carob from its fruits. Currently human activities have deforested large populations of algarrobo in the dry forest, so it is very important a massive propagation for large-scale reforestation plans in these ecosystems, in order to conserve the species and also the genetic characteristics of elite individuals. The objective of the research was to establish a protocol for the in vitro propagation of algarrobo. Before in vitro propagation, It was evaluated the effect of three seeds treatments to accelerate seeds germination in order to get more propagation material. Then, the effect of woody plants medium with the addition four concentrations (0, 0.5, 1.0 and 1.5 mg /L) of cytokinins (BAP and ZEA) on the in vitro propagation of Prosopis was evaluated and it has performed three trials for the ineffective budding of apical buds, resulting in a better absence of cytokinins in apical buds with their two cotyledons, and using cotton lids in the test tubes. In this stage the number of nodes, seedling height and number of shoots was evaluated. For rooting, it was tested with three concentrations (0, 0.5 and 1.0 mg /L) of auxins (NAA, IBA and IAA), and the percentage of rooting, root length and amount of roots was evaluated, obtaining better results with 0.5 mg / L IBA. In the acclimation phase, the acclimation percentage was evaluated using two substrates, and the best results were commercial substrate of peat and vermiculite.

ABSTRACT

Objetivou-se avaliar os efeitos da manipulação da temperatura de incubação sobre a resposta imune de codornas desafiadas termicamente após eclosão. Para isso, foram utilizados 540 ovos, distribuídos em três incubadoras, com temperatura de 37,8°C e umidade de 60%. A partir do sexto dia de incubação até a eclosão, as temperaturas foram ajustadas em 37,8°C (padrão), 38,5°C (intermediária) e 39,5°C (alta). Após a eclosão as codornas foram pesadas e distribuídas, em delineamento inteiramente ao acaso, com três temperaturas de incubação (37,8, 38,5 e 39,5°C) e duas temperaturas de ambiente (estresse e termoneutro). Aos 10, 20, 30 e 40 dias, quatro codornas por tratamento foram eutanasiadas para coleta da bolsa cloacal, do fígado e do coração, para se determinar o peso absoluto (g), o peso relativo (%) e a área dos folículos bursais. Sangue foi coletado para realização do hemograma, do leucograma e da bioquímica sérica. Os dados foram analisados e as diferenças entre as médias foram determinadas pelo teste de Tukey a 5%. O estresse térmico por calor, a partir dos 20 dias, promove redução no peso absoluto do fígado, do coração, da bolsa cloacal e na área dos folículos bursais, além de heterofilia, linfopenia e aumento da relação heterófilo/linfócito. Em conclusão, o estresse térmico por calor após 10 dias de idade pode causar imunossupressão.(AU)


The objective of this study was to evaluate the effects of manipulation of the incubation temperature on the immune response of quails challenged thermally after hatching. For this, 540 eggs were distributed in three incubators, with temperature of 37.8°C and 60% humidity. From the 6th day of incubation to hatching the temperatures were adjusted to 37.8°C (standard), 38.5°C (intermediate) and 39.5°C (high). After hatching the quails were weighed and distributed in a completely randomized design with three incubation temperatures (37.8, 38.5 and 39.5°C) and two ambient temperatures (stress and thermoneutral). At 10, 20, 30 and 40 days four quail per treatment were euthanized to collect the cloacal burse, liver and heart to determine the absolute weight (g), relative weight (%) and area of the bursal follicles. Blood was sampled for determination of hemogram, leukogram and serum biochemistry. The data were analyzed and the differences between the means were determined by the Tukey test at 5%. Heat stress from 20 days onwards promotes a reduction in the absolute weight of the liver, heart, cloacal sac and in the area of the follicles. In addition, there was heterofilia, lymphopenia and increased heterophile/lymphocyte ratio. In conclusion, heat stress after 10 days of age can cause immunosuppression.(AU)

ABSTRACT: Sincoraea mucugensis (Wand. & A.A. Conc.) LOUZADA & WAND, an endangered bromeliad, is confined to the central region of the Chapada Diamantina, in the municipality of Mucugê, Brazil. From various researches, it is evident that for the propagation of this species, the in vitro technique is a feasible option. However, due to the low multiplication rates reported in various papers, this study aimed to establish a micropropagation protocol of direct organogenesis for S. mucugensis. First, the inoculation of the stem explants was done in MS ½ culture medium which contained different levels of BAP (0.00; 6.66; 8.88; 11.10; 13.20 µM) and NAA (0.00; 2.60; 5.20 µM). These shoots were then subjected to a couple of distinct rooting periods (of 30- and 60-day duration) using activated charcoal; finally, these microplants were transferred to a greenhouse for acclimatization, and covered with transparent plastic cups, as a water loss prevention test method. All the data were submitted to the analysis of variance (ANOVA), and the means were subjected to regression analysis or compared using the Tukey test. The findings revealed that the S. mucugensis stem explants raised in the NAA-rich medium (6.42 to 7.43 shoots/explants) showed high multiplication rates; the shoot rooting was done for 30 days using activated charcoal with the medium. Acclimatization, which was performed by directly exposing the microplants to the ex vitro environment, showed 95% survival rate.


RESUMO: Sincoraea mucugensis (Wand. & A.A. Conc.) LOUZADA & WAND é uma bromélia vulnerável de ocorrência restrita ao município de Mucugê, Chapada Diamantina. Estudos indicam que a cultura de tecidos é uma alternativa viável para a propagação in vitro desta espécie. Contudo, em função das baixas taxas de multiplicação obtidas em estudos anteriores, objetivou-se estabelecer um protocolo de micropropagação via organogênese direta para S. mucugensis. Explantes caulinares foram inoculados em meio de cultura MS ½ contendo diferentes concentrações de BAP (0,00; 6,66; 8,88; 11,10; 13,20 µM) e ANA (0,00; 2,60; 5,20 µM). Os brotos obtidos foram submetidos a diferentes períodos de enraizamento (30 e 60 dias) com carvão ativado; posteriormente as microplantas foram aclimatizadas em casa de vegetação, testando-se o efeito da cobertura com copos plásticos transparentes como estratégias contra perda de água. Os dados foram submetidos à análise de variância (ANOVA) e as médias analisadas por regressão ou comparadas pelo Teste de Tukey. Os resultados demonstram que altas taxas de multiplicação de S. mucugensis são geradas a partir de explantes caulinares cultivados em meio contendo ANA (6,42 a 7,43 brotos/explantes); o enraizamento dos brotos é realizado por 30 dias em meio com carvão ativado e a aclimatização é feita com exposição direta das microplantas ao ambiente ex vitro com 95% de sobrevivência.

ABSTRACT

A regeneration protocol for castor bean plant (Ricinus communis) was successfully developed using epicotyl sections obtained from in vitro seedlings. The maximum number of induced shoots (4.3 shoots/explant) and highest shoots frequency (75,56%) was obtained in WPM medium supplemented with TDZ (1 mg/L) and zeatin (0.5 mg/L), whereas the minimum number (0.8 shoots/explant) and lowest shoots frequency (37,78%) was obtained in medium containing TDZ (1 mg/L) and BAP (0.5 mg/L). The highest percentage of rooting (93.3%) was obtained in a medium containing IBA (1 mg/L). These plants were transplanted in a mesh house and achieved a high adaptability to acclimatization, reaching 77% survival. On the other hand, the maximum elongation (height) during this stage was 7.9 cm in plants supplemented with WPM nutrients, whereas it was only 4.38 cm in control plants


Foi desenvolvido com sucesso um protocolo de regeneração para a planta de Mamona (Ricinus communis) utilizando seções de epicótilos, obtidas a partir de mudas in vitro. O número máximo de brotações induzidas (4.3 brotos/explante), assim como a maior frequência de brotações (75,56%), foi obtido em meio WPM suplementado com TDZ (1 mg/L) e zeatina (0,5 mg/L). Enquanto que o número mínimo (0,8 brotos/explante), como a menor freqüência de rebentos (37,78%), foi obtido em meio contendo TDZ (1 mg/L) e BAP (0,5 mg/L). Adicionalmente, a maior percentagem de enraizamento (93,3%) foi obtida em um meio contendo IBA (1 mg/L). Depois da regeneração, as plantas foram transplantadas em casa de vegetação e conseguiram uma alta adaptabilidade e aclimatização, atingindo 77% de sobrevivência. Por outro lado, oalongamento máximo (altura) durante este estágio foi de 7,9 cm em plantas suplementadas com nutrientes de WPM, enquanto as plantas de controle presentaram apenas 4,38 cm

ABSTRACT

Aim: To study the effect of various plant growth regulators (PGRs) for induction of somatic embryogenesis and plantlet formation from cotyledon and leaflet explants in S. nigrum (night shade) an important medicinal plant used in treatment of digestive problems and skin infections. Place and Duration of Study: Department of Biotechnology, Kakatiya university, Warangal. Telangana, India, 3 years. Methodology: Cotyledon (0.8 cm2) and leaflet explants (0.8-1.0 cm2) from 3 week and 4 week old were cultured on MS medium supplemented with 30 g/L sucrose along with different concentrations of 0.5 mg/L BAP+NAA (0.5 – 6.0 mg/L) . Results: Maximum percentage of somatic embryogenesis was observed in cotyledon(89%) and leaf (98%) explants on MS medium augmented with 0.5mg/L BAP in combination with 2.0 mg/L NAA whereas the highest number of somatic embryos per explant (86 ± 0.19) was formed in leaflet explant. Conclusion: Somatic embryogenesis was induced from both cotyledon and leaf explants. Since it is threatened and medicinally important species S. nigrum, the present protocol can be used for its conservation and genetic transformation experiments.

ABSTRACT

Runner tips explants of strawberry give rise to multiple shoots when cultured on MS medium supplemented with different concentrations and combinations of BAP with KIN or NAA or GA3.The highest response of shoot multiplication was obtained on MS containing 2.5 mgl-1 BAP + 0.5 mgl-1 Kin + 0.5 mgl-1 GA3. The maximum frequency of rooting (83%) and highest number of roots (3.49) was produced in medium containing 1.0 mgl-1 IBA. The well grown rooted plantlets were acclimatized and successfully established in autoclaved vermiculate soil and as well as natural condition. Using our established protocol, it is also possible to provide large numbers of micropropagated plantlets of this cultivars to produce high quality strawberry fruit for commercial cultivation practices.

ABSTRACT

In vitropropagation increases the supply and commercialisation of products of interest. For this, optimising the growing conditions and the composition of the culture medium is crucial to benefit the full development of the plants. Thus, the objective was to evaluate the in vitropropagation of Aloysia triphyllaon different culture media, with varying agar and sucrose concentrations. The experiment was conducted as a completely randomised design, 3×3×3 factorial scheme, with three culture media (MS, JADS and WPM), three sucrose concentrations (8, 10 and 12 g L-1) and three agar concentrations (15, 30 and 45 g L-1), with three replicates each and experimental units composed of one plant per replicate. After 25 days of cultivation, the fresh and dry mass of the plants, numbers of leaves, numbers of nodes, plant lengths, numbers of oxidised leaves, hyperhydricity and acclimatization percentages were evaluated. The WPM medium resulted in a reduced fresh mass, reflecting in the low hyperhydricity observed in the explants, and favoured the acclimatization of the plants. Thus, the WPM medium with sucrose (15 g L-1) and agar (12 g L-1) is recommended as the medium most suitable for the in vitroregeneration of Aloysia triphylla.

ABSTRACT

The objective of this work was to investigate the use of Palm Male Inflorescence (PMI) and river-sand as substrate for the acclimatization of plantain. Plantlets from three plantain cultivars (Batard, Ebanga and French Clair) were obtained after 16 weeks of tissue cultures and the plantlets were subjected to routine acclimatization under screen house conditions using two different substrates mixed in different ratios (100% Sand, 100% PMI, 75% PMI, 60% PMI and 50% PMI). The experiment was arranged in a completely randomized design with ten (10) replications; each replicate consisting of one micro-pot. The different substrates used significantly influenced the performance of the cultivars. The best medium for acclimatization for French Clair was 60% PMI in terms of percentage survival of plantlets (96.88%), plantlet height (6.03 cm), diameter (0.60 cm), number of leaves (4.42 leaves), leaf area (20.23 cm2), leaf emergence rate (1.64), number of roots (7.70 roots), and root length (18.86 cm). Ebanga plantlets had the best results with 75% PMI in terms of percentage survival of plantlets (96.88%), plantlet height (6.18 cm), diameter (0.62 cm), number of leaves (4.39 leaves), leaf area (20.48 cm2), leaf emergence rate (1.76), and total fresh weight (10.05 g). Meanwhile with Batard cultivar, 50% PMI was the best substrate in terms of percentage survival of plantlets (96.88%), plantlet height (4.41 cm), diameter (0.55 cm), number of leaves (4.55 leaves), leaf area (12.96 cm2), leaf emergence rate (1.55), and number of roots (5.73 roots). This study clearly show that PMI can be a viable substrate to use with sand in plantlet acclimatization; however, the different plant cultivars had optimal result at different proportions of PMI.

ABSTRACT

Se estudió la capacidad de aclimatación de 6 variedades de Chenopodium quinoa Willd. (Kancolla, Salcedo, Ccoito, Negra collana, Pasancalla e Illpa) a alta temperatura. Las plantas de quinua se obtuvieron a partir de semillas, las que crecieron en macetas, en un sombreadero, hasta el estado fenológico de 6 hojas verdaderas. Estas fueron expuestas a un tratamiento por calor en una cámara de crecimiento con termostato a 34/32 °C día/noche (D/N) por 3 días y a baja intensidad luminosa. Las no aclimatadas crecieron en las mismas condiciones pero a la temperatura de 22/20 °C D/N. La tolerancia a la alta temperatura se determinó exponiendo discos de hojas a diferentes tiempos (8, 16, 32 y 64 min) a 50 °C (baño maría), midiendo la termoestabilidad de la membrana celular, por la pérdida electrolitos utilizando un conductimetro. El grado de termotolerancia fue hallado según el TL50(tiempo letal al 50% de daño). Los resultados muestran que las variedades Pasancalla e Illpa presentaron capacidad de aclimatación, la variedad más termotolerante fue la variedad Illpa TL50= 64 min cuando fue aclimatada y la más sensible fue la variedad Pasancalla TL50= 8 min cuando no fue aclimatada. También se observó que el porcentaje de daño celular se incrementó a medida que el tiempo de exposición a la alta temperatura fue mayor


The capacity of acclimatization at high temperature of 6 varieties (Kancolla, Salcedo, Ccoito, Negra collana, Pasancalla and Illpa) of Chenopodium quinoa Willd was studied. Quinoa plants were obtained from seeds, which grew in pots, in a grennhouse, until the phenological stage of 6 true leaves. These were exposed to a heat treatment in a growth chamber with a thermostat at 34/32 °C day/night (D/N) for 3 days and at low light intensity. The non-acclimated plants grew under the same conditions but at temperature of 22/20 °C D/N. The tolerance to high temperature was determined by exposing leaf discs at different times (8, 16, 32 and 64 min) at 50 °C (warm bath), measuring the thermostability of the cell membrane trough the loss of electrolytes with a conductimeter. The degree of thermotolerance was found according to the TL50 (time lethal to 50% of damage). The results showed that the varieties Pasancalla and Illpa presented acclimatization capacity, the most thermotolerant variety was the variety Illpa TL50 = 64 min when it was acclimatized and the most sensitive was Pasancalla TL50 = 8 min when it was not acclimatized. It was also observed that the percentage of cell damage increased as the exposure time to the high temperature was higher.

ABSTRACT: The aim of the present study was to evaluate the effects of carbohydrate supplementation on the propagation of the orchid Cattleya schilleriana. The 120-d-old seedlings were subcultured in fructose-, glucose-, or sucrose-supplemented (0, 15, 30, and 45g L-1) ½ MS culture medium (half-strength macronutrient concentrations), using a completely random design with four repetitions per treatment. After 120d of treatment, root number and length, leaf number and length, and fresh weight were evaluated, and seedling survival was evaluated after 75d of acclimatization in a greenhouse. The in vitro growth data were submitted to regression analysis, whereas the percentage survival data were analyzed using ANOVA and Tukey's test. Both in vitro growth and ex vitro survival were lowest when the plantlets were grown in the absence of a carbohydrate source and highest (>90% survival) when supplemented with glucose. According to our findings, the addition of either glucose (30g L-1) or sucrose (30g L-1) is recommended for mass propagation of C. schilleriana.


RESUMO: O objetivo deste estudo foi avaliar a adição de doses de carboidratos em meio de cultura ½ MS para a propagação da orquídea Cattleya schilleriana. Plântulas com 120 dias após semeadura foram recultivadas em 12 tratamentos, onde foram investigadas a adição de diferentes doses de frutose, glicose e sacarose (0, 15, 30 e 45g L-1). O delineamento utilizado foi inteiramente casualizado com quatro repetições por tratamento. Após 120 dias nos tratamentos, foram mensurados o número de raízes, comprimento da maior raiz, número de folhas, comprimento da maior folha e massa fresca total. Depois as plântulas foram aclimatizadas em casa de vegetação e avaliada a taxa de sobrevivência após 75 dias. Os dados obtidos foram submetidos ao teste de Tukey a 5% de probabilidade e ajustadas curvas de regressão. A ausência de carboidrato reduziu o crescimento das plântulas entre todas as características avaliadas in vitro e ex vitro. As doses de glicose promoveram maior eficiência para o crescimento in vitro e sobrevivência acima de 90% após a aclimatização. Assim, a adição de glicose (30g L-1) ou sacarose (30g L-1) são recomendadas para a propagação massal de C. schilleriana.

ABSTRACT

Objective To investigate the effects of heat acclimatization training on the inflammatory reaction and multiple organ dysfunction syndrome (MODS) in patients with exertional heat stroke (EHS). Methods 600 officers and soldiers from a special team who participated in 5 km armed wild training from June to July 2017 were selected as the research object, while 30 healthy officers and men who did not participate in armed wild training during the same period were selected as normal controls. The officers and soldiers who participated in 5 km armed wild training (ambient temperature > 35 ℃, humidity > 65%, 2-3 times a week for 3 weeks) were divided into heat acclimatization group and non-acclimatization group, with 300 in each group. The heat acclimatization group first took part in the heat acclimatization training of wild or long distance running (the initial temperature was 30 ℃, gradually transferred to the hot time of 37 ℃), 2 hours each time, twice a day, and 5 days a week, for a total of 2 weeks. Venous blood was taken before and after heat acclimatization training, before armed wild training, and after the last training or EHS onset, and the contents of serum interleukin (IL-1β, IL-10), tumor necrosis factor-α (TNF-α) and γ-interferon (IFN-γ) were detected by enzyme linked immunosorbent assay (ELISA). The occurrence of EHS and MODS in EHS patients were recorded. Results There was no significant difference in serum inflammatory factors between the officers and soldiers who participated in the training and the healthy control group before heat training or cross-country training. Compared with those before heat training, IL-1β, TNF-α, IFN-γ were significantly increased in all participants of heat acclimatization training while IL-10 was significantly decreased. For those who experienced premonitory (6 cases) and mild (2 cases) heatstroke during training, they could return to normal without severe heatstroke or EHS within 10-30 minutes after being immediately put in a cool and ventilated place and given anti- heatstroke drugs and other interventions. Compared with those before wild training, the levels of inflammatory factors in the two groups of officers and soldiers also changed after wild training, but the increase or decrease of inflammatory indexes in the heat acclimatization group were significantly smaller than those in the non-acclimatization group [IL-1β (ng/L): 10.65±5.18 vs. 12.13±7.91, TNF-α (ng/L): 14.60±5.79 vs. 16.27±8.52, IFN-γ (ng/L): 13.66±5.43 vs. 15.33±8.71, IL-10 (ng/L):8.22±2.68 vs. 7.13±2.63, all P < 0.05]. During armed wild training, a total of 27 cases of EHS occurred. The incidence of EHS in the heat acclimatization group was significantly lower than that in the non-acclimatization group [2.67% (8/300) vs. 6.33% (19/300), χ2= 4.693, P = 0.030]. In patients with EHS, IL-1β, TNF-α, IFN-γ after wild training in the heat acclimatization group were also significantly lower than those in the non-acclimatization group, and IL-10 was significantly higher [IL-1β (ng/L): 34.50±3.74 vs. 39.53±4.51, TNF-α (ng/L): 43.75±2.87 vs. 46.79±2.66, IFN-γ (ng/L): 40.25±1.75 vs. 46.58±1.92, IL-10 (ng/L): 7.50±2.45 vs. 5.42±1.80, all P < 0.01], and the incidence of MODS and organ involvement of EHS patients in the heat acclimation group were significantly lower than that in the non-acclimatization group [50.00% (4/8) vs. 89.47% (17/19), χ2= 5.075, P = 0.024; 28.13% (9/32) vs. 47.79% (65/136), χ2=4.066, P=0.044]. Conclusion Heat acclimatization training before high strength training in high temperature and humidity environment can effectively reduce the degree of inflammation reaction of EHS, protect the physiological functions of EHS organs, and reduce the incidence of MODS.

Objective To evaluate the efficacy of tinnitus retraining therapy (TRT) on anxiety , depression , and symptoms of sleep disorders in patients with chronic subjective tinnitus. Methods Eighty patients with chronic subjective tinnitus visiting The Affiliated Hospital of Hangzhou Normal University from January 2016 to December 2017 were recruited ,and were divided into an observation group and a control group through computer generated random numbers. Patients in the control group were given drug treatment only ,while those in the observation group received TRT in addition to drug treatment. Clinical efficacy was evaluated using Tinnitus Handicap Inventory (THI) , Pittsburgh Sleep Quality Index (PSQI) ,Hamilton Depression Scale (SDS) ,and Hamilton Anxiety Scale (SAS ) for both groups. Results Patients in the observation group were associated with significantly lower scores of THI ,PSQI ,SAS and SDS ,compared with those in the control group 3 months after treatment (each P<0.05). Furthermore ,the effectiveness rate was markedly higher (82.5% or 33 cases vs. 55.0% or 22 cases ,χ2=7.040 ,P<0.01) in the observation group than in the control group 6 months after treatment. Conclusion Tinnitus retraining therapy combined with conventional therapy can help to reduce the severity of tinnitus handicap ,ameliorate negative emotions such as anxiety and depression ,and improve sleep quality in patients with chronic subjective tinnitus.
