ABSTRACT

Objective: This paper aims to provide the experimental foundation for quality control of Aurantii Immaturus Fructus from different habitats by analyzing the multi-component qualitatively and quantitatively. Methods: Fingerprint pattern and QAMS were combined in this experiment, and HPLC analysis was performed on a Waters X-Brige C18 (200 mm × 4.6 mm, 5 μm) column with mobile phase composed of acetonitrile (A) -0.1% methanoic acid solution at a flow rate of 1.0 mL/min in gradient elution mode. The column temperature was maintained at 30 ℃, the injection volume was 10 μL, and the detection wavelengths were set at 283 and 330 nm. Results: The HPLC-UV fingerprint of Aurantii Immaturus Fructus from different habitats was established with 21 common peaks, 9 of them were identified as narirutin, naringin, hesperidin, neohesperidin, naringenin, hesperetin, sinensetin, nobiletin and tangeretin, and the similarity of 23 batches of samples were greater than 0.9, CA and PCA show the classification is consistent with the habitats distribution of the medicine. Then, the QAMS result showed some differences with neohesperidin to be the internal standard. Conclusion: From the perspective of composition, there are some differences in the quality of Aurantii Immaturus Fructus from different habitats, and the method of QAMS is accurate, efficient and feasible, which has obvious advantages than standard curve method and external standard method. In a word, it can be applied to quality evaluation and provides the experimental foundation for quality control of Aurantii Immaturus Fructus from different habitats.

ABSTRACT

Objective To identify whole ingredients in Niuhuang Qingwei Pills (NQP) and give primary grade evaluation of NQP. Methods Firstly, microscopic characteristics specified by the statutory standard of NQP were summarized. Then new microscopic identification methods were established for Sennae Folium and Gypsum Fibrosum while new thin layer chromatography (TLC) identification methods were established for Bovis Calculus Artifactus, Borneolum Syntheticum, and Rhei Radix et Rhizoma. In addition, microscopic identification method for Menthae Haplocalycis Herba was improved. In this way, whole-ingredient identification of 17 materials were realized. NQP reference drug was developed and applied as accompanying control in whole-ingredient identification and primary grade evaluation of the samples. Results According to the results of 48 samples from 18 manufactures by primary grade evaluation, 10, 37, and 1 samples were classified as superior, qualified, and unqualified, respectively. Conclusion The proposed methods are accurate, simple, and objective, which offers a more comprehensive approach for quality control of NQP. And the methods provide research strategy and experimental basis for further work on establishment of grade standard.

ABSTRACT

Objective: To establish a method for simultaneously determination for the content of four flavones (narirutin, naringin, hesperidin, and neohespridin) in Aurantii Immaturus Fructus by single marker, which is feasible and accurate. Methods: Taking hesperidin as internal standard substance, to establish a method for quantitative analysis of multi-components with single marker. To determinate the content of narirutin, naringin, hesperidin, and neohespridin in Aurantii Immaturus Fructus. The contents in 10 batches of samples were determined by both external standard method and QAMS. The The scientificity and feasibility of the method were evaluated by comparison of the quantitative results between external standard method and QAMS. Results: The relative correction factor (RCF) was perfect. The results calculated with a single marker were consistent with the results by the external standard method. Conclusion: The method with a single marker is accurate and feasible to evaluate the quality of Aurantii Immaturus Fructus.
