ABSTRACT

A non specific acid phosphatase from Aspergillus oryzae NRRL447 catalyzes the phosphate hydrolysis from nicotinamide adenine dinucleotide forming nicotinamide riboside, adenosine and Pi as the final products of the reaction. The enzyme was purified to homogeneity by a sequential treatment of acetone fractionation, DEAE-cellulose chromatography and gel filtration chromatography. The enzyme was purified 400-fold. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) of the purified enzyme showed a single protein band of MW 52 kDa. The enzyme displayed maximum activity at pH 5.0 and 40 °C with NAD as substrate. The enzyme activity appeared to be stable over pH 2.0–5.0 and up to 40 °C. The enzyme activity was enhanced slightly by Mg2+, Ca2+ whereas inhibited strongly by F-, Mo04 -, Cu2+ and Fe2+. The enzyme hydrolyzes several phosphate esters, suggesting a probable non-specific nature. The substrate concentration-activity relationship is the hyperbolic type and the apparent Km for NAD+ was 6.25 x 10-4 M.

ABSTRACT

Estimation of time since death is one of the primary objectives of an autopsy. Forensic Scientists and researchers have been persevering hard to find out methods of accurate determination of postmortem interval since long. However, the concept of “Postmortem Clocking” so far seems to be a distant dream only. The favorite biological fluids, to study postmortem biochemical changes, have been those which withstand putrefactive changes for longer duration, like vitreous humor, cerebrospinal fluid, pericardial fluid etc. In blood, markers like electrolytes, urea, creatinine, glucose etc have been more commonly studied. Enthusiastic studies have been undertaken by various researchers to find out reasonably reliable methods of estimating postmortem interval by studying serial quantitative changes in serum levels of various enzymes and to extrapolate the data obtained therefore in terms of duration of death. However, the accuracy of such an opinion remains big area of concern even today, as the range of duration is mostly too wide to be practically useful.

ABSTRACT

Se realizó un estudio para determinar la actividad de la fosfatasa ácida y alcalina en 180 cepas fúngicas aisladas desde suelo rizósferico y la rizósfera de tres plantas forrajeras (Dactilys glomerata, Lolium perenne y Trifollium repens) cultivadas en una pradera en rotación y una pradera permanente. Las fosfatasas fueron determinadas por el método desarrollado por Tabatabai & Bremner (1969) y se leyeron en un espectrofotómetro a 400 nm. Los resultados obtenidos se sometieron a un análisis de varianza (ANDEVA). En las cepas fúngicas ensayadas se determinó actividad para fosfatasa ácida y alcalina. Los mayores valores tanto para fosfatasa ácida (139.68 mg/mL*g micelio) y alcalina (127.12 mg/mL*g micelio) los registró la cepa 14-2R de Penicillium chrysogenum aislada de la rizófera de L. perenne cultivado en pradera permanente. Entre las mejores cepas evaluadas de la pradera permanente existió una mejor concordancia entre los valores determinados para fosfatasa ácida y alcalina.

ABSTRACT

La relación que existe entre la flora microbiana y la actividad de la fosfatasa ßcida (FAc) en los tejidos animales es poco conocida y su acción sobre los tejidos orales de animales inmunes de gÚrmenes (GF) es poco estudiada. El objetivo de este trabajo es verificar la influencia de la flora indÝgena sobre la actividad de FAc en el masetero, lengua, riñón e hÝgado de animales GF y convencionales (CV). Fueron seleccionados camundongos con 45 dÝas, machos y hembras: 10 GF y 10 CV. Para el estudio fueron removidos el masetero, la lengua, el riñón y el hÝgado y posteriormente teñidos histoquÝmicamente para la actividad de FAc. Anßlisis de los resultados mostraron una reacción enzimßtica fuertemente positiva en los animales CV, en cuanto que los tejidos de los animales GF se presentó francamente positiva. Este estudio sugiere que la microbiota indÝgena es importante para la reaccion de FAc en los tejidos de los animales estudiados

ABSTRACT

Se revisan la aplicación y utilidad diagnóstica de los principales métodos citoquímicos en diferentes trastornos hematológicos. Se describen las técnicas siguientes: hemosiderina y sideroblastos, fosfatasa alcalina leucocitaria, mieloperoxidasa, reacción con ácido peryódico-Schiff (PAS), fosfatasa ácida, esterasas específicas e inespecíficas; con indicaciones sobre procedimientos e interpretación de resultados acorde con la experiencia de los autores. Se propone un algoritmo para el estudio de leucemias agudas y se expone un amplio material fotográfico que abarca a todas las técnicas empleadas

ABSTRACT

The acid phosphatase in ungerminated conidia from Colletotrichum graminicola, a corn pathogen, was investigated using spectrophotometric and cytochemical methods. Acid phosphatase activity was studied in a homogenate obtained by fragmentation of ungerminated conidia. With p-nitrophenylphosphate as substrate, the apparent Vmax and Km were 1.000 nmol p-nitrophenol/mg of protein/min and 0.631 mM, respectively. The pH and temperature optima were 5.5 and 60 graus Celsius, respectively. A cytochemical ultrastructural assay showed deposition of the reaction product inside vacuoles but not extracellularly on the cell surface. The permeabilization of conidia with Triton X-100 increased acid phosphatase activity eight fold. Compared to other procedures, our method was fast, easy to perform and gave consistent results.

ABSTRACT

Se investigó la presencia de la 5 isoenzima de la fosfatasa ácida leucocitaria tartrato resistente (FATRE) en los monocitos de sangre periférica humana en 32 muestras: 26 normales, 4 plaquetopenias, 1 anemia y 1 tricoleucemia. Se empleó el separador celular Cobe Spectra Versión 4 en 3 muestras y en las demás se obtuvo la concentración celular por centrifugación sin y con partículas de látex, para estudiar monocitos y macrófagos, respectivamente. Empleando el Kit Sigma para las dos reacciones de fosfatasa ácida total y de FATRE, se demostró la presencia de dos poblaciones de monocitos, una minoritaria para FATRE y otra negativa. Con la adición de látex los monocitos se transformaron en macrófagos haciéndose fuertemente positivos para FATRE. En consecuencia se concluye que la FATRE debe desempeñar un papel principal en la función macrofágica y por ende en la inmunidad celular humana.

ABSTRACT

Nonspecific phosphatase activities were surveyed comparing major species of the family Enterobacteriaceae. The strains were subjected to a whole cell assay system with paranitrophenyl phosphate as substrate over a wide pH range and with a standardized number of bacterial cells. The overall results suggest that the general shape of the pH activity curve and the location of peaks (pH optimum) can be employed as a supplementary criterion to characterize species of Enterobacteriaceae.
