ABSTRACT

OBJECTIVE@#To study DNA quantification and STR typing of samples pre-treated with pyramidon.@*METHODS@#The blood samples of ten unrelated individuals were anticoagulated in EDTA. The blood stains were made on the filter paper. The experimental groups were divided into six groups in accordance with the storage time, 30 min, 1 h, 3 h, 6 h, 12 h and 24h after pre-treated with pyramidon. DNA was extracted by three methods: magnetic bead-based extraction, QIAcube DNA purification method and Chelex-100 method. The quantification of DNA was made by fluorescent quantitative PCR. STR typing was detected by PCR-STR fluorescent technology.@*RESULTS@#In the same DNA extraction method, the sample DNA decreased gradually with times after pre-treatment with pyramidon. In the same storage time, the DNA quantification in different extraction methods had significant differences. Sixteen loci DNA typing were detected in 90.56% of samples.@*CONCLUSION@#Pyramidon pre-treatment could cause DNA degradation, but effective STR typing can be achieved within 24 h. The magnetic bead-based extraction is the best method for STR profiling and DNA extraction.

ABSTRACT

This paper reports the establishment of a method for rapid identification 15 effective components of anti common cold medicine (paracetamol, aminophenazone, pseudoephedrine hydrochloride, methylephedrine hydrochloride, caffeine, amantadine hydrochloride, phenazone, guaifenesin, chlorphenamine maleate, dextromethorphen hydrobromide, diphenhydramine hydrochloride, promethazine hydrochloride, propyphenazone, benorilate and diclofenac sodium) with MRM by LC-MS/MS. The samples were extracted by methanol and were separated from a Altantis T3 column within 15 min with a gradient of acetonitrile-ammonium acetate (containing 0.25% glacial acetic acid), a tandem quadrupole mass spectrometer equipped with electrospray ionization source (ESI) was used in positive ion mode, and multiple reaction monitoring (MRM) was performed for qualitative analysis of these compounds. The minimum detectable quantity were 0.33-2.5 microg x kg(-1) of the 15 compounds. The method is simple, accurate and with good reproducibility for rapid identification many components in the same chromatographic condition, and provides a reference for qualitative analysis illegally added chemicals in anti common cold medicine.

ABSTRACT

Traditional medicines, in particular herbal products, have been used abundantly over the years in curing several diseases. Pharmacological interactions of herbal products with modern drugs, however, remain to some extent unknown. Herein, we examined whether co-administration of Faizol Ubat Batuk [FUB], a mixture of aqueous extract of different plants, modifies the metabolism of aminopyrine, a conventional analgesic drug, in rat liver. We used rat hepatocytes outfitted by collagenase perfusion technique. Determination of aminopyrine n-demethylase activity was performed using the Nash colorimetric method, by measuring the amount of formaldehyde produced. Compared to control treatment, FUB significantly increased the hepatic metabolism of aminopyrine in healthy adult male rats. In contrast, the hepatic metabolism of aminopyrine in adult female rats was decreased. Besides, a biphasic effect in n-demethylase activity was observed in young male rats treated with FUB. In a subsequent experiment, FUB did not change the metabolism of aminopyrine in streptozotocin [STZ]-diabetic adult male rats. In conclusion, administration of FUB could affect phase I aminopyrine metabolism in rat heptocytes. In addition, the effects of FUB on hepatic n-demethylase activity were gender and disease dependent

ABSTRACT

The aminopyrine breath [ABT] was done in 25 patients with hepatosplenic schistosomiasis scheduled for splenectomy and left gastric artery ligation and in 15 patients with normal livers and spleens requiring upper abdominal surgical intervention. The relevant radiological and laboratory investigations indicate significant liver involvement in the schistosomal patients. The mean ABT score correlated well with the Child's class where the mean score for class A> class B> class C. However, in this group the mean score for patients with pure schistosomal fibrosis was very close to normal while that for patients with mixed cirrhosis was 40% lower. The incidence of post-operative complications was much higher in schistosomal patients who had lower ABT scores. For determining the probability of postoperative morbidity, the ABT may be of value in the framework of an overall clinico laboratory regime for diagnosis

ABSTRACT

Se desarrolló un modelo de diabetes experimental en ratas , analizaron los parámetros fisiológicos, el contenido de citocromo p-450 hepático y el metabolismo in vitro Fase I y II de estructuras moleculares modelos. Se observaron alteraciones significativas en el contenido de proteína microsomal hepática en las ratas diabéticas en tanto el tratamiento con insulina a estas ratas determinó una disminución del contenido de citocromo p-450 hepático. Las ratas diabéticas presentaron una capacidad metabólica in vitro Fase 1 mayor que las ratas controles y la administración de insulina produjo diversos efectos según sustrato empleado. El metabolismo in vitro Fase II de las ratas diabéticas resultó similar al obtenido en las ratas controles, en cambio el tratamiento con insulina produjo variaciones en la afinidad de la enzima por el sustrato y en la cantidad de la enzima que cataliza la reacción. Los resultados sugieren que el estado de diabetes y su tratamiento con insulina, modifica en forma diferente las isoenzimas citocromo P-450 y reduce selectivamente algunas enzimas UDP glucuronil transferasas.

ABSTRACT

Las pruebas de funcionamiento hepático son instrumento fundamental en el estudio de pacientes con sospecha de enfermedad hepática para precisar el diagnóstico, conocer la gravedad, establecer el pronóstico decidir la terapéutica más adecuada; existen pruebas que se clasifican en dos grandes grupos: las pruebas bioquímicas de función hepática perse y, los marcadores de afección hepatobiliar. Ninguna de ellas en forma individual cumple con estos objetivos, es necesario la integración de varias. En los últimos años, no se ha promovido el uso de pruebas ya existentes y se han innovado algunos aspectos de ellas para hacerlas más accesibles y sencillas. La tendencia actual, estimulada por los programas de trasplante hepático, es evaluar la capacidad metabólica del injerto y disponer de parámetros más cuantitativos del grado de función. La pruebas actuales permanecen como estudios muy útiles en hepatología.

ABSTRACT

Se informa que entre los productos que contienen aminas terciarias se encuentra la aminopirina, conocida como el analgésico piramidón, la cual es fácilmente nitrosable en condiciones de pH moderadamente ácido. Se expresa que el compuesto resultante de la nitrosación es la dimetilnitrosamina (DMN), un agente altamente hepatotóxido y carcinogénico, especialmente al nivel del hígado, donde es metabolizado. Se señala que el modelo experimental, mediante la utilización de ratas albinas hembras con un peso de 100 a 140 g, a las cuales les fue administrada diariamente la aminopirina y el nitrito de sodio aisladamente y en combinación, durante 35 días, confirma la hepatotoxicidad de la combinación, de acuerdo con los cambios hísticos observados que corresponden a alteraciones del tipo cirrótico

Out of a consecutive series of 4000 patients 23 were thought to have a 'fixed drug eruption', representing a far higher incidence than in comparable Western studies. The possible reasons are discussed; the widespread ingestion of the dipyrone drug Novalgin was considered to be the most important single cause. The dangers of dipyrone are emphasized and the wisdom of the health authorities in completely banning such preparations is commended
