ABSTRACT

Este artigo propõe estudar os primeiros 12 anos de existência do Instituto de Radium de Minas Gerais, fundado em 1922. Sua atuação na luta contra o câncer no Brasil, ainda pouco conhecida, começa a ser esboçada pelo estudo de documentação institucional inédita. Através de um banco de dados elaborado com informações constantes em seu livro de registro de pacientes, foram feitos levantamentos estatísticos dos tipos de câncer e das formas de tratamento existentes entre 1923 e 1935. Esse livro faz parte de um conjunto de outros cinco recentemente descobertos no Centro de Memória da Medicina/UFMG. A documentação permite resgatar os primórdios das intervenções de radioterapia no país e acompanhar seu desenvolvimento e a influência exercida por esse hospital modelo.


This article proposes to study the first 12 years of the Minas Gerais Radium Institute, founded in 1922. Its work in the fight against cancer in Brazil, albeit still little known, is coming to light as its institutional documents are studied. A database has been prepared using information from its patient register, based on which statistical analyses have been done to identify the types of cancer and treatments available there between 1923 and 1935. This register is one of five recently unearthed at the Medicine Memory Center of the Universidade Federal de Minas Gerais. Through them, the earliest experiments in radiotherapy in Brazil can be reconstituted, and its development and the influence of this model hospital can be mapped out.

ABSTRACT

Genotoxic effects of benznidazole were studied by the induction of homozygosis of genes previously present in heterozygous. UT448//A757 diploid strain was used in the benznidazole's recombinagenesis test. Although toxic effects on growth of colonies were not observed, 75 and 100 æM benznidazole induced an increasing of mitotic recombination events in diploid strain. Results were related to the induction of chromosomal breaks by the antiparasitic drug.

ABSTRACT

As a preliminary step towards characterizing genes encoding ATP-binding cassette (ABC) transporters that confer pleiotropic drug resistance in Aspergillus, we used a PCR-based approach to isolate four DNA fragments corresponding to different ABC type transporter genes. DNA sequencing and Southern blot analysis confirmed that they were distinct genes, which were designated abcA-D. One of these genes, abcD, was cloned and characterized. It was found to have a predicted 1,452-amino acid translation product with a calculated molecular mass of 147,467 kDa. The abcD gene specifies a single transcript of approximately 5.0 kb; there was a two- to six-fold enhancement of mRNA levels following exposure to miconazole, camptothecin, methotrexate, and ethidium bromide

ABSTRACT

Alcoholic extract of dry nuts of S. anacardium showed dose dependent antifungal activity in vitro against Aspergillus fumigatus and Candida albicans. At 400 mg/ml concentration, growth of both the fungi was inhibited and considerable reduction in size of cells and hyphae was observed. Sporulation also decreased.

ABSTRACT

Microtúbulos säo filamentos compostos por dímeros das tubulinas alfa e beta e têm uma variedade de funçöes nas células vivas. Em fungos, os corpúsculos polares dos fusos säo geralmente considerados os centros organizadores dos microtúbulos. Com o objetivo de contribuir para uma melhor compreensäo dos processos de nucleaçäo dos microtúbulos no fungo filamentoso A. nidulans, nós utilizamos a droga antimicrotúbulo Benomil em experimentos de bloqueio e liberaçäo para depolimerizar e repolimerizar os microtúbulos. Após 20 segundos de reincubaçäo em meio sem Benomil, pequenos microtúbulos foram formados a partir de pontos distribuídos pela célula, sugerindo que os pontos de nucleaçäo de microtúbulos säo aleatoriamente distribuídos pelas hifas de A. nidulans. Como em A. nidulans o movimento nuclear é dependente de microtúbulos foi analisado se mutantes defectivos na distribuiçäo de núcleos ao longo das hifas (mutantes nud) possuíam algum defeito evidente nos microtúbulos. Os microtúbulos citoplasmáticos, dos fusos e astrais estäo presentes e aparentam-se normais em todos os mutantes nud, mas foi observada uma pequena distorçäo na proporçäo de fusos mitóticos longos e curtos nestes mutantes, comparados com o controle. Isto sugere que alguns núcleos de mutantes nud näo alcançam a fase tardia da divisäo celular, em temperatura näo restritiva.

ABSTRACT

El objetivo de este trabajo es estudiar la actividad genética como inductores de recombinación mitótica y de delecciones, de dos medicamentos: meprobamato y pirimetamina, utilizando el ascomiceto Aspergillus nidulans como organismo de ensayo. Para el primer caso se utilizó una cepa diploide heterocigótica para marcadores del color de los conidios, y en el segundo caso, la cepa llevaba una duplicación de parte del genoma en el grupo de ligamento II. Para cada medicamento se probaron varias concentraciones diferentes; 6 para el meprobamato y 4 para la pirimetamina. Los resultados obtenidos indican que el meprobamato induce delecciones a las dosis de 10mM, 5mM y 2,5mM. La pirimetamina, a su vez, presentó un efecto positivo como inductora de recombinación mitótica a la dosis de 50*g/ml. Estos resultados indican que estos medicamentos, de amplio uso en nuestro país, presentan cierto tipo de actividad genética, detectada en las condiciones experimentales usadas por nosotros en este trabajo
