ABSTRACT

The objective of the present study was to investigate the different plasma metabolites between anestrus and estrus postpartum dairy cows and to provide a theoretical basis for prevention of anestrus in dairy farm cows. In the experiment, one hundred and sixty-seven Holstein dairy cows were selected with similar age and parity. According to the concentration of ß-hydroxybutyric acid, non-esterified fatty acids and glucose in plasma during 14 to 21 days in milk, all dairy cows were determined as having a status of energy balance. According to the results of clinical symptom, rectal and B ultrasound examination at 60 to 90 days postpartum, these cows were divided into twenty estrus and twenty-four anestrus group, other dairy cows were removed. 1H nuclear magnetic resonance technology was utilized to detect the plasma metabolites changes and screen different plasma metabolites between anestrus and estrus cows. Ten different metabolites including alanine, glutamic acid, asparagine, creatine, choline, phosphocholine, glycerophosphocholine, low-density lipoprotein, and very-low-density lipoprotein were significantly decreased in anestrous cows compared with estrous cows. Metabolic pathway analyses indicated that differential metabolites were primarily involved in amino acid and glycerophospholipid metabolism. These metabolites and their enrichment pathways indicate that reduced steroid hormone synthesis precursors result in lower levels of estradiol and progesterone and cause anestrus in negative energy balance. These data provide a better understanding of the changes that may affect estrus of postpartum dairy cows at NEB status and lay the ground for further research.(AU)


O objetivo do presente estudo foi investigar os diferentes metabolitos do plasma entre o cio e o cio pós-parto de vacas leiteiras e fornecer uma base teórica para a prevenção do cio de vacas em fazendas de leite. No experimento, foram selecionadas 127 vacas leiteiras Holstein com idade e paridade similares. De acordo com a concentração de ß- ácido hidroxibutírico, ácidos graxos não esterificados e glicose no plasma entre 14 e 21 dias no leite, todas as vacas leiteiras foram determinadas em estado de equilíbrio energético. De acordo com os resultados dos sintomas clínicos, do exame de ultra-som retal e B aos 60 a 90 dias pós-parto, estas vacas foram divididas em vinte cios e vinte e quatro grupos de cio, outras vacas leiteiras foram removidas. A tecnologia de ressonância magnética nuclear 1H foi utilizada para detectar as alterações dos metabólitos plasmáticos e para triar diferentes metabólitos plasmáticos entre as vacas do cio e do cio. Dez diferentes metabólitos incluindo alanina, ácido glutâmico, asparagina, creatina, colina, fosfocholina, glicerofosfocolina, lipoproteína de baixa densidade e lipoproteína de muito baixa densidade foram significativamente diminuídos nas vacas antróficas em comparação com as vacas estro. As análises da via metabólica indicaram que os metabólitos diferenciais estavam principalmente envolvidos no metabolismo de aminoácidos e glicerofosfolipídios. Estes metabólitos e suas vias de enriquecimento indicam que a redução dos precursores da síntese de hormônios esteróides resulta em níveis mais baixos de estradiol e progesterona e causa anestros no balanço energético negativo. Estes dados fornecem uma melhor compreensão das mudanças que podem afetar o cio das vacas leiteiras pós-parto no estado de NEB e preparam o terreno para mais pesquisas.(AU)

ABSTRACT

El objetivo de este manuscrito es realizar una revisión y actualización de la literatura de la insuficiencia ovárica primaria (IOP) en población adolescente, a partir del diagnóstico, manejo y seguimiento de un caso clínico. La insuficiencia ovárica primaria se define como la menopausia en una mujer antes de los 40 años, acompañada de amenorrea, hipogonadismo hipergonadotrópico e infertilidad. Su prevalencia varía entre 1 a 2%, y en mujeres menores de 20 años su prevalencia es un caso de cada 10,000. Aunque se sabe que muchas afecciones pueden llevar a una IOP, la más común es la causa idiopática. La presentación clínica es diversa, y varios trastornos diferentes pueden también, llevar a esta condición. CASO CLÍNICO: Se presenta el caso de una adolescente de 17 años, previamente sana, con historia de amenorrea secundaria, no embarazada, con examen físico general y ginecológico normal. Se solicita estudio analítico complementario resultando con niveles de hormona folículo estimulante (FHS), estradiol (E2) y hormona antimülleriana (AMH) compatibles con una insuficiencia ovárica como la observada en la posmenopausia. Se inicia terapia hormonal (TH) clásica con estradiol y progesterona, siendo posteriormente reemplazada por anticoncepción hormonal combinada (AHC) oral, coincidente con el inicio de vida sexual, con respuesta favorable y sangrados regulares. La IOP tiene graves consecuencias para la salud incluyendo trastornos psicológicos como angustia, síntomas depresivos o depresión, infertilidad, osteoporosis, trastornos autoinmunes, cardiopatía isquémica, y un mayor riesgo de mortalidad. La enfermedad de Hashimoto es el trastorno autoinmune más frecuente asociado a la IOP. Su tratamiento y diagnóstico deben establecerse de forma precoz para evitar consecuencias a largo plazo. La terapia con estrógenos es la base del tratamiento para eliminar los síntomas de la deficiencia de estrógenos, además de evitar las consecuencias futuras del hipogonadismo no tratado. También el manejo debe incluir los siguientes dominios: fertilidad y anticoncepción, salud ósea, problemas cardiovasculares, función psicosexual, psicológica y neurológica, informando a los familiares y a la paciente sobre la dimensión real de la IOP y la necesidad de tratamiento multidisciplinario en muchos casos. CONCLUSIÓN: El caso presentado, pese a ser infrecuente, permite abordar de manera sistematizada el diagnostico de IOP y evaluar alternativas de manejo plausibles para evitar graves consecuencias en la salud, así como conocer respuesta clínica y de satisfacción de la adolescente.


The objective of this manuscript is to review and update the literature on primary ovarian insufficiency (POI) in an adolescent population, based on the diagnosis, management and follow-up of a clinical case. Primary ovarian insufficiency is defined as menopause in a woman before the age of 40, accompanied by amenorrhea, hypergonadotropic hypogonadism, and infertility. Its prevalence varies between 1 to 2%, and in women under 20 years of age its prevalence is one case in every 10,000. Although it is known that many conditions can lead to POI, the most common is the idiopathic cause. The clinical presentation is diverse, and several different disorders can also lead to this condition. CLINICAL CASE: The case of a 17-year-old adolescent, previously healthy, with a history of secondary amenorrhea, not pregnant, with a normal general physical and gynecological examination is presented. A complementary analytical study is requested, resulting in levels of follicle stimulating hormone (FHS), estradiol (E2) and anti-müllerian hormone (AMH) compatible with ovarian insufficiency such as that observed in postmenopause. Classic hormonal therapy (HT) with estradiol and progesterone was started, later being replaced by combined hormonal contraception (CHC), coinciding with the beginning of sexual life, with a favorable response and regular bleeding. POI has serious health consequences including psychological disorders such as distress, depressive symptoms or depression, infertility, osteoporosis, autoimmune disorders, ischemic heart disease, and an increased risk of mortality. Hashimoto's disease is the most common autoimmune disorder associated with POI. Its treatment and diagnosis must be established early to avoid long-term consequences. Estrogen therapy is the mainstay of treatment to eliminate the symptoms of estrogen deficiency, in addition to avoiding the future consequences of untreated hypogonadism. Management should also include the following domains: fertility and contraception, bone health, cardiovascular problems, psychosexual, psychological and neurological function, informing family members and the patient about the real dimension of POI and the need for multidisciplinary treatment in many cases. CONCLUSION: The case, although infrequent, allows a systematic approach to the diagnosis of POI and evaluate plausible management alternatives to avoid serious health consequences, as well as to know the clinical response and satisfaction of the adolescent.

ABSTRACT

The various studies on the manipulation of the estrous cycle culminated in the development of estrous and ovulation synchronization protocols, to the point of defining an optimum moment for artificial insemination. The induction of ovulation is a primordial part for the determination of the moment of the TAI, so to study which hormones and how they behave allows a better understanding of this physiological process and, to manipulate it efficiently. The aim of the present study was to compare the use of estradiol benzoate (EB) and cypionate (ECP) as ovulation inducer in Girolando cows submitted to Timed Artificial Insemination (TAI). Ovulations of 108 Girolando cows were synchronized and the initial day of treatment, recorded as D0, which was when the animals received an intravaginal device containing 1g of progestogen and an intramuscular application (im) of 2mg of EB. After 8 days (D8), the device was removed and 500µg of cloprostenol (im) was administered to all females. Then, the animals were randomly divided into two treatments: BG Group (n = 52) and CG Group (n = 56). The CG animals received 1mg of ECP (im) as the ovulation inducer at the time of device removal, while BG Group cows received 1mg of EB (im) 24 h later (D9). TAI occurred at D10. After device removal, ultrasound evaluations were performed every 12 h up to ovulation. The following reproductive parameters were evaluated: interval from intravaginal device removal to ovulation - IDO (hours); interval from ovulation to TAI-IOT (hours); diameter of the largest follicle at intravaginal device removal (mm); maximum diameter of the DF (mm) at D10; the dominant follicle growth rate (mm/day); synchronization rate - SR (%); ovulation rate - OR (%) and PR - pregnancy rate (%). Only the ovulation rate presented a statistical difference (p<0.05). In conclusion, despite the ovulation rate difference, both estradiol esters administered were effective and presented similar pregnancy rates in Girolando cows submitted to TAI.


Os diversos estudos sobre manipulação do ciclo estral culminaram com o desenvolvimento de protocolos de sincronização do estro e da ovulação, a ponto de se definir um momento ótimo para a inseminação artificial. A indução da ovulação é parte primordial para a determinação do momento da IATF. Dessa forma, estudar quais hormônios e como eles se comportam permite compreender melhor esse processo fisiológico e, ainda, manipulá-lo de forma eficiente. O objetivo do presente estudo foi comparar o uso de benzoato de estradiol (BE) e cipionato (ECP) em vacas Girolando submetidas à inseminação artificial em tempo fixo (IATF). As ovulações de 108 vacas Girolando foram sincronizadas e no início do tratamento, D0, os animais receberam um dispositivo intravaginal contendo 1g de progestágeno e uma aplicação intramuscular (im) de 2mg de BE. No D8, o dispositivo foi removido e 500µg de cloprostenol (im) foram administrados a todas as fêmeas. Depois disso, os animais foram divididos aleatoriamente em dois tratamentos: Grupo GB (n = 52) e Grupo GC (n = 56). Os animais do Grupo GC receberam 1mg de ECP (im) como indutor de ovulação no momento da retirada do dispositivo, enquanto as vacas do Grupo GB receberam 1mg de BE (im) 24 horas depois (D9). IATF ocorreu em D10. Após a remoção do dispositivo, as avaliações ultrassonográficas foram realizadas a cada 12 horas até a ovulação. Os parâmetros reprodutivos avaliados foram: intervalo entre a retirada do dispositivo intravaginal de P4 e a ovulação- IRD (horas); intervalo entre a ovulação e a IATF- IOI (horas); diâmetro do maior folículo na retirada do dispositivo intravaginal de P4 (mm); taxa de crescimento do FD-TCFD (mm/dia); taxa de sincronização - TS (%); taxa de ovulação- TO (%) e taxa de concepção - TC (%). Apenas a taxa de ovulação apresentou diferença estatística (p<0.05). Em conclusão, apesar da diferença na taxa de ovulação, ambos os ésteres de estradiol administrados foram eficazes e apresentaram taxas de prenhez semelhantes em vacas Girolando submetidas a IATF.

ABSTRACT

Foram avaliados os efeitos tóxicos do hormônio 17β-estradiol (E2) livre e complexado à β-ciclodextrina (CD) sobre o comportamento e a fisiologia de tilápia (Oreochromis niloticus). Os peixes foram observados por 30 dias, em dois estágios do desenvolvimento (alevino e juvenil), pelo método ad libitum, para a confecção de um etograma. Posteriormente, juvenis foram divididos em três grupos: controle e expostos ao E2 (10ng/L) livre e complexado à β-ciclodextrina (β-CD:E2) por 90 dias. Foram avaliados o comportamento pelo método de varredura instantânea, o consumo de ração, o ganho de peso e a mortalidade em diferentes intervalos. Os alevinos e os juvenis apresentaram frequências de exibição comportamentais diferentes (P<0,05) nos eventos: Afastar (4,7±1,3 e 3,6±0,6%) e Ondulação de repulsão (2,3±0,9 e 1,3±1,0%). Os juvenis expostos ao complexo β-CD:E2 apresentaram aumento (P<0,05) na exibição dos comportamentos agressivos, como Afastar, Ataque caudal, Confronto prolongado, Perseguição, Fuga, e menor mortalidade, quando comparados ao grupo exposto ao E2 livre e controle. Pode-se concluir que a complexação do E2 com a β-CD alterou a toxicidade do E2, pois promoveu um aumento na frequência de exibição dos comportamentos agressivos e interferiu na mortalidade dos animais.(AU)


Toxic effects of free and complexed 17β-estradiol (E2) hormone into β-cyclodextrin (CD) on the behavior and physiology of tilapia (Oreochromis niloticus) were evaluated. The fish were observed for 30 days in two stages of development (fingerling and juvenile) by the ad libitum method to make an ethogram. After this, juveniles were divided into three groups: control and exposed to free E2 (10ng/L) and complexed into β-cyclodextrin (β-CD:E2) for 90 days. The behavior was evaluated through scan sampling method, feed intake, body mass and mortality at different intervals. The fingerlings and juveniles showed behavioral patterns with different display frequencies (P<0.05) for events: Move Away (4.7±1.3 and 3.6±0.6%) and Waving Repulsion (2.3±0.9 and 1.3±1.0%). The juveniles exposed to β-CD:E2 complex showed a significant increase (P<0.05) in the frequency of display of aggressive behaviors as Move Away, Caudal Attack, Clash Extended, Chase, Escape and decrease of mortality when compared to group exposed to free E2 and control. In conclusion, complexation of E2 into β-CD modified E2 toxicity, because it promoted an increase in the frequency of display of aggressive behaviors and it affected the mortality of animals.(AU)

ABSTRACT

Background: CDC25 is a dual-specificity phosphatase that was first identified in the yeast Schizosaccharomyces pombe as a cell cycle-defective mutant. Although CDC25 is involved in the cell cycle of ovarian granulosa cells, the CDC25 signaling pathway has not been clarified fully. To explore the role of CDC25C in the cell cycle of goat ovarian granulosa cells, a CDC25C-overexpressing vector, pCMV-HA-CDC25C, was constructed and transfected into granulosa cells from adult and young white goats from Jiangsu Nantong. RT-PCR was used to measure CDC25C, CDK1, and WEE1 gene expression levels, and flow cytometry was used to distinguish ovarian granulosa cells in different phases of the cell cycle. Progesterone and estradiol levels in transfected ovarian granulosa cells were also measured. Results: In adult goat follicular granulosa cells transfected with pCMV-HA-CDC25C, CDC25C expression increased significantly, which greatly increased the relative gene expression levels of both CDK1 and WEE1. Additionally, progesterone and estradiol levels were increased in goat follicular granulosa cells overexpressing CDC25C. And the cell cycle results showed that transfection of pCMV-HA-CDC25C leads to a higher proportion of cells in S phase compared to the no vector-transfected groups. Conclusions: The results of this study indicated that the overexpression of CDC25C may increase the gene expression levels of both WEE1 and CDK1 in S phase and accelerate the transition of cells from G1 phase to S phase.

Muitas vezes, torna-se um grande desafio para o ginecologista a identificação daquelas com maior ou menor chance de concepção. Vários marcadores laboratoriais e ultrassonográficos, conhecidos conjuntamente como testes de avaliação da reserva ovariana, são estudados há décadas com a intenção de se buscar uma ferramenta para a predição do potencial reprodutivo. E, embora ainda se busquem os marcadores ideais para aplicação clínica, mais difícil do que os definir é definir quando eles estão indicados. Este artigo de atualização, assinado pela Comissão Nacional Especializada em Ginecologia Endócrina da Febrasgo, pretende oferecer ao leitor as ferramentas necessárias para o uso racional dos testes de avaliação da reserva ovariana no cotidiano.(AU)


Often, it becomes a great challenge for the gynecologist to identify women with a greater or lesser chance of conception. Several laboratory and ultrasound markers, known jointly as ovarian reserve evaluation tests, have been studied for decades with the intention of seeking a tool for the prediction of reproductive potential. And, while the ideal markers for clinical application are still sought, defining them is as harder as defining when they are indicated. This update article, signed by the National Specialized Committee on Gynecologic Endocrinology, Febrasgo, intends to offer the reader the necessary tools for the rational use of ovarian reserve evaluation tests in daily practice.(AU)

ABSTRACT

The present study investigated the hormonal profile and expression of prostaglandin F2α (PGF2α), oxytocin and estrogen receptors in uterine tissues of postpartum cows treated with cloprostenol. Twenty Holstein-Zebu crossbred cows were treated with saline solution (treatment CONT) or cloprostenol (treatment CLO), both administered two and five days postpartum. Blood samples were collected on days two, seven, 14, 21 and 28 postpartum for progesterone, PGF2α metabolite (PGFM) and estradiol determination, and endometrial biopsy was performed in order to quantify the expression of oxytocin receptor (OXTR), prostaglandin F receptor (PTGFR) and estrogen receptor 1 (ERS1) genes. In the CLO treatment, expression of OXTR was reduced (P<0.05) but no difference (P>0.05) between treatments was found for PTGFR and ERS1 expression. Estrogen concentrations increased progressively until day 14 (P<0.05) and the highest OXTR expression and lowest PTGFR expression were observed on day 14 (P<0.05) in both treatments. Serum PGFM concentrations were high throughout the experiment. In conclusion, cloprostenol administration at days two and five of postpartum seems to reduce OXTR expression in the endometrium in crossbred cows.(AU)


O presente estudo avaliou o perfil hormonal e a expressão gênica de receptores de prostaglandina F2α (PGF2α), ocitocina e estrógeno no endométrio de vacas pós-parto tratadas com cloprostenol. Vinte vacas mestiças Holandês-Zebu foram tratadas com solução salina (tratamento CONT, n = 10) ou cloprostenol (tratamento CLO, n = 10), ambos administrados dois e cinco dias após o parto. Amostras de sangue foram coletadas nos dias dois, sete, 14, 21 e 28 pós-parto para mensuração de progesterona, de metabólito de PGF2α (PGFM) e de estradiol, e foram obtidas biópsias endometriais para quantificar a expressão de PTGFR, OXTR e ESR1. No tratamento CLO, a expressão gênica de receptores de ocitocina foi menor (P<0,05). As concentrações de estrógeno aumentaram progressivamente até o dia 14 (P<0,05). A maior expressão de OXTR foi observada no dia 14 (P<0,05). A expressão de ESR1 foi semelhante entre os tratamentos (P>0,05). Os níveis de PGFM foram altos durante todo o estudo. Conclui-se que a administração de cloprostenol nos dias dois e cinco pós-parto parece diminuir a expressão de OXTR no endométrio em vacas mestiças.(AU)

ABSTRACT

In this study, four crab-eating fox females (Cerdocyon thous) maintained at the Federal University of Mato Grosso Zoo, Cuiabá, Brazil, were investigated for 12 months, using feces measurement of estradiol and progesterone concentrations. Fecal collections were performed three times a week for hormone extraction. Two methods of analysis, Elisa (EIA) and Radioimmunoassay (RIA), were used in the measurement of progesterone (P4) and estradiol (E2) metabolites. The aim of this study was to compare and validate two different methods of hormone measurement for C. thous. There were no differences regarding the method used. The Radioimmunoassay technique proved to be more sensitive, however, both showed similar results.(AU)


Neste estudo, quatro fêmeas de cachorros-do-mato (Cerdocyon thous) mantidas no Zoológico da Universidade Federal de Mato Grosso, Cuiabá, MT, Brasil, foram investigadas pelo período de 12 meses, mediante a mensuração de concentrações de estradiol e progesterona em fezes. Coletas de fezes foram realizadas três vezes por semana para posterior extração hormonal. Dois métodos de análise de metabólitos fecais, elisaimunoensaio (EIA) e radioimunoensaio (RIA), foram utilizados na mensuração dos metabólitos de progesterona (P4) e estradiol (E2). O objetivo deste estudo foi comparar e validar dois diferentes métodos de mensuração hormonal para C. thous. Não houve diferença significativa com relação ao método empregado. A técnica de radioimunoensaio demonstrou ser mais sensível, no entanto ambas apresentaram resultados semelhantes.(AU)

Aims and Objectives : The study was done to assess the hormones namely Estradiol, Testosterone, and Dehydroepiandrosterone-sulphate (DHEAs) one day before operation and on ninth post-operative day following surgical menopause. Materials and Methods : This is a cross sectional observational study. The study was done amongst the thirty four women aged between 40-48 years with functioning uterus and at least one ovary, not using any exogenous hormone preparations affecting ovarian function for last three months and having at least one menstrual period in three previous months were included in this study. They had under gone hysterectomy with bilateral salpingooophorectomy due to non-ovarian pathology. Fasting venous blood samples were taken one day before operation and on ninth post-operative day of surgical menopause Serum concentration of estradiol, testosterone, and DHEAS were determined. Results : The circulating estradiol level decreased significantly (p = 0.043) from 161 pg/ml preoperatively to 108 pg/ml. on ninth post-operative day after surgical menopause. In spite of reduction in mean testosterone level from 0.11 ng/ml. to 0.09 ng/ml. following surgical menopause, which is statistically insignificant (p = 0.247).There was no significant difference between the serum DHEA-S level before and after surgical menopause. A significant positive correlation was observed between pre-operative circulatory levels of DHEA-S with that of estradiol while there was absence of any significant co-relations corelations between any of the other pairs of values. Conclusion : The circulating estradiol level decreased significantly on ninth day after surgical menopause and significant positive correlation between pre-operative circulatory levels of DHEA-S with that of estradiol, but there was no significant co-relation between post-operative circulating estradiol with that of DHEA-S. Testosterone did not show any significant relation with estradiol neither in pre-operative period nor in post-operative condition.

Objetivo: Relacionar los niveles de hormonas esteroideas foliculares con el ciclo de estimulación ovárica y sus resultados globales. Métodos: Se incluyeron pacientes < 38 años, con esterilidad de causa masculina, tubárica o desconocida, que recibieron un protocolo largo con agonistas de GnRH y rFSH. Se recogieron las muestras de la primera y segunda aspiración folicular de cada ovario y se realizó un quimioinmunoanálisis de estradiol, progesterona, testosterona y DHEAS. Resultados: Se obtuvieron cifras menores de DHEAS folicular en las pacientes con más días de frenado con agonistas de GnRH (p=0,0003). Cuantos más días de rFSH administrados, mayores fueron los niveles de testosterona y DHEAS folicular (p=0,03; p=0,03). En los resultados globales del ciclo, se obtuvo una correlación negativa entre las cifras de testosterona folicular y el número de complejos puncionados (r= -0,360; p=0,002) y entre la testosterona folicular y el número de embriones de calidad D (r= -0,233; p=0,047). El número de ovocitos maduros fue menor en pacientes con mayores niveles de testosterona folicular (p=0,008). La progesterona folicular fue superior en ovocitos de buena calidad frente a los de calidad no destacable (p=0,006) y muy mala calidad (p=0,04). Conclusiones: Las cifras altas de testosterona folicular se correlacionaron con menor número de complejos puncionados, ovocitos maduros y embriones de calidad D. La buena calidad ovocitaria se asoció a niveles de progesterona folicular superiores.


Objective: To relate the levels of follicular steroid hormones with the ovarian stimulation cycle and its overall results. Method: It was included patients < 38 years old with sterility of male, tubaric or unknown origin who underwent a long protocol with GnRH agonists and rFSH. Samples were obtained from the first and second follicular aspiration of each ovary. A chemiluminescent immunoassay of estradiol, progesterone, testosterone and DHEAS was performed. Results: Figures of follicular DHEAS decreased as the days of treatment with GnRH agonists increased (p=0.0003) and levels of follicular testosterone and DHEAS increased along with the days of treatment with rFSH (p=0.03, p=0.03). In regard to the outcomes of the overall cycle it was found a negative correlation between follicular testosterone levels and the number of punctured complexes (r= -0.360; p=0.002) and between follicular testosterone and the number of D quality embryos (r= -0.233; p=0.047). The number of mature oocytes was lower in patients with higher levels of follicular testosterone (p=0.008). Follicular progesterone was higher in good quality oocytes as compared to those of no remarkable quality (p=0.006) and very poor quality (p=0.04). Conclusions: High levels of follicular testosterone were correlated with a fewer number of punctured complexes, mature oocytes and D quality embryos. Good oocyte quality was associated with higher follicular progesterone levels.

Most studies investigating the impact of oral contraceptives have been performed some years ago, when the level of sexual hormones was greater than the actual formulations. Objective: The aim of this study was to evaluate the effects of current combined oral contraceptives (COC) on periodontal tissues, correlating the clinical parameters examined with the total duration of continuous oral contraceptive intake. Material and methods: Twenty-five women (19-35 years old) taking combined oral contraceptives for at least 1 year were included in the test group. The control group was composed by 25 patients at the same age range reporting no use of hormone-based contraceptive methods. Clinical parameters investigated included pocket probing depth (PD), clinical attachment level (CAL), sulcular bleeding index (SBI) and plaque index (Pl.I). Data were statistically evaluated by unpaired t test, Pearson’s correlation test and Spearman’s correlation test. Results: The test group showed increased PD (2.228±0.011 x 2.154±0.012; p<0.0001) and SBI (0.229±0.006 x 0.148±0.005, p<0.0001) than controls. No significant differences between groups were found in CAL (0.435±0.01 x 0.412±0.01; p=0.11). The control group showed greater Pl.I than the test group (0.206±0.007 x 0.303±0.008; p<0.0001). No correlation between the duration of oral contraceptive intake, age and periodontal parameters was observed. Conclusions: These findings suggest that the use of currently available combined oral contraceptives can influence the periodontal conditions of the patients, independently of the level of plaque accumulation or total duration of medication intake, resulting in increased gingival inflammation.

ABSTRACT

OBJETIVO: avaliar a concentração dos hormônios esteroides no fluido folicular (FF) de folículos pequenos (10-14 mm) e grandes (> 18 mm) de mulheres com síndrome dos ovários policísticos (SOP) submetidas à hiperestimulação ovariana controlada (HOC) e ciclos de fertilização in vitro (FIV). MÉTODOS: estudo caso-controle foi conduzido em 13 mulheres inférteis com SOP (17 ciclos) e 31 mulheres inférteis por fator masculino - Grupo Controle (31 ciclos). Os FF foram aspirados individualmente e dividos em 4 grupos: G1 (FF pequeno do Grupo Controle), G2 (FF pequeno do grupo SOP), G3 (FF grande do Grupo Controle) e G4 (FF grande do grupo SOP). A metodologia utilizada para as dosagens de estradiol, progesterona e β-hCG foi a quimioluminescência, e de testosterona e androstenediona o radioimunoensaio. Para a análise das dosagens hormonais no FF entre os grupos SOP e Controle utilizou-se o teste t não-pareado, e para a comparação entre os quatro grupos, o ANOVA. Para a taxa de gravidez, foi utilizado o teste exato de Fisher. RESULTADOS: os folículos pequenos dos dois grupos tiveram valores menores de progesterona (8.435±3.305 ng/mL) comparados aos grandes (10.280±3.475 ng/mL), com valor de p<0,01. Os níveis de progesterona de todos os folículos do grupo SOP (8.095±4.151 ng/mL) foram inferiores ao Controle (9.824±3.128 ng/mL), com valor de p=0,03. Os níveis de testosterona diferiram entre G1 (326,6±124,4 ng/dL) e G3 (205,8±98,91 ng/dL), com valor de p<0,001, e entre G3 (205,8±98,91 ng/dL) e G4 (351,10±122,1 ng/dL), com valor de p<0,001. Os folículos pequenos (508,9±266 ng/dL) apresentaram valores superiores de testosterona comparados aos grandes (245,10±123 ng/dL), com valor de p<0,0001. As taxas de gravidez não diferiram entre os grupos SOP (5/13, 38,5 por cento) e Controle (9/31, 40,9 por cento), com valor de p=072. CONCLUSÕES: mulheres com SOP apresentam altas concentrações de testosterona no FF, independentemente do estágio de desenvolvimento folicular, e níveis de progesterona diminuídos, sugerindo que fatores parácrinos podem inibir sua secreção pelas células foliculares. As taxas de gravidez mostraram que o tratamento de HOC e FIV é uma boa opção para mulheres com infertilidade secundária à SOP.


PURPOSE: to evaluate the concentration of steroid hormones in follicular fluid (FF) of small (10-14 mm) and large (> 18 mm) follicles of women with polycystic ovary syndrome (PCOS) submitted to controlled ovarian hyperstimulation (COH) and in vitro fertilization (IVF) cycles. METHODS: a case-control study was conducted on 13 infertile women with PCOS (17 cycles) and 31 infertile women due to male factor - Control Group (31 cycles). FF was aspirated individually and divided into four groups: G1 (FF of small follicles of the Control Group), G2 (FF of small follicles of the PCOS group), G3 (FF of large follicles of the Control Group) and G4 (FF of large follicles of the PCOS group). Estrogen, progesterone and β-hCG were determined by chemiluminescence, and testosterone and androstenedione by radioimmunoassay. The unpaired t-test was used to compare the hormone determinations in the FF of the PCOS and Control Groups, and the four groups were compared by ANOVA. Fisher's exact test was used to compare the pregnancy rates. RESULTS: the small follicles of the two groups had lower progesterone levels (8,435±3,305 ng/mL) than large follicles (10,280±3,475 ng/mL), p-value <0.01. The progesterone levels of all follicles of group PCOS (8,095±4,151 ng/mL) were lower than Control (9,824±3,128 ng/mL), p-value =0.03. Testosterone differed between G1 (326.6±124.4 ng/dL) and G3 (205.8±98.91 ng/dL), p-value <0.001, and between G3 (205.8±98.91 ng/dL) and G4 (351.10±122.1ng/dL), p-value <0.001. Small follicles had higher testosterone levels (508.9±266 ng/dL) than large follicles (245.10±123 ng/dL), p-value <0.0001. The pregnancy rates did not differ between the PCOS (5/13, 38.5 percent) and the Control groups (9/31, 40.9 percent), p-value =072. CONCLUSIONS: women with PCOS had high testosterone concentrations in the FF, regardless of the stage of follicle development, and reduced progesterone levels, suggesting that paracrine factors may inhibit the secretion of the latter by follicular cells. The pregnancy rates showed that treatment with COH and IVF is a good option for women with infertility secondary to PCOS.

ABSTRACT

El objetivo de la investigación fue comparar los efectos de la salpingectomía sobre la función hormonal y el flujo sanguíneo útero-ovárico. Se incluyeron 33 mujeres sanas con ciclos menstruales regulares. La salpingectomía fue realizada inmediatamente en los 5 días siguientes a la menstruación. Se tomaron muestras de sangre venosa para determinar las concentraciones de las hormonas folículo estimulante y luteinizante, estradiol y progesterona antes y luego de 3 meses de la cirugía. El flujo sanguíneo fue evaluado usando Doppler color y de pulso para medir el índice de resistencia, índice de pulsatilidad y velocidad sistólica pico en la vasculatura ovárica y las arterias uterinas. Se observó un aumento estadísticamente significativo en las concentraciones de hormonas folículo estimulante y luteinizante después de 3 meses de la salpingectomía (P<0,05). No se encontraron diferencias estadísticas significativas en las concentraciones de estradiol y progesterona (P=ns). Se observaron variaciones estadísticas significativas en los valores de velocidad sistólica pico, índice de resistencia e índice de pulsatilidad de la arteria ovárica al comparar los valores antes y 3 meses después de la salpingectomia (P<0,05). También se observó un aumento estadísticamente significativo en el volumen de los ovarios después de 3 meses de la cirugía (P<0,05). Se concluye que luego de 3 meses, la salpingectomía produce cambios en las concentraciones de gonadotropinas, tamaño de los ovarios y en los parámetros ecográficos Doppler de las arterias ováricas


The objetive of research was to compare the effects of salpingectomy over hormonal function and uterine and ovary blood flow. Thirty-three healthy women with normal menstrual cycles were included. Salpingectomy was performed immediately in five days following menstruation. A venous blood sample was taken to determine folicular stimulant and luteinizant hormones, estradiol and progesterone before and after three months of surgery. Bood flow was evaluated using color and pulse Doppler to measure resistance index, pulsatility index and peak systolic flow in ovary vasculare and uterine artery. There were statically significant increased in folicular stimulant and luteinizant hormones concentrations after three months of salpingectomy (P<0.05). There were not significant differences in estradiol and progesterone concentrations (P=ns). There were statistically significant variations in values of peak systolic flow, resistance index and pulsatility index of ovarian arteries when values before and three moths after salpingectomy were compared (P<0.05). There was also a statistically significant increase in ovary volume after three months of surgery (P=0.05). It is concluded that after three months, salpingectomy produces changes in gonadotropins concentrations, ovary size and ultrasound Doppler parameters of ovarian artery

ABSTRACT

Objetivo: establecer la correlación entre los niveles séricos de grelina y los hallados en fluido folicular, el IMC, y los niveles séricos de estradiol y progesterona. Establecer la asociación entre los niveles plasmáticos de grelina y los resultados reproductivos en cueanto a tasa de embarazo. Diseño: estudio prospectivo de cohortes. Materiales y métodos: se incluyeron para el análisis los procedimientos de alta complejidad realizados entre junio y diciembre de 2007 (n=492). Todas las pacientes tuvieron un IMC menor de 26 kg/m2, ritmos menstruales conservados, FSH basal menor de 12 mUI/ml y edad menor de 40 años. Se excluyeron los ciclos con síndrome de ovario poliquístico y/o insulinorresistencia, antecedente de falla previa de fertilización in vitro o ciclos con muestras de espermatozoides provenientes de biopsia de testículo. El grupo de estudio (Grupo A) fue definido por aquellas pacientes con un IMC menor de 20 kg/m2 y se logró incluir un total de 19 ciclos. El grupo control (Grupo B) quedó definido por ciclos con IMC entre 20-25 kg/m2 , los ciclos del grupo B fueron seleccionados de los realizados en el mismo día del grupo A en una relación !:!. De una muestra tomada en el día de la punción ovárica, se realizó el dosaje sérico de grelina, estradiol y progesterona; se utilizó el fluido folicular del primer folículo aspirado libre de medio de lavado. Se evaluó causa de esterilidad, FSH basal, IMC, ovocitos captados, ovocitos metafase II (M II), ovocitos fertilizados, calidad embrionaria, tasas de implantación y embarazo. Resultado: se halló una correlación positiva entre los niveles séricos de grelina y fluido folicular (r=0,72m p<0,05), así como una correlación indirecta significativa entre los niveles de grelina y estradiol y progesterona (r=0,44 y r=0,43 respectivamente, ambos con p<0,05). ABSTRACT

Various evidences show a higher prevalence of temporomandibular disorder [TMD] in women. The purpose of this study was to evaluate T.M.D etiologic factors in women referring to Mashhad Dental School. Twenty-five patients with jaw clicking and 19 with M.P.D syndrome were randomly selected among female patients referring to Mashhad dental school. Seventeen women with no signs and symptoms of temporomandibular disorder were used as the control group. The women were between the ages of 18 to 40. All of them were examined for TMD and etiologic factors such as Parafunctional habits [Bruxism and Clenching] and premature contacts in eccentric movements. The level of blood estradiol and progesterone status were also recorded. The data were analyzed by Kruskal Wallis and Chi-square tests through SPSS software. Although there was a difference in level of blood estradiol and progesterone between TMD patients and control group, this difference was not significant. There was a significant difference between the two groups regarding parafunctional habits [P<0.001]. There was no significant difference between the two groups regarding eccentric premature contacts [P=0.098] In this study, the most important TMD etiologic factor was parafunctional habits [Bruxism and clenching] and the second effective factor was eccentric premature contacts. There was no relation between the level of blood estradiol and progesterone and the incidence of TMD in women

OBJETIVO: examinar a hipótese de que o nível sérico do hormônio anti-Mülleriano (HAM) reflete o status folicular ovariano. MÉTODOS: Desenho: estudo prospectivo. Pacientes: foram incluídas 101 candidatas à FIV-TE submetidas à estimulação ovariana controlada com agonista de GnRH e FSH. Depois de atingir a supressão da hipófise e antes da administração de FSH (dia basal), os níveis séricos de HAM, inibina B e FSH foram avaliados. O número de folículos antrais foi determinado pela ultra-sonografia (dia basal) (folículo antral precoce; 3-10 mm). RESULTADOS: as médias do nível sérico de HAM, inhibina B, E2, P4 e FSH (dia basal) foram 3,4±0,14 ng/mL, 89±4,8 pg/mL, 34±2,7 pg/mL, 0,22±0,23 ng/mL e 6,6±0,1 mUI/mL, respectivamente, e a média do número de folículos antrais precoces foi 17±0,39. O nível sérico do HAM foi negativamente correlacionado com a idade (r= -0,19, p<0,04) e positivamente correlacionado com o número de folículos antrais precoces (r=0,65, p<0,0001), mas isto não se aplicou aos níveis séricos de inibina B, E2 e FSH. CONCLUSÕES: esse dado demonstra a associação do HAM com a quantidade de folículo antral, sendo aquele, portanto, um provável biomarcador do status folicular ovariano.


PURPOSE: to examine the hypothesis that serum anti-Müllerian hormone (AMH) levels reflect the ovarian follicular status. METHODS: Design: prospective study. Patients: we studied 101 IVF-ET candidates undergoing controlled ovarian hyperstimulation with GnRH agonist and FSH. After the achievement of pituitary suppression and before FSH administration (baseline), serum AMH, inhibin B, and FSH levels were measured. The number of antral follicles was determined by ultrasound at baseline (early antral follicles; 3-10 mm). RESULTS: at baseline, median serum levels of AMH, inhibin B, E2, P4 and FSH were 3.42±0.14 ng/mL, 89±4.8 pg/mL, 34±2.7 pg/mL, 0.22±0.23 ng/mL and 6.6±0.1 mIU/mL, respectively, and the mean number of early antral follicles was 17±0.39. Serum levels of AMH were negatively correlated with age (r=-0.19, p<0.04), and positively correlated with number of antral follicles (r=0.65, p<0.0001), but this did not apply to serum levels of either inhibin B, E2 or FSH. CONCLUSION: the data demonstrate an association between AMH and antral follicular counts. Therefore, AMH is probable a biomarker of ovarian follicular status.

ABSTRACT

Estradiol, the most important hormone in females, was complexed with hydroxypropyl-beta-cyclodextrin (HP-beta-CD) and randomly methylated beta-cyclodextrin (RM-b-CD). After obtaining the inclusion complexes, they were characterized by DSC, ¹H-NMR and solubility studies. The enhancing effect of CDs on estradiol skin permeation/retention was investigated in vitro using Franz diffusion cells. Both CDs increased estradiol aqueous solubility, but in different proportions. DSC and NMR-H¹ analyses showed that estradiol was complexed with both CDs and RM-beta-CD has stronger interactions with the hormone than HP-beta-CD. Moreover, complexes formation increased estradiol flux through the skin (P<0.01), but the hormone retention in the stratum corneum (SC) only increased after complexation with HP-beta-CD. On the other hand, only RM-b-CD was able to modify estradiol retention in the SC after skin pretreatment with CDs. The results suggest that the enhancing effect of CDs on estradiol flux through the skin may be mainly described as an increase in drug availability on the skin surface due to inclusion complexation. Furthermore, the formation of a significant reservoir of estradiol in the SC due to HP-beta-CD complexation makes it an interesting approach for estradiol transdermal delivery.


Estradiol, o mais importante hormônio feminino, foi complexado com hidroxipropil-beta-ciclodextrina (HP-beta-CD) e metil-beta-ciclodextrina randômica (RM-beta-CD). Após a obtenção dos complexos, os mesmos foram caracterizados por estudos de solubilidade, CDV e RMN-H¹. O efeito das CDs sobre a absorção/retenção do estradiol na pele foi investigado in vitro em células de difusão de Franz. As CDs aumentaram a solubilidade aquosa do estradiol em diferentes proporções. As análises de CVD e RMN-H¹ comprovaram a complexação do estradiol com ambas CDs, sendo que RM-beta-CD apresentou interação mais forte com estradiol do que HP-beta-CD. Mais adiante, a formação de complexos aumentou o fluxo de estradiol através da pele (P<0,01), mas o aumento de fármaco no estrato córneo (EC) foi observado somente após complexação com HP-beta-CD. Por outro lado, somente a RM-beta-CD foi capaz de aumentar a retenção do fármaco no EC após o pré-tratamento da pele com CDs. Os resultados sugerem que o efeito promotor das CDs no fluxo de estradiol pode ser descrito principalmente como aumento da disponibilidade de fármaco na superfície da pele devido à complexação. Além disso, a formação de um significativo reservatório do hormônio no EC torna a complexação com HP-beta-CD uma estratégia interessante para liberação transdérmica do estradiol.

ABSTRACT

Avaliaram-se as concentrações hormonais e os parâmetros de desenvolvimento folicular de vacas leiteiras expostas ao calor sazonal e agudo. Dividiram-se os animais em quatro grupos: verão (n=5), outono (n=5), inverno com hipertermia aguda (grupo câmara climática, (CC), n=5) e inverno (n=9). Os animais foram abatidos no sétimo dia após a ovulação, e os parâmetros de desenvolvimento folicular avaliados. O líquido folicular do maior folículo foi aspirado e armazenado para posterior análise de hormônios esteróides e inibina. O número de células da granulosa vivas no verão e no outono foi 40 e 45 por cento respectivamente, menor que no inverno (P<0,05). A concentração de estradiol (E2) no inverno foi 62 por cento maior que no outono (P<0,05) e 34 por cento superior ao grupo verão (P<0,06). Houve um aumento na quantidade de androstenediona no verão em relação aos grupos inverno (P<0,08) e outono (P<0,05). A concentração de inibina foi maior no inverno do que no verão e CC (P<0,05). A exposição ao calor sazonal e agudo modificou os parâmetros de desenvolvimento do folículo e as concentrações hormonais no líquido folicular, podendo explicar em parte a queda nas taxas de concepção no verão.


The present study evaluated the seasonal and acute heat stress on follicular development and steroid and inhibin concentrations in follicular fluid, in bovine dominant follicle. Cows were distributed into four treatments: summer (n=5), autumn (n=5), animals heat stressed during the winter (n=5) and winter (n=9). On day 7 of the estrous cycle, animals were slaughtered and parameters related to follicle development were evaluated. The follicular fluid (FF) was aspirated and stored for further hormonal analysis. During the summer, the number of viable granulosa cells was 40 percent lower than during the winter, and there was a 45 percent decrease in this parameter during the autumn (P<0.05). In the winter, estradiol concentration was 62 percent higher than during the autumn (P<0.05) and 42 percent higher than during the summer (P<0.06). There was an increase in androstenedione concentration in summer group, when compared to winter (P<0.08) and autumn (P<0.05) groups. Inhibin concentration was higher in winter groups than summer and winter heat stressed groups (P<0.05). Seasonal and acute heat stress altered developmental parameters in dominant follicle and hormonal concentration in follicular fluid, those effects can partially explain the decrease in conception rates during summer.

ABSTRACT

The present study was carried out to assess the possibility of measuring fecal steroid hormone metabolites as a noninvasive technique for monitoring reproductive function in the three-toed sloth, Bradypus variegatus. Levels of the estradiol (E2) and progesterone (P4) metabolites were measured by radioimmunoassay in fecal samples collected over 12 weeks from 4 captive female B. variegatus sloths. The validation of the radioimmunoassay for evaluation of fecal steroid metabolites was carried out by collecting 10 blood samples on the same day as defecation. There was a significant direct correlation between the plasma and fecal E2 and P4 levels (P < 0.05, Pearson's test), thereby validating this noninvasive technique for the study of the estrous cycle in these animals. Ovulation was detected in two sloths (SL03 and SL04) whose E2 levels reached 2237.43 and 6713.26 pg/g wet feces weight, respectively, for over four weeks, followed by an increase in P4 metabolites reaching 33.54 and 3242.68 ng/g wet feces weight, respectively. Interestingly, SL04, which presented higher levels of E2 and P4 metabolites, later gave birth to a healthy baby sloth. The results obtained indicate that this is a reliable technique for recording gonadal steroid secretion and thereby reproduction in sloths.
