ABSTRACT

BACKGROUND: Salep is obtained by grinding dried orchid tubers and used as a valuable ingredient in the food industry. Because of the glucomannan content of salep, it is thought to have prebiotic potential. However, there is little information in studies concerning the fermentation characteristics and potential prebiotic properties of salep. The objective of this study was to investigate the effect of salep on bifidobacterial growth by measuring the highest optical density (OD), calculating the specific growth rates, and determining the production of lactic acid and short-chain fatty acids (acetic, propionic, and butyric acid) as a result of bacterial fermentation. RESULT: The OD and pH values obtained in this study showed that salep was utilized as a source of assimilable carbon and energy by the Bifidobacterium species (BS). All Bifidobacterium strains produced lactic, acetic, propionic, and butyric acid, indicating that salep is readily fermented by these bacteria. Salep at 1% (w/v) showed a similar effect on bifidobacterial growth as that promoted by 1% (w/v) glucose used as a traditional carbon source. CONCLUSIONS: Bifidobacterium species can develop in media containing salep as well as in glucose and exhibit the potential to be used as new sources of prebiotics.

ABSTRACT

Nowdays it is established that ischemic brain damage like ischemic stroke is one of the leading cause of death and disability in the population that assumes relevance development of anti-ischemic drugs. The work studied the anti-hypoxic and anti-ischemic effect of 7 plant extracts. Antihypoxic activity was assessed on models of hypobaric, hypercapnic, histotoxic, hematotoxic hypoxia. Anti-ischemic activity of test-extracts was studied on the focal cerebral ischemia model. Administration of Tagetes patula, Gaillardia pulchella, Sorbaria sorbifolia, Grossularia reclinata, Ribes nigrum, Rubus caesius and Lysimachia punctata extracts contributed to the necrosis zone reduction by 56.6% (p<0.05); 37.3% (p<0.05); 73.2% (p<0.05); 49.4% (p<0.05); 42.5% (p<0.05); 85.5% (p<0.05); 44.2% (p<0.05) and also restored aerobic metabolism in brain tissue. Test - objects increased of the animal lifespan under hypoxia conditions. Based on the data obtained, it is assumed that further studies of North Caucasus flora plant extracts as cerebro-protective agents are promising.


Hoy en día, se ha establecido que el daño cerebral isquémico, como el accidente cerebrovascular isquémico, es una de las principales causas de muerte y discapacidad en la población lo cual hace relevante el desarrollo fármacos antiisquémicos. En este trabajo se estudió el efecto antihipóxico y antiisquémico de siete extractos de plantas. La actividad antihipóxica se evaluó en modelos de hipoxia hipocrática, hipercápnica, histotóxica y hematotóxica. La actividad antiisquémica de los extractos de prueba se estudió en el modelo de isquemia cerebral focal. La administración de los extractos de Tagetes patula; Gaillardia pulchella; Sorbaria sorbifolia; Grossularia reclinata; Ribes nigrum; Rubus caesius y Lysimachia punctata contribuyeron a la reducción de la zona de necrosis en un 56,6% (p<0,05); 37,3% (p<0,05); 73,2% (p<0,05); 49,4% (p<0,05); 42,5% (p<0,05); 85,5% (p<0,05); 44.2% (p<0.05), respectivamente, además, de restaurar el metabolismo aeróbico en el tejido cerebral. Comparado con el control, se observó un aumento en el tiempo de sobrevida del animal en condiciones de hipoxia. Sobre la base de los interesantes datos obtenidos, se sugiere estudios adicionales de extractos de plantas de la flora del Cáucaso Norte como agentes protectores del cerebro.

ABSTRACT

Background: Plastic waste is a serious problem because it is difficult to degrade, thereby leading to global environment problems. Poly(lactic acid) (PLA) is a biodegradable aliphatic polyester derived from renewable resources, and it can be degraded by various enzymes produced by microorganisms. This study focused on the scale-up and evaluated the bioprocess of PLA degradation by a crude microbial enzyme produced by Actinomadura keratinilytica strain T16-1 in a 5 L stirred tank bioreactor. Results: PLA degradation after 72 h in a 5 L bioreactor by using the enzyme of the strain T16-1 under controlled pH conditions resulted in lactic acid titers (mg/L) of 16,651 mg/L and a conversion efficiency of 89% at a controlled pH of 8.0. However, the PLA degradation process inadvertently produced lactic acid as a potential inhibitor, as shown in our experiments at various concentrations of lactic acid. Therefore, the dialysis method was performed to reduce the concentration of lactic acid. The experiment with a dialysis bag achieved PLA degradation by weight loss of 99.93%, whereas the one without dialysis achieved a degradation of less than approximately 14.75%. Therefore, the dialysis method was applied to degrade a commercial PLA material (tray) with a conversion efficiency of 32%, which was 6-fold more than that without dialysis. Conclusions: This is the first report demonstrating the scale-up of PLA degradation in a 5 L bioreactor and evaluating a potential method for enhancing PLA degradation efficiency.

ABSTRACT

This study was aimed to determine and compare the chemical and microbiological properties of yoghurts made from different types of milk and their mixtures (35%, 65%, and 100%) during their storage at 4 °C for 28 days. For this purpose, chemical and microbiological properties of yoghurts during storage at 4 °C for 28 days were investigated. The total amount of dry matter, fat, pH and protein of yoghurt made from the buffalo and cow milk mixtures was significantly higher than that of pure buffalo milk (P<0.01). Also, storage time has led to significant differences in these components. Considering the results of microbiological analysis, a significant (P<0.01) difference was found between yoghurt samples in terms of total count of mesophilic aerobic bacteria, lactobacilli, lactococcus and yeast and mould. Hence, it is concluded that the addition of buffalo milk to that of cow improves the composition of yoghurt made from cow milk, which indicated the possibilities of processing and marketing of both types of milk especially because the health benefits of cow milk and the fermented products are well documented(AU)


El objetivo de este estudio fue determinar y comparar las propiedades químicas y microbiológicas de los yogures hechos con diferentes tipos de leche y sus mezclas (35%, 65% y 100%) durante su almacenamiento a 4° C por 28 días. La cantidad total de materia seca, grasa, pH y proteínas del yogur hecho con las mezclas de leche de búfala y vaca fue significativamente mayor que la de la leche de búfala pura (P <0.01). Además, el tiempo de almacenamiento generó diferencias significativas en estos componentes. De acuerdo con los resultados del análisis microbiológico, se encontró una diferencia significativa (P<0.01) entre las muestras de yogur en términos de bacterias mesófilas aerobias totales, lactobacilos, lactococcus y recuentos totales de levadura y mohos. Por lo tanto, se concluye que la adición de leche de búfala a la de vaca mejora la composición del yogur hecho de leche de vaca, lo que indica las posibilidades de procesamiento y comercialización de ambos tipos de leche, especialmente porque los beneficios para la salud de la leche de vaca y de los productos fermentados están bien documentados(AU)

ABSTRACT

Lactate modulates the expression of lactate oxidation complex (LOC)-related genes and cardiac blood flow under physiological conditions, but its modulatory role remains to be elucidated regarding pathological cardiac stress. The present study evaluated the effect of lactate on LOC-related genes expression and hemodynamics of hearts submitted to myocardial infarction (MI). Four weeks after MI or sham operation, isolated hearts of male Wistar rats were perfused for 60 min with Na+-lactate (20 mM). As expected, MI reduced cardiac contractility and relaxation with no changes in perfusion. The impaired cardiac hemodynamics were associated with increased reactive oxygen species (ROS) levels (Sham: 19.3±0.5 vs MI: 23.8±0.3 µM), NADPH oxidase (NOX) activity (Sham: 42.2±1.3 vs MI: 60.5±1.5 nmol·h−1·mg−1) and monocarboxylate transporter 1 (mct1) mRNA levels (Sham: 1.0±0.06 vs MI: 1.7±0.2 a.u.), but no changes in superoxide dismutase (SOD), catalase, NADH oxidase (NADox), and xanthine oxidase activities. Lactate perfusion in MI hearts had no additional effect on ROS levels, NADox, and NOX activity, however, it partially reduced mct1 mRNA expression (MI-Lactate 1.3±0.08 a.u.). Interestingly, lactate significantly decreased SOD (MI-Lactate: 54.5±4.2 µmol·mg−1·min−1) and catalase (MI: 1.1±0.1 nmol·mg−1·min−1) activities in MI. Collectively, our data suggest that under pathological stress, lactate lacks its ability to modulate the expression of cardiac LOC-related genes and the perfused pressure in hearts submitted to chronic MI. Together, these data contribute to elucidate the mechanisms involved in the pathogenesis of heart failure induced by MI.

Abstract AIM The aim of this study was to investigate the lactate response in physically inactive hypertensive women submitted to the treadmill maximal lactate steady state (MLSS) protocol. METHODS Twenty-two hypertensive women (40 - 64 years) performed a familiarization period of walking on the treadmill following by one incremental test for estimating the initial workload for exercise testing. MLSS protocol was composed by walking in a treadmill during thirty minutes with fixed velocity in 5.5 km/h. Incline was used for determination of the intensity of each volunteer. Blood samples were collected from the ear lobe in the rest period, minute 10th and at the end of the test (minute 30th or at exhaustion time point) for lactate analysis. RESULTS Hypertensive women showed a lower lactate concentration at MLSS (3.25 ± 0.81 mmol/L) as compared with data obtained in the literature (4 mmol/L), approximately 18.8%. Neither inclines nor age affected MLSS parameters in the population. A positive and strong correlation was found between incline and MLSS, as well as incline and lactate level at minute 30th, even when adjusted by age factor. CONCLUSION Physically inactive hypertensive women show a lower MLSS than the average established in the literature but within the range of variations previously reported. Furthermore, a higher MLSS incline correlates positive and directly with higher lactate concentrations for the same aerobic capacity regardless of age.

RESUMO Objetivo: Avaliar o Escore para Meningite Bacteriana (EMB) isolado e associado ao valor do lactato no líquor para diferenciar meningite bacteriana (MB) e meningite asséptica (MA). Métodos: Foram selecionadas crianças com meningite atendidas em hospital terciário privado entre janeiro de 2011 e dezembro de 2014. Os dados foram obtidos na admissão. Utilizou-se o EMB com: coloração de Gram no líquor (2 pontos); neutrófilos no líquor ≥1.000 células/mm3 (1 ponto); proteína no líquor ≥80 mg/dL (1 ponto); neutrófilos no sangue periférico ≥10.000 células/mm3 (1 ponto); e convulsão durante/antes da chegada (1 ponto). Analisou-se também o lactato no líquor (elevado: ≥30 mg/dL). Avaliaram-se sensibilidade, especificidade e valor preditivo negativo de diversos valores de corte do EMB e do EMB associado ao lactato elevado para prever MB. Resultados: Dos 439 pacientes elegíveis, 94 não tinham todos os dados necessários para o escore, sendo 345 pacientes selecionados: 7 no grupo de MB e 338 no de MA. Como preditivos de MB, o EMB ≥1 mostrou sensibilidade de 100% (intervalo de confiança de 95% - IC95% 47,3-100), especificidade de 64,2% (58,8-100) e valor preditivo negativo de 100% (97,5-100), enquanto o EMB ≥2 ou EMB ≥1 associado a lactato liquórico ≥30 mg/dL mostrou sensibilidade de 100% (47,3-100), especificidade de 98,5% (96,6-99,5) e valor preditivo negativo de 100% (98,3-100). Conclusões: O EMB com 2 pontos associado à dosagem de lactato no líquor manteve a sensibilidade e o valor preditivo negativo, ao passo que aumentou a especificidade para identificar meningites bacterianas em relação à utilização do EMB com 1 ponto.


ABSTRACT Objective: To evaluate Bacterial Meningitis Score (BMS) on its own and in association with Cerebrospinal Fluid (CSF) lactate dosage in order to distinguish bacterial from aseptic meningitis. Methods: Children diagnosed with meningitis at a tertiary hospital between January/2011 and December/2014 were selected. All data were obtained upon admission. BMS was applied and included: CSF Gram staining (2 points); CSF neutrophil count ≥1,000 cells/mm3 (1 point); CSF protein ≥80 mg/dL (1 point); peripheral blood neutrophil count ≥10,000 cells/mm3 (1 point) and seizures upon/before arrival (1 point). Cutoff value for CSF lactate was ≥30 mg/dL. Sensitivity, specificity and negative predictive value of several BMS cutoffs and BMS associated with high CSF lactate were evaluated for prediction of bacterial meningitis. Results: Among 439 eligible patients, 94 did not have all data available to complete the score, and 345 patients were included: 7 in bacterial meningitis group and 338 in aseptic meningitis group. As predictive factors of bacterial meningitis, BMS ≥1 had 100% sensitivity (95%CI 47.3-100), 64.2% specificity (58.8-100) and 100% negative predictive value (97.5-100); BMS ≥2 or BMS ≥1 associated with high CSF lactate also showed 100% sensitivity (47.3-100); but 98.5% specificity (96.6-99.5) and 100% negative predictive value (98.3-100). Conclusions: 2 point BMS in association with CSF lactate dosage had the same sensitivity and negative predictive value, with increased specificity for diagnosis of bacterial meningitis when compared with 1-point BMS.

ABSTRACT

The aim of the current study was to investigate biogenic amines and mycotoxins concentrations in baled silage (mainly Poaceae family grasses) prepared in organic and conventional farms and to relate these parameters to fermentative parameters. The mean dry matter (DM) content was 364.10±93.31 and 424.70±95.93g/kg in the silage from organic and conventional farms respectively. The silage samples from organic farms had 17.00% higher (P≤ 0.05) tyramine (TY) than the silage from conventional farms. Conventional farm samples were characterized by 46.00% higher histamine (HIS) (P≤ 0.05), 9.80% higher putrescine (PUT) (P≤ 0.05), 17.30% higher cadaverine (CAD) (P≤ 0.05). Aflatoxins (AFL) (total) and zearalenone (ZEN), T-2/HT-2 concentrations were higher respectively 16.00% (P≤ 0.05) and 13.40% (P≤ 0.05), 1.80% (P≤ 0.05) in the silage prepared in organic farms. Deoxynivalenol (DON) concentration was higher 42.40% (P≤ 0.05) in silage from conventional farms. Volatile fatty acids (VFA), lactic acid, ethanol, pH and ammonia nitrogen showed that the silage samples from organic and conventional farms were of good quality. Our study suggests differences in biogenic amine formation or mycotoxins content in silage from organic and conventional farming, but, overall, the measured values are too low to be relevant for animal health. Furthermore, these differences might as well be due to the difference in dry matter content and plant maturity between the organic and conventional silage samples.(AU)


O objetivo do presente estudo foi investigar concentrações de aminas biogênicas micotoxinas em silagem embalada (principalmente gramíneas da família Poaceae) preparada em fazendas orgânicas e convencionais e relacionar esses parâmetros a parâmetros fermentativos. A massa seca média (MS) foi 364,10±93,31 e 424,70±95,93g/kg na silagem de fazendas orgânicas e convencionais, respectivamente. As amostras de silagem de fazendas orgânicas tinham 17% a mais de tyramina (TY) (p≤ 0,05) que as de fazendas convencionais. As amostras de fazendas convencionais foram caracterizadas por histamina (HIS) 46,00% mais alta (P≤ 0,05), 9,80% putrecina (PUT) mais alta (P≤ 0,05), 17,30% de cadaverina (CAD) mais alta (P≤ 0,05). Aflatoxnas (AFL) (total) e zearalenone (ZEN), T-2/HT-2 tinham concentrações mais altas em respectivamente 16,00% (P≤ 0,05) e 13,40% (P≤ 0,05), 1,80% (P≤ 0,05) na silagem preparada em fazendas orgânicas. Deoxinivalenol (DON) tinha concentração mais alta 42,40% (P≤ 0,05) na silagem de fazendas convencionais. Ácidos graxos voláteis (AGV), ácido lático, etanol, pH e nitrogênio de amônia mostraram que as amostras de silagem de fazendas orgânicas e convencionais tinham boa qualidade. Nosso estudo sugere diferenças na formação biogênica de amônia ou micotoxinas em silagem de fazendas orgânicas ou convencionais mas, em geral, os valores medidos foram muito baixos para serem relevantes à saúde animal. Ademais, essas diferenças podem ser devido à diferença na matéria sólida e maturidade da planta entre as amostras de silagem orgânica e convencional.(AU)

Abstract AIMS This study was designed to investigate the effects of 14 water environment adaptation days on motor behavior and physiological condition of swimming rats. METHODS Sixty male Wistar rats were divided into four groups-baseline (Bl) and control (Co) groups-which did not perform the water environment adaptation; and sub (SubAnT) and (SupraAnT) anaerobic threshold groups, which performed 14 water environment adaptation days with sub or supra anaerobic threshold progressive loads (from the tenth day), respectively. The climbing-swimming prevalence (i.e. motor behavior) was analyzed during the water environment adaptation days. Lactate minimum test (LMT) parameters and muscular/hepatic glycogen content in addition to serum creatine kinase were also measured. RESULTS Animals from SubAnT and SupraAnT groups presented a lower climbing-swimming pattern throughout the extent of the experiment (p=0.000), especially after the 5th session. These results were achieved without an improvement in the LMT results or glycogen/creatine kinase. In addition, improvements of 26.6% and 25% for the LMT success rate (i.e. LMT reliability) were obtained only for SubAnT and SupraAnT animals. CONCLUSION Overall, we demonstrated that a water environment adaptation period is necessary for lowering the climbing-swimming pattern without physiological improvement.(AU)

ABSTRACT

Fundamentos: A insuficiência cardíaca é uma doença multissistêmica que inclui disfunção autonômica. Objetivo: Avaliar os efeitos agudos da Estimulação Elétrica Funcional (EEF) e do Treinamento Muscular Inspiratório (TMI) sobre o controle autonômico, a função endotelial e os níveis de citocinas inflamatórias em pacientes portadores de IC. Métodos: Estudo randomizado cruzado que incluiu 12 pacientes submetidos a três intervenções randomizadas: EEF, TMI, e EEF + TMI, com intervalo de 1 semana entre as sessões. O TMI foi realizado durante 15 minutos, com 30% da pressão inspiratória máxima. A EEF foi realizada nos músculos vasto lateral e vasto medial, a uma frequência de 20Hz durante 30 minutos. O controle autonômico foi medido através de monitorização de pressão batimento por batimento (Finapres); a função endotelial, através da técnica de dilatação mediada por fluxo (DMF); e os níveis de citocinas inflamatórias foram medidos antes e depois de cada sessão. Resultados: O controle autonômico após EEF diminuiu em termos de BF/AF (p=0,01) e BFn.u (p=0,03), e aumentou em termos de RR médio (p=0,005). Observou-se um aumento do RR médio após o TMI (p=0,005) e após EEF+TMI (p=0,02). Não houve diferenças na DMF e na concentração de lactato sérico. Quanto às citocinas, a EEF promoveu uma redução nos níveis de TNF-α (pré versus pós 24 horas, p = 0,05). O TMI resultou em níveis aumentados de IL-10 (pré versus 24 horas pós, p=0,05) e em níveis diminuídos de TNF-α (1 hora pós versus 24 horas pós, p = 0,03). Não houve diferenças quando as duas intervenções foram associadas. Conclusão: EEF, TMI, e EEF + TMI alteraram o controle autonômico, mas não a função endotelial. A EEF e o TMI isoladamente alteraram os níveis de citocinas inflamatórias. Ensaios Clínicos: NCT01325597


Background: Heart Failure is a multisystem disorder, which includes autonomic dysfunction. Objective: To evaluate the acute effects of Functional Electrical Stimulation (FES) and Inspiratory Muscle Training (IMT) on autonomic control, endothelial function and inflammatory cytokine levels in patients with HF. Methods: Randomized crossover trial including 12 patients undergoing three randomized interventions: FES, IMT, and FES+IMT, with a 1-week interval between sessions. IMT was performed for 15 minutes with 30% of the maximal inspiratory pressure. FES was performed in the vastus lateralis and vastus medialis muscles, at 20Hz for 30 minutes. The autonomic control was measured using beat-to-beat blood pressure monitoring (Finapres); the endothelial function, using the flow-mediated dilation technique (FMD); and inflammatory cytokine levels were assessed before and after the sessions. Results: Autonomic control after FES decreased regarding LF/HF (p=0.01) and LFn.u (p=0.03), and increased regarding mean RR (p=0.005). Increased mean RR was observed after IMT (p=0.005) and after FES+IMT (p=0.02). No differences were found in FMD and blood lactate concentration. As regards the cytokines, FES led to a decrease in TNF-α levels (pre vs. 24 hours post, p = 0.05). IMT resulted in increased IL-10 levels (pre vs. 24 hours post, p=0.05) and decreased TNF-α levels (1 hour post vs. 24 hours post, p = 0.03). No difference was observed when the two interventions were associated. Conclusion: FES, IMT, and FES+IMT changed the autonomic control without changing the endothelial function. ABSTRACT

This study evaluated the effects of varying amounts of melon with high sugar content offered to sheep without prior melon experience and that were not adapted to consuming it. We used 12 eight-month-old, rumen-cannulated crossbred sheep weighing 25 kg each. The animals received a base diet of roughage, and then half were randomly selected to have 25% of their diet replaced with melon (G25%) and the other half had 75% of their diet replaced with melon (75%). Ruminal fluid was collected before administration of melon and at 0, 3, 6, 12, 18, and 24 h after the administration of the fruit. Sheep from the G25% group presented volatile fatty acid ruminal acidosis (sub-acute) between 3 and 6 h after consumption. This acidosis was characterized by a rumen pH slightly lower than 5.6, increased discrete L-lactic acid content, and increased redox potential (RP) and methylene blue redox (MBR) time of the ruminal fluid. The G75% group presented lactic ruminal acidosis at T6h, characterized by a rumen pH lower than 5.0, high lactate-L content, increased RP and MBR time, and increased ruminal fluid osmolarity. Therefore, offering large amounts of melon (75% of dry matter (DM)) is not recommended but 25% of DM of this fruit can be used safely.


O presente estudo avaliou o efeito da adição à dieta de duas diferentes quantidades de melão com alto teor de açucares oferecidos subitamente à ovinos não adaptados. Foram utilizados 12 ovinos mestiços da raça Santa Inês, com 8 meses de idade e pesando cerca der 25 kg providos de cânula ruminal. Os animais recebiam dieta à base de volumoso e foram aleatoriamente divididos em dois grupos iguais que receberam 25 ou 75% de adição de melão picado à dieta. Foram coletadas amostras de conteúdo ruminal nos momentos zero e após 3, 6, 12, 18, e 24 horas da administração do melão. Ovinos do grupo G25% apresentaram acidose por ácido graxo voláteis (subaguda) entre 3 e 6 horas, caracterizada por pH ruminal ligeiramente abaixo de 5,6, aumento discreto na concentração de ácido láctico L e aumento do potencial redox (PR) e tempo de redução do azul de metileno (TRAM) do fluido ruminal. O grupo G75% apresentou acidose láctica ruminal com pH menor que 5,0, alta concentração de Lactato-L e aumento do PR e TRAM e da osmolaridade ruminal. Em conclusão, o oferecimento de grandes quantidades de melão (75% da MS) não é recomendado, mas 25% da MS desta fruta pode ser utilizada na dieta de ovinos com segurança.

ABSTRACT

Lactic acid bacteria species were molecularly identified in milk from Lacaune, Santa Inês and crossbred sheep breeds and their in vitro probiotic potential was evaluated. The species identified were Enterococcus faecium (56.25%), E. durans (31.25%) and E. casseliflavus (12.5%). No other lactic acid bacteria species, such as lactobacilli, was identified. Most of the isolated enterococci were resistant to gastric pH (2.0) and to 0.3% oxgall. All tested enterococci were resistant to ceftazidime, oxacillin and streptomycin and sensible to clindamycin, erythromycin and penicillin. The resistance to ciprofloxacin, gentamicin, tetracycline and vancomycin varied among tested species. All tested enterococci strongly inhibited (P<0.05) Escherichia coli and Listeria monocytogenes, moderately inhibited E. faecalis and Staphylococcus aureus and did not inhibit Pseudomonas aeruginosa, Salmonella enterica var. Typhimurium and also one E. durans sample isolated from sheep milk. Four samples of E. faecium, one of E. durans and one of E. casseliflavus presented the best probiotic potential...


Espécies de bactérias ácido-lácticas foram identificadas em nível molecular em leite das raças ovinas Lacaune, Santa Inês e suas mestiças, e o seu potencial probiótico in vitro foi avaliado. As espécies identificadas foram Enterococcus faecium (56,25%), E. durans (31,25%) e E. casseliflavus (12,5%). Nenhuma outra espécie de bactéria ácido-láctica, como Lactobacillus sp., foi identificada. A maioria dos enterococos isolados foi resistente ao pH gástrico (2.0) e a 0,3% de oxgall. Todos os enterococos testados foram resistentes à ceftazidima, oxacilina e estreptomicina e sensíveis à clindamicina, eritromicina e penicilina. A resistência à ciprofloxacina, gentamicina, tetraciclina e vancomicina variou entre as amostras. Todos os enterococos testados inibiram fortemente (P<0,05) Escherichia coli e Listeria monocytogenes, inibiram moderadamente E. faecalis e Staphylococcus aureus e não inibiram Pseudomonas aeruginosa, Salmonella enterica var. ABSTRACT

The anaerobic threshold is a physiologic event studied in various species. There are various methods for its assessment, recognized in the human and equine exercise physiology literature, several of these involving the relationship between blood lactate concentration (LAC) and exercise load, measured in a standardized exercise test. The aim of this study was to compare four of these methods: V2, V4, individual anaerobic threshold (IAT) and lactate minimum speed (LMS) with the method recognized as the gold standard for the assessment of anaerobic threshold, maximal lactate steady-state (MLSS). The five tests were carried out in thirteen trained Arabian horses, in which velocities and associated LAC could be measured. The mean velocities and the LAC associated with the anaerobic threshold for the five methods were respectively: V2 = 9.67±0.54; V4 = 10.98±0.47; V IAT = 9.81±0.72; V LMS = 7.50±0.57 and V MLSS = 6.14±0.45m.s-1 and LAC IAT = 2.17±0.93; LAC LMS = 1.17±0.62 and LAC MLSS = 0.84±0.21mmol.L-1. None of the velocities were statistically equivalent to V MLSS (P<0.05). V2, V4 and V LMS showed a good correlation with V MLSS , respectively: r = 0.74; r = 0.78 and r = 0.83, and V IAT did not significantly correlate with V MLSS. Concordance between the protocols was relatively poor, i.e., 3.28±1.00, 4.84±0.30 and 1.43±0.32m.s-1 in terms of bias and 95% agreement limits for V2, V4 and LMS methods when compared to MLSS. Only LAC LMS did not differ statistically from LAC MLSS. Various authors have reported the possibility of the assessment of anaerobic threshold using rapid protocols such as V4 and LMS for humans and horses. This study corroborates the use of these tests, but reveals that adjustments in the protocols are necessary to obtain a better concordance between the tests and the MLSS.


O limiar anaeróbio é um evento fisiológico estudado em várias espécies. Sua mensuração possui vários métodos reconhecidos na literatura da fisiologia do exercício humano e equino, muitos deles envolvendo a relação entre a concentração sanguínea de lactato (LAC) e a carga de exercício. O objetivo do presente estudo foi comparar quatro desses métodos: V2 , V4 , limiar anaeróbio individual (LAI) e o teste do lactato mínino (LM) com o método reconhecido na literatura como o padrão ouro para a mensuração do limiar anaeróbio, a máxima fase estável do lactato (MFEL). Os cinco testes foram realizados em treze equinos árabes treinados, nos quais as velocidades e suas respectivas LAC puderam ser quantificadas. As velocidades médias e LAC associadas ao limiar anaeróbio aferido pelos cinco métodos foram respectivamente: V2 = 9,67±0,54; V4 = 10,98±0,47; V LAI = 9.81±0.72; V LM = 7,50±0,57 e V MFEL = 6,14±0,45m.s-1 ; e LAC LAI = 2,17±0,93; LAC LM = 1,17±0,62 e LAC MFEL = 0,84±0,21mmol.L-1. Nenhuma dessas velocidades foi estatisticamente igual à V MFEL (P<0,05). A V2 , a V4 e a V LM mostraram uma boa correlação com a V MFEL , respectivamente r = 0,74; r = 0,78 e r = 0,83, e a V LAI não se correlacionou significativamente com a V MFEL. A concordância entre os protocolos foi relativamente fraca, sendo 3,28±1,00; 4,84±0,30 e 1,43±0,32m.s-1 em termos de viés e limites de concordância a 95% para os métodos V2 , V4 e LM comparados à MFEL. Muitos autores relataram a possibilidade da mensuração do limiar anaeróbio pelo uso de protocolos rápidos, como a V4 e o LM, para humanos e equinos. O presente estudo corrobora a utilização desses testes, mas revela que ajustes nos protocolos são necessários para se obter uma melhor concordância entre os mesmos e a MFEL.

ABSTRACT

PURPOSE: To assess whether the Modified Early Warning Score (MEWS) predicts the need for intensive care unit (ICU) transfer for patients with severe sepsis or septic shock admitted to general wards. METHODS: A retrospective chart review of 100 general ward patients with severe sepsis or septic shock was implemented. Clinical information and MEWS according to point of time between ICU group and general ward group were reviewed. Data were analyzed using multivariate logistic regression and the area under the receiver operating characteristic curves with SPSS/WIN 18.0 program. RESULTS: Thirty-eight ICU patients and sixty-two general ward patients were included. In multivariate logistic regression, MEWS (odds ratio [OR] 2.02, 95% confidence interval [CI] 1.43-2.85), lactic acid (OR 1.83, 95% CI 1.22-2.73) and diastolic blood pressure (OR 0.89, 95% CI 0.80-1.00) were predictive of ICU transfer. The sensitivity and the specificity of MEWS used with cut-off value of six were 89.5% and 67.7% for ICU transfer. CONCLUSION: MEWS is an effective predictor of ICU transfer. A clinical algorithm could be created to respond to high MEWS and intervene with appropriate changes in clinical management.

ABSTRACT

O potencial probiótico in vitro de 12 amostras de bactérias ácido-láticas (11 Lactobacillus spp. e uma Weissella paramesenteroides), isoladas de queijo-de-minas artesanal da Serra da Canastra, foi investigado. Essas amostras foram caracterizadas in vitro quanto à susceptibilidade a antimicrobianos, antagonismo contra microrganismos de referência patogênicos e não patogênicos e sensibilidade a pH gástrico e sais biliares. Apenas L. rhamnosus B4, W. paramesenteroides C10 e L. rhamnosus D1 apresentaram resistência a menor número de antimicrobianos (ceftazidima, oxacilina, estreptomicina e vancomicina). Todas as amostras apresentaram atividade antagonista frente a todas as bactérias patogênicas testadas e não inibiram as bactérias não patogênicas. Todas as amostras foram resistentes ao pH gástrico (2.0). Alguns microrganismos mostraram pouca inibição do crescimento em presença de sais biliares, enquanto outros foram moderadamente ou altamente inibidos. L. rhamnosus B4, W. paramesenteroides C10 e L.rhamnosus D1 apresentaram o melhor potencial probiótico de acordo com os testes in vitro realizados.


The in vitro probiotic potential of twelve samples of lactic acid bacteria (eleven Lactobacillus spp. and one Weissella paramesenteroides) isolated from Minas artisanal cheese from Serra da Canastra was evaluated for their antimicrobial susceptibility, antagonistic activity against pathogenic and lactic acid bacteria, as well as for their tolerance to gastric pH and biliary salts. Only L. rhamnosus B4, W. paramesenteroides C10, and L. rhamnosus D1 showed resistance to tested antimicrobials (ceftazidime, oxacillin, streptomycin, and vancomycin). All samples showed antagonism against all pathogenic bacteria tested and did not inhibit the growth of nonpathogenic samples. Regarding gastric pH tolerance, all samples showed low inhibition at pH 2. The tolerance to biliary salts varied from low to great among tested samples. L. rhamnosus B4, W. paramesenteroides C10, and L. rhamnosus D1 showed the best probiotic potential revealed by their in vitro test results.

ABSTRACT

Avaliar o comportamento do lactato sanguíneo pós-exercício aeróbico contínuo (EAC) e pós-exercício resistido intervalado (ERI). Estudo analítico tipo cross-over, do qual participaram voluntários com doença de Chagas crônica divididos em dois grupos, grupo sintomático (GS), com seis voluntários, e grupo assintomático (GA) com oito voluntários. Os sujeitos foram submetidos a um teste cardiopulmonar em esteira ergométrica para determinar o limiar de anaerobiose (LA) e o VO² pico, e 24 horas depois a um teste de carga máxima (CM). Decorridos sete dias, os voluntários participaram de uma sessão de EAC e, sete dias posteriormente, a uma sessão de ERI. O EAC foi realizado em esteira ergométrica durante 20 minutos (min), sendo 10 min a 40% de VO² pico, 7 min no LA e a mais 3 min em intensidade decrescente. O ERI teve duração aproximada de 20 min e foi realizado com pesos livres a 30% da CM em duas séries de quatro movimentos. Foram coletados amostras sanguíneas para dosagem da lactacemia nos tempos 0 (repouso), 5 e 10 min após as sessões de EAC e ERI. O GA apresentou maior variação do lactato em relação ao repouso (p=0,05) no ERI ao final dos 10 min de atividade quando comparado ao GS, tendo esse último apresentado maior variação do lactato no EAC, sem diferença significativa. O ERI parece ser mais confortável e seguro que o EAC quando aplicado em indivíduos chagásicos sintomáticos. Já para indivíduos assintomáticos, parece ser indiferente o emprego do EAC ou do ERI.


To evaluate blood lactate post continuous aerobic exercise (CAE) and post resistance exercise interval (REI). Analytical study crossover which involved volunteers with chronic Chagas disease divided into two groups, the symptomatic group (SG) with six volunteers and asymptomatic group (AG) with eight volunteers. First, the subjects underwent cardiopulmonar exercise testing on a treadmill to determine the anaerobic threshold (AT) and VO² peak, and 24 hours after a maximum load test (ML). After seven days, the volunteers underwent a session of CAE and seven days later at a session of REI. The CAE was performed on a treadmill for 20 min, 10 min and 40% of VO² peak, 7 min in LA and decreasing in intensity over 3 min. The REI lasted approximately 20 minutes and was carried out with free weights to 30% of ML in two series of four movements. Blood samples were collected to determine lacticemia at 0 (rest), 5 and 10 min after the sessions of CAE and REI. The AG varied more lactate in relation to rest (p=0.05) in the REI at the end of 10 minutes of activity compared to SG, the later presented a greater variation of lactate in the CAE, without significant difference. The REI appears to be more comfortable and secure than CAE when applied to symptomatic chagasic individuals. Whereas asymptomatic individuals seem to be indiferent to the use of the CAE or the REI.

ABSTRACT

High hydrostatic pressure (HHP) has been investigated and industrially applied to extend shelf life of meat-based products. Traditional ham packaged under microaerophilic conditions may sometimes present high lactic acid bacteria population during refrigerated storage, which limits shelf life due to development of unpleasant odor and greenish and sticky appearance. This study aimed at evaluating the shelf life of turkey ham pressurized at 400 MPa for 15 min and stored at 4, 8 and 12 ºC, in comparison to the non pressurized product. The lactic acid bacteria population up to 10(7) CFU/g of product was set as the criteria to determine the limiting shelf life According to such parameter the pressurized sample achieved a commercial viability within 75 days when stored at 4 ºC while the control lasted only 45 days. Predictive microbiology using Gompertz and Baranyi and Roberts models fitted well both for the pressurized and control samples. The results indicated that the high hydrostatic pressure treatment greatly increased the turkey ham commercial viability in comparison to the usual length, by slowing down the growth of microorganisms in the product.

ABSTRACT

This study was performed to determine the accuracy of proton magnetic spectroscopy (1H-MRS) lipid peak as a noninvasive tool for quantitative in vivo detection of brain cell death. Seven day-old Sprague Dawley rats were subjected to 8% oxygen following a unilateral carotid artery ligation. For treatment, cycloheximide was given immediately after hypoxic ischemia (HI). Lipid peak was measured using 1H-MRS at 24 hr after HI, and then brains were harvested for fluorocytometric analyses with annexin V/propidium iodide (PI) and fluorescent probe JC-1, and for adenosine-5'-triphosphate (ATP) and lactate. Increased lipid peak at 1.3 ppm measured with 1H-MRS, apoptotic and necrotic cells, and loss of mitochondrial membrane potential (DeltaPsi) at 24 hr after HI were significantly improved with cycloheximide treatment. Significantly reduced brain ATP and increased lactate levels observed at 24 hr after HI showed a tendency to improve without statistical significance with cycloheximide treatment. Lipid peak at 1.3 ppm showed significant positive correlation with both apoptotic and necrotic cells and loss of DeltaPsi, and negative correlation with normal live cells. Lipid peak at 1.3 ppm measured by 1H-MRS might be a sensitive and reliable diagnostic tool for quantitative in vivo detection of brain cell death after HI.

Avaliaram-se a concentração de lactato e a glicemia em sete cadelas em trabalho de parto submetidas à cesariana e em seus respectivos neonatos. O lactato materno foi mensurado utilizando-se medidor portátil e pelo método enzimático colorimétrico. Nos neonatos avaliou-se também o peso ao nascimento. Os valores de lactato materno e neonatal foram estatisticamente diferentes, sendo mais elevado nos neonatos. Ao nascimento, as cadelas e os neonatos encontravam-se normoglicêmicos, e não se observou correlação entre a glicemia de ambos. Os valores de lactato obtidos pelo o analisador portátil (3,5±0,6mmol/L) foram mais altos do que os obtidos pelo método enzimático colorimétrico (1,6±0,6mmol/L). Observou-se correlação significativa entre o peso do neonato e a concentração de lactato, isto é, os filhotes de menor peso apresentaram maior concentração de lactato.


This work evaluated the concentration of lactate and blood glucose in seven bitches in labor subject to caesarean section and in their newborns. The lactate concentration was measured using a portable analyzer and enzymatic colorimetric method. In neonates, the birth weight was evaluated. The maternal and neonatal lactate values were statistically different, being higher in newborns. At birth, the bitches and the newborns had normal glucose status, and no correlation was observed between blood glucose in either. The lactate concentration obtained by the portable analyzer (3.5±0.6mmol/L) was higher than that obtained with the enzymatic colorimetric method (1.6±0.6mmol/L). There was significant correlation between the neonate weight and lactate concentration and in puppies with lighter weight there was a higher concentration of lactate.
