ABSTRACT

Abstract Introduction: Spills of flammable liquids can lead to serious accidents, mainly in industrial plants and on roads. To prevent the spread of spills, various forms of collection are used, such as absorption with porous solids. Agroindustrial waste can be used as sorbent materials for flammable liquids. Objective: To determine the sorption capacity of the residual empty-fruit bunch of oil-palm (Elaeis guineensis) and the macaw palm (Acrocomia sp.) nutshell for four organic flammable liquids. Methods: The residual biomasses of E. guineensis and Acrocomia sp. were assessed as sorbents for spilled fuels (diesel, jet fuel, commercial kerosene, and gasoline). Volumetric measurement of liquid-fuel absorption at 24 ºC was taken during a week. Desorption was measured at 50 ºC as the drying kinetics, by using moisture scales. Results: The sorption capacity of the Acrocomia sp. material was not satisfactory, compared to the E. guineensis residual material, due to differences in the residual architecture of the organic material. This last can absorb 2.4 ± 0.2 cm3 g-1 at 24 ºC, during a one-week period. Diatomite absorbs greater quantities of the organic liquids but, the fluids diffusion at 50 ºC is 0.26 ± 0.09 times more slowly in the mineral matrix, because of the greater pore tortuosity in this mineral matrix. Conclusions: The oil-palm empty fruit bunch of E. guineensis, showed lesser but adequate performance than the sorbing behavior for fire hazard mitigation of diatomite. The nutshell of macaw palm (Acrocomia sp.) did not prove to be useful for this recovery operation.

ABSTRACT

The study was designed to investigate the effect of Coconut Oil on the levels of some liver and hematological parameters in carbon tetrachloride intoxicated rabbits. Also the antioxidant capacity of Coconut Oil for various concentrations was assessed on the basis of percent scavenging of (DPPH) free radical. Experimental animals were divided into five groups, eight rabbits in each group. These were: group A (Normal control), group B (Toxic control), group C (Standard control), group D (Treated with Coconut Oil 50 mL/kg body weight after CCl4 intoxication), group E (Treated with Coconut Oil 200 mL/kg body weight after CCl4 intoxication). The effects observed were compared with a standard hepatoprotective drug silymarine (50 mL/kg body weight). The Coconut Oil (200 mL/kg body weight) significantly (P<0.05) reduced the elevated serum levels of alanine transaminase (ALT), aspartate transaminase (AST) and alkaline phosphatase (ALP) when compared to a toxic control rabbits. The results of extract treated rabbits were similar to silymarine administered rabbits group. Treatment with Coconut Oil root and silymarine caused no significant changes in RBC, Platelets, (Hb), (MCH) concentration and (HCT) values. However, significant (P<0.05) increase was observed in the total WBC count. The present study suggested that Coconut Oil can be used as an herbal alternative (need further exploration i.e to detect its bioactive compound and its efficacy) for hepatoprotective activit.

ABSTRACT

Alzheimer's disease (AD) is a neurodegenerative disease whose main characteristic is the progressive loss of cognitive functions, affecting millions of people worldwide. In this sense, there is a continuous search for therapeutic tools that act in the prevention and treatment of the disease. In addition, research using coconut oil as a therapeutic tool has demonstrated a potent anti-inflammatory effect of this substance. Thus, a literature review was conducted to investigate the possible effects of coconut oil on AD. This article is a literature review using the PubMed, Science Direct, SciELO and LILACS databases. From the search with specific descriptors, studies were included referring to the last five years, published in English, Portuguese and Spanish with pre-clinical and clinical data. The included preclinical studies demonstrated that coconut oil interferes by inhibiting important metabolic pathways that act in the promotion of neuroinflammation, amyloid plaque formation, neurotransmitter imbalance, and signaling pathway dynamics. In addition, the use of coconut oil increased the levels of ketone bodies. To some extent, many clinical trials have already shown favorable support for the use of coconut oil as an adjuvant in the treatment of AD. However, there is still a need for more randomized, controlled clinical trials that evaluate the optimal dosage, as well as whether or not coconut oil is effective against Alzheimer's disease.

ABSTRACT

Aims@#This study was designed to examine the enzyme activity of selected virulent isolates of Ganoderma boninense against oil palm. In a separate in vitro assessment, the effect of macronutrients on the mycelial growth of four selected Ganoderma spp. was also tested.@*Methodology and results@#The study involved a comparison of ligninolytic enzymes; lignin peroxidase (LiP), manganese peroxidase (MnP) and laccase (Lac) profiling of eight isolates of G. boninense, categorized into three levels of aggressiveness, with two control isolates (G. boninense PER71 and G. tornatum NPG1) using solid-state fermentation (SSF). The Principal Component Analysis (PCA) revealed that the isolates had a significant production of ligninolytic enzymes on day 80. The most aggressive isolate, ET61 had the highest Lac production. As for the macronutrient test, mycelial growth for all the Ganoderma spp. was highly affected by potassium (K).@*Conclusion, significance and impact of study@#The findings of this study elucidated the characteristics of G. boninense in relation to enzyme production for the degradation of oil palm lignin and the identification of essential nutrients involved in the survival and growth of Ganoderma spp. The study provides vital information on the pathogenic characteristics of G. boninense isolates involved in biomass degradation along with the role of nutrient on the growth of Ganoderma spp. that may influence basal stem rot (BSR) management in the field.

ABSTRACT

Due to rainfall and high temperatures, the Amazonian soil undergoes changes in its source material and leaching of base cations. This results in deep, infertile, and acidic soil. Aluminum present in acidic soil impairs plant growth and development by inhibiting root formation, enzymatic reactions, absorption, transport, and nutrient utilization. This study aimed to evaluate the effects of aluminum dosage on the metabolism of the oil palm Elaeis guineensis Jacq. The study was conducted in a greenhouse at the Federal Rural University of Amazonia. The experimental design was randomized, with five replications, in which dosages of 0, 10, 20, 30, and 40 mg L-1 aluminum chloride (AlCl3.6H2O) were administered. Electrolyte leakage, nitrate, nitrate reductase, free ammonium, soluble amino acids, proline content, and soluble proteins were analyzed in the leaves and roots of the oil palm. The highest concentration of aluminum was found in the roots. AlCl3 treatment at 40 mg L-1 increased electrolyte leakage, nitrate, ammonium, and proline concentrations in the roots, and amino acid concentrations in both the leaves and roots. Furthermore, a decrease in nitrate reductase enzyme activity was observed in the roots. This study demonstrates that the oil palm has mechanisms of tolerance to aluminum toxicity.

ABSTRACT

The concentrations of nutrients in the leaves allow a better understanding of the nutritional status of oil palm plants, making it a tool for diagnosing the origins of nutritional problems and assisting in fertilizer recommendations. In this sense, the objective of the current research was to evaluate the leaf concentrations of N, P, K, Ca, Mg and S in the oil palm under fertilization with phosphate, potassium and magnesium in edaphoclimatic conditions of Eastern Amazon. The experiment was conducted at the AGROPALMA® Company in the municipality of Tailândia, state of Pará, Brazil. The experimental design used was randomized blocks in a factorial scheme 4 x 2 x 3 x 2, with four levels of phosphorus, two sources of phosphorus (natural phosphate and triple superphosphate), three levels of potassium and two levels of magnesium. Phosphorus fertilization increased the leaf concentrations of N, P, Ca and Mg, providing higher levels when triple superphosphate was applied. However, in adult oil palm plants (12 years old), there was different on leaf concentrations of N, P, K and S between phosphorus sources aphid. Potassium and magnesium fertilization only increased the leaf concentrations of K and Mg, respectively. Fertilization with P, K and Mg promotes adequate leaf concentrations of K, Ca, Mg e S in oil palm plants grown in the Eastern Amazon.

ABSTRACT

Aims@#Insect pests are one of the major constraints of oil palm production in Malaysia. However, synthetic chemical insecticides are the most common method for its control, despite their negative effects on non-target organisms and the development of resistance. Therefore, the present study is aimed to identify novel indigenous isolates of entomopathogenic fungi (EPF) in oil palm soil as part of integrated pest management (IPM) of oil palm insect pests.@*Methodology and results@#The potential of EPF were isolated from the soil collected from the oil palm plantation in UPM using a mealworm beetle larva (Tenebrio molitor) as an insect bait. Seven Metarhizium anisopliae and two Purpureocillium lilacinum isolates were identified by morphological characterization (macroscopic and microscopic observation) and molecular identification using internal transcribed spacer (ITS) region (ITS region amplification).@*Conclusion, significance and impact of study@#To our knowledge, this is the first time P. lilacinum has been found in the soil of an oil palm plantation or any other host in Malaysia. Furthermore, both of the isolates M. anisopliae and P. lilacinum may be potentially considered as biological control candidates for major insect pests in oil palm.

ABSTRACT

Aims@#Intercropping system in oil palm plantation is recognized as one of a nature-based solution as well as a promising sustainable practice. This study aimed to observe the advantages of existing intercropping system in one of North Sumatra’s oil palm plantation. It is achieved by analyzing the population of soil bacteria and fungi in oil palm intercropping fields with sorghum and cassava, compared with the non-intercropping field that using Mucuna bracteata (MB) as a common legume cover crop in oil palm plantations.@*Methodology and results@#Soil samples were collected from the weeded circle and windrow area (the area between palms within the row). The results showed that the highest and the lowest soil bacteria populations were in sorghum (1.7 ± 1.4 × 108 CFU/g) and MB (1.7 ± 0.4 × 107 CFU/g), while the highest and the lowest soil fungi populations were in sorghum (4.3 ± 2.9 × 106 CFU/g) and cassava (2.1 ± 0.8 × 106 CFU/g).@*Conclusion, significance and impact of study@#The intercropping system in this study showed a significant difference in the bacteria population, while the fungi population had no difference compared to the non-intercropping system. The bacterial and fungi population results also indicate that the intercropping system potentially enhances the soil's biological activity as an indicator of improved soil health. It is also followed by a slightly higher soil organic carbon value in intercropping system. This research suggests that further studies should be done to identify specific soil functional microbes (nutrients fixers and solubilizers). The future research will be used as a reference for promising biofertilizer agents in supporting sustainable crop production.

ABSTRACT

Aims@#The primary aim of this study was to utilize abundant palm oil mill effluent (POME) waste and turn it into a value-added product of biomass fuel with high calorific energy value (CEV) via fermentation and drying process, then simultaneously reduce abundant liquid waste.@*Methodology and results@#POME is available abundantly in Malaysia and only a small portion of it is utilized to produce other value-added products. In this study, fermentation of POME in the presence of bacteria (Lysinibacillus sp.) and fungus (Aspergillus flavus) separately at 37 °C, 180 rpm for 5 days, followed by overnight oven-drying at 85 °C was conducted. Four fermentation medium conditions were performed, viz.: (1) autoclaved POME, (2) autoclaved POME with the addition of Lysinibacillus sp., (3) autoclaved POME with the addition of A. flavus and (4) POME as it is (non-sterile).@*Conclusion, significance and impact of study@#Among all conditions, fermentation utilizing autoclaved POME in the presence of A. flavus evinced the highest CEV of 25.18 MJ/kg. The fermentation in the presence of Lysinibacillus sp. strain revealed high COD and BOD removal efficiency of 59.20% and 320.44 mg/L as well as the highest reduction of oils and grease among other groups with the value of 15.84%. Future research directions are proposed for the elucidation of co-fermentation in the presence of both Lysinibacillus sp. and A. flavus.

ABSTRACT

Aims@#Palm kernel cake (PKC) is a high-protein, high-energy food that is widely utilized in the animal feed business. However, the high fibre and limited amino acid content of untreated PKC were the main issues for it to be used as animal feed, particularly in non-ruminants. To improve the quality of PKC, this study combined the use of solid-state fermentation (SSF) and consortia of fungi and bacteria to treat the PKC.@*Methodology and results@#Two fungi, Emericella nidulans (4DP5) and Cladosporium herbarum (7DF12) and three strains of bacteria, Bacillus subtilis, which were active mannanase producers, were used in different combinations to reduce the hemicellulose content and improve the crude protein content of PKC in a lab-scale solid-state fermentation. PKC inoculated separately with five types of mixed culture treatments were allowed to ferment. The fermentation conditions were 20% inoculum (w/v), 85-92% humidity, pH 7.0 and PKC particle size 0.8 mm. PKC treatments with two fungi, E. nidulans (4DP5) and C. herbarum (7DF12), as well as a fungus-bacterium combination, E. nidulans (4DP5) and B. subtilis, outperformed the other three treatments. The crude protein levels were increased by 3.34% and 1.86%, respectively, due to these treatments. Furthermore, the level of aflatoxins produced increased marginally but remained within the permissible limits.@*Conclusion, significance and impact of study@#The treated PKC has more sugar and crude protein and less than 20 parts per billion (ppb) of aflatoxin, making it appropriate for animal consumption. The SSF technique of combining fungi and Bacilli enhanced the nutritional and market value of PKC substantially, which can be upscaled.

ABSTRACT

The present in vitro study determined the antibacterial effect of Coconut (Cocus nucifera) oil on Streptococcus mutans ATCC 25175. Cross-sectional and experimental study. Streptococcus mutans strain ATCC 25175 was seeded on Müeller- Hinton Blood Agar. The effect was determined by inhibition halo and the minimum inhibitory concentration, using procaine penicillin G as a positive control and standard suspension of Streptococcus mutans as a negative control. Twelve replicates per Coconut oil concentration (25 %, 50 % and 75 %) were performed. Coconut oil concentration at 25 % generated an inhibitory mean of 17 mm and 2.23x102 CFU, at 50 % a mean of 21.75 mm and 0.17 x 102 CFU, at 75 % a mean of 22 mm and 0 CFU, procaine penicillin G a mean of 14.25 mm and 0 CFU, the negative control gave a mean of 2.8 x 10 5 CFU. The Kruskal-Wallis non-parametric test detected a highly significant statistical difference of the three concentrations of Coconut oil (p < 0.01). The Mann-Whitney test with Bonferroni adjustment determined that the 50 % and 75 % concentration had similar inhibitory action and that both the 75 % concentration and procaine penicillin G gave a mean of 0 CFU. All concentrations of Coconut oil showed inhib itory action on Streptococcus mutans ATCC 25175. 75 % concentration showed the highest inhibitory mean and was the minimum inhibitory concentration that completely eliminated CFU.

ABSTRACT

@#Background and Objectives. Neuroprotection agents may help improve the outcomes of large vessel ischemic stroke. This study aims to explore the role of Virgin Coconut Oil (VCO), with its well-documented anti-oxidant properties, in neuroprotection after transient occlusion of the extracranial internal carotid artery in a rat model of stroke. Methods. Twenty-three Sprague-Dawley rats were randomized into two groups: 1) control group (n=11) given distilled water, and 2) treatment group (n=12) given virgin coconut oil at 5.15 ml/kg body weight for seven days. Subsequently, the rats underwent transient right extracranial internal carotid artery occlusion (EICAO) for 5 minutes using non-traumatic aneurysm clips. At 4 and 24 hours after EICAO, the animals were examined for neurologic deficits by an observer blinded to treatment groups, then sacrificed. Eight brain specimens (4 from each group) were subjected to histopathologic examination (H & E staining) while the rest of the specimens were processed using triphenyltetrazolium chloride (TTC) staining to determine infarct size and area of hemispheric edema. Results. VCO treatment significantly improved the severity of neurologic deficit (1.42 ± 2.31) compared to the control distilled water group (4.09 ± 2.59) 24 hours after EICAO. Whereas, infarct size and percent hemispheric edema did not significantly differ between the two groups. Conclusion. Prophylactic treatment of VCO is protective against EICAO-induced neurologic deficits in a rat model. VCO shows great potential as a neuroprotective agent for large vessel ischemic stroke. However, more studies are necessary to elucidate the neuroprotective mechanisms of VCO therapy in ischemic stroke.

ABSTRACT

Aims@#The development of an effective biocontrol formulation for inhibition of Ganoderma boninense, a well-known destructive pathogen in oil palm plantation is important to prolong the palm’s lifespan and reduce the losses due to this disease. In this paper, we present some new bioformulations with combination of different types of biocontrol agents in managing basal stem rot (BSR) disease. @*Methodology@#The effectiveness of the treatments designed as T1 (Trichoderma harzianum + Lecanicillium spp. + Streptomyces sundarbansensis + Pseudomonas aeruginosa), T2 (Penicillium simplicissimum + Lecanicillium sp. + S. sundarbansensis + P. aeruginosa), T3 (P. simplicissimum + P. aeruginosa) and T4 (LEStani®) was evaluated through treatment on the oil palm seedlings artificial infected by G. boninense and the results were expressed in disease severity index (DSI), bole severity index (BSI) and ergosterol content.@*Conclusion, significance and impact of study@#All tested treatments (T1-T4) managed to control the severity of the Ganoderma infection from continuously increasing when the treatments were applied either one month before or after artificial inoculation. The disease severity of infected seedlings without treatments had increased for almost 2-fold at the end of the trial. Moreover, T1 had the greatest inhibition of G. boninense with the lowest ergosterol content (a bioindicator of Ganoderma colonization) detected (676.67 g/mL), which is about 1.9-fold lower than control (1273.33 ug/mL) without treatments and a BSI score of 1. Based on the effectiveness among the four tested biocontrol formulations, T1 is the most promising formulation to be further evaluated in the field for control of BSR disease. However, more research is needed in the future to estimate the effective amount for application in open environment.

ABSTRACT

Abstract Rhamnolipid is a potent biodegradable surfactant, which frequently used in pharmaceutical and environmental industries, such as enhanced oil recovery and bioremediation. This study aims to engineer Escherichia coli for the heterologous host production of rhamnolipid, to characterize the rhamnolipid product, and to optimize the production using autoinduction medium and POME (palm oil mill effluent). The construction of genes involved in rhamnolipid biosynthesis was designed in two plasmids, pPM RHLAB (mono-rhamnolipid production plasmid) and pPM RHLABC (di-rhamnolipid production plasmid). The characterization of rhamnolipid congeners and activity using high-resolution mass spectrometry (HRMS) and critical micelle concentration (CMC). In order to estimate rhamnolipid yield, an oil spreading test was performed. HRMS and CMC result show E. coli pPM RHLAB mainly produced mono-rhamnolipid (Rha-C14:2) with 900 mg/L and 35.4 mN/m of CMC and surface tension value, whereas E. coli pPM RHLABC mainly produced di-rhamnolipid (Rha-Rha-C10) with 300 mg/L and 34.3 mN/m of CMC and surface tension value, respectively. The optimum condition to produce rhamnolipid was at 20 h cultivation time, 37 oC, and pH 7. In this condition, the maximum rhamnolipid yield of 1245.68 mg/L using autoinduction medium and 318.42 mg/L using 20% (v/v) of POME. In conclusion, the characteristics of the rhamnolipid by recombinant E. coli is very promising to be used in industries as the most economical way of producing rhamnolipid.

ABSTRACT

Digestive disorders in cattle are associated with the breeding system and feed provided to the animals. Abomasal compaction is primarily related to the ingestion of forage with elevated levels of lignin, low quality, and difficult digestibility. In addition, the excess of fibrous food in the diet can lead to phytobezoars that may be responsible for intestinal obstruction disorders. This study aimed to describe pathological and clinical aspects of an outbreak of digestive disorders associated with the consumption of palm fiber (Elaeis guineensis). The outbreak struck a herd of 499 animals raised in a feedlot system after a change in diet that included an increase in the amount of palm fiber. Forty (8.01%) animals showed clinical signs such as fattening and regurgitation during rumination, and 21 (4.2%) animals died later. The cattle affected presented with apathy, emaciation, dehydration, distended abdomen, incomplete or absent ruminal movements, and congestive mucosa. Three animals were submitted to necropsy, and distended rumen and reticulum has a large amount of brownish liquid, long and tangled vegetable fibers with sand and stones. In two animals, the omasum had many rounded structures measuring approximately 5cm in diameter, made of vegetable fiber (phytobezoars). Abomasum of animals had similar material to the rumen, and one animal had compressed content. In two animals, dilatation was observed in the small intestine, and in the opening, the total obstruction of the lumen by phytobezoar was observed. During the follow-up of the slaughter of 76 cattle, 15 (19.7%) had phytobezoars of different sizes in the omasum and abomasum. The increased amount of oil palm fiber in animal feeding favored the occurrence of compression abomasum and intestinal obstruction phytobezoa, causing significant economic losses.(AU)

ABSTRACT

HIGHLIGHTS Production of lipid nanoemulsions (<100 nm) of industrial interest with low energy demand. The antioxidant properties of babassu oil have been improved and the nanoemulsions are not cytotoxic. Babassu oil is a food and medicinal product. The nanoemulsion is strategic for the developed of new antioxidants phytotherapeutics.

Abstract Background: Babassu oil is an extract from a Brazilian native coconut (Orbignya phalerata Martius) and is used both as a food and a medicinal product. Methods: we produced two babassu oil nanoemulsions and evaluated them regarding their nanoscopic stability, antioxidant activity and cytotoxicity. The nanoemulsions were characterized by Dynamic Light Scattering, and their stability was investigated for 120 days. The antioxidant activity was assessed by Spectroscopy Electron Paramagnetic Resonance, and the cytotoxicity was assessed by a colorimetric method (MTT) with the NIH/3T3 cell lineage. Results: the results showed nanoemulsions with average hydrodynamic diameter lower than 100 nm (p(0.001).and a polydispersity index of less than 0.3 (p(0.001), indicating monodisperse systems and good stability at room temperature. The exposure of nanoemulsions at varying pH revealed that the isoelectric point was at 3.0, and the images obtained by Transmission Electron Microscopy showed spherical droplets with a size 27 nm. The antioxidant activity showed that the babassu nanoemulsions exposed to free radicals had a better response when compared to the oil free samples. The cell viability assays showed low toxicity of the formulation with viability over 92% (p(0.05). Conclusion: babassu oil nanoformulations showed low polydispersity and kinetic stability with effective antioxidant action. Therefore, they can be promising for application in the food industry or as antioxidant phytotherapeutics.

ABSTRACT

Abstract Background: The thousand peso palm tree (Oenocarpus bataua) is a promising species, both for its oil-rich fruits of excellent properties and for its unique adaptation to poor soils. Objective: Analyze the fatty acid profile and physicochemical properties of oil from the thousand peso palm tree. Materials and Methods: The physicochemical properties and the percentage of saturation ere found using AOAC techniques, and the fatty acid profile was analyzed by liquid gas chromatography with flame ionization detector (GC-FID). Results: The oil analyzed had a fatty acid profile similar to olive oil, with the following percentage distribution of fatty acids: palmitic (11.9±1.1), oleic (76.1±1), linoleic (3±0.4), α-linolenic (1.9±0.3), with a predominance of monounsaturated fats (77.0±1.0%), moderate contribution of saturated (18.1±1.2%) and low polyunsaturated (4.9±0.6%). Only the polyunsaturated profile would constitute a limitation for use as a sole source of fat for food preparation, otherwise thousand peso palm oil complies with the Colombian standard for high oleic palm oil in terms of the fatty acid profile and its physicochemical characteristics. Conclusions: The oil studied is potentially marketable for human consumption and has cardiovascular health benefits for consumers.

ABSTRACT

BACKGROUND: Liquid wax esters are widely used in cosmetic as well as pharmaceutical and other industries. The demand of organic and natural products is increasing nowadays. Coconut oil contains benefit fatty acids and has been mainly used for oil-based and moisturizer products. Liquid wax esters from coconut oil and unsaturated fatty alcohol can be synthesized by enzymatic reaction; and it is interesting for using as an alternative natural ingredient in these industries. RESULTS: Optimal condition for coconut oil based wax ester synthesis by immobilized lipase EQ3 was 10 U of enzyme, temperature at 30°C and molar ratio of coconut oil to oleyl alcohol at 1:3 (mol/mol) (0.33X) dissolved in isooctane for 12 h, while for Lipozyme RM IM optimal condition was 10 U of enzyme, temperature at 45°C and oil/alcohol molar ratio at 1:3 (0.33X) dissolved in isooctane for 3 h. Percentage of wax esters synthesized by both lipases reached more than 88%. Both immobilized lipases catalyzed high yield of wax esters within the 2nd batch; after that, the immobilized lipases showed reduced activity and synthesized b60% of wax esters from the 3rd to 5th batch. The main composition of wax esters was ~48% oleyl laurate with 10% degradation at ~250°C. CONCLUSIONS: The liquid wax ester synthesis by commercial Lipozyme RM IM had higher effect than immobilized lipase EQ3, but both catalysts were stable within 2 batches in the optimum condition. The characteristic properties of wax esters showed potential for use as components in cosmetics and skin care products.

ABSTRACT

Obesity is characterized by the accumulation of body fat associated with low-grade chronic inflammation, which is related to an increased risk of developing bone loss, and consequently, osteoporosis. Bone loss occurs due to several factors, including the presence of an inflammatory response. Chronic diseases such as obesity show an increase in the synthesis and secretion of inflammatory mediators that could favor bone loss. Although dietary approaches are proposed to treat bone loss, they are still poorly explored. Virgin coconut oil (VCO) is a functional food due to its significant amount of medium-chain fatty acids. In recent years, VCO has been widely used to treat several diseases, including Alzheimer's, heart disease, obesity, among others. However, knowledge about VCO supplementation in the treatment of bone loss is still incipient. This study aimed to evaluate the effect of dietary supplementation with VCO in the treatment of bone loss in mice fed a 45% high-fat (HF) diet. Male C57BL / 6 mice were initially divided into two groups and fed either the AIN-93M (C) control diet or the HF diet for eight weeks. At the 9th week, the mice fed the HF diet were regrouped in four groups until the 12th week: (i) HF diet; or HF diet supplemented with different doses of VCO, (iii) 1000 mg / kg, (iv) 3000 mg / kg or (v) 9000 mg / kg. Although the weight gain does not present a statistical difference between the groups, the final body weight was higher in the HF group than the control group, but without changes in those treated with VCO in relation to the control. There was no significant difference in food intake between the groups evaluated. When bone parameters were evaluated, the serum concentrations of RANKL, a bone loss marker, and OPG, opposed to this signaling, did not change between the groups. However, only animals that received the medium dose of VCO showed a tendency towards a lower RANKL / OPG ratio, which was the dose chosen for bone microarchitecture evaluation. In general, the HF group showed lower bone mineral density and bone volume, thinner trabeculae with greater space between them, characterizing increased bone resorption in the femur and maxilla's bone structure. When treated with the medium dose of VCO, there was a worsening of bone mineral density and trabecular separation in the maxilla, with no change in other parameters when compared with the HF group. However, the bone loss present in the femur was not altered. As expected, adiposity, area of adipocytes and serum leptin concentrations were higher in the group fed with HF compared to the control group. In the group treated with the high dose of VCO an increase in the area of adipocytes was observed, but in the other parameters, no changes were observed after the different treatments with the VCO. The glucose intolerance observed in the HF group was not altered with the addition of VCO to the HF diet, and they were also shown to be hyperglycemic. Despite being changed in the HF group, serum concentrations of total cholesterol and triglycerides did not change with treatments. Therefore, the use of VCO supplementation in mice fed the HF diet does not seem to be beneficial for treating bone loss, obesity, and even the associated metabolic disorders.

ABSTRACT

The cocoa and palm oil agro-industries active in the state of Bahia, Brazil, generate high quantities of lignocellulosic wastes that could be recycled through their use in the formulation of substrates to cultivate edible mushrooms. Pleurotus ostreatus, also known as oyster mushroom, is the second most cultivated mushroom in the world due to its highly appreciated gastronomic, nutritional, and medicinal characteristics. This work evaluated the vertical mycelium growth, biological efficiency, mushroom yield, and nutritional composition of P. ostreatus produced in substrates formulated with a combination of palm oil fruit mesocarp (POFM) and cocoa almond peels (CAP) processing wastes. The substrates were formulated with the following POFM/CAP proportions (%/%): S1 86.4/9.6; S2 76.8/19.2; S3 67.2/28.8; S4 57.6/38.4, and S5 48.0/48.0. Substrates also received 3% powdered charcoal and 1% calcium carbonate. Substrates S1, S2, S3, and S4 were superior for vertical mycelium growth. S2 promoted the best biological efficiency (148.8%) and yield (560.5g·kg-1).The mushrooms produced in all substrates presented good nutritional values, although mushrooms produced using the S2 presented the highest crude protein content. Overall, S1 is the recommended substrate as it results in higher yields of nutrient rich mushrooms. Production of P. ostreatus in substrates composed of POFM and CAP represents a good alternative for recycling these wastes with potential economic and ecological benefits to regions where palm oil and cocoa are grown.(AU)
