J Strength Cond Res 33(1): 217-224, 2019-A transient augmentation in the metabolic efficiency of skeletal muscle is the purported basis for dietary nitrate supplementation amongst competitive and recreational athletes alike. Previous studies support the ergogenic effects of nitrate supplementation, as findings indicated improved microvascular blood flow, exercise economy, and performance with relatively short-term supplementation. As with most ergogenic aids, the optimum duration of supplementation before performance or competition, i.e., loading phase, is a critical determinant for efficacy. Therefore, the purpose of this study was to investigate the effects of long-term vs. single dosing nitrate supplementation on skeletal muscle oxygenation and cycling performance. In a randomized, placebo controlled, double blind, parallel design study, healthy, recreationally active men (n = 15) and women (n = 14) subjects (age = 18-29 years) completed an 8 km (5 mi) simulated cycling time trial before and after a 14-day supplementation period with either a nitrate supplement (Multi-Day Dosing Group) (n = 14) or placebo (Single Pre-Exercise Dosing Group; SD) (n = 15). Both groups consumed a single dose of the nitrate supplement 2 hours before the post-treatment time trial. In addition, skeletal muscle oxygenation was measured via near-infrared spectroscopy during each time trial. Multiday nitrate supplementation significantly decreased time to completion (p = 0.01) and increased average power (p = 0.04) and speed (p = 0.02) from pre-to post-treatment, while a single dosing produced no significant changes to these measures. There were no significant differences over time and across treatments for any other measures including muscle oxygenation variables. Overall, long-term nitrate supplementation appears to have an advantage over a single pre-exercise dosing on cycling performance and metabolic efficiency as indicated by an increase in power output with no change in oxygenation.

This study investigated the capability of near-infrared spectroscopy (NIRS) to predict the concentration of Zygosaccharomyces rouxii in apple and kiwi fruit juices. The yeast was inoculated in fresh kiwi fruit juice ( n = 68), reconstituted kiwi juice ( n = 85), and reconstituted apple juice ( n = 64), followed by NIR spectra collection and plate counting. A principal component analysis indicated direct orthogonal signal correction preprocessing was suitable to separate spectral samples. Parameter optimization algorithms increased the performance of support vector machine regression models developed in a single variety juice system and a multiple variety juice system. Single variety juice models achieved accurate prediction of Z. rouxii concentrations, with the limit of quantification at 3 to 15 CFU/mL ( R2 = 0.997 to 0.999), and the method was also feasible for Hanseniaspora uvarum and Candida tropicalis. The best multiple variety juice model obtained had a limit of quantification of 237 CFU/mL ( R2 = 0.961) for Z. rouxii. A Bland-Altman analysis indicated good agreement between the support vector machine regression model and the plate counting method. It suggests that NIRS can be a high-throughput method for prediction of Z. rouxii counts in kiwi fruit and apple juices.

At present, scanning laser Doppler imaging uses a 633-nm helium-neon laser (RED) as the only light source, but this restricts its ability to measure blood flow (i) at darkly pigmented skin and (ii) from deeper or subdermal structures. Because near-infrared (NIR) light is known to penetrate deeper into tissue and to be less absorbed than RED, two imagers were adapted to include a NIR laser diode source (one of 830 nm for UK studies; one of 780 nm for leprosy field trials) in parallel with the existing RED source. In human hands representing a range of skin pigmentations, RED scans were unobtainable at the darkest areas of skin, but intact NIR scans could be collected in all cases. In experiments at the rat knee and the dorsal human hand, NIR and RED values were similar on normal skin. Over underlying vessels, however, NIR values greatly exceeded RED values, an effect abolished by occlusion. Similarly, in patients with leprosy and in healthy controls in Spain, fingerpulp NIR values exceeded RED values to the greatest degree when thermoregulatory flow was highest, i.e., when the deeper-lying arteriovenous anastomoses were open. Over areas of experimental inflammation, NIR gave higher values and also exhibited a greater degree of spatial heterogeneity than RED. We conclude that some current limitations of laser Doppler imaging technology can be overcome by the use of NIR laser diode sources.
