RESUMO

Citrus leprosis is the most important viral disease affecting citrus. The disease is caused predominantly by CiLV-C and is transmitted by Brevipalpus yothersi Baker mites. This study brings some insight into the colonization of B. yothersi in citrus [(Citrus × sinensis (L.) Osbeck (Rutaceae)] previously infested by viruliferous or non-viruliferous B. yothersi. It also assesses the putative role of shelters on the behavior of B. yothersi. Expression of PR1 and PR4 genes, markers of plant defense mechanisms, were evaluated by RT-qPCR to correlate the role of the plant hormonal changes during the tri-trophic virus-mite-plant interplay. A previous infestation with either non-viruliferous and viruliferous mites positively influenced oviposition and the number of adult individuals in the resulting populations. Mite populations were higher on branches that had received a previous mite infestation than branches that did not. There was an increase in the expression of PR4, a marker gene in the jasmonic acid (JA) pathway, in the treatment with non-viruliferous mites, indicating a response from the plant to their feeding. Conversely, an induced expression of PR1, a marker gene in the salicylic acid (SA) pathway, was observed mainly in the treatment with viruliferous mites, which suggests the activation of a plant response against the pathogen. The earlier mite infestation, as well as the presence of leprosis lesions and a gypsum mixture as artificial shelters, all fostered the growth of the B. yothersi populations after the second infestation, regardless of the presence or absence of CiLV-C. Furthermore, it is suggested that B. yothersi feeding actually induces the JA pathway in plants. At the same time, the CiLV-C represses the JA pathway and induces the SA pathway, which benefits the mite vector.

BACKGROUND: The invasive pest, Drosophila suzukii attacks fresh soft-skinned fruit. Broad-spectrum insecticides are implemented for control but there is a need to reduce environmental risks and insecticide residues on fruits. Hanseniaspora uvarum is a yeast frequently found on ripe fruits and associated with D. suzukii. We aim to exploit the ecological association and attraction of D. suzukii to H. uvarum by developing an attract-and-kill strategy, with spray-application on canopy but not fruit. We therefore investigated D. suzukii attraction, egg-laying and mortality when exposed to insecticidal yeast-based formulations. RESULTS: Hanseniaspora uvarum strongly attracted D. suzukii when applied on leaves of grapevine, Vitis vinifera. Notably, this attractiveness was competitive to ripe grape berries that were susceptible to D. suzukii infestation. Moreover, adding H. uvarum enhanced the efficacy of insecticidal formulations against D. suzukii. Flies exposed to leaves treated with yeast-insecticide formulations showed higher mortality and laid a lower number of eggs compared to flies exposed to insecticide alone. In a wind tunnel, all treatments containing H. uvarum alone or in combination with insecticides, caused similar upwind flight and landing at the odor source, which provides evidence that the addition of insecticide did not reduce D. suzukii attraction to yeast. CONCLUSION: Hanseniaspora uvarum can be used to manipulate the behavior of D. suzukii by attracting flies to insecticide formulations. Yeast attraction is competitive to grape berries and improves insecticide effectiveness, suggesting that sprays covering canopy only, could reduce residues on fruit without compromising management efficacy.

Vinegar flies (Diptera: Drosophilidae) are well known to be associated with yeasts, which provide important nutrients and emit attractive semiochemicals. Drosophila suzukii (Matsumura) has become a major pest of berries and cherries around the world, requiring intensive management to maintain fruit quality. Although insecticides remain a dominant control approach, disruption of fly-yeast-host interactions remains a promising avenue for reducing the economic impact of this pest. We conducted field and laboratory experiments to explore whether a crop sterilant (peroxyacetic acid and hydrogen peroxide) developed for disease control can affect D. suzukii. In 2 yr of field tests in highbush blueberries, we found significantly lower infestation by D. suzukii in plots treated with the crop sterilant, both alone and in a rotation program with zeta-cypermethrin. When shoots from treated plots were tested in no-choice bioassays, crop sterilant treatments did not affect adult mortality or oviposition, but they reduced infestation. To explore the mechanisms in the laboratory, we found that the crop sterilant did not affect adult mortality, nor oviposition on treated fruit under no-choice settings, but adult flies settled and oviposited less on treated fruit in choice settings. When the crop sterilant was applied to colonies of Hanseniaspora uvarum (Niehaus) (Saccharomycetales: Saccharomycodaceae) and Issatchenkia terricola (Van der Walt) (Saccharomycetales: Saccharomycetacea) yeasts that are attractive and provide nutrition to D. suzukii, there was a dose-dependent inhibition of their growth. We highlight the potential for microbial management as a component of integrated pest management programs and prioritize research needs to incorporate this approach into control programs.

Two commercial strains, Hy-Line W-36 and DeKalb XL, were moved to a laying house at 18 wk of age. They were housed 6 hens/layer cage at 2 densities (361 and 482 cm2/bird) with 2 replications each per strain/density combination. The high-density treatment contained 24 hens/replication and the low-density treatment contained 18 hens/replication for a total of 168 hens. Production parameters were measured during the first egg production cycle, the molt period, and the first 4 wk of the second lay cycle (20 to 68 wk of age). Behavioral observations were taken during 2 consecutive d at 26, 34, 43, 51, 62, 64, and 68 wk of age to examine behavioral patterns. Modified Hansen's tests were conducted concurrently to provide indication of the fearfulness levels of hens at the various stages of production. The production characteristics were similar for both strains. The hens kept at the higher density had lower (P < 0.01) hen-day production and (P < 0.05) daily egg mass. Appetitive behaviors were not affected by strain or density but were affected by the age of the hen and by molting. During the molt, feeding and drinking behavioral acts were fewer (P < 0.05) at 0.018 and 0.013 acts per bird/min, respectively, and standing behavior was highest. The results indicated that the frequencies of pecking inedible objects during the molt period were similar to the frequencies at 26 and 34 wk. Hens performed more acts of standing, and crouching and had lower frequency of movement during the molt. Those kept at a low density performed more movement acts. Feather pecking decreased as hens aged and increased when they molted but was not affected by strain or density. The frequency of aggression and submissive acts was significantly lower during the molt period. Behaviors were affected by strain, density, bird age, and molting; however, the patterns and number of aggressive acts did not increase to compromise the welfare status of the hens. Behaviors during the molt appeared consistent with mechanisms for conservation of body reserves.

There have been many methods proposed to induce molting. Some worked very well in practice, but others were detrimental to the health and welfare of the hens. The most effective methods use some level of feed restriction and daylength manipulation to reduce body weight (Hansen, 1966; Ruszler, 1974, 1984, 1996; Swanson and Bell, 1974; Brake and Carey, 1983). Weight reduction is necessary for rest and rejuvenation of body tissues. Other methods evaluated incorporated dietary imbalances using either zinc, iodine, or sodium. Pharmaceuticals have been used but have not been cost effective. In recent years there have been those who question whether molting techniques are humane. Therefore, interest has been heightened in alternate methods to induce molting. Research reported to date has been inadequate to accurately determine which methods of induced molting are the least stressful, if they in fact, cause any more stress than that experienced by the hen during a natural molt. The three or four most highly refined methods being used commercially are not generally detrimental to the health and welfare of today's laying hen, provided that they are managed in accordance with proper husbandry practices.
