RESUMO

The specific fatty acid (FA) profile of colostrum may indicate a biological requirement for neonatal calves. The objective of this study was to characterize the FA profile and yields in colostrum, transition milk, and mature milk in primiparous (PP) and multiparous (MP) cows. Colostrum was milked from 10 PP and 10 MP Holstein cows fed the same pre- and postpartum rations. Milkings (M) 2 to 5 and 12 were respectively termed transition and mature milk. Overall, short-chain FA (C4:0 and C6:0) were 61 and 50% lower in colostrum than mature milk, respectively. A parity by milking interaction was also present, with higher C4:0 for PP cows at M2 and for MP cows at M12. Additionally, higher concentrations of C6:0 were present for PP cows at M2 through M4 and for MP cows at M12. Palmitic (C16:0) and myristic (C14:0) acids were 38% and 19% higher in colostrum than mature milk, respectively. However, total saturated FA remained relatively stable. Branched-chain FA were 13% lower in colostrum than mature milk and higher in PP than MP cows throughout the milking period. The proportion of trans-monounsaturated FA (MUFA) was 72% higher in PP cows throughout the milking period, as well as 13% lower in colostrum than mature milk. In contrast, cis-MUFA and total MUFA were not affected by milking nor parity. Linoleic acid (LA) was 25% higher in colostrum than transition and mature milks, but α-linolenic acid (ALA) did not differ. Consequently, the ratio of LA to ALA was 29% higher in colostrum than mature milk and 33% higher in MP cows. Linoleic acid was also 15% higher in MP cows, whereas ALA was 15% higher in PP cows. Conjugated linoleic acid (CLA, cis-9,trans-11) was 2.7-fold higher in PP cows, and no differences between colostrum and mature milk were detected. Overall, polyunsaturated FA (PUFA) from the n-6 and n-3 series were over 40% higher in colostrum compared with transition and mature milk. Milking by parity interactions were present for arachidonic acid (ARA), eicosapentaenoic acid (EPA), docosapentaenoic acid (DPA), docosahexaenoic acid (DHA), and total n-3 PUFA, translating to higher proportions in PP cows in M1 to M3, whereas proportions remained relatively stable throughout the milking period in MP cows. Despite increasing milk yields throughout the subsequent milkings, higher yields of EPA, ARA, DPA, and DHA were present in colostrum than in mature milk. Greater proportions and yields of n-3 and n-6 FA in colostrum may translate to specific requirements for newborn calves. Differences were also observed between PP and MP cows and may reflect different nutrient requirements and partitioning.

Overall, polyunsaturated FA (PUFA) from the n-6 and n-3 series were over 25% higher in colostrum compared with transition and mature milk. The present study was designed to determine if feeding steers extruded flaxseed and hay (25 and 75%; DM basis) together as a total mixed ration (TMR), or sequentially (non-TMR) would result in different enrichments of polyunsaturated fatty acids (PUFA) and their biohydrogenation intermediates (BHI) in beef adipose tissues [subcutaneous (SC) vs perirenal (PR) fat]. Forty-eight Angus cross steers (325 ± 16 kg) were stratified by weight to six pens, and pens were randomized to either TMR or non-TMR and fed ad libitum for an average of 242 days. The concentrations of α-linolenic acid increased by 18 mol% in both SC and PR in non-TMR steers compared to TMR steers (P < 0.01). trans 18:1 isomers were more concentrated in PR than SC (14.4 vs 9.5 mol%; P < 0.01) and increased by 10 mol% in both fat depots for non-TMR (P < 0.01). Other BHI including non-methylene-interrupted 18:2 (atypical dienes), conjugated linoleic acids and conjugated linolenic acids (CLnA) were affected by diet × tissue interactions (P < 0.01). The CLnA and CLA contents were higher in both fat depots when feeding the non-TMR, but the effect of diet was more pronounced in PR than in SC (P < 0.01). Atypical dienes were highest in PR from non-TMR and lowest in TMR fed steers (4.3 and 3.6 mol%) with SC contents being intermediate. The sequential feeding of lipid supplement can thus profoundly affect the enrichment of PUFA and their BHI in beef fat and their differentially enrichment is also fat depot dependant.

The objective of this research was to study the delta-9 desaturation of individual trans (t) fatty acids that can be found in ruminant fat or partially hydrogenated vegetable oils (PHVO) and determine their effects on lipogenic gene expression in adipocytes. It was hypothesized that delta-9 desaturation and lipogenic properties of t-18:1 isomers depend on the position of double bond. Differentiated 3T3-L1 adipocytes were treated with 200 µM of t6-18:1, t9-18:1, t11-18:1, t13-18:1 or t16-18:1, cis (c)-9 18:1 or bovine serum albumin (BSA) vehicle control for 48 h. Cells were then harvested for fatty acid and gene expression analyses using gas chromatography and quantitative PCR respectively. Among t-18:1 isomers, t13-18:1 and t11-8:1 had the greatest percent delta-9 desaturation (44 and 41 % respectively) followed by t16-18:1 and t6-18:1 (32 and 17 % respectively), while c9-18:1 and t9-18:1 did not undergo delta-9 desaturation. Trans9-18:1 up-regulated (P < 0.05) the expression of lipogenic genes including fatty acid synthase and stearoyl-CoA desaturase-1 (P < 0.05), whereas the expression of these genes were not affected with other t-18:1 isomers (P > 0.05). Consistent with gene expression results, t9-18:1 increased the de novo lipogenic index (16:0/18:2n-6) compared with control cells and increased delta-9 desaturation index (c9-16:1/18:0) compared to other t-18:1 isomers (P < 0.05). The current study provides further evidence that the predominant trans fatty acid in PHVO (t9-18:1) has isomer specific lipogenic properties.

Polyunsaturated fatty acids typically found in cattle feed include linoleic (LA) and α-linolenic acid (ALA). In the rumen, microbes metabolize these resulting in the formation of biohydrogenation products (BHP), which can be incorporated into meat and milk. Bioactivities of LA-BHP, including conjugated linoleic acid (cis (c) 9,trans (t) 11-18:2 and t10,c12-18:2) and trans fatty acid isomers (t9-, t10- and t11-18:1) have been investigated, but effects of several BHP unique to ALA have not been extensively studied, and most ALA-BHP are not commercially available. The objective of the present research was to develop methods to purify and collect ALA-BHP using silver ion (Ag(+)) chromatography in sufficient quantities to allow for convenient bioactivity testing in cell culture. Fatty acid methyl esters (FAME) were prepared from perirenal adipose tissue from a cow enriched with ALA-BHP by feeding flaxseed. These were applied to Ag(+)-solid phase extraction, and eluted with hexane with increasing quantities of acetone (1, 2, 10, 20%) or acetonitrile (2%) to pre-fractionate FAME based on degree of unsaturation and double bond configuration. Fractions were collected, concentrated and applied to semi-preparative Ag(+)-high performance liquid chromatography (HPLC) for the isolation and collection of purified isomers, which was accomplished using isocratic elutions with hexane containing differing amounts of acetonitrile (from 0.015 to 0.075%). Purified trans-18:1 isomers collected ranged in purity from 88 to 99%. Purity of the ALA-BHP dienes collected, including c9,t13-18:2, t11,c15-18:2 and t10,c15-18:2, exceeded 90%, while purification of other dienes may require the use of other complementary procedures (e.g. reverse phase HPLC).

Steers were fed grass hay or red clover silage based diets containing flaxseed or sunflower seed as sources of 18:3n-3 and 18:2n-6 respectively. Hamburgers were made from triceps brachii and perirenal or subcutaneous fat. Perirenal-hamburgers contained more polyunsaturated fatty acids (PUFA), several PUFA biohydrogenation intermediates (BHI), and 18:0 (P<0.05). Oxidative stability was similar across hamburgers (P>0.05). Sensory differences were found due to hamburger fat source, but were < one panel unit. Within perirenal-hamburgers, feeding flaxseed increased 18:3n-3 and its BHI (P<0.05), and feeding sunflower seed increased 18:2n-6 and its BHI (P<0.05). Feeding flaxseed increased off-flavour intensity and oxidation in perirenal-hamburgers (P<0.05). Feeding oilseeds in forage based diets while using perirenal fat to make hamburgers provides opportunities to increase PUFA and BHI with potential to impact human health, but control measures need to be explored to limit oxidation and off-flavours when feeding flaxseed.

This study compared carcass, meat quality and fatty acid profiles of longissimus thoracis (LT) from feedlot cattle fed barley grain with or without oilseed (OS). Six diets containing no oilseed (No-OS), 10% ground flaxseed (FS), 10% high oleate sunflower seeds (SS) with or without 30% triticale dried distiller's grain (DDGS) were prepared. Feeding DDGS increased chroma at 24 and 144 h post mortem. Feeding FS increased weight% of LT PUFA (P<0.05) compared to No-OS or SS. An OS by DDGS interaction occurred for 18:3n-3 (P<0.05) where FS increased weight% of 18:3n-3 (P<0.05), a response accentuated (P<0.05) by DDGS. Feeding DDGS increased weight% of LT 18:2n-6 (P<0.05), but neither OS nor DDGS affected conjugated linoleic acid (CLA, t7,c9 &c9,t11-18:2). Feeding FS increased weight% of n-3 FA, and both FS and SS increased t10-18:1 with no effect on CLA or t11-18:1. Combination feeding of DDGS and FS further increased weight% of n-3 FA and tempered increases in t10-18:1 with no effect on CLA or t11-18:1. The findings suggest a new strategy to increase beef omega-3 fatty acids efficiently through inclusion of a combination of DDGS and FS in feedlot diet.

NIRS potential to estimate the proportion of PUFA and their biohydrogenation products in adipose tissues from cattle fed sunflower or flaxseed was tested. Immediately after skinning, perirenal and subcutaneous fat samples from 63 steers were collected, scanned intact at 37°C and 33°C, respectively, over a NIR spectral range from 400 to 2498nm using benchtop equipment and then analyzed for fatty acid composition. NIRS calibrations in perirenal fat showed high predictability for total and major omega-6 and omega-3, conjugated linolenic acids, t,t-conjugated linoleic acids (CLA), non-CLA dienes and trans-monounsaturated fatty acids, with R(2) (RMSECV, %) of 0.88-0.89 (0.16-0.20), 0.89-0.91 (0.07-0.08), 0.86-0.89 (0.01-0.09), 0.82 (0.07), 0.89 (0.46) and 0.86-0.88 (0.87-1.29), respectively. NIRS predictions in subcutaneous fat were less reliable, probably due to lower fatty acid variability. The results show NIRS to be a useful technique for the early, fast and relatively inexpensive estimation of proportions of fatty acids with potential human health effects in cattle perirenal fat.

The current study was conducted to determine the effect of different α-tocopherol (vitamin E) inclusion levels on trans(t)-18:1 and conjugated linoleic acid (CLA) profiles in subcutaneous and intramuscular fat of steers fed a barley-based diet. Fifty-six feedlot steers were offered a barley-based finisher diet (73% steam rolled barley, 22% barley silage and 5% supplement as-fed basis) with four levels of supplementary dl-α-tocopheryl acetate (340, 690, 1040 or 1740 IU/steer per day) for 120 days. Adding vitamin E to the diet had little effect on the overall fatty acid composition of intramuscular fat. The proportion of individual and total t,t- and cis(c),t-CLA, n-3 fatty acids, total polyunsaturated fatty acids (PUFA), mono-unsaturated fatty acids and saturated fatty acids to PUFA ratio in subcutaneous fat were not influenced (P > 0.05) by dietary vitamin E supplementation. Increasing levels of vitamin E led to linear reductions in t6-/t7-/t8-18:1 and t10-18:1 (P < 0.05), and linear increase in t11-/t10-18:1 ratio (P < 0.05) in subcutaneous fat. The content of 20:3n-6 and total n-6 in subcutaneous fat decreased (P < 0.05) linearly with increasing amounts of vitamin E. The subcutaneous fat n-6:n-3 ratio showed a quadratic (P < 0.05) response to vitamin E. In conclusion, although vitamin E supplementation has some potential to reduce t10-18:1 formation and increase t11-/t10-18:1 ratio in subcutaneous fat of cattle fed barley-based diets, the changes in the present study were limited and may not have been sufficient to impact on human health.

RESUMO

The fatty acid composition of beef and pork has been stigmatized due to their relationships with several diseases from cardiovascular disease to cancer. Meat lipids are, however, one of the few components of meat that can be modified in content and composition, and can present opportunities for value added production and health promotion. Until regulations and policies are in place to define requirements for fatty acid enrichment, however, the process remains relatively academic. Once practical goals are in place for fatty acid enrichment in meat, both theory and practice need to converge for successful production of fatty acid enriched meat. The present review covers aspects of policy in Canada, and requirements for research networks to respond to theoretical and practical challenges associated with production of fatty acid enriched meat. Finally, needs for education and marketing are outlined which must be in place to truly realize a transition of meat lipids from perceived disease and waste to health and opportunity.

Dietary lipid effect, as a consequence of protein supplement, on lamb m. longissimus dorsi fatty acid composition was investigated, with emphasis on biohydrogenation intermediates. Crossbred lambs (White Swedish Landrace × Texel) were fed a barley-based diet without (CON) or with protein supplements including peas (PEA), rapeseed cake (RC) or hempseed cake (HC). The HC diet resulted in the highest muscle 22:6n-3 proportion, with the RC diet being similar (P<0.05). Protein supplement did not affect the c9,t11 conjugated linoleic acid (CLA) proportion, however the HC diet increased some minor CLA isomers, including t10,c12 CLA (P<0.05). The t10-18:1 and total trans-18:1 were lowest for the RC diet (P<0.05), likely relating to rumen conditions and precursor availability. The saturated, monounsaturated and branched-chain fatty acids were largely unaffected by protein supplement. In conclusion, feeding the RC diet lowered the t10-18:1 and total trans-18:1 in meat, and modestly increased 22:6n-3 content. The direction of these changes would be beneficial, making the RC diet the preferred protein supplement; however the magnitude of the changes in the present experiment may not be sufficient to have an impact on human health.

The objective of the study was to determine temporal fat deposition and fatty acid profiles in beef cows fed hay- or barley silage-based diets, with or without flaxseed. Crossbred cull beef cows (n = 64, >30 mo of age, 620 ± 5 kg) were removed from grassland pastures, randomly assigned to 16 pens, and given ad libitum access to 50:50 (wt/wt, DM basis) forage:concentrate diets containing 0 or 15% ground flaxseed (DM basis, 5.2% added fat). Diets consisted of hay control (HC), hay+flaxseed (HF), barley silage control (SC), and silage+flaxseed (SF). Backfat biopsies were obtained from each cow at 0, 6, and 12 wk, and at slaughter (~20 wk) to assess fatty acid composition. With the exception of feed efficiency, flaxseed × forage interactions were not significant for backfat accumulation or performance parameters. Flaxseed improved (P < 0.01) feed conversion when supplemented to hay-based diet and increased ADG (P = 0.03), resulting in a heavier (P = 0.02) BW. Compared with hay, barley silage increased (P < 0.01) DMI, ADG, and feed efficiency. Subcutaneous fat contained 0.68% n-3 fatty acids at wk 0, and reached 0.68, 0.81, and 0.94% in HF cows after 6, 12, and 20 wk, respectively (Y(n-3) = 0.0133X + 0.6491, r = 0.87). It was 0.67% at wk 0, and reached 0.65, 0.77, and 0.90% in SF cows after 6, 12, and 20 wk, respectively (Y(n-3) = 0.0121X + 0.6349, r = 0.75). In contrast, weight percentage of n-3 fatty acids decreased in HC cows from 0.63, 0.50, and 0.47, to 0.43%, and in SC cows from 0.63, 0.40, and 0.36, to 0.33% over the 20 wk. A forage × flaxseed interaction (P < 0.05) occurred for many of the α-linolenic acid (ALA) biohydrogenation intermediates, including vaccenic acid (C18:1 trans-11) and CLA (combined C18:2 trans-7,cis-9 and cis-9,trans-11) in plasma, and in subcutaneous fat this also included non-CLA dienes. Concentrations of most α-linolenic acid biohydrogenation intermediates were greater when feeding flaxseed with hay. In conclusion, forage source altered plasma concentrations and rate of accumulation of ALA biohydrogenation products in subcutaneous fat from beef cows fed flaxseed. Factors responsible for this response are yet to be defined, but may include forage-mediated changes in ruminal biohydrogenation of ALA, as well as alterations in fatty acid metabolism and deposition.

This study was conducted to investigate changes in fatty acid profiles of beef cows fed grass hay or barley silage based diets, with or without flaxseed supplementation. Both flaxseed and hay feeding increased levels of α-linolenic acid (LNA; 18:3n-3) in longissimus thoracis and backfat (P<0.001). A forage type by flaxseed level interaction was observed for most LNA biohydrogenation intermediates (P<0.05) that indicated feeding hay combined with flaxseed led to the greatest levels of total conjugated linolenic acid, total conjugated linoleic acid, total non-conjugated dienes and total trans-18:1. Predominant biohydrogenation intermediates included t11,c15-18:2, rumenic acid (c9,t11-18:2) and vaccenic acid (t11-18:1).

To evaluate meat quality of beef with different α-tocopherol tissue levels, 55 feedlot steers were fed a barley-based finisher diet with four vitamin E supplementation levels (0, 350, 700 and 1400 IU DL-α-tocopheryl acetate/animal per day) for 120 days. Although the increase in oxidation levels overtime was much smaller (P < 0.001) in the high-medium and high groups, α-tocopherol tissue levels did not affect (P > 0.05) pH, proximate analysis, drip and cooking losses, and shear force of steaks. No effect of α-tocopherol tissue levels was found in retail evaluation of steaks after a short ageing time of 6 days, but with 21 days of ageing, a delay in formation of metmyoglobin (P = 0.008) was observed in steaks with higher tissue levels of α-tocopherol. Similar results were found for ground beef (25% fat) prepared from 6-day aged meat. Thus, higher α-tocopherol tissue levels protect ground beef and long-aged steaks from discolouration and lipid oxidation.

To examine the effect of co-extrusion on subsequent n-3 fatty acids in pig tissues, 8 pigs (barrows and gilts) were assigned to either a control treatment or one of nine treatments arranged in a 3 x 3 factorial design with 3 levels of co-extruded flaxseed (5%, 10% and 15%) and 3 durations of feeding (4, 8 and 12 weeks). Feed conversion improved slightly (P=0.01) with increasing dietary flaxseed but feeding flax for more than 8 weeks reduced average daily gain (P=0.02). In general, the duration and level of co-extruded flaxseed feeding affected (P<0.05) most fatty acids except for 22:6n-3 (P>0.05). Increasing the duration of flax feeding led to significant quadratic effects in backfat 18:3n-3 (P<0.001) and total n-3 fatty acids (P=0.002) when feeding 5% co-extruded flaxseed. Those increases were linear (P<0.001) when feeding 10% and 15% co-extruded flaxseed. Consequently feeding higher levels of flax for shorter periods vs. lower levels for longer periods appears to be more efficient at increasing n-3 fatty acids in pig backfat, but increases appeared to be less consistent. Moreover the addition of a 50:50 mix of extruded flax/peas to pig diets provided a highly available source of 18:3n-3 yielding n-3 fatty acid enrichments in backfat comparable to studies where extracted flaxseed oil was fed. Feeding flax co-extruded with field peas can be used to optimize consistent enrichments of n-3 fatty acids in back fat and relatively small amounts of this fat could be used to manufacture pork products to meet Canadian standards for n-3 fatty acid enrichment.

The common practice in North American feedlot industries is to add antibiotics to the diet to prevent disease and improve both BW gain and feed efficiency. In this study, 240 crossbred steer calves were backgrounded on a 54% silage diet for 80 d and fed a finishing diet consisting of 81% barley grain, 10% barley silage, and 7.5% supplement (DM basis) with and without in-feed antibiotics for approximately 120 d. Calves were assigned to 1 of 5 treatments: a control with no antibiotics, 11 mg/kg of chlortetracycline, 44 mg/kg of chlortetracycline, 44 mg/kg of chlortetracycline plus 44 mg/kg of sulfamethazine, and 11 mg/ kg of tylosin phosphate. A combination of GLC and silver-ion HPLC methods was used to analyze the fatty acid composition of brisket adipose tissue, with emphasis on trans-18:1 and CLA isomers. The inclusion of nonionophore antibiotics in the diet had little effect on the fatty acid composition, except that feeding either 44 mg/kg of chlortetracycline or 11 mg/kg of tylosin caused small increases in 9c-14:1 and 16:0 relative to the control (0.26 and 0.9 g/100 g of total fatty acids, respectively). Likewise, profiles of trans-18:1 and CLA isomers were unchanged by antibiotics, but across treatments the predominant trans-18:1 isomer was 10t-18:1 (where t = trans; 3.22%) at 3 times the concentration of the second most abundant isomer (11t-18:1; vaccenic acid, 1.05%). Rumenic acid (9c,11t-18:2, where c = cis) was the major CLA isomer at 61% of total CLA, followed by 7t,9c-18:2 at 9%. Because no other effects on fatty acid composition were evident, data for trans-18:1 and CLA were pooled across treatments to investigate possible relationships among rumen PUFA metabolites. The total trans-18:1 content in brisket adipose tissue was positively correlated with 10t-18:1, but not with 11t-18:1, whereas the total CLA was positively correlated with 9c,11t-18:2, but not with 7t,9c-18:2. The 7t,9c-18:2 was, however, positively correlated with 10t-18:1 and 6t/7t/8t-18:1 but was negatively correlated with rumenic acid. These metabolic interrelationships suggest the presence of bacterial populations with distinct pathways for PUFA biohydrogenation in which either 10t-18:1 or 11t-18:1 predominate. Overall, the nonionophore antibiotics tested did not appreciably change adipose tissue composition and consequently could not be used to improve the trans-18:1 or CLA profile (i.e., increase vaccenic and rumenic acids at the expense of 10t-18:1).

The effects of supplementing a wheat-based diet for weaner pigs with exogenous xylanase and phospholipase on ileal and faecal nutrient digestibilities and on the level of microbial metabolites in ileal digesta were examined. Fourteen piglets, weaned at 11 days, were fitted with a simple T-cannula at the distal ileum. The pigs were offered a control diet or diets supplemented with xylanase and phospholipase individually or in combination, in a two period crossover design. The combination of xylanase and phospholipase tended to increase the ileal recovery of the amino sugar galactosamine, whereas the concentration expressed in mg/kg dry matter intake of glucosamine was slightly decreased (p < 0.10). There was neither an effect of enzyme supplementation on ileal and faecal digestibility of the other nutrients and energy, nor was there an effect on pH and on the level of microbial metabolites in ileal digesta. However, an increase in ileal and faecal nutrient and energy digestibility with increasing age was observed. The ileal and faecal digestibility coefficients (except for ether extract) were significantly (p < 0.05) higher in experimental period I than in period II. These higher values may be attributed to a lower feed intake during period I. Since a lower level of feed intake is generally associated with a slower rate of passage and a longer retention time of digesta, a positive impact on digestion and absorption of nutrients can be assumed, which, on the other hand, limits the potential of additional enzyme effects.
