In floor-raised broilers, coccidiosis is responsible for reducing the use of nutrients, mainly by impairing intestinal tissue function and activating the immune system. Understanding and quantifying how balanced dietary protein (BP) is used when birds are challenged will allow nutritionists to make decisions regarding challenged flocks. This study aimed to determine the effects of Eimeria maxima on broiler performance and body composition, and to calculate changes in the maintenance and efficiency of protein utilisation (Ep). A total of 2 400 male 14-day-old Cobb500 broiler chickens were randomly allotted to ten groups with six replications of 40 birds each, with a 5 × 2 factorial arrangement of treatments. Five levels of BP in reference to digestible lysine (3.6, 7.2, 10.8, 14.4, and 18.0 g/kg) were fed to unchallenged (NCH) and challenged (CH) broilers with 7 × 103E. maxima sporulated oocysts from 14 to 28 days of age. Performance and body deposition were measured using a comparative slaughter technique to compare BP maintenance requirements and Ep. ANOVA followed by a posthoc test was performed to compare the effects of BP levels, challenge, and their interactions. A monomolecular model describing the responses of NCH and CH broilers to BP intake, maintenance, and maximum protein deposition was compared. There were significant interactions between body weight gain and digestible lysine intake among the factors studied. Infection had a negative impact on all variables analysed, proving the efficacy of the challenge. The maintenance did not differ between the CH and NCH groups. Increased levels of dietary BP did not recover the maximum protein deposition in CH broilers. Eimeria maxima significantly reduced Ep by a factor of 0.09 times on Ep compared to the control group. The Eimeria maxima challenge was responsible to modify the use of BP altering the body composition and impairing broilers performance.

The aim of this study was to evaluate two sources of oil (soybean and fish) and four additional levels of vitamin E (0, 150, 250 and 350mg/kg diet) in breeder diets between the 42nd and 56th week of age and its effect on performance and meat quality of offspring. The supplementation of fish oil in the maternal diet increased the deposition of DHA and CLA in egg yolk. From 1 to 35 days of age, intermediate levels of vitamin E resulted in lower weight gain and feed intake. To feed conversion, the best result was obtained supplementing the maternal diet with soybean oil, regardless of vitamin inclusion. In addition, the evaluation of meat quality, the fish oil-supplemented-diet resulted in lower moisture loss of the breast fillets. For maternal diet supplemented with fish oil, the inclusion of vitamin E resulted in a linear increase of the pH value of the meat. Greater levels of vitamin E in the maternal diet influenced the functional properties of meat according to the type of oil that was added to the diet. The deposition of different fatty acids in the embryonic egg yolk can directly influence the meat quality of offspring.(AU)


O objetivo deste estudo foi avaliar duas fontes de óleo (soja e peixe) e quatro níveis adicionais de vitamina E (0, 150, 250 e 350mg/kg dieta), em dietas de matrizes entre a 42º e a 56º semana de idade, sobre o desempenho e a qualidade de carne da progênie. A suplementação de óleo de peixe na dieta das matrizes aumentou a deposição de DHA e CLA na gema dos ovos. Entre um e 35 dias de idade, níveis intermediários de vitamina E resultaram em menor ganho de peso e consumo de ração. Para a conversão alimentar, o melhor resultado foi obtido suplementando a dieta materna com óleo de soja, independentemente da inclusão vitamínica. Dietas suplementadas com óleo de peixe resultaram em menor perda de umidade nos filés de peito. Para a dieta materna suplementada com óleo de peixe, a inclusão de vitamina E resultou em um aumento linear do valor de pH da carne. Níveis mais altos de vitamina E na dieta materna influenciaram as propriedades funcionais da carne de acordo com o tipo de óleo adicionado à dieta. A deposição de diferentes ácidos graxos na gema de ovos embrionados pode influenciar diretamente a qualidade da carne da progênie.(AU)

O objetivo deste estudo foi avaliar o efeito do fornecimento de dietas pós-eclosão suplementadas com diferentes fontes de gordura insaturada e adicionadas ou não de taurina e glicina sobre o desempenho produtivo, a biometria e a morfometria do intestino delgado de pintos de corte de um a 21 dias de idade. Foram utilizados 480 pintos de corte machos de um dia de idade da linhagem Cobb. O delineamento foi inteiramente ao acaso, em esquema fatorial 2 x 4, com e sem suplementação de taurina e glicina e quatro dietas (controle, óleo de peixe, de soja e de girassol), totalizando oito tratamentos com seis repetições de 10 aves cada. As rações experimentais foram fornecidas de zero a quatro dias de idade. O desempenho zootécnico foi avaliado ao alojamento e aos quatro, sete e 21 dias de idade. Nestas mesmas datas, foram sacrificadas duas aves por unidade experimental para biometria do intestino e histomorfometria da mucosa do intestino. A adição de diferentes fontes de gordura e a suplementação de glicina e taurina às dietas de transição não influenciaram o desempenho produtivo de um a 21 dias. A suplementação das dietas com glicina e taurina alterou a morfologia da mucosa intestinal, principalmente do duodeno, resultando em maior comprimento do vilo e relação vilo:cripta. Entretanto, parte dos efeitos positivos depende do tipo de óleo adicionado, mostrando que dietas pós-eclosão acrescidas de fontes de lipídios podem ser benéficas no desenvolvimento da capacidade funcional do intestino de frangos de corte.(AU)


The aim of this study was to assess post-hatch diets supplemented with different sources of unsaturated fat and added or not with taurine and glycine on the productive performance, biometry and morphology of small intestine of chicks from 1 to 21 days of age. Four hundred and eighty (480) one day old male broiler Cobb chicks were used. The experimental design was completely randomized in a factorial 2 x 4, with and without supplemental taurine and glycine and 4 diets (control, fish, soy and sunflower oil), totaling six treatments with six repetitions of 10 birds each. The experimental diets were supplied from 0 to 4 days old. The performance was evaluated in housing and 4, 7 and 21 days of age. On these same dates, 2 birds per experimental unit were sacrificed for gut biometrics and histomorphometry of intestinal mucosa. The addition of different sources of fat, glycine and taurine supplementation on transition diets did not influence productive performance from 1 to 21 days. Supplementation of diets with glycine and taurine altered the morphology of the intestinal mucosa, mainly of the duodenum, resulting in greater length of villi and villi: crypt ratio. However, the positive effects depend on the type of oil added, showing that post-hatch diets increased with lipid sources may be beneficial in the development of the functional capacity of the intestine of broilers.(AU)

The impact on cell wall (CW) of the deficiency in nitrogen (-N), phosphorus (-P) or sulphur (-S), known to impair essential metabolic pathways, was investigated in the economically important fruit species Vitis vinifera L. Using cuttings as an experimental model a reduction in total internode number and altered xylem shape was observed. Under -N an increased internode length was also seen. CW composition, visualised after staining with calcofluor white, Toluidine blue and ruthenium red, showed decreased cellulose in all stresses and increased pectin content in recently formed internodes under -N compared to the control. Using CW-epitope specific monoclonal antibodies (mAbs), lower amounts of extensins incorporated in the wall were also observed under -N and -P conditions. Conversely, increased pectins with a low degree of methyl-esterification and richer in long linear 1,5-arabinan rhamnogalacturonan-I (RG-I) side chains were observed under -N and -P in mature internodes which, in the former condition, were able to form dimeric association through calcium ions. -N was the only condition in which 1,5-arabinan branched RG-I content was not altered, as -P and -S older internodes showed, respectively, lower and higher amounts of this polymer. Higher xyloglucan content in older internodes was also observed under -N. The results suggest that impairments of specific CW components led to changes in the deposition of other polymers to promote stiffening of the CW. The unchanged extensin amount observed under -S may contribute to attenuating the effects on the CW integrity caused by this stress. Our work showed that, in organized V. vinifera tissues, modifications in a given CW component can be compensated by synthesis of different polymers and/or alternative linking between polymers. The results also pinpoint different strategies at the CW level to overcome mineral stress depending on how essential they are to cell growth and plant development.

The effects of using plant ingredients in Senegalese sole (Solea senegalensis) diet on immune competence and intestine morphology and microbial ecology are still controversial. Probiotics or immunostimulants can potentially alter the intestinal microbiota in a way that protects fish against pathogens. The current study aimed to examine the intestine histology and microbiota and humoral innate immune response in juvenile sole fed diets with low (35 %) or high (72 %) content of plant protein (PP) ingredients supplemented with a multispecies probiotic bacteria or autolysed yeast. Fish fed the probiotic diet had lower growth performance. Lysozyme and complement activities were significantly higher in fish fed PP72 diets than in their counterparts fed PP35 diets after 17 and 38 days of feeding. At 2 days of feeding, fish fed unsupplemented PP72 showed larger intestine section area and longer villus than fish fed unsupplemented PP35. At 17 days of feeding, fish fed unsupplemented PP72 showed more goblet cells than the other dietary groups, except the group fed yeast supplemented PP35 diet. High dietary PP level, acutely stimulate fish innate immune defence of the fish after 2 and 17 days of feeding. However, this effect does not occur after 73 days of feeding, suggesting a habituation to dietary treatments and/or immunosuppression, with a reduction in the number of the goblet cells. Fish fed for 38 days with diets supplemented with autolysed yeast showed longer intestinal villus. The predominant bacteria found in sole intestine were Vibrio sp. and dietary probiotic supplementation caused a reduction in Vibrio content, regardless of the PP level.

The aim of this study was to evaluate the regeneration of the intestinal mucosa in Eimeria and E. coli challenged broilers supplemented with glutamine, arginine, and threonine. Six hundred male broilers at one d of age from the Cobb strain were utilized. The design was completely randomized using a 2×3 factorial design (unchallenged and challenged and 3 diets). A commercial diet was used as a control and 2 other diets were formulated with glutamine (1.5 and 3% Aminogut®), arginine (1 and 2% L-Arginine), and threonine (1 and 2% L-threonine). The animals that consumed diets supplemented with amino acids presented better (P<0.05) feed conversion in the period from one to 42 d of age. The ability of cell proliferation and the villus:crypt ratio in response to enteric challenge were greater (P<0.05) for broilers that received diets supplemented with amino acids. High levels of amino acids in the experimental feeds reflected in greater protein levels in poultry house litter, and they did not interfere with ammonia production. The supplementation of diets with trophic amino acids can positively contribute to the regeneration and proliferation of the intestinal mucosa in broilers and to the maintenance of zootechnical performance when submitted to enteric challenges.

Toxic effects of ultraviolet (UV) radiation on skin include protein and lipid oxidation, and DNA damage. The latter is known to play a major role in photocarcinogenesis and photoaging. Many plant extracts and natural compounds are emerging as photoprotective agents. Castanea sativa leaf extract is able to scavenge several reactive species that have been associated to UV-induced oxidative stress. The aim of this work was to analyze the protective effect of C. sativa extract (ECS) at different concentrations (0.001, 0.01, 0.05 and 0.1 µg/mL) against the UV mediated-DNA damage in a human keratinocyte cell line (HaCaT). For this purpose, the cytokinesis-block micronucleus assay was used. Elucidation of the protective mechanism was undertaken regarding UV absorption, influence on (1)O2 mediated effects or NRF2 activation. ECS presented a concentration-dependent protective effect against UV-mediated DNA damage in HaCaT cells. The maximum protection afforded (66.4%) was achieved with the concentration of 0.1 µg/mL. This effect was found to be related to a direct antioxidant effect (involving (1)O2) rather than activation of the endogenous antioxidant response coordinated by NRF2. Electrochemical studies showed that the good antioxidant capacity of the ECS can be ascribed to the presence of a pool of different phenolic antioxidants. No genotoxic or phototoxic effects were observed after incubation of HaCaT cells with ECS (up to 0.1 µg/mL). Taken together these results reinforce the putative application of this plant extract in the prevention/minimization of UV deleterious effects on skin.

The toxicity of organic plant extracts to Sitophilus zeamais Motschulsky (Coleoptera: Curculionidae) was assessed for three tropical plant species: branches, leaves, and seeds of Annona montana (Annonaceae), branches of Aristolochia paulistana (Aristolochiaceae), and leaves and branches of Casearia sylvestris (Salicaceae). The screening assay resulted that the extracts of A. montana seeds obtained with hexane (LC50 = 534.75 mg kg(-1) and LT50 = 6.10 days) and with dichloromethane (LC50 = 424.67 mg kg(-1) and LT50 = 5.03 days) were the most promising treatments, followed by the extract prepared from A. montana leaves with hexane (LC50 = 837.70 mg kg(-1) and LT50 = 4.90 days). Moreover, extracts (at 1,500 mg kg(-1)) prepared from C. sylvestris branches with dichloromethane and A. paulistana with hexane caused significant mortality (37% and 41.5%, respectively) beyond sublethal effects on S. zeamais. Therefore, based on the biological assays, extraction yield, and evaluation of the chromatographic profile of the crude extracts by TLC, the hexane extract of A. montana seeds was selected and fractioned using liquid-liquid partitioning. The hydroalcoholic fraction caused mortality of 55.5%, significantly superior to dichloromethane fraction, which caused 35.5% of mortality. Chemical analyses ((1)H NMR, HPLC, and TLC) were performed, and the results showed the presence of alkaloids and acetogenins in both active fractions, which have been associated with bioactivity. Therefore, extracts prepared from A. montana seeds (especially) is an interesting source of new compounds with promising grain-protectant properties.

Pejerrey (Odontesthes bonariensis) is a native species from Rio Grande do Sul, Uruguay and Argentina where it is of great economic importance for artisanal fishing. One difficulty in laboratory research with pejerrey is related to its sensitivity, as it presents higher basal cortisol levels than other freshwater species. For this reason, the aim of this work was to evaluate the efficiency of benzocaine and clove oil as anesthetics for pejerrey fingerlings. Two experiments were done where fingerlings (57±7.8mm and 1.1±0.44g) were exposed to benzocaine with concentrations between 40mgL-1 and 120mgL-1 and to clove oil with concentrations between 12mgL-1 and 75mgL-1. Survival, anesthesia induction time and recovery time for each pharmaceutics were evaluated. Both benzocaine and clove oil pharmaceutics showed efficiency as anesthetics for pejerrey fingerlings, with negative correlation between the dose of anesthetics and the anesthesia induction time. For benzocaine, the concentrations between 80mgL-1 and 100mgL-1 showed better results, as for clove oil the optimal concentrations were between 25mgL-1 and 50mgL-1. On the other hand, the anesthesia recovery time did not present significant variation on the different concentrations of the tested products. The tested products are highly metabolizable by pejerrey.


O peixe-rei (Odontesthes bonariensis) é uma espécie nativa do Rio Grande do Sul, Uruguai e Argentina, onde tem grande importância econômica para a pesca artesanal. Uma dificuldade da pesquisa em laboratório com peixe-rei está relacionada à sua sensibilidade, pois apresenta níveis basais de cortisol mais elevados que outras espécies de água doce. Este trabalho avaliou a eficiência da benzocaína e do óleo de cravo como anestésicos para alevinos de peixe-rei. Foram realizados dois experimentos em que alevinos (57±7,08mm e 1,1±0,44g) foram expostos à concentração entre 40mg-1 e 120mgL-1 de benzocaína e entre 12mgL-1 e 75mgL-1 de óleo de cravo. Avaliaram-se a sobrevivência, o tempo de anestesia e o tempo de recuperação para cada um dos fármacos. Ambos os fármacos, benzocaína e óleo de cravo, mostraram eficiência para anestesiar peixe-rei, com correlação negativa entre a dose e o tempo de indução de anestesia. Para benzocaína, concentrações entre 80mgL-1 e 100mgL-1 mostraram melhor resultado, enquanto para óleo de cravo as melhores concentrações ficaram entre 25mgL-1 e 50mgL-1. Por outro lado, o tempo de recuperação do estado de anestesia não apresentou variação significativa nas diferentes concentrações testadas. O peixe-rei tem elevada capacidade de metabolização dos produtos testados.

Acitretin is currently used alone or combined with PUVA (psoralen + UVA) or with narrow-band ultraviolet B (NBUVB), to treat moderate and severe psoriasis. However, little is known about the potential genotoxic/carcinogenic risk and the cytostatic/cytotoxic effects of these treatments. Our aim was to study the cytotoxic and genotoxic effects of acitretin - alone or in combination with PUVA or NBUVB - by performing studies with blood from patients with psoriasis vulgaris who were treated with acitretin, acitretin+PUVA or acitretin+NBUVB for 12 weeks, and in vitro studies with blood from healthy volunteers, which was incubated with acitretin at different concentrations. The cytotoxic and genotoxic effects were evaluated by the cytokinesis-blocked micronucleus test and the comet assay. Our results show that psoriatic patients treated with acitretin alone or with acitretin+NBUVB, did not show genotoxic effects. In addition, these therapies reduced the rate of proliferation and induced apoptosis and necrosis of lymphocytes; the same occurred with lymphocyte cultures incubated with acitretin (1.2-20µM). The acitretin+PUVA reduced also the proliferation rate, and increased the necrotic lymphocytes. Our studies suggest that therapy with acitretin alone or combined with NBUVB, as used in psoriatic patients, does not show genotoxic effects, reduces the rate of proliferation and induces apoptosis and necrosis of lymphocytes. The combination of acitretin with PUVA also reduces the proliferation rate and increases the number of necrotic lymphocytes. However, as it induced slight genotoxic effects, further studies are needed to clarify its genotoxic potential.

Determinou-se a melhor relação vitamina E:vitamina C em dietas para frangos de corte, visando ao melhor desempenho produtivo e à melhor qualidade da carne das aves submetidas ao estresse pré-abate. Utilizaram-se 800 pintos de corte, machos, distribuídos em delineamento inteiramente ao acaso, em esquema fatorial 2x4, com dois níveis de suplementação de vitamina E - 0 e 250mg/kg - e quatro de vitamina C - 0, 150, 300 e 450mg/kg. Aos 42 dias de idade, 12 horas ante mortem, amostras de aves de cada tratamento foram submetidas ao estresse por calor e, em seguida, pelo transporte. Foram avaliadas características de desempenho - peso vivo, consumo de ração e conversão alimentar - , bem como rendimento de carcaça e qualidade da carne de peito e coxas - perda de água, cor e pH. Os níveis de vitaminas avaliadas não influenciaram nas características de desempenho avaliadas. Houve menor rendimento de peito (34,2 vs. 34,9%) e maior pH inicial (6,3 vs. 6,1) dos cortes, para aves que sofreram estresse em relação àquelas que não foram submetidas ao estresse pré-abate.


The best relationship for vitamin E:vitamin C in diets for broilers regarding growth performance and meat quality of birds submitted to pre-slaughter stress was determined. 800 male chicks at one day of age were distributed in a completely randomized design in a 2x4 factorial scheme, with two levels of vitamin E supplementation - 0 and 250mg/kg - and four of vitamin C - 0, 150, 300 and 450mg/kg. At 42 days of age, 12 hours ante-mortem, samples of birds from each treatment were submitted to heat stress and then transportation. The performance characteristics evaluated were body weight, feed intake and feed:gain ratio, carcass yield and meat quality of breast and thighs, water loss, color and pH. The levels of vitamins evaluated did not influence the performance characteristics measured. There was a lower breast yield (34.2 vs 34.9%) and higher initial pH (6.3 vs 6.1) in the cuts for birds that suffered stress than for those who did not undergo pre-slaughter stress.

The nutcracker syndrome (NCS) is a rare cause of haematuria. It embraces an extended nonpathognomonic spectrum of symptoms that imply a difficult diagnosis. Ultimately it may be associated with substantial morbidity and even life-threatening events. We report a rare cause if a 12-year-old boy who presented with a history of frequent intermittent episodes of painless constant haematuria. The cystoscopy showed a bloody urine ejaculate from the left ureter meatus. The Doppler ultrasonography showed turbulent pattern of venous blood flow of the posterior renal vein branch behind the aorta. The abdominopelvic computer tomography (apCT) revealed left renal vein (LRV) duplication with a dilated retroaortic branch, entrapped between the aorta and the vertebral column, promoting the renal nutcracker syndrome. The patient was initially hospitalized and managed with oral iron supplements and continuous saline bladder irrigation, not requiring additional treatment. The child is currently asymptomatic, with haemoglobin value returning to normal and therefore proposed to conservative management with close followup. The authors present a case report of episodic haematuria caused by a rare entity-posterior nutcracker syndrome with renal vein duplication.

Determinaram-se os níveis nutricionais de cálcio (Ca) para aves, machos e fêmeas, da linhagem ISA Label, nas fases inicial (um a 28 dias), crescimento (28 a 56 dias) e final (56 a 84 dias). Foram realizados três ensaios, um para cada fase, e, em cada ensaio, 480 aves com idade correspondente à fase de criação foram alojadas em 24 unidades experimentais com áreas de abrigo e de pastejo. Foi utilizado delineamento inteiramente ao acaso, em esquema fatorial 4x2 (Ca e sexo), totalizando oito tratamentos com três repetiç ões de 20 aves. Avaliaram-se: ganho de peso (GP); consumo de dieta (CD); conversão alimentar (CA); teores de fósforo (PT), de cálcio (CaT) e de cinzas na tíbia (CT) e resistência à quebra óssea (RQO). Na fase inicial, recomenda-se 1,16 por cento de Ca na dieta, para aves de ambos os sexos, na fase de crescimento, 0,78 e 0,88 por cento de Ca para machos e fêmeas, respectivamente, e, na fase final, 0,69 por cento de Ca na dieta para ambos os sexos.


The nutritional calcium (Ca) levels were determined for ISA Label broilers in the starter (1 to 28 days), growing (28 to 56 days), and finishing (56 to 84 days) phases. Three trials were conducted, one for each phase and in each trial, 480 birds with age corresponding to the phase were housed in 24 experimental units with shelter and pasture areas. The experimental design was completely randomized in a factorial arrangement of 4x2 (Ca levels and sex), totaling eight treatments with three replicates of 20 birds. Body weight gain (BWG); feed intake (FI); feed per gain (FG); contents of phosphorus (TP), calcium (TCa) and ash (TA) in tibia; and bone breaking strength (BSB) were evaluated. At the starter phase, 1.16 percent of Ca is recommended in the diet for birds of both sexes. At the growing phase, 0.78 and 0.88 percent of Ca for males and females, respectively, are recommended and at the finishing phase, the level of 0.69 percent of Ca is recommended in diet for both sexes.

Intense physical activity is associated with biological adaptations involving hormones and trace elements. Zinc supplementation may affect plasma copper concentration, thyroid-stimulating hormone (TSH), thyroid hormones, insulin, and glucose homeostasis, but data in athletes are scarce. The aim of this study was to evaluate in competitive athletes (cyclists, n = 7, 32 ± 8 years) the effect of zinc supplementation (22 mg/day as zinc gluconate) during 30 days, and discontinuation using placebo (maltodextrin) during the following 30 days, on plasma zinc and copper concentrations, serum thyroid hormones, insulin and glucose levels, and HOMA2-IR. Compared to baseline, plasma zinc and Zn:Cu plasma ratio increased, but plasma copper decreased after zinc supplementation (day 30) and discontinuation (day 60) (p < 0.05). Zn supplementation and discontinuation had no effect on TSH, T3, and T4. Fasting serum insulin and HOMA2-IR increased (27% and 47%, respectively) on day 60 compared to baseline (p = 0.03), suggesting a delayed effect of zinc supplementation. Moreover, plasma zinc was positively associated with serum insulin (r = 0.87, p = 0.009) and HOMA2-IR (r = 0.81, p = 0.03) after zinc supplementation (day 30), indicating that supplemental zinc may impair glucose utilization in cyclists.

Foram utilizados 240 alevinos de carpa húngara, com peso médio de 3,42±0,32g, distribuídos em delineamento inteiramente ao acaso, com quatro tratamentos e quatro repetições. A dieta basal foi formulada com ingredientes de origem vegetal (36 por cento de proteína bruta e 3.200kcal de energia digestível). Os tratamentos corresponderam a quatro níveis de fitase ativa (UF) por kg na dieta: zero (controle), 500, 1000 e 1500UF/kg. A cada 15 dias, foi realizada uma biometria e, ao término do período experimental (45 dias), 5 por cento dos animais foram eutanasiados para determinação do rendimento de carcaça e composição químico-bromatológica. Os parâmetros de desempenho avaliados foram: peso final, ganho de peso, comprimento total, taxa de crescimento específico e sobrevivência. A suplementação de fitase não influiu no desempenho produtivo, entretanto melhorou significativamente as características da carcaça.

Basal diet consisted of vegetal ingredients (36 percent crude protein and 3,220kcal of digestible energy). Treatments corresponded to four levels of active phytase (UF) per kg of diet: zero (control), 500, 1,000, and 1,500. Every 15 days, a biometry was made and at the end of the experimental period (45 days) 5 percent of animals were euthanized to determine carcass yield and chemical-bromatologic composition. The following variables were evaluated: final weight, weight gain, total length, specific growth rate, and survial. There was no effect of phytase on productive performance; however, it significantly improved carcass characteristics.

The current drug research techniques, combinatorial synthesis and high throughput screening, enabled the obtaining and pre-evaluation of thousands of compounds in short time. In order to chose the best hits to become leads, observation of drug-likeness tries to optimize this selection. Probably, the most widely used filter is Lipinski's Rule-of-five, which proposes that molecules with poor permeation and oral absorption have molecular weight > 500, Clog P > 5, hydrogen-bond donor > 5 and hydrogen-bond acceptor > 10. In order to evaluate the Rule-of-five, the top pharmaceutical products in 2007 were analyzed. Among 60 drugs, 7 (atorvastatin, montelukast, docetaxel, telmisartan, tacrolimus, leuprolide and olmesartan) did not fit the rule, and 5 failed only one of the threshold values. It was possible to conclude that the rule is very useful to select better compounds in chemolibraries, but it must be used carefully and with criteria, to avoid a possible exclusion of promising compounds.

This work aimed at evaluating the effects of the supplementation of starter diet with Arg on breast muscle development in broilers and the activation of satellite cells and the aggregation of myofibrillar protein. Male Cobb chicks (n = 990) were randomly assigned to 1 of 5 treatments in a complete random design. Measurements of 33 chicks per treatment were made in 6 repetitions. The treatments consisted of a basal diet with 1.390% digestible Arg (without supplementation) and 4 dietary levels of Arg (1.490, 1.590, 1.690, and 1.790%) with Arg:Lys ratios of 1.103, 1.183, 1.262, 1.341, and 1.421, respectively. Arginine supplementation was used only in the starter phase (1 to 21 d). Dietary supplementation with Arg had a positive effect (P < 0.05) on breast and breast fillet weight on d 7 and 21 and on myofiber diameter on d 14 and 21. However, no effect was observed (P > 0.05) on the protein:DNA ratio, which demonstrates that Arg does not interfere with the mitotic activity of the satellite cells. Independently from mechanism, Arg affected muscle growth in the starter phase positively. Dietary supplementation with Arg in the starter phase had no effect (P > 0.05) on the carcass yield of broilers on d 42. Diet supplementation with Arg at levels above the ones recommended for the starter phase may be necessary for improved muscle development in broilers.

Triplicate groups of juvenile suribim were fed for 183 days one of four different isonitrogenous (47.6% crude protein) and isolipidic (18.7% lipid) diets formulated using three different lipid sources: 100% fish oil (FO, diet 1); 100% pig lard (L, diet 2); 100% soybean oil (SO, diet 3), and FO/L/SO (1:1:1, w/w/w; diet 4). The tissue levels of fatty acids 18:2n-6 and 18:3n-3 decreased relative to corresponding dietary fatty acid values. The 20:5n-3 and 22:6n-3 composition of muscle and liver neutral lipids were linearly correlated with corresponding dietary fatty acid composition. In contrast, the 22:6n-3 composition of the brain and eye were similar among treatments. The 22:6n-3 level was enriched in all tissues, particularly in the neural tissues. Similar results were observed for tissue polar lipids: fatty acids content reflected dietary composition, with the exception of the 22:6n-3 level, which showed enrichment and no differences between groups. Given these results, the importance of the biochemical functions (transport and/or metabolism) of 22:6n-3 in the development of the neural system of surubim warrants further investigation.

An improved gas chromatography-mass spectrometry (GC-MS) method to determine acrylamide (AA) in coffee and coffee products was developed. The method was based on two main purification steps: the first with ethanol and Carrez solutions in order to precipitate polysaccharides and proteins, respectively; and the second with a layered solid-phase extraction (SPE) column which proved to be efficient in the elimination of the main chromatographic interferences. The method is applicable to a wide range of coffee products. Twenty-six samples of different coffee products were analysed. The levels of AA were in the range 11.4-36.2 microg l-1 for 'espresso coffee' and 200.8-229.4 microg l-1 for coffee blends with cereals. The results indicate that the presence of cereals significantly increased the levels of AA.

Though used occasionally, systemic therapies in severe childhood psoriasis have not been systematically investigated. Cyclosporin A (CysA) is effective in adults with severe psoriasis but there are no extensive data regarding the efficacy and safety of its use in childhood psoriasis. In this paper, we describe six children aged between 11 months and 13 years (average: 7.6 years) treated with CysA microemulsion formulation for severe psoriasis, who had been unresponsive to other treatments. The CysA dose ranged from 2 to 4 mg/kg/day, for periods varying from 8 to 105 weeks (mean: 54 weeks). Dose tapering was gradual after lesion improvement and adjusted according to clinical response. Adjuvant therapy with topical steroids, vitamin D3 ointments, coal tar preparations or anthralin was used in all children. Acitretin was used in three patients for short periods. The children were regularly monitored for serum renal and liver function and blood pressure. Improvement of skin lesions was achieved after between 4 and 30 (mean: 12) weeks of treatment, with complete remission in three children. Relapse of lesions occurred in the other children during CysA reduction, but they responded to a dose increase. The treatment was found to be well tolerated and with no significant side-effects. CysA can be used in carefully selected and monitored patients and may represent an alternative tool for severe episodes of psoriasis in children, when other therapies are unsuccessful.
