RESUMO

Aflatoxin B1 (AFB1), a ubiquitous mycotoxin in corn-based animal feed, particularly in tropical regions, impairs liver function, induces oxidative stress and disrupts cellular pathways, potentially worsening bone health in modern broilers. A 19-day experiment was conducted to investigate the effects of feeding increasing levels of AFB1-contaminated feed (<2, 75-80, 150, 230-260 and 520-560 ppb) on bone mineralization markers in broilers (n = 360). While growth performance remained unaffected up to Day 19, significant reductions in tibial bone ash content were observed at levels exceeding 260 ppb. Micro-computed tomography results showed that AFB1 levels at 560 ppb significantly decreased trabecular bone mineral content and density, with a tendency for reduced connectivity density in femur metaphysis. Moreover, AFB1 above 230 ppb reduced the bone volume and tissue volume of the cortical bone of femur. Even at levels above 75 ppb, AFB1 exposure significantly downregulated the jejunal mRNA expressions of the vitamin D receptor and calcium and phosphorus transporters. It can be concluded that AFB1 at levels higher than 230 ppb negatively affects bone health by impairing bone mineralization via disruption of the vitamin D receptor and calcium and phosphorus homeostasis, potentially contributing to bone health issues in broilers.

Endoplasmic reticulum stress (ERS) and apoptosis of nucleus pulposus (NP) cells are considered to be the main pathological factors of intervertebral disc degeneration (IDD). Fucoxanthin (FX), a marine carotenoid extracted from microalgae, has antioxidant, anti-inflammatory, and anticancer properties. The aim of this study was to investigate the effect of FX on NP cells induced by oxidative stress and its molecular mechanism. Primary NP cells of the lumbar vertebrae of rats were extracted and tested in vitro. qRT-PCR, western blot, immunofluorescence, and TUNEL staining were used to detect apoptosis, ERS, extracellular matrix (ECM), and Sirt1-related pathways. In vivo experiments, the recovery of IDD rats was determined by X-ray, hematoxylin and eosin, Safranin-O/Fast Green, Alcian staining, and immunohistochemistry. Our study showed that oxidative stress induced ERS, apoptosis, and ECM degradation in NP cells. After the use of FX, the expression of Sirt1 was up-regulated, the activation of PERK-eIF2α-ATF4-CHOP was decreased, and apoptosis and ECM degradation were decreased. At the same time, FX improved the degree of disc degeneration in rats in vivo. Our study demonstrates the effect of FX on improving IDD in vivo and in vitro, suggesting that FX may be a potential drug for the treatment of IDD.

An experiment was conducted to evaluate effects of phytase and coccidial vaccine on growth performance, bone ash, bone 3-D microstructure, nutrient digestibility, and gene expression of intestinal biomarkers in broilers fed a regular or nutrient-reduced diet. The experiment was conducted in a 2 × 4 factorial arrangement with 6 replicates per treatment and 10 birds per replicate. Two main factors were coccidial vaccine and dietary treatments. The dietary treatments included: 1) a positive control (PC; normal nutrient levels); 2) a negative control (NC; with a reduction of 0.15% of Ca and avP and 5% of essential amino acid (EAA) and crude protein relative to PC); 3) NC + 500 FTU/kg of phytase; and 4) NC + 1,500 FTU/kg of phytase. No interaction effect of phytase and coccidial vaccine on growth performance, bone ash, and apparent ileal digestibility (AID) was observed. For the main effect, birds fed the NC diet showed lower (P = 0.007) BWG during d 0 to 21 compared to PC birds, whereas supplementing 500 or 1,500 FTU/kg phytase increased BWG to the similar level to the PC. During d 0 to 21, vaccinated birds had a lower (P < 0.001) FI and better (P = 0.045) FCR compared to unvaccinated birds. Birds fed the NC diet resulted a decrease in tibia fat-free dry bone weight (P = 0.012), ash weight (P = 0.005), ash percentage (P < 0.001), and ash concentration (P = 0.019) compared to the PC group at d 21, whereas supplementing phytase at 500 or 1,500 FTU/kg in NC diet was able to improve these bone parameters to the similar level to the PC; however, vaccination did not have any effect on bone ash. Similarly, birds fed with the NC diet showed had significant lower bone microstructure levels including bone volume, bone mineral density, and bone mineral content (P < 0.001), and supplementing phytase at 1,500 FTU/kg improved these parameters. Vaccination improved AID of nitrogen (P < 0.001). Birds from the NC and both phytase supplementation groups had a higher (P = 0.001) AID of Ca compared to the PC. Supplementing phytase at 500 FTU/kg or 1,500 FTU/kg improved (P < 0.001) AID of P compared to the NC. Additionally, the NC had a lower AID of DM than the PC, whereas supplementing phytase at 500 FTU/kg or 1,500 FTU/kg improved DM digestibility (P = 0.0299). In conclusion, supplementation of phytase at 500 or 1,500 FTU/kg improved growth performance, bone mineralization, and nutrient digestibility regardless of vaccination, with a more pronounced effect when supplementing phytase at 1,500 FTU/kg.

BACKGROUND: Propofol-based sedations are widely used in elderly patients for endoscopic retrograde cholangiopancreatography (ERCP) procedure, but respiratory depression and cardiovascular adverse events commonly occur. Magnesium administered intravenously can alleviate pain and decrease propofol requirements during surgery. We hypothesized that intravenous magnesium was used as adjuvant to propofol might be beneficial in elderly patients undergoing ERCP procedures. METHODS: Eighty patients aged from 65 to 79 years who were scheduled for ERCP were enrolled. All patients were intravenously administered 0.1 µg/kg sufentanil as premedication. The patients were randomized to receive either intravenous magnesium sulfate 40 mg/kg (group M, n = 40) or the same volume of normal saline (group N, n = 40) over 15 min before the start of sedation. Intraoperative sedation was provided by propofol. Total propofol requirement during ERCP was the primary outcome. RESULTS: The total propofol consumption were reduced by 21.4% in the group M compared with the group N (151.2 ± 53.3 mg vs. 192.3 ± 72.1 mg, P = 0.001). The incidences of respiratory depression episodes and involuntary movement were less in the group M than those in the group N (0/40 vs. 6/40, P = 0.011; 4/40 vs. 11/40, P = 0.045; respectively). In the group M, the patients experienced less pain than those in the group N at 30 min after the procedure (1 [0-1] vs. 2 [1-2], P < 0.001). Correspondingly, the patients' satisfaction was clearly higher in the group M (P = 0.005). There was a tendency towards lower intraoperative heart rate and mean arterial pressure in group M. CONCLUSIONS: A single bolus of 40 mg/kg of intravenous magnesium can significantly reduce propofol consumption during ERCP, with higher sedation success and lower adverse events. OBJECTIVE: To observe the clinical efficacy and safety of the short-term administration of different doses of calcium polystyrene sulfonate in the treatment of hyperkalemia in patients with stage 3-5 non-dialysis chronic kidney disease. METHODS: A prospective, open, randomized, controlled, single-center clinical observation was conducted. In total, 107 patients were randomly assigned to receive calcium polystyrene sulfonate at 15 (group A) or 30 mg/day (group B) for 1 week. Patients were assessed on days 0, 3, and 7. RESULTS: After 3 days of treatment, the serum potassium levels in groups A and B had decreased by 0.68 ± 0.46 and 0.75 ± 0.43 mmol/L, respectively. After 7 days, the serum potassium levels in groups A and B had decreased by 0.64 ± 0.37 and 0.94 ± 0.49 mmol/L, respectively. Conversely, serum sodium, phosphorus, and calcium levels did not significantly change during the treatment period. Constipation was the most common adverse drug reaction, and no treatment-related serious adverse events were observed. CONCLUSION: Calcium polystyrene sulfonate administered at a dose of 15 or 30 g/day can rapidly reduce potassium levels in patients with stage 3-5 non-dialysis chronic kidney disease without adverse effects on sodium, phosphorus, or calcium levels.

This study aimed to investigate the effect of metabolizable energy (ME) levels and exogenous emulsifier supplementation on growth performance, apparent ileal digestibility (AID), body composition, and carcass yield in broilers. The experiment was designed as a 2  ×  2 factorial arrangement with ME levels (control ME vs. reduced 100 kcal/kg ME) and exogenous emulsifier supplementation (0 vs. 0.05 %). A total of 1,000 one-day-old male Cobb 500 broilers were randomly allocated into 4 treatments with 10 replicates and 25 birds per floor pen for 42 d (starter, d 0-14; grower, d 14-28; and finisher, d 28-42). Growth performance was measured biweekly, and AID was evaluated using the indigestible indicator method during d 21 to 28. Body composition was measured at d 35 using Dual-Energy X-Ray Absorptiometry (DXA), and carcass yield was evaluated at d 42. Data were analyzed using the GLM procedure for 2-way ANOVA. Results indicated reduced ME decreased body weight gain and feed intake (P < 0.05). Exogenous emulsifier supplementation improved FCR during the finisher and overall periods (P < 0.05). Reduced ME decreased AID of dry matter (DM), fat, and gross energy (P < 0.05) but increased AID of Val (P = 0.013). Exogenous emulsifier supplementation increased AID of DM, crude protein, His, Ile, Lys, Thr, Val, Pro, Ala, and Tyr (P < 0.05). Reduced ME decreased dressing rate and the relative weight of abdominal fat (P < 0.05). DXA results indicated that reduced ME decreased bone mineral density and fat (P < 0.001) but increased bone mineral contents and muscle (P < 0.05). Therefore, a reduction of 100 kcal/kg ME in the diet had adverse effects on the growth performance and carcass characteristics, but the use of exogenous emulsifier supplementation improved growth performance and nutrient digestibility.

The response to amino acid (AAs) supplementation on broiler production, carcass and meat traits and nitrogen in the excreta during the early growth period was evaluated. Two experiments were performed during 1-28 d (245 birds, experiment 1) and 1-21 d of age (455 birds, experiment 2). In both experiments, the positive control (PC) diet had 22.5% crude protein (CP) and the negative control group (NC) diet had around 18% CP with the same methionine (Met) plus lysine (Lys) concentration as the PC diet. In experiment 1, the NC diet was fed to the other five groups supplemented with synthetic amino acids, such as L-arginine (Arg), L-threonine (Thr), L-valine (Val), L-isoleucine (Ile) or all these AAs, respectively. In experiment 2, the NC diet was formulated to contain 18% CP with either corn-soybean meal and animal protein or with only vegetable protein. Both NC diets were offered to the other ten groups with synthetic amino acids such as L-Arg, L-Thr, L-Val, L-tryptophan (Trp) or a combination of all these AAs plus L-isoleucine (Ile), respectively. In conclusion, broilers fed 18% CP supplemented with DL-Met plus L-Lys showed lower performance and a European production efficiency value (EPEV); Arg, Thr and Val addition improved growth, the feed conversion ratio and EPEV of the diets containing animal protein only, but broiler performance and EPEV was lower than with PC, indicating that DL-Met, L-Lys, L-Arg, L-Thr and L-Val supplementation may be limited in low-protein diets. Furthermore, a low-protein diet supplemented with amino acids did not affect the survivability of broilers up to 28 days of age.

A study was conducted to evaluate effects of phytase and coccidial vaccine on growth performance, bone mineralization, nutrient digestibility, and intestinal gene expression of broiler chickens. The experiment was conducted in a 2 × 4 completely randomized factorial arrangement with 6 replicates per treatment and 10 birds each. Applications of coccidiosis vaccine and different dietary treatments were the 2 main factors in the current study. The dietary treatments included 1) a positive control (PC; 0.90% Ca and 0.45% available P: avP); 2) a negative control (NC; 0.75% Ca and 0.30% AvP); 3) NC + 500 FTU/kg of phytase (NC + 500PHY); and 4) NC + 1500 FTU/kg of phytase (NC + 1500PHY). Data were analyzed using SAS by 2-way ANOVA via GLM procedure. The statistical significance was set at P ≤ 0.05, and means were further separated using Tukey's Test. The results indicated that vaccination had no effect on growth performance except for feed intake from 0 to 14 d but negatively (P < 0.05) regulated bone ash and Ca digestibility. Birds fed with the Ca and P-reduced diet (NC) showed a lower BWG and bone ash compared to birds fed with the normal diet (PC), but supplementing phytase mitigated the negative effects on those birds. Broilers fed the NC diet had higher (P < 0.05) total Ca and P digestibility, and phytate degradation; supplementing phytase further increased P digestibility and phytate degradation of the broilers. A significant interaction (P < 0.05) between phytase and vaccination was observed, suggesting the vaccinated birds fed the PC diet and the unvaccinated birds fed the NC + 1500PHY increased calcium-sensing receptor gene expression compared with the unvaccinated birds fed the PC diet. In conclusion, in spite of coccidiosis vaccine, supplementing phytase at 1,500 FTU/kg alleviated the negative effects on growth performance, bone mineralization, and apparent ileal digestibility of P and phytate.

This study was conducted to investigate the effects of different dosages of tannic acid (TA) on growth performance, nutrient digestibility, gut health, immune system, oxidative status, microbial composition, volatile fatty acids (VFA), bone mineral density, and fat digestion and accumulation in broilers and to find optimal dosages of TA for efficient growth and gut health in broilers. A total of 320 male Cobb500 broilers were randomly distributed to 4 treatments with 8 replicates including 1) tannic acid 0 (TA0): basal diet without TA; 2) tannic acid 0.5 (TA0.5): basal diet with 0.5 g/kg TA; 3) tannic acid 1.5 (TA1.5); and 4) tannic acid 2.5 (TA2.5). Supplemental TA at levels greater than 972 mg/kg tended to reduce BW on D 21 (p = 0.05). The TA2.5 had significantly lower apparent ileal digestibility (AID) of crude protein compared to the TA0 group. The AID of ether extract tended to be reduced by TA at levels greater than 525 mg/kg (p = 0.08). The jejunal lipase activities tended to be reduced by TA at levels less than 595.3 mg/kg (p = 0.09). TA linearly decreased goblet cell density in the crypts of the jejunum (p < 0.05) and reduced mRNA expression of mucin two at levels less than 784.9 mg/kg and zonula occludens two at levels less than 892.6 mg/kg (p < 0.05). The TA0.5 group had higher activities of liver superoxide dismutase compared to the TA0 group (p < 0.05). Bone mineral density and contents tended to be linearly decreased by TA (p = 0.05), and the ratio of lean to fat was linearly decreased (p < 0.01). Total cecal VFA production tended to be linearly reduced by TA at levels greater than 850.9 mg/kg (p = 0.07). Supplemental TA tended to increase the relative abundance of the phylum Bacteroidetes (p = 0.1) and decrease the relative abundance of the phylum Proteobacteria (p = 0.1). The relative abundance of the family Rikenellaceae was the lowest at 500 mg/kg TA, and the relative abundance of the family Bacillaceae was the highest at 1,045 mg/kg TA. Collectively, these results indicate that the optimum level of supplemental TA would range between 500 and 900 mg/kg; this range of TA supplementation would improve gut health without negatively affecting growth performance in broilers under antibiotic-free conditions.

Osteoarthritis (OA) is an age-related degenerative disease. Oxidative stress (OS) modulates OA pathogenesis by enhancing chondrocyte apoptosis and extracellular matrix (ECM) degeneration via activation of the endoplasmic reticulum (ER) stress. Prior studies revealed that safranal plays a critical role in multiple diseases treatments, but there are no reports on its effect on OA. Therefore, investigating the effect of safranal on OA is needed. As a compound that can lead excessive reactive oxygen species (ROS) accumulation, tert-butyl hydroperoxide (TBHP) was used to induce OS and OS-mediated endoplasmic reticulum (ER) stress for imitating OA in vitro. Besides, the bilateral medial meniscus was removed to induce joint instability and excessive friction of the joint surface to establish destabilization of medial meniscus for imitating the initiation and progression of OA in vivo. We, next, conducted Western blot and RT-PCR analyses to identify biomarkers of the underlying signaling pathway. Our results demonstrated that 30 µM safranal strongly upregulated Sirt1 expression, suppressed TBHP-mediated ER stress, and, in turn, prevented chondrocyte apoptosis and ECM degeneration. Furthermore, compared with the other two classic signaling pathways of ER stress, safranal can inhibit the PERK-eIF2α-CHOP axis at the lower concentration (5 and 15 µM). In vivo, using Safranin O staining, X-ray, immunofluorescence (IF), and immunohistochemical (IHC) staining, we demonstrated that OA progression can be postponed with intraperitoneal injection of 90 and 180 mg/kg safranal in an OA mouse model. Taken together, our analyses revealed that safranal can potentially prevent OA development.

This review provides insight into the effects of the branched-chain amino acids (BCAA: leucine, isoleucine, and valine) on the growth, production performance, immunity, and intestinal health of poultry. Besides providing nitrogen substrates and carbon framework for energy homeostasis and transamination, BCAA also function as signaling molecules in the regulation of glucose, lipid, and protein synthesis via protein kinase B and as a mechanistic target of the rapamycin (AKT-mTOR) signaling pathway that is important for muscle accretion. The level of leucine is generally high in cereals and an imbalance in the ratio among the 3 BCAA in a low protein diet would produce a negative effect on poultry growth performance. This occurs due to the structural similarity of the 3 BCAA, which leads to metabolic competition and interference with the enzymatic degradation pathway. Emerging evidence shows that the inclusion of BCAA is essential for the proper functioning of the innate and adaptive immune system and the maintenance of intestinal mucosal integrity. The recommended levels of BCAA for poultry are outlined by NRC (1994), but commercial broilers and laying hen breed standards also determine their own recommended levels. In this review, it has been noted that the requirement for BCAA is influenced by the diet type, breed, and age of the birds. Additionally, several studies focused on the effects of BCAA in low protein diets as a strategy to reduce nitrogen excretion. Notably, there is limited research on the inclusion ratio of BCAA in a supplemental form as compared to the ingredient-bound form which would affect the dynamics of utilization in different disease-challenged conditions, especially those affecting digesta passage ratio. In summary, this review encompasses the role of BCAA as functional AA and discusses their physiological effects on the productivity and health of poultry. The observations and interpretations of this review can guide future research to adjust the recommended levels of BCAA in feeding programs in the absence of subtherapeutic antibiotics in poultry.

A study was conducted to investigate the effects of tannic acid (TA) supplementation on growth performance, gut health, antioxidant capacity, gut microbiota, and meat yield and quality in broilers raised for 42 days. A total of 700 one-day-old male broiler chickens (Cobb500) were allocated into 5 treatments with 7 replicates of 20 birds per pen. There were five treatments: 1) tannic acid 0 (TA0: basal diet without TA); 2) tannic acid 0.25 (TA0.25: basal diet+0.25 g/kg TA); 3) tannic acid 0.5 (TA0.5: basal diet+0.5 g/kg TA); 4) tannic acid 1 (TA1: basal diet+1 g/kg TA); and 5) tannic acid 2 (TA2: basal diet+2 g/kg TA). The dietary phases included starter (D 0 to 18; crumble feed), grower (D 18 to 28; pellet feed), and finisher (D 28 to 42; pellet feed). On D 18, the supplementation of TA linearly reduced body weight (BW) and average daily feed intake (ADFI) (p < 0.05), and on D 28, the supplementation of TA linearly reduced BW, average daily gain (ADG), and feed conversion ratio (FCR) (p < 0.05). Relative mRNA expression of genes related to mucin production (MUC2), tight junction proteins (CLDN2 and JAM2), and nutrient transporters (B0AT1 and SGLT1) was linearly increased by the supplementation of TA (p < 0.05). The supplementation of TA tended to linearly increase the relative abundance of the family Enterobacteriaceae (p = 0.08) and quadratically increased the relative abundance of the families Lachnospiraceae and Ruminococcaceae in the cecal microbial communities (p < 0.05). On D 36, the ratio of the phyla Firmicutes and Bacteroidetes was quadratically reduced by the supplementation of TA (p < 0.05). On D 42, bone mineral density and the lean to fat ratio were linearly decreased by the supplementation of TA (p < 0.05). On D 43, total chilled carcass weight was linearly reduced (p < 0.05), and proportion of leg weight was increased by supplementation of TA (p < 0.05). The supplementation of TA linearly reduced pH of the breast meat (p < 0.05) and linearly increased redness (a*) (p < 0.05). ß-mannan is a nonstarch polysaccharide found in hulled and dehulled soybeans that can survive drying-toasting phase of processing soybeans and have antinutritive effects in poultry. ß-mannanase is an active enzyme (endohydrolase) that can hydrolyze ß-mannan to reduce its antinutritional effects. Two experiments were conducted to evaluate the effects of ß-mannanase supplementation in low energy/protein diets on egg production, egg quality, and apparent ileal digestibility of the dry matter (DM), crude protein (CP), and amino acids in 21-week-old Single Comb White Leghorn hens (Hy-Line W-36). A total of 192 hens (8 replicates of 6 hens per treatment) for a production study (Exp. 1) and a total of 64 hens (8 replicates of 2 hens per treatment) for a digestibility study (Exp. 2) were randomly allocated to 4 experimental treatments in a 2 × 2 factorial arrangement. Four dietary treatments were control (CS) based on corn and 44% CP soybean meal (ME: 2,850 kcal/kg CP: 18.5%) and CS-low energy/protein (CSL) (ME: 2,750 kcal/kg CP: 17.5%), with or without 0.05% ß-mannanase enzyme. Hens were fed the experimental diets for 14 d for the digestibility study and 8 wk for the production study. Hen-day egg production (HDEP), weekly feed intake, FCR, and biweekly egg quality parameters were measured. Significant interaction on feed intake (P < 0.01) was observed between energy/protein and enzyme. At 3, 6 and 8 wk, the feed intake and FCR of CSL with enzyme were significantly lower than those of CSL without enzymes. The main effects indicated that birds fed diets without inclusion of ß-mannanase had higher feed intake than those fed diets with enzymes at 4, 7, and 8 wk. The inclusion of ß-mannanase significantly increased (P < 0.05) HDEP at 2, 3, 5, and 7 wk. However, there was no significant effect of nutrient density or enzyme supplementation on egg quality parameters. The digestibility study showed that the inclusion of ß-mannanase significantly improved (P < 0.01) apparent ileal digestibility of lysine, histidine and tryptophan in the diet. The results of these experiments indicate that supplementation of ß-mannanase could reduce the feed intake and FCR and improve HDEP and apparent ileal digestibility of key amino acids in corn/soy diets fed to laying hens.

BACKGROUND: The aerial parts of Salvia miltiorrhiza, which was considered to be the waste part and discarded during the root harvest, is rich in protocatechuic aldehyde (PAI). This study investigated the health-promoting effects of extracts and PAI from the aerial parts of Salvia miltiorrhiza, including its anti-inflammatory effects and the underlying mechanisms of action in vitro and in vivo. METHOD: Purification of the sample paste of Salvia miltiorrhiza was accomplished using HPLC analysis. TheMTT (Methylthiazolyldiphenyl-tetrazolium bromide) assay was employed to determine the cell viability. The production of inflammatory factors was detected by ELISA assays. The histopathological analysis was used to analyse the lungs and livers of mice treated with PAI. Western blot was performed to reveal the mechanism of PAI in anti-inflammatory. RESULTS: The extracts and PAI from the aerial parts of Salvia miltiorrhiza inhibited TNF-α, IL-6 production and promoted the production of IL-10 in vivo in mice and in vitro in the macrophage cell line RAW264.7. NF-κB and MAPKs kinase phosphorylation were also suppressed by PAI in vivo and in vitro, indicating that PAI exhibited an anti-inflammatory effect. CONCLUSION: These findings suggest that the aerial parts of Salvia miltiorrhiza extract may serve as potential protective agents for inflammatory.

Vancomycin (VCM) is an effective chemotherapeutic agent commonly used against gram-positive microorganisms but has serious nephrotoxic side effects that limit its effectiveness. New therapeutics and strategies are urgently needed to combat VCM associated nephrotoxicity. In this study, we determined the protective effect of chlorogenic acid (CA) in a rat model of VCM-induced nephrotoxicity. VCM administration led to markedly elevated blood urea nitrogen and serum creatinine levels that could be prevented with CA co-administration. VCM-mediated oxidative stress was also significantly attenuated by CA as reflected by decreased malondialdehyde and nitric oxide in VCM-treated kidneys. CA administration also prevented the VCM-mediated decrease in the renal antioxidative enzyme activities of glutathione reductase, glutathione peroxidase, and catalase and led to increased levels of reduced glutathione that had been depleted by VCM. Moreover, CA administration clearly inhibited VCM-induced expression of nuclear factor-kappa B, inducible nitric oxide synthase and the downstream pro-inflammatory mediators tumor necrosis factor-α and interleukins 1ß and 6. Apoptotic markers were also markedly down-regulated with CA. Overall, CA treatment mitigated VCM-induced oxidative and nitrosative stresses and countered the apoptotic and inflammatory effects of VCM. Notably, CA did not affect the antibacterial activity of VCM in vitro.

The objective of this study was to evaluate the effect of low-protein diets with amino acid supplementation on growth performance, carcass yield, meat quality and nitrogen excretion of broilers raised under hot climate conditions during the finisher period. In trial 1, broilers from 28 to 49 days of age were fed 18% crude protein (CP) as a positive control or 15% CP supplemented with (1) DL-methionine (Met) + L-lysine (Lys), (2) Met + Lys + L-Arginine (Arg), or (3) Met + Lys + L-Valine (Val). In trial 2, broilers from 30 to 45 days of age, were fed an 18% CP diet as a positive control or 15% CP supplemented with Met, Lys, Arg, Val, L-Isoleucine (Ile) or combination with glycine (Gly) and/or urea as nitrogen sources: (1) Met + Lys, (2) Met + Lys + Arg, (3) Met + Lys + Val, (4) Met + Lys + Ile, (5) Met + Lys + Arg +Val + Ile + Gly, and (6) Met+ Lys + Arg + Val + Ile + Gly + urea. Protein use was improved by feeding low-protein amino acid-supplemented diets as compared to the high-protein diet. Feeding 15% crude protein diet supplemented with only methionine and lysine had no negative effects on carcass yield, CP, total lipids and moisture% of breast meat while decreasing nitrogen excretion by 21%.

ETHNOPHARMACOLOGY RELEVANCE: Danggui Buxue Tang has been used in China to treat clinical anemia for more than 800 years. However, there is no scientific report on its effect on bone marrow stromal cells. AIM OF THE STUDY: Here, we aimed to explore the effect of Danggui Buxue Tang on bone marrow stromal cell adhesion and migration. MATERIALS AND METHODS: Bone marrow stromal cells were used as a model to evaluate the effect of Danggui Buxue Tang on the adhesion and migration of bone marrow stromal cells. RNA-sequencing, quantitative polymerase chain reaction, and western blotting were used to detect and confirm the expression of genes related to the focal adhesion pathway before and after drug delivery. RESULTS: Danggui Buxue Tang significantly increased the number of bone marrow stromal cells. After 12 days of 16 mg/mL Danggui Buxue Tang treatment, bone marrow stromal cells were significantly increased (by 0.527 ±â€¯0.008 fold; p < 0.001) as compared to the control group (0.180 ±â€¯0.019). The effect was not due to enhanced cell proliferation, as there was no difference in the cell cycle (p > 0.05). The adhesion area of a single cell was doubled by Danggui Buxue Tang treatment (p < 0.001), and the time required for cell adhesion to a Petri dish was shortened. Thus, Danggui Buxue Tang increases the number of bone marrow stromal cells by promoting adhesion. Danggui Buxue Tang also significantly promoted bone marrow stromal cell migration (p <  0.001). Transcript analysis revealed that the focal adhesion and PI3K-Akt signaling pathways were activated. Expression analysis confirmed that the gene and protein expression of focal adhesion-related factors were upregulated. CONCLUSION: Danggui Buxue Tangaffects bone marrow stromal cell adhesion and migration by enhancing the focal adhesion pathway in vitro, and bone marrow stromal cells are a target of DBT-regulated hematopoiesis, and the active ingredients of DBT involved in the effects require further investigation.

This study aimed to investigate the effect of dietary cysteamine hydrochloride (CSH) on the growth performance, oxidation, inflammation, and gene expression of cytoskeleton and tight junction in the intestinal mucosa of broiler chickens challenged with Clostridium perfringens (C. perfringens). A total of 360 one-day-old broiler chicks were randomly distributed into 5 groups for a negative control (NC, without C. perfringens challenge), a positive control (PC, with C. perfringens challenge), and PC plus CSH at 100, 150, or 200 mg/kg of diet. The results showed that average daily gain, gain:feed, cecal population and enterotoxin of C. perfringens were negatively affected (P < 0.05) by the C. perfringens challenge, but were conversely affected (P < 0.05) by the CSH supplementation, and G:F reached to the level of NC group. The PC group increased (P < 0.05) serum diamine oxidase, malondialdehyde, protein carbonyl, interleukin-6, interleukin-1ß, and tumor necrosis factor-α, whereas the supplementation of CSH decreased (P < 0.05) these parameters. Moreover, the C. perfringens challenge worsened the disruption of intestinal mucosal cytoskeleton and tight junction by downregulating (P < 0.05) the mRNA levels of actin protein of muscle Z-line alpha, syncoilin, synemin, tubulin, claudin-1, and zona occludens protein-2, while these parameters were partially compensated (P < 0.05) by CSH supplementation. For the dose trends of CSH, there were linear and quadratic (P < 0.05) effects on gain:feed, enterotoxins, tumor necrosis factor-α, tubulin alpha 1c, syncoilin, and synemin. In conclusion, the CSH can be an alternative against C. perfringens infection by beneficially regulating gut pathogenic bacteria and enterotoxins, oxidation, inflammation, cytoskeleton, and tight junction in broiler chickens.
