RESUMEN

BACKGROUND: Scutellaria baicalensis Georgi is a famous traditional Chinese medicine, which is widely used in treating fever, upper respiratory tract infection and other diseases. Pharmacology study showed it can exhibit anti-bacterial, anti-inflammation and analgesic effects. In this study, we investigated the effect of baicalin on the odonto/osteogenic differentiation of inflammatory dental pulp stem cells (iDPSCs). METHODS AND RESULTS: iDPSCs were isolated from the inflamed pulps collected from pulpitis. The proliferation of iDPSCs was detected by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2,5-tetrazolium bromide (MTT) assay and flow cytometry. Alkaline phosphatase (ALP) activity assay, alizarin red staining, Real-time reverse transcription-polymerase chain reaction (RT-PCR) and Western blot assay were conducted to examine the differentiation potency along with the involvement of nuclear factor kappa B(NF-κB) and ß-catenin/Wnt signaling pathway. MTT assay and cell-cycle analysis demonstrated that baicalin had no influence on the proliferation of iDPSCs. ALP activity assay and alizarin red staining demonstrated that baicalin could obviously enhance ALP activity and calcified nodules formed in iDPSCs. RT-PCR and Western blot showed that the odonto/osteogenic markers were upregulated in baicalin-treated iDPSCs. Moreover, expression of cytoplastic phosphor-P65, nuclear P65, and ß-catenin in iDPSCs was significantly increased compared with DPSCs, but the expression in baicalin-treated iDPSCs was inhibited. In addition, 20 µM Baicalin could accelerate odonto/osteogenic differentiation of iDPSCs via inhibition of NF-κB and ß-catenin/Wnt signaling pathways. CONCLUSION: Baicalin can promote odonto/osteogenic differentiation of iDPSCs through inhibition of NF-κB and ß-catenin/Wnt pathways, thus providing direct evidence that baicalin may be effective in repairing pulp with early irreversible pulpitis.

Synthetic systems of nonequilibrium self-assembly have made considerable progress; however, the achievement of innovative materials with self-regulated functions analogous to living systems remains a grand challenge. Herein, a versatile nonequilibrium system of polymer brushes with spatiotemporally programmable properties and functions driven by chemical fuels is reported. By combining a responsive polymer with an autonomous pH regulator, the polymer brushes self-regulate their swelling and deswelling process with tunable lifetimes. By using patterned copolymer brushes with pH-responsive fluorescence moiety, an autonomous fluorescence modulator is created that self-regulates its fluorescence in spatiotemporally programmable fashion driven by a chemical or an enzymatic reaction. Furthermore, a self-regulated wettability switcher of polymer brushes both in air and in an aqueous solution is implemented. The methodology and results in the work provide a useful avenue into the exploration of nonequilibrium synthetic materials with programmable functions and would accelerate the transformative developments of nonequilibrium materials and systems in practical applications.

RESUMEN

A bionanocomposite with artificial binding pockets for a DNA repair enzyme has been developed by in situ assembly of an affinity protein with a surrounding contact surface of polydopamine on the surface of silica coated magnetic nanoparticles via molecular imprinting reactions. The obtained nanoparticles exhibited antibody-like binding behavior toward the target enzyme with highly specific and efficient inhibition effect. Moreover, the binding and inhibition could be flexibly tuned by the addition of metal ions such as Mn2+ and Mg2+, which provided a convenient tool to regulate enzyme activity with artificially engineered nanoinhibitors.

Repair of spinal cord injury will require enhanced recruitment of endogenous neural stem cells (NSCs) from the central canal region to the lesion site to reestablish neural connectivity. The strategy toward this goal is to provide directional cues, e.g., alignment topography and biological gradients from the rostral and caudal ends toward the center. This study demonstrates a facile method for fabrication of continuous gradients of stromal-cell-derived factor-1α (SDF1α) embedded in the radially aligned electrospun collagen/poly (ε-caprolactone) mats. Gradients can be readily produced in a controllable and reproducible fashion by adjusting the collection time and collector size during electrospinning. To get a long-term gradient, the SDF1α is fused with a unique peptide of collagen-binding domain (CBD), which can bind to collagen specifically. Aligned CBD-SDF1α gradients show stable, sustained, and gradual release during 7 d. Further, the effect of aligned CBD-SDF1α gradients on the guidance of NSCs is investigated. It is found that the CBD-SDF1α gradient scaffolds direct and enhance NSC migration from the periphery to the center along the aligned electrospun fibers. Taken together, the tubular conduits based on radially aligned electrospun fibers with continuous SDF1α gradient show great potential for guiding nerve regeneration.

Formaldehyde (FA) is widely used as an adhesion promoter and dyeing aid in industrial production. Ingestion of a certain amount of formaldehyde may cause corrosive burns in the mouth, throat, and digestive tract. Therefore, it is very necessary to use simple and effective detection methods to ensure human health and food safety. Herein, a novel fluorescent probe NFD based on naphthalimide for the detection of formaldehyde in food was designed and synthesized. The probe had a remarkable fluorescence response to formaldehyde at 554 nm. And it exhibited fascinating advantages of good selectivity, high sensitivity, and low detection limit. In addition, the solid sensor prepared by loading the probe on the filter paper was successfully realized the visual detection of liquid and gaseous formaldehyde. More importantly, the probe possessed excellent stability in the detection of formaldehyde in real food samples and animal serum samples.

In this study, a new rapid and inexpensive filter solid-phase extraction method (FSPE) was developed for the extraction and separation of five perfluorinated carboxylic acids (PFCAs) (namely, perfluorooctanoic acid, perfluorononanoic acid, perfluorodecanoic acid, perfluoroundecanoic acid, and perfluorododecanic acid) from milk samples. Commercial nylon 66 syringe filters were used as adsorbents without additional modification. The proposed method could achieve the fast adsorption of analytes by benefitting from the advantages of using the nylon 66 filter membrane on PFCAs with multiple adsorption interactions, namely, hydrophobic interactions and hydrogen bonds. The FSPE was designed to achieve the rapid isolation of analytes based on the rapid solid-liquid separation while using the solid-phase extraction disk. PFCAs residues in milk were extracted by first flowing the samples through a nylon 66 syringe filter from top to bottom. Double contact desorption with methanol was then accomplished by pulling and pushing the syringe, followed by derivatization with acetyl chloride-methanol. Finally, the extracted PFCAs residues were monitored using gas chromatography-tandem mass spectrometry (GC-MS/MS) quantification. Under optimal conditions, the established method presented good linearity (correlation coefficient ≥ 0.9996), precision (relative standard deviation ≤ 13%), accuracy (81-105%), sensitivity (limits of quantification: 4-18 ng kg-1) and extraction performance (10 min).

Theranostic agents with fluorescence in the second near-infrared (NIR-II) window, especially in its long-wavelength region, and NIR-II-excitable photothermal effect is promising but challenging in tumor diagnosis and therapy. Here, we report a simple but effective strategy to develop semiconducting polymer nanoparticles-based theranostic agents (PBQx NPs) and demonstrate their applications for long-wavelength NIR-II fluorescence imaging beyond 1400 nm and photothermal therapy (PTT) of tumors upon excitation at 1064 nm. Both experimental results and theory calculations show that the brightness and photothermal performance of PBQx NPs can be simultaneously improved by simply increasing the repeating unit number of semiconducting polymers. For example, PBQ45 NPs have 5-fold higher brightness than PBQ5 NPs and 6.7-fold higher photothermal effect (based on PCE × Îµ) than PBQ3 NPs, and exhibit promising applications in long-wavelength NIR-II fluorescence abdomen imaging, image-guided tumor resection, and image-guided PTT. This study demonstrates the effectiveness and importance of repeating unit numbers in regulating the theranostic performance, which has not received enough attention before.

An imidazolium ionic-liquid-modified phenolic resin (ILPR) was synthesized using 3-aminophenol as a functional monomer, glyoxylic acid as a green cross-linker, and polyethylene glycol 6000 as a porogen. The obtained ILPR showed better extraction of benzoylurea plant hormones thidiazuron and forchlorfenuron than the unmodified phenolic resin because the imidazolium IL provides more interaction modes with the analytes. ILPR, as a tailored adsorbent for solid-phase extraction, was coupled with high-performance liquid chromatography (ILPR‒SPE‒HPLC) for the simultaneous determination of thidiazuron and forchlorfenuron in cucumbers. Good linearity of the ILPR‒SPE‒HPLC method was obtained, ranging from 0.0100 to 5.00 µg g-1 with a correlation coefficient (r) ≥ 0.9999. The recoveries of spiked samples ranged from 91.4% to 100.7% with a relative standard deviation of ≤ 6.0%.
