ABSTRACT

Heat-killed cell suspension of several Propionibacterium acnes strains were prepared and studied for their protective activity in viral infection of mice and immunomodulating properties. Majority of the strains caused marked spleen enlargement in the treated mice. These changes persisted for several weeks. Only some of the tested strains enhanced significantly primary humoral immune response against sheep red blood cells. There was no increase, however, in neutralizing or hemagglutination-inhibition antibody production against vaccinia virus in mice treated with Propionibacteria. No evidence in the increase of spleen lymphocytes migration inhibition of mice infected with vaccinia virus and treated with P. acnes or in hypersensitivity reaction to oxazolone have been found. Significant resistance enhancement of mice pretreated with P. acnes against vaccinia virus or herpes simplex virus type 1 infection was observed. Activity of Propionibacteria depended on the applied strain, dose and scheme of administration.

ABSTRACT

The aim of the present study was to examine the influence of the thiosemicarbazone-group of compounds with antiviral activity on the humoral and cellular immune response in R III, CBA and Balb/c mice, immunized with sheep erythrocytes or sensitized with oxazolon. The plaque forming cells test, according to Jerne's method and the oxazolon hypersensitivity test were performed. All of the examined compounds, inhibited the humoral immune response, and three of them decreased also the cellular response. The relationship between the antiviral activity and their immuno-suppressive activity has been discussed. Low antiviral activity of compound Hoe 105 could be due to its immunosuppressive action. Moreover, the mechanism of antiviral activity of one of the tested compounds, which displayed strong activity against vaccinia virus, was examined and compared with the activity of methisazone.

ABSTRACT

Determination of the mechanism of action of FK506 and cyclosporin A has yielded new molecular targets involved in signal transduction during T cell activation. A common target of FK506 and cyclosporin A is inhibition of activation of the NFAT transcription factor, for which a specific binding region is present in the promoter of the IL-2 gene. A reporter gene assay has been used to screen for agents that interfere with this early step in T cell activation. Simple aromatic compounds that block NFAT-dependent transcription and show in vitro immunosuppressive activity were isolated from the broth and mycelia of two Streptomyces sp. fermentations. The compounds were active at concentrations that were not directly cytotoxic.

ABSTRACT

The novel calcineurin inhibitor, dibefurin, has been isolated from the fungal culture AB 1650I-759. The isolation was bioactivity-directed fractionation using an assay which measures the phosphatase activity of calcineurin. The compound was purified by countercurrent, reverse phase and gel filtration chromatographies. Several studies, including crystallographic, NMR and MS, revealed that dibefurin is a novel dimeric compound of a unique structural type.

ABSTRACT

Dipyridamole in the dose of 50 mg/kg body weight stimulated the primary humoral response in BALB/c and C57Bl mice. The cell-mediated immune response was not influenced in BALB/c mice.

ABSTRACT

In the work a case of an excessive functional activity of core of hyper suprarenal gland is presented. It is caused by atypical overgrowth of the left suprarenal gland. The cyst observed on the left kidney covering pathologically the changed suprarenal gland was an additional diagnostic difficulty.
