RESUMEN

Toxoplasma gondii is an apicomplexan protozoan parasite that has been associated with reproductive failure in small ruminants. Although T. gondii infections in ruminants and humans have been recorded in several Egypt's governorates, but little is known about the risk factors associated with T. gondii infections. In this study, 350 sheep and 290 goat serum samples from three governorates in Egypt were examined for presence of antibodies against T. gondii, and evaluate the associate risk factors for the infection. The seroprevalence in sheep and goats was 24% and 38.28%, respectively. In sheep and goats, age, sex, the presence of cats, and hygienic conditions were identified as risk factors for T. gondii infection. In addition, the prevalence rates were significantly higher in older animals more than 2 years old, females, among animals contacted with cats, and animals living in bad hygienic condition. In conclusion, sheep and goats in the examined regions are commonly infected with T. gondii. The identification of risk variables defines the sort of actions to be implemented in order to decrease, and prevent T. gondii infection in small ruminant animals and, as a result, human infection.

Theileria annulata (T. annulata) is an intracellular protozoan, transmitted by ixodid ticks of the genus Hyalomma and affects camels. There are few epidemiological data on T. annulata infection and its associated risk factors in Egyptian camels. Thus, the present study aimed to determine the prevalence of T. annulata in camels using PCR and assess the associated risk factors for infection. A total of 380 blood samples were collected from camels raising in three Egyptian governorates and examined by PCR assay targeting 30-kDa gene to detect the presence of T. annulata infection, beside statistical analysis of associated factors. The results revealed presence of T. annulata with overall prevalence of 21.1%. In addition, the univariate analysis revealed significant (P<0.05) association between prevalence of T. annulata in camels and locality, age, sex, tick infestation, and application of acaricides. Whereas the prevalence of T. annulata was higher in camels of age group >6 to 10 years (38%), females (25.7%) and in infested camels with ticks (29%) and in case of absence of acaricides application (25%). In contrast, the body condition of camels had not significant effect on prevalence of theileriosis in camels. The current study concluded that T. annulata is prevalent in Egyptian camels and that a tick control program is required to reduce the risk of infection.

Fasciola hepatica is a trematode parasite that affects wide range of mammalian hosts including horses. There are no epidemiological data on fasciolosis in Egyptian horses. Hence, the present study aimed to evaluate the seroprevalence of F. hepatica in horses using ELISA as well as assess the associated risk factors for fasciolosis infection. A total of 400 sera samples of horses from three Egyptian governorates were examined. The overall seroprevalence rate in horses was 13% and the disease was more prevalent in Qalubia (14.17%) and Gharbia (16.67%) which are located at Nile Delta. The likelihood of having F. hepatica seropositivity was higher in mixed-breed animals and stallions (OR = 3.97, 95%CI: 1.10-14.28 and OR = 2.07, 95%CI: 1.09-3.93, respectively), and in older horses more than 10 years (OR = 5.69, 95% CI: 1.98-16.36). Horses were more likely to be seropositive with F. hepatica when kept in outdoor housing (OR= 1.53, 95%CI: 0.78-3), during autumn (OR = 3.80, 95%CI: 1.17-12.32) and in the presence of snails (OR = 2.72, 95%CI: 1.34-5.52). Therefore, the obtained data contributes to the understanding of the epidemiology of Fasciola species, assess in establishment of management programmes to reduce disease and public health risks.

Neospora caninum (N. caninum) is one of the causative agents that causing cattle abortion, and severe economic losses. Due to the scarcity of data on N. caninum infection in Egyptian cattle, the purpose of this study was to estimate the seroprevalence and determine the risk factors for parasite infection. In four governorates in northern Egypt, 540 blood samples from cattle were taken, and tested using a commercial ELISA kit. The overall seroprevalence of N. caninum in examined cattle was 28.89%. A multivariate logistic regression model determined that age (OR = 2.63, P < 0.001), manual milking (OR = 1.39, P = 0.14), abortion history (OR = 2.78, P < 0.0001), repetition of estrus (OR = 2.31, P < 0.0001), and contact with dogs (OR = 2.57, P < 0.0001) were significant risk factors. The findings proved that N. caninum infection was one of the factors contributing to abortion and financial losses in dairy cattle in Egypt. Therefore, the application of sanitary security and control programs is very important in dairy farms.

Cryptosporidium is one of the causative parasitic agents that causes gastrointestinal diseases in calves. The parasite poses a zoonotic risk to immunocompromised individuals and children. Thus, this study aimed to determine the prevalence of Cryptosporidium infection in calves in three Egyptian governorates situated in Nile Delta and assess the associated risk factors. The Cryptosporidium oocysts were detected in 81 out of 430 calves (18.84%). In addition, the univariant analysis showed that age, feeding source, hygienic status, presence of diarrhea and contact with other animals were significantly (P < 0.05) associated with Cryptosporidium prevalence in calves. Furthermore, the risk factors related with Cryptosporidium prevalence were age (OR 1.96, 95%CI 0.97-3.94), feeding on milk and pasture (OR 2.07, 95%CI 1.15-3.72), poor hygienic condition (OR 2.25, 95%CI 1.28-3.94), presence of diarrhea (OR 2.47, 95%CI 1.23-4.96) and contact with other domestic animals (OR 2.08, 95%CI 1.24-3.50). In addition, the PCR assay targeting 18srRNA showed that the most prevalent species among calves was C. parvum. Although additional researches are required to understand the most effective steps that farmers and veterinary professionals should take to decrease the occurrence of Cryptosporidium infection.

Neosporosis is a parasitic disease that causes reproductive disorders in animals, making it a barrier to maximum efficiency. The purpose of this study was to determine the seroprevalence of Neospora caninum (N. caninum) antibodies in water buffaloes from four governorates in northern Egypt. A commercial indirect-ELISA test was used to detect antibodies against N. caninum in the serum of 450 water buffaloes. The total seroprevalence of N. caninum in water buffaloes from Egypt was 31.3%, and the highest prevalence was observed in Gharbia governorate. The identified risk factors for N. caninum infections in water buffaloes were sex (OR = 1.96, 95%CI: 1.22-4.17), buffaloes more than 4 years of age ( OR = 5.80, 95%CI: 2.26-14.86), abortion in second trimester (OR = 16.48, 95%%CI: 2.99-34.03), history of abortion (OR = 3.45, 95%CI: 1.58-7.52) and contact with dogs (OR = 2.55, 95%CI: 1.51-4.32). Thus, more studies are needed to determine the role of buffaloes in the epidemiology of neosporosis in Egypt.

BACKGROUND: Toxoplasma gondii is classified as intracellular protozoa and is one of the major zoonotic parasites. Most warm-blooded intermediate hosts, including humans, are commonly infected by this parasite. The epidemiology of T. gondii infection in Egyptian horses is currently poorly understood. METHODS: 420 blood samples were randomly collected from horses raised in four governorates in Northern Egypt (110 each from Giza and Kafr El Sheikh, and 100 each from Qalyubia and Gharbia) to investigate the existence of antibodies against T. gondii using a commercial ELISA kit, and to ascertain the risk factors for the infection. RESULTS: the antibodies for T. gondii were found in 16.2% (68/420) of the examined horses, with no significant differences among the four studied governorates. The highest prevalence rate was observed in Giza. The results revealed that sex, breed, age, and contact with domestic ruminants or cats were recognized as potential risk factors. The high prevalence rate was found in mixed breed horses (OR = 2.63, 95% CI: 0.95-7.26), mares (OR = 2.35, 95% CI: 1.31-4.19), and horses aged over 10 years (OR = 2.78, 95% CI: 1.30-3.44). Moreover, the likelihood of seropositivity for T. gondii infection was higher in horses raised in environments with cats (OR = 1.97, 95% CI: 1.13-3.44, p = 0.017) or domestic ruminants (OR = 2.16, 1.21-3.86, p = 0.010). This report confirms that horses in Northern Egypt are exposed to T. gondii and thus raises the possibility that people and other animals could contract the disease. CONCLUSIONS: routine examination and management of T. gondii infection in horses in these governorates is advised.

Surra, a wasting disease caused by Trypanosoma evansi, is one of the major animal health burdens in camel-rearing countries, imposing significant economic losses due to reduced fertility and high mortality rates. The present study used inactivated T. evansi (from the Card Agglutination Test for Trypanosomes/Trypanosoma evansi; CATT/T. evansi) and flow cytometry to investigate their binding and activation potential toward camel leukocyte subsets. Labeling T. evansi with propidium iodide (PI) enabled their flow cytometric enumeration and identification with forward scatter (FSC; indicative for cell size) and side scatter (SSC; indicative for cell internal complexity) characteristics that are comparable with values reported for Trypanosoma cruzi. The incubation of PI-labeled non-opsonized T. evansi with camel leukocyte populations revealed that camel monocytes have the highest potential to bind T. evansi, followed by granulocytes and lymphocytes. The identification of pattern recognition receptors (PRRs) on camel immune cells and the pathogen-associated molecular patterns (PAMPs) in T. evansi that are responsible for this different binding capacity requires further studies. Stimulation of camel neutrophils with Trypanosoma evansi induced shape change, reactive oxygen species (ROS) production, and neutrophil extracellular traps (NET)-formation. To ensure that T. evansi, in the parasite concentration used in this study, is not apoptotic or necrotic to camel leukocytes, we evaluated cell apoptosis and necrosis after stimulation with T. evansi. The results revealed no impact of T. evansi stimulation for 2 h on the cell viability of camel leukocytes. Subsequent work may focus on the diagnostic employment of labeled T. evansi and flow cytometry for the detection of anti-Trypanosoma antibodies in camel serum. In addition, more efforts should be deployed to investigate the host-pathogen interaction mechanisms and the escape mechanisms of T. evansi in camels. To complete these data, further studies using the living or freshly killed parasites could also be implemented in camels and/or horses.

(1) Background: Cystic echinococcosis is a zoonotic helminth disease that causes severe economic losses. The study aimed to assess the prevalence and viability of cystic echinococcosis in examined camels. In addition, assessing the histological, morphological, oxidative, and antioxidant state related to the cystic echinococcosis infection; (2) Methods: The study was performed on 152 slaughtered dromedary camels between March and September 2022 at El-Basatin abattoir in Cairo Governorate, Egypt; (3) Results: The results revealed that the prevalence of hydatidosis was 21.7% in slaughtered camel and the highest infection rate observed in lungs was 87.87%, while it was 9% in livers. Camels' liver infections were rare, whereas their lung infections were more common. By comparing to non-infected camels, the level of MAD was significantly increased with hydatid cysts infection, while the level of GSH, SOD and CAT was significantly decreased. Histopathological section of camel cyst revealed layered membranes surrounded by a zone of cellular infiltration and an outermost fibrous tissue reaction. In addition, there was evidence of atelectasis, emphysema, hemorrhage, congestion, and fibrosis in the surrounding tissues. Nonetheless, the degeneration and necrosis of hepatocytes and other pathological alterations in liver cyst sections were remarkably comparable to those seen in the lungs. Furthermore, calcification was detected.

Toxoplasmosis is a global zoonotic disease caused by Toxoplasma gondii (T. gondii). The primary aim of this study was to identify hygienic and cat management practices that could affect the occurrence of T. gondii in cats and their owners in Sharqia Governorate, Egypt. T. gondii infection was evaluated in 80 pregnant women and 29 domestic cats using Real-time PCR. A questionnaire was administered to obtain information regarding the risk factors associated with T. gondii infection. Blood samples were collected from enrolled pregnant women, and fecal samples were collected from their pet cats. Firth logistic regression model complemented with random forest (RF) analysis was used to evaluate the association of different hygiene and cat management practices with T. gondii infection in pregnant women. T. gondii infection was identified in 27.5% of pregnant women and 17% of domestic cats. Drinking raw milk and contacting stray and pet cats were significantly associated with higher odds of T. gondii infection. The proportion of T. gondii-positive women who ate raw meat (60.0%) was higher than those ate heat-cooked meat (25.3%). Moreover, women who did not wash their hands after contact with cats were 6 times (OR 6.12; CI: 3.03-9.21) more likely to experience T. gondii infection than those washed their hands after cat contact. The RF analysis showed that washing hands constitute a serious yet preventable public health concern that requires targeted, community-specific efforts. Cat owners, particularly pregnant women, need to be aware about the risk of T. gondii infection, while handling cat and pet's owner should be advised to take necessary hygienic measures to prevent its infection.

Acinetobacter baumannii (A. baumannii) is an opportunistic pathogen associated with nosocomial infections. In this study, 100 raw milk samples were collected from Qena, Egypt, and subjected to conventional and molecular assays to determine the presence of A. baumannii and investigate their antimicrobial resistance and biofilm formation. Our findings revealed that, among the 100 samples, Acinetobacter spp. were found in 13 samples based on CHROM agar results. We further characterized them using rpoB and 16S-23SrRNA sequencing and gyrB multiplex PCR analysis and confirmed that 9 out of the 13 Acinetobacter spp. isolates were A. baumannii and 4 were other species. The A. baumannii isolates were resistant to ß-lactam drugs, including cefotaxime (44%), ampicillin-sulbactam and levofloxacin (33.3% for each), imipenem, meropenem and aztreonam (22.2% for each). We observed different antimicrobial resistance patterns, with a multi-antibiotic resistant (MAR) index ranging from 0.2 to 0.3. According to the PCR results, blaOXA-51 and blaOXA-23 genes were amplified in 100% and 55.5% of the A. baumannii isolates, respectively, while the blaOXA-58 gene was not amplified. Furthermore, the metallo-ß-lactamases (MBL) genes blaIMP and blaNDM were found in 11.1% and 22.2% of isolates, respectively, while blaVIM was not amplified. Additionally, eight A. baumannii isolates (88.8%) produced black-colored colonies on Congo red agar, demonstrating their biofilm production capacity. These results showed that, besides other foodborne pathogens, raw milk should also be examined for A. baumannii, which could be a public health concern.

From January 2017 - December 2019, 75 out of 850 (8.8 %) great hammerhead sharks from the Arabian Gulf had skin lesions of black irregular discolorations on the ventral surface of the head. The lesions consisted of pencil-like lineations often advancing forward by about 2 mm in back-and-forth looped scribbles often forming a relatively linear bands of about 5-7 cm wide. Similar lesions were also found in the blacktip reef shark from the same area within the same period, and consisted of straight to irregular black lines, extended indiscriminately across the skin of the sharks. Microscopic examination of the skin revealed the presence of dark-brown eggs exhibiting the spindle or ellipsoidal eggs characteristic of Huffmanela sp. The morphometrics of eggs from both hosts were similar (62.9-89.9 µm long and 29.3-56.1 µm wide). The eggshells were smooth with polar plugs protruding or not, with an abruptly truncated crown-like or shoulder-like collar surrounding the plug. The eggs were only found in the epidermal layer of the skin. Based on the unique morphometrics of the eggs, we report a new species, named: Huffmanela selachii n. sp.. This appears to be the first report of Huffmanela from either the great hammerhead shark or the blacktip reef shark, and the third reported Huffmanela in sharks from the Arabian Gulf. It is also one of few species reported from connecting waters of the greater Indian Ocean. This new finding contributes to our understanding of the diversity and ubiquity of Huffmanela sp. in marine creatures.

The present study was designed to investigate the prevalence of gastrointestinal (GI) parasites in cattle and buffaloes of Lower Dir Khyber Pakhtunkhwa, Pakistan. The presence of the eggs, cysts, and oocysts of GI parasites in fecal samples were detected using direct smear methods and concentration techniques including floatation, centrifugation, and sedimentation. Identification of recovered fecal stages were determined by morphology using size and appearance of the recovered eggs, cysts, and oocysts. A total of 314 fecal samples were collected from the different Tehsils (Administrative Districts) and analyzed through microscopy. A higher prevalence was observed in the buffalo than the cow population. A total of 184 samples were positive for GI parasites of which 109/196 (55.61%) were from cattle, whereas 75/118 (63.55%) were from buffaloes. The minimum number of strongyle eggs detected in all the samples were 136.39 eggs/g (EPG). The mean EPG in cattle was 143.30 and 122.56 in buffaloes. The open-source water prevalence of GI parasites was higher than the other sources in cattle and the second highest after tap water in buffaloes. The seasonal prevalence of GI parasites ranged from 32.39% (23/71), in spring to 68.8% (86/125) in summer in cattle. In was For buffaloes the infection prevalence was 52.94% (27/51) and 71.64% (48/67) in spring and summer, respectively. Gastrointestinal parasites are a serious problem in cattle and buffaloes in the lower district of Dir Khyber Pakhtunkhwa Pakistan. In general, the burden of parasitic infection was low in most animals that received previous anti-parasitic treatment.

Phytogenic feed additives have been gaining considerable interest due to their ability to improve gut health and thereby performance of broiler chickens. The impact of Glycyrrhiza glabra (licorice) extract (GE) on expression of genes coding for tight junction proteins and gut protection and Campylobacter jejuni colonization in broilers has not been discussed until now. Thus, the current study assessed the effective dose of GE for maximum growth in broiler chickens, clear-cut molecular mechanisms related to integrity and health of intestine, and controlling C. jejuni colonization. Over a 35-day feeding period, a total of 500 Ross broiler chicks were allocated to five groups; the first group was fed a control diet without GE and the second group to the fifth group were fed a control diet with GE (0.25, 0.5, 1, and 2 g/kg of diet); each group comprised 100 chicks with 10 replicates (10 birds/replicate). Birds fed GE had an improved body weight gain and feed conversion ratio. Furthermore, the highest body weight gain was observed in the group that received 1 g/kg of GE (P < 0.05). The expression of genes coding for tight junction proteins [occludin and junctional adhesion molecules (JAM)] was upregulated in all groups supplemented with GE. Moreover, birds fed 1 g/kg of GE exhibited the maximum gene expression of occludin and JAM [0.2 and 0.3 fold change, respectively (P < 0.05)]. In relation to enterocyte protective genes [glucagon-like peptide (GLP-2) and fatty acid-binding protein (FABP-6)], use of GE significantly upregulated expression of GLP-2 gene with 0.8 fold change in 2 g/kg of the GE supplemented group (P < 0.05) while the expression of FABP-6 gene was not affected by GE supplementation (P > 0.05). After challenge with C. jejuni, the expression of mucin (MUC-2) gene was upregulated and the inflammatory markers such as Toll-like receptors (TLR-4) and interleukin (IL-1ß) were downregulated with increasing level of supplemented GE (P < 0.05). The mean log10 count of C. jejuni in cecal samples after 7 days post-infection by culture and real-time qPCR was decreased in groups fed GE in a dose-dependent manner (P < 0.05). In addition, the highest reduction of C. jejuni count in cecal samples by culture and real-time qPCR was observed in the group fed 2 g/kg of GE (2.58 and 2.28 log10 CFU/g, respectively). Results from this study suggested that G. glabra extract (1 g/kg) improved growth performance of broiler chickens, as well as influenced the maintenance of intestinal integrity and reduced C. jejuni shedding from infected birds.
