Medicinal signaling cells (MSC) hold promise for regenerative medicine due to their ability to repair damaged tissues. However, their effectiveness can be affected by how long they are cultured in the lab. This study investigated how passage number influences key properties for regenerative medicine of pig bone marrow MSC. The medicinal signiling cells derived from pig bone marrow (BM-MSC) were cultured in D-MEM High Glucose supplemented with 15% foetal bovine serum until the 25th passage and assessed their growth, viability, ability to differentiate into different cell types (plasticity), and cell cycle activity. Our findings showed that while the cells remained viable until the 25th passage, their ability to grow and differentiate declined after the 5th passage. Additionally, cells in later passages spent more time in a resting phase, suggesting reduced activity. In conclusion, the number of passages is a critical factor for maintaining ideal MSC characteristics. From the 9th passage BM-MSC exhibit decline in proliferation, differentiation potential, and cell cycle activity. Given this, it is possible to suggest that the use of 5th passage cells is the most suitable for therapeutic applications.

Brazil is strategic in controlling neglected zoonoses, such as glanders, in its territory. Among the Brazilian states, Piauí is a strategic state for the spread of the disease in the country. The present study aimed to evaluate the spatial and temporal distribution of official cases of glanders in Piauí between 2015 and 2022. The glanders cases were located in the municipalities of the north and central-north mesoregions, mainly in Campo Maior, Teresina and Altos. The highest incidence risk (IR) occurred in of Altos (IR = 257.9), Sussuapara (IR = 158.4), and Teresina (IR = 157.7). A primary cluster was formed with a relative risk of 14.88 between 2019 and 2022, encompassing 34 municipalities in the north and central-north regions. In Piauí, glanders is well localized, with the potential for spread across borders. This is the first study demonstrating the distribution of reported cases of glanders in the state of Piauí.

OBJECTIVE: This study aimed to assess the impact of ß-caryophyllene (BC) supplementation in the extender on the post-cryopreservation quality of semen from Dorper rams. METHODS: Six Dorper rams were utilized for semen collection over 16 weeks, with BC concentrations determined via the MTT test. Animals were divided into a control group and three treatment groups receiving BC at concentrations of 1.0mM, 2.0mM, and 3.0mM in the Trisegg yolk diluent. Semen was cryopreserved and stored in liquid nitrogen for at least 15 days. After thawing, in vitro assessments including CASA, acrosomal integrity, plasma membrane integrity, mitochondrial membrane potential, and thermo-resistance tests were conducted. Additionally, the TBARS assay was performed to evaluate oxidative stress. RESULTS: While BC supplementation did not significantly affect sperm motility, it notably improved mitochondrial potential and mitigated oxidative stress in cryopreserved ram semen. CONCLUSIONS: Incorporating ß-caryophyllene into the extender exhibited beneficial effects on the quality of Dorper ram semen post-cryopreservation, enhancing mitochondrial functionality and reducing oxidative stress.

The Brazilian Buriti oil presents low extraction costs and relevant antioxidant properties. Thus, this work aimed to analyze Buriti oil biomaterial (BB), within its physicochemical properties, biocompatibility and cellular integration, with the purpose to the use as a growth matrix for Goat Wharton's jelly mesenchymal stem cells. Biomaterials were produced from Buriti oil polymer (Mauritia flexuosa), for it's characterization were performed Infrared Region Spectroscopy (FTIR) and Thermogravimetric Analysis (TG and DTG). The biointegration was analyzed by Scanning Electron Microscopy (SEM) and histological techniques. In order to investigate biocompatibility, MTT (3-(4,5-dimetil-2-tiazolil)-2,5-difenil-2H-tetrazólio) test and hemolytic activity tests were performed. The activation capacity of immune system cellswas measured by phagocytic capacity assay and nitric oxide synthesis . The BB presented an amorphous composition, with high thermal stability and high water expansion capacity, a surface with micro and macropores, and good adhesion of Wharton's jelly mesenchymal stem cells (MSCWJ). We verified the absence of cytotoxicity and hemolytic activity, in addition, BB did not stimulate the activation of macrophages. Proving to be a safe material for direct cultivation and also for manufacturing of compounds used for in vivo applications.

OBJECTIVE: This study aimed to evaluate the inflammatory effect and bone formation in sterile surgical failures after implantation of a collagen sponge with mesenchymal stem cells from human dental pulp (hDPSCs) and Aloe vera. MATERIAL AND METHODS: Rattus norvegicus (n=75) were divided into five experimental groups according to treatment: G1) control (blood clot); G2) Hemospon®; G3) Hemospon® in a culture medium enriched with 8% Aloe vera; G4) Hemospon® in a culture medium containing hDPSCs and G5) Hemospon® in a culture medium enriched with 8% Aloe vera and hDPSCs. On days 7, 15 and 30, the animals were euthanized, and the tibia was dissected for histological, immunohistochemistry and immunofluorescence analyses. The results were analyzed using nonparametric Kruskal-Wallis test and Dunn's post-test. RESULTS: On days 7 and 15, the groups with Aloe vera had less average acute inflammatory infiltrate compared to the control group and the group with Hemospon® (p<0.05). No statistically significant difference was found between the groups regarding bone formation at the three experimental points in time. Osteopontin expression corroborated the intensity of bone formation. Fluorescence microscopy revealed positive labeling with Q-Tracker® in hDPSCs before transplantation and tissue repair. CONCLUSION: The results suggest that the combination of Hemospon®, Aloe vera and hDPSCs is a form of clinical treatment for the repair of non-critical bone defects that reduces the inflammatory cascade's effects.

Morphological features are essential to assess neonates' viability. In order to characterize the morphology and biometry of newborn agouti, 36 animals born at 103 days of gestation under human care in Teresina-PI, Brazil were evaluated. After birth, the newborns were weighed on digital precision scales, measured with a flexible tape measure and their external morphology was assessed. The animals observed had coat color trait of adults, open eyes, hairs in the nasal region and four incisor teeth. The forelimbs had developed four digits and the hind limbs three digits, with white edged claws and a short hairless tail. The following were observed: weight of 144.58 ± 33.39g and lengths: crown-rump 14.73 ± 1.82cm, total 19.88 ± 1.52cm, head to tail 16.95 ± 1.55cm, head 7.33 ± 0.59cm, thoracic girth 11.78 ± 1.36cm, abdominal girth 10.73 ± 1.21cm, tail length 1.18 ± 0.25cm, eye diameter 1.27 ± 0.27cm and ear length 0.21 ± 0.35cm. The agoutis presented morphological maturity at birth with external morphology very similar to that of the adult animals. Most of the births in this species are the double type and newborns that weigh more than 90g have better chances of survival in captivity.


As características morfológicas são essenciais para avaliar a viabilidade dos neonatos. Para caracterizar a morfologia e biometria de cutias recém-nascidas, foram avaliados 36 animais nascidos aos 103 dias de gestação sob cuidados em Teresina-PI, Brasil. Após o nascimento, os recém-nascidos foram pesados em balança digital de precisão, medidos com fita métrica flexível e avaliada sua morfologia externa. Os animais observados apresentavam traço de pelagem de adultos, olhos abertos, pelos na região nasal e quatro dentes incisivos. Os membros anteriores desenvolveram quatro dedos e os membros posteriores três, com garras brancas afiadas e uma cauda curta e sem pêlos. Foram observados: peso de 144,58 ± 33,39g e comprimentos: garupa 14,73 ± 1,82cm, total 19,88 ± 1,52cm, cabeça a cauda 16,95 ± 1,55cm, cabeça 7,33 ± 0,59cm, perímetro torácico 11,78 ± 1,36cm, abdominal perímetro 10,73 ± 1,21 cm, comprimento da cauda 1,18 ± 0,25 cm, diâmetro do olho 1,27 ± 0,27 cm e comprimento da orelha 0,21 ± 0,35 cm. As cutias apresentaram maturidade morfológica ao nascimento com morfologia externa muito semelhante à dos animais adultos. A maioria dos nascimentos nesta espécie são do tipo duplo e recém-nascidos que pesam mais de 90g têm melhores chances de sobrevivência sob cuidados humanos.

PURPOSE: To assess the efficacy of allogeneic mesenchymal stem cells and xenogenic platelet rich plasma in the treatment of bone failure of osteoporotic rabbits secondary to estrogenic deprivation and iatrogenic hypercortisolism. METHODS: Eight female rabbits underwent ovarian resection and corticoid therapy to induce clinical status of osteoporosis. Four failures were produced in the tibiae, with each failure being treated with hemostatic sponge, allogenic mesenchymal stem cells, xenogenic platelet-rich plasma and the association between both. The animals were divided into two groups, evaluated radiographically and histopathologically at 30 and 60 days post treatment. RESULTS: A radiographically confirmed consolidation of bone failures treated with allogeneic mesenchymal stem cells, associated with the histopathological image of mature and immature bone tissue, without evidence of osteopenia, was compared with the other groups, in which radiolucent failures with osteopenia and fibrosis were still present, denoting the satisfactory effect of the first treatment in detriment to the others. CONCLUSION: The treatment of bone failures of rabbits with secondary osteoporosis with allogeneic mesenchymal stem cells induced greater bone consolidation with mature and immature bone tissue production (p<0.01), when compared to the other treatments.

PURPOSE: The use of the collared peccary as an experimental model for ischemic nephropathy. METHODS: A total of 12 collared peccary (Tayassu tajacu) was used and ischemic nephropathy was induced in six of these animals that constituted the experimental group (G1) while the other six formed the control group (G2). Ischemic nephropathy was induced surgically by partial occlusion of the left renal artery. The disease course was assessed by hematological tests, serum chemistry, urinalysis, ultrasound (US) and doppler ultrasound function of the renal artery before induction, and at five, 10, 15 and 20 days after surgery. Twenty days after the occlusion, unilateral nephrectomy and histopathological examination were performed to assess renal morphology. RESULTS: Statistical analysis by Fischer's test showed a significant difference (p<0.05) between the control group and the experimental group. The histopathological examination showed glomerular, tubular and interstitial lesions. In the experimental group, 83.3% (5 /6) showed moderate renal lesions and only 16.7% (1/6) were classified with no lesions. The ultrasound examination of the right kidney presented statistical difference between day 5 and day 10 post occlusion. CONCLUSION: The collared peccary as a good experimental model for ischemic renal disease, because it could be manipulated during the research time without death, with health conditions that permit any subsequent procedure for disease therapy.

Stem cells are present in the adult tissues of most diverse species. Bone marrow is recognized to be the most exploited site to obtain stem cells and cell progenitors. The objective of the present study was to characterize hematopoietic progenitor (HP) morphology and analyze the performance of adherent cell progenitors (ACPs) cultivated in vitro from black-rumped agouti bone marrow (Dasyprocta prymnolopha). Bone marrow aspirates were obtained from tibia crest and used to prepare histological slides and identify cell morphology. Cells were also scattered on culture plates for later isolation, expansion, and quantification. Smears obtained from bone marrow demonstrated HPs at different stages of maturity. In culture, these cells showed fibroblastoid morphology and a strong tendency to form colonies, demonstrated by the presence of cell aggregates, cytoplasmic elongations lying side by side. An 80% cell confluence was observed at 18 days in culture and progressive reduction in the percentage of nonadherent mononuclear cells. After eight passes, a mean cell viability of 96.07% was observed, from a pool of 1.6 × 10(7) cells (ACP). Thirteen 25-cm(2) culture bottles were trypsinized, resuspended in freezing medium, stored in 14 criotubes at a concentration of 1 × 10(6) cells per milliliter, and placed in liquid nitrogen at -196°C. Agouti bone marrow demonstrated high plasticity, moreover different HP lines, and a population of adherent cells demonstrated morphology similar to mesenchymal stem cells in culture.

The aim of this estudy was to establish the levels of serum total protein, albumin, globulin, alanine aminotransferase (ALT), aspartate aminotransferase (AST), calcium, phosphorus, urea, creatinine, bilirubin and glucose during pregnancy in agoutis. Animals: Twelve pregnant agouti from the Center for the Study and Preservation of Wild Animals (CSPWA) of the Federal University of Piauí (UFPI) were used in this research. After identification of the estrus, the day of the coverage was confirmed by means of vaginal cytology with the visualization of spermatozoa (day zero) and confirmation of pregnancy by ultrasonographic examination after 15 days. Blood samples were collected by lateral saphenous vein puncture after physical restraint, every 10 days until the end of pregnancy, for biochemical analyzes. A completely randomized experimental design was used and the means compared by the Duncan test at 5% probability using the SAS (Statistical Analysis System). The results of the biochemical analysis of total protein, albumin, globulin, urea, creatinine, calcium, phosphorus, serum ALT, glucose, AST, total bilirubin, direct bilirubin and indirect bilirubin in pregnant agouti (Dasyprocta prymnolopha) did not differ when compared to nonpregnant females. The serum biochemical levels during pregnancy in agoutis, except for calcium and phosphorus, were unchanged compared to those found in the non-pregnant adult animal, as occurs in other species. The changes during pregnancy reflect the physiology and biology of wild species, elucidating information about the biochemical parameters during pregnancy, thus characterizing the animal as a benchmark for comparisons with other species, extolling its importance both for nature conservation and production in capivity.


O estudo objetivou estabelecer os níveis séricos de proteínas totais, albumina, globulina, Alanina Aminotransferase (ALT), Aspartato Aminotransferase (AST), cálcio, fósforo, ureia, creatinina, bilirrubina e glicose durante a gestação em cutias. A pesquisa foi desenvolvida utilizando-se 12 cutias fêmeas criadas no Núcleo de Estudos, Produção e Preservação de Animais Silvestres da Universidade Federal do Piauí. Após a identificação do estro, o dia da cobertura foi confirmado por meio de citologia vaginal com a visualização de espermatozoides (dia zero) e confirmação da gestação por exame ultrassonográfico após 15 dias. Confirmada a gestação, foram coletados 03 mL de sangue mediante punção da veia pudenda interna, após contenção física, a cada 10 dias, até o final da gestação. Foi feita a análise de variância para um delineamento inteiramente casualizado com teste de Duncan para comparação das médias a 5% de probabilidade utilizando-se do programa estatístico SAS (Statistical Analysis System). Os resultados obtidos por meio da análise bioquímica de proteína total, albumina, globulina, ureia, creatinina, cálcio, fósforo, ALT séricas, glicose, aspartato aminotransferase (AST), bilirrubina total, bilirrubina direta e bilirrubina indireta de cutias gestantes (Dasyprocta prymnolopha) diferem de forma absoluta quando comparados a fêmeas não gestantes. Os níveis bioquímicos séricos durante a gestação em cutias, com exceção do cálcio, fósforo, sofrem alterações comparadas ao animal adulto não prenhe, como ocorre em outras espécies. Os níveis nas cutias gestantes sofrem alterações de acordo com o tempo de gestação, com maiores mudanças no período inicial e final da prenhez. As mudanças durante a gravidez refletem a fisiologia e a biologia da espécie silvestre, elucidando informações sobre os parâmetros bioquímicos durante a gestação, caracterizando o animal como referência para comparações com outras espécies, exaltando a importância tanto para sua conservação quanto para a sua produção em cativeiro.

The histopathological examination showed glomerular, tubular and interstitial lesions. In the experimental group, 83.3% (5 /6) showed moderate renal lesions and only 16.7% (1/6) were classified with no lesions. The ultrasound examination of the right kidney presented statistical difference between day 5 and day 10 post occlusion. CONCLUSION: The collared peccary as a good experimental model for ischemic renal disease, because it could be manipulated during the research time without death, with health conditions that permit any subsequent procedure for disease therapy. .
