RESUMEN

The antimalarial efficacy of the most important vector control interventions-long-lasting insecticidal nets (LLINs) and indoor residual spraying (IRS)-primarily protect against mosquitoes' biting people when they are in bed and indoors. Mosquito bites taken outside of these times contribute to residual transmission which determines the maximum effectiveness of current malaria prevention. The likelihood mosquitoes feed outside the time of day when LLINs and IRS can protect people is poorly understood, and the proportion of bites received outdoors may be higher after prolonged vector control. A systematic review of mosquito and human behavior is used to quantify and estimate the public health impact of outdoor biting across Africa. On average 79% of bites by the major malaria vectors occur during the time when people are in bed. This estimate is substantially lower than previous predictions, with results suggesting a nearly 10% lower proportion of bites taken at the time when people are beneath LLINs since the year 2000. Across Africa, this higher outdoor transmission is predicted to result in an estimated 10.6 million additional malaria cases annually if universal LLIN and IRS coverage was achieved. Higher outdoor biting diminishes the cases of malaria averted by vector control. This reduction in LLIN effectiveness appears to be exacerbated in areas where mosquito populations are resistant to insecticides used in bed nets, but no association was found between physiological resistance and outdoor biting. Substantial spatial heterogeneity in mosquito biting behavior between communities could contribute to differences in effectiveness of malaria control across Africa.

BACKGROUND: Large-scale use of insecticide-treated nets and indoor residual spraying have contributed to a significant decrease in malaria transmission worldwide. Further reduction and progress towards elimination, however, require complementary control measures which can address the remaining gaps in protection from mosquito bites. Following the development of novel pyrethroids with high knockdown effects on malaria vectors, programmatic use of spatial repellents has been suggested as one potential strategy to fill the gaps. This report explores social and contextual factors that may influence the relevance, uptake and sustainable use of a spatial repellent in two remote villages in Mondulkiri province, Cambodia, with endemic malaria transmission. The repellent consisted of polyethylene emanators, held in an open plastic frame and impregnated with 10% metofluthrin. RESULTS: In a baseline survey, 90.9% of households in Ou Chra (n = 30/33) and 96.6% in Pu Cha (n = 57/59) were interviewed. Behavioural data were collected for all household occupants (n = 448). In both villages, there were times and places in which people remained exposed to mosquito bites. Prior to the installation of the repellent, 50.6 and 59.5% of respondents noted that bites occurred "very often" inside the house and in the outdoor area surrounding the house, respectively. Indoor biting was reported to occur more frequently in the evening, followed by at night, while outdoor biting occurred more frequently in the early morning. In a follow-up survey, spatial repellents were well received in both villages, although 63.2% of respondents would not replace bed nets with repellents. Most participants (96.6%) were willing to use the product again; the mean willingness to pay was US$ 0.3 per unit. A preference for local procurement methods emerged. CONCLUSION: Widespread use of spatial repellents would not fill all protective gaps, but, if their entomological efficacy can be ascertained, outdoor application has the potential to enhance vector control strategies in Cambodia. Successful implementation would require subsidisation and integration with the existing national malaria control strategy. It is hoped that this study, while contributing to a better understanding of the social contexts of residual malaria transmission, will generate further interest in the evaluation of spatial repellents for malaria control.

RESUMEN

Disease surveillance, including entomological surveillance, serves as the basis for all vector control program activities. How to do this in the most ecologically sensible way, so that the most suitable, naturalistic method, of control for that population can be identified, should be a priority. Here we describe a set of techniques, whose only energy requirement is a torch (flashlight), that can be used to collect both endo and exophagic and endo and exophilic malaria vectors. The data obtained over a number of years from an individual sentinel house in a village in Mozambique and from a village in Cambodia using these kinds of collection techniques, is presented.

Treating both male and female Anopheles gambiae as if they are "boids" (a computer program that mimics flocking in birds) explains much of the swarming and mating behavior in this important group of malaria vectors. It is suggested that species specific swarm sites act as the mate recognition system in anophelines and it is proposed that virgin females respond to the swarm site per se rather than the swarm itself. Given the high operational sex ratio and the inability of any male to dominate all females within the swarm, it is considered that chance, rather than sexual selection, is the most important determinant of mating. The male being in the swarm may be a sufficiently strong signal to the female of his fitness, so that more elaborate sexual selection is unnecessary. The possibility of alternative mechanisms for mating may also exist but need to be investigated further. Given the importance of swarms as the isolating mechanism between species, emphasis should be placed on determining the characteristics of swarm sites and markers between them.

BACKGROUND: With the possible implications of global warming, the effect of temperature on the dynamics of malaria vectors in Africa has become a subject of increasing interest. Information from the field is, however, relatively sparse. We describe the effect of ambient temperature over a five-year period on the dynamics of An. funestus and An. gambiae s.l., collected from a single village in southern Mozambique where temperatures varied from a night-time minimum of 6 °C in the cool season to a daytime maximum of 35 °C in the hot season. RESULTS: Mean daily air temperatures varied from 34 °C to 20 °C and soil temperatures varied from 26 °C to 12 °C. Diurnal variation was greatest in the cooler months of the year and were greater in air temperatures than soil temperatures. During the study 301, 705 female An. funestus were collected in 6,043 light-trap collections, 161, 466 in 7,397 exit collections and 16, 995 in 1,315 resting collections. The equivalent numbers for An. gambiae s.l. are 72,475 in light-traps, 33, 868 in exit collections and 5,333 from indoor resting collections. Numbers of mosquito were greatest in the warmer months. Numbers of An. gambiae s.l. went through a one hundredfold change (from a mean of 0.14 mosquitoes a night to 14) whereas numbers of An. funestus merely doubled (from a mean of 20 to 40 a night). The highest environmental correlations and mosquito numbers were between mean air temperature (r2 = 0.52 for An. funestus and 0.77 for An. gambiae s.l.). Numbers of mosquito collected were not related to rainfall with lags of up to four weeks. Numbers of both gravid and unfed An. gambiae complex females in exit collections continued to increase at all temperatures recorded but gravid females of An. funestus decreased at temperatures above 28 °C. Overall the numbers of gravid and unfed An. funestus collected in exit collections were not correlated (p = 0.07). For an unknown reason the number of An. gambiae s.l. fell below monitoring thresholds during the study. CONCLUSIONS: Mean air temperature was the most important environmental parameter affecting both vectors in this part of Mozambique. Numbers of An. gambiae s.l. increased at all temperatures recorded whilst An. funestus appeared to be adversely affected by temperatures of 28 °C and above. These differences may influence the distribution of the vectors as the planet warms.

BACKGROUND: Clarifying the role of the innate immune system of the malaria vector Anopheles gambiae is a potential way to block the development of the Plasmodium parasites. Pathogen recognition is the first step of innate immune response, where pattern recognition proteins like GNBPs play a central role. RESULTS: We analysed 70 sequences of the protein coding gene GNBPB2 from two species, Anopheles gambiae (s.s.) and An. coluzzii, collected in six African countries. We detected 135 segregating sites defining 63 distinct haplotypes and 30 proteins. Mean nucleotide diversity (π) was 0.014 for both species. We found no significant genetic differentiation between species, but a significant positive correlation between genetic differentiation and geographical distance among populations. CONCLUSIONS: Species status seems to contribute less for the molecular differentiation in GNBPB2 than geographical region in the African continent (West and East). Purifying selection was found to be the most common form of selection, as in many other immunity-related genes. Diversifying selection may be also operating in the GNBPB2 gene.

Isolated areas, such as the 2 × 7 km peninsula of Linga Linga in Mozambique, are the places where malaria might be most easily eliminated. Currently available control strategies include long-lasting insecticidal bednets impregnated with pyrethroid insecticides (LLINs), rapid diagnostic tests (RDTs) for diagnosis and artemisinin based combination therapy (ACT) for treatment and these were applied on the peninsula. In 2007, following a census of the population and mapping of 500 households, five annual all-age prevalence surveys were conducted. Information on LLIN use, house construction, and animal ownership was obtained. A spatially structured generalized additive model indicated that malaria risk was greatest towards the northern end of the peninsula and that people living in houses with grass or thatch roofs had a greater risk of malaria than those living in houses with corrugated iron roofs. Incidence peaked nine weeks after rainfall (r (2) = 0.34, p = 0.0002). From 2009 incidence was measured at a centrally based project clinic. The proportion of under nine-year-old resident attendees diagnosed with malaria decreased significantly from 48% in 2009, to 35% in 2010 and 25% in 2011. At the same time, there was a shift in the peak age of cases from 1-4 year olds to 5-9 year olds. Nevertheless, in order to further reduce malaria transmission in an area such as Linga Linga, additional vector control measures need to be considered.

The evolutionary processes at play between island and mainland populations of the malaria mosquito vector Anopheles gambiae sensu stricto are of great interest as islands may be suitable sites for preliminary application of transgenic-based vector control strategies. São Tomé and Príncipe, located off the West African coast, have received such attention in recent years. This study investigates the degree of isolation of An. gambiae s.s. populations between these islands and the mainland based on mitochondrial and ribosomal DNA molecular data. We identify possible continental localities from which these island populations derived. For these purposes, we used F ST values, haplotype networks, and nested clade analysis to estimate migration rates and patterns. Haplotypes from both markers are geographically widespread across the African continent. Results indicate that the populations from São Tomé and Príncipe are relatively isolated from continental African populations, suggesting they are promising sites for test releases of transgenic individuals. These island populations are possibly derived from two separate continental migrations. This result is discussed in the context of the history of the African slave trade with respect to São Tomé and Príncipe.

How often insecticide resistance mutations arise in natural insect populations is a fundamental question for understanding the evolution of resistance and also for modeling its spread. Moreover, the development of resistance is regarded as a favored model to study the molecular evolution of adaptive traits. In the malaria vector Anopheles gambiae two point mutations (L1014F and L1014S) in the voltage-gated sodium channel gene, that confer knockdown resistance (kdr) to DDT and pyrethroid insecticides, have been described. In order to determine whether resistance alleles result from single or multiple mutation events, genotyping of the kdr locus and partial sequencing of the upstream intron-1 was performed on a total of 288 A. gambiae S-form collected from 28 localities in 15 countries. Knockdown resistance alleles were found to be widespread in West Africa with co-occurrence of both 1014S and 1014F in West-Central localities. Differences in intron-1 haplotype composition suggest that kdr alleles may have arisen from at least four independent mutation events. Neutrality tests provided evidence for a selective sweep acting on this genomic region, particularly in West Africa. The frequency and distribution of these kdr haplotypes varied geographically, being influenced by an interplay between different mutational occurrences, gene flow and local selection. This has important practical implications for the management and sustainability of malaria vector control programs.
