PREMISE OF THE STUDY: Microsatellite markers were developed in Veratrum maackii (Melanthiaceae) to assess the pattern of population genetic structures across the species' distribution. METHODS AND RESULTS: Ten polymorphic loci were isolated from V. maackii using 454 shotgun pyrosequencing and the biotin capture method. The number of alleles per locus ranged from three to 19. The observed heterozygosity and expected heterozygosity ranged from 0.143 to 0.929 and 0.269 to 0.904, respectively. CONCLUSIONS: These newly developed microsatellite markers are useful for assessing the genetic diversity, population structure, and demographic history of V. maackii across its distribution range.

Although dendritic cells (DCs) play an important role in tumor immunity, there have been no reports on their role in cholangiocellular carcinoma (CCC). In 26 formalin-fixed, paraffin-embedded tissue sections from patients with CCC, cells positive for CD83 (a marker of mature DCs), CD1a (a marker of immature DCs), and CD8 and CD4 (T cell markers) were counted, and expression of glucose-regulated protein (grp) 94, which is considered to participate in the maturation of DCs, was evaluated by immunohistochemistry and Western blot analysis to study the relationship between their expression and patients' disease outcome. The number of CD83-positive DCs at the invasive margin of CCCs correlated significantly with the number of CD8-positive or CD4-positive T cells in the cancerous region and was significantly higher in grp94-positive cancer than in grp94-negative cancer (P = 0.0006). CD83-positive patients (positive cells in invasive margin > 12.4/field) had both a significantly lower incidence of lymph node metastasis (23.1% vs 69.2%; P = 0.0206) and a better outcome than CD83-negative patients (P <0.001). We conclude that mature DCs are distributed predominantly at the invasive margin of cancers, and a significantly higher number of mature DCs at the invasive margin are observed in patients with grp94-positive cancer cells. Mature DCs may enhance CD8- and CD4-positive cell infiltration into cancers and improve prognosis in patients with CCC, due in part to abatement of lymph node metastasis.

RESUMEN

We report three patients with recurrent gastric cancer responding to TS-1 therapy after combination chemotherapy with 5-fluorouracil, mitomycin C and cisplatin. All 3 cases had undergone total gastrectomy with lymphadenectomy for advanced gastric cancer. Postoperative follow-up computed tomography (CT) showed liver metastases (cases 1 and 3), peritoneal dissemination (case 2) and enlargement of paraaortic lymph nodes (case 1) due to cancer recurrence. After 2 to 4 courses of combined treatment with 5-fluorouracil (500-750 mg/body/day, days 1-5, civ), mitomycin C (6-8 mg/body, day 6) and cisplatin (60-80 mg/body, day 7), CT revealed considerable reduction of the metastatic tumors. Subsequently oral administration of TS-1 (80-100 mg/body/day) for 4 weeks was performed. All 3 patients are well without any signs of increase in tumor size.

We have recently proposed a new general concept regarding amphiphilic materials that have been named as "active interfacial modifier (AIM)." In emulsion systems, an AIM is essentially insoluble in both water and organic solvents; however, it possesses moieties that are attracted to each of these immiscible liquid phases. Hence, an AIM practically stays just at the interface between the two phases and makes the resulting emulsion stable. In this study, the effects of silicone oil species on the dispersion stability of water-in-oil (W/O) emulsions in the presence of an AIM sample were evaluated in order to understand the destabilization mechanism in such emulsion systems. The AIM sample used in this study is an amphiphilic polymer consisting of a silicone backbone modified with hydrocarbon chains and hydrolyzed silk peptides. The Stokes equation predicts that the sedimentation velocity of water droplets dispersed in a continuous silicone oil phase simply depends on the expression (ρ - ρ0)/η assuming that the droplet size is constant (where ρ is the density of the dispersed water phase, ρ0 is the density of the continuous silicone oil phase, and η is the viscosity of the oil phase). The experimental results shown in this paper are consistent with the Stokes prediction: i.e., in the low-viscous genuine or quasi-Newtonian fluid region, the dispersion stability increases in the following order: dodecamethylpentasiloxane (DPS) < decamethylcyclopentasiloxane (D5) ≤ dodecamethylcyclohexasiloxane (D6). This order agrees well with the order obtained by using the expression (ρ - ρ0)/η as DPS > D5 > D6. This indicates that our emulsion system experiences destabilization through sedimentation, but hardly any coalescence occurs owing to the presence of an additional third phase consisting of the AIM that stabilizes the silicone oil/water interface in the emulsions.
