With petroleum exploration and development expanding in the Arctic (AMAP, 2007) there is a need to obtain additional information on the ecotoxicology of Arctic organisms. Here we perform 192 h laboratory exposure experiments on the keystone Arctic zooplankton species, Calanus finmarchicus. We trace the accumulation and depuration of two polycyclic aromatic hydrocarbons (PAHs): phenanthrene and benzo[a]pyrene (B[a]P) using (14)C labeled PAH compounds. Copepods were not fed during the experiment, limiting uptake to diffusion processes alone. The lighter PAH compound, phenanthrene, accumulated rapidly in C. finmarchicus, reaching steady state within 96 h. The heavier PAH compound, B[a]P, accumulated more slowly and steady state was not reached within the 192 h exposure period. As expected, the bioconcentration factor (BCF) for B[a]P was higher than for phenanthrene in accordance with a higher octanol/water partition coefficient for B[a]P (log K(ow)=6.04) compared to phenanthrene (log K(ow)=4.53). However, for both compounds, log BCF was lower than log K(ow) that may indicate active biotransformation and excretion of the selected PAH compounds. These findings on the bio-uptake kinetics for petroleum hydrocarbons are essential for evaluating the potential consequences of an oil spill in the Arctic.

Reliable risk assessment approaches for Arctic environments are requested to manage potential impacts associated with increased activities in Arctic regions. We performed toxicity tests on Arctic and temperate species exposed to the narcotic acting oil component, 2-methyl naphthalene. The experimental results were used to quantify concentration causing lethality to 50% of exposed individuals and no-effect concentration (individual level). For estimates at community level, the hazardous concentrations affecting 5% and 50% of the species were calculated from sensitivity distribution curves. These survival metrics were then used to elucidate whether temperate toxicity data used in risk assessment are sufficiently representative for the Arctic. Taking data uncertainty into consideration, we found no regional difference in tolerances to 2-methyl naphthalene either at the species level or at the community level. Hence these data support a conclusion that values of survival metrics for temperate regions are transferrable to the Arctic for the chemical 2-methyl naphthalene, as long as extrapolation techniques are properly applied and uncertainties are taken into consideration.

Concentrations and biomagnification of total mercury (TotHg) and methyl mercury (MeHg) were studied in selected species from the pelagic food web in Kongsfjorden, Svalbard. Twelve species of zooplankton, fish and seabirds, were sampled representing a gradient of trophic positions in the Svalbard marine food web. TotHg and MeHg were analysed in liver, muscle and/or whole specimens. The present study is the first to provide MeHg levels in seabirds from the Svalbard area. The relative MeHg levels decreased with increasing levels of TotHg in seabird tissues. Stable isotopes of nitrogen (delta(15)N) were used to determine the trophic levels and the rate of biomagnification of mercury in the food web. A linear relationship between mercury levels and trophic position was found for all seabird species combined and their trophic level, but there was no relationship within species. Biomagnification factors were all >1 for both TotHg and MeHg, indicating biomagnification from prey to predator. TotHg levels in the different seabirds were similar to levels detected in the Kongsfjorden area in the 1990s.

Stress caused by environmental factors evokes dynamic changes in plant phenotypes. In this study, we deciphered simultaneously the reaction of plant growth and chlorophyll fluorescence related parameters using a novel approach which combines existing imaging technologies (GROWSCREEN FLUORO). Three different abiotic stress situations were investigated demonstrating the benefit of this approach to distinguish between effects related to (1) growth, (2) chlorophyll-fluorescence, or (3) both of these aspects of the phenotype. In a drought stress experiment with more than 500 plants, poly(ADP-ribose) polymerase (PARP) deficient lines of Arabidopsis thaliana (L.) Heynh showed increased relative growth rates (RGR) compared with C24 wild-type plants. In chilling stress, growth of PARP and C24 lines decreased rapidly, followed by a decrease in Fv/Fm. Here, PARP-plants showed a more pronounced decrease of Fv/Fm than C24, which can be interpreted as a more efficient strategy for survival in mild chilling stress. Finally, the reaction of Nicotiana tabacum L. to altered spectral composition of the intercepted light was monitored as an example of a moderate stress situation that affects chlorophyll-fluorescence related, but not growth-related parameters. The examples investigated in this study show the capacity for improved plant phenotyping based on an automated and simultaneous evaluation of growth and photosynthesis at high throughput.
