RESUMEN

Model-based testing (MBT) has been extensively researched for software-intensive systems but, despite the academic interest, adoption of the technique in industry has been sparse. This phenomenon has been observed by our industrial partners for MBT with graphical models. They perceive one cause to be a lack of evidence-based MBT guidelines that, in addition to technical guidelines, also take non-technical aspects into account. This hypothesis is supported by a lack of such guidelines in the literature. Objective: The objective of this study is to elicit, and synthesize, MBT experts' best practices for MBT with graphical models. The results aim to give guidance to practitioners and aspire to give researchers new insights to inspire future research. Method: An interview survey is conducted using deep, semi-structured, interviews with an international sample of 17 MBT experts, in different roles, from software industry. Interview results are synthesised through semantic equivalence analysis and verified by MBT experts from industrial practice. Results: 13 synthesised conclusions are drawn from which 23 best-practice guidelines are derived for the adoption, use and abandonment of the technique. In addition, observations and expert insights are discussed that help explain the lack of wide-spread adoption of MBT with graphical models in industrial practice. Conclusions: Several technical aspects of MBT are covered by the results as well as conclusions that cover process- and organizational factors. These factors relate to the mindset, knowledge, organization, mandate and resources that enable the technique to be used effectively within an organization. The guidelines presented in this work complement existing knowledge and, as a primary objective, provide guidance for industrial practitioners to better succeed with MBT with graphical models.

The Japanese red and pink corals are known to be precious because of their commercial value resulting from their use in ornaments, jewelry, and medicine. Precious corals are very interesting models for biomineralization studies and possess two different skeletal structures: an axial skeleton and an endoskeleton (sclerites). Although it has long been known that the organic matrix proteins existing in coral skeletons are critical for the oriented precipitation of CaCO3 crystals, these proteins in moderate deep-sea Japanese precious corals remain uncharacterized. Therefore, in this study, we performed skeletal whole proteome analyses using 1D and 2D electrophoresis, nano-LC, and MALDI-TOF-TOF MS. We identified a total of 147 functional coral skeletal organic matrix proteins (120 from the sclerites and 36 from the axial skeleton), including two calcium-binding calmodulin. Among the organic matrix proteins identified, nine key proteins are highly typical and common in both skeletons. Strong glycosylation activity, which is essential for skeletal formation in calcifying organisms, was detected in both skeletons. This work demonstrates unique biomineralization-related proteins in precious corals and provides the first description of the major proteinaceous components of CaCO3 minerals in precious corals, enabling the comparative investigation of biocalcification in other octocorals.

RESUMEN

A 41-year-old man with repaired tetralogy of Fallot and a single coronary artery (CA) arising anteriorly presented with dyspnea in the setting of moderate right ventricle-pulmonary artery conduit (RV-PAC) stenosis and moderate-to-severe extrinsic left main CA compression between the aorta and RV-PAC. His CA stenosis resolved after successful RV-PAC replacement.

BACKGROUND: Findings regarding the protective effect of Angiotensin II receptor blockers (ARBs) against Alzheimer's disease and related dementias (AD/ADRD) and cognitive decline have been inconclusive. METHODS: Individuals with hypertension who do not have any prior ADRD diagnosis were included in this retrospective cohort study from Optum's de-identified Clinformatics® Data Mart. We identified antihypertensive medication (AHM) drug classes and subclassified ARBs by blood-brain barrier (BBB) permeability. We compared baseline characteristics and used the Kaplan-Meier (KM) survival curve and adjusted Cox proportional hazards (PH) model for survival analyses. FINDINGS: From 6,390,826 individuals with hypertension, there were 1,839,176 ARB users, 3,366,841 non-ARB AHM users, and 1,184,809 AHM non-users. The unadjusted KM curve showed that ARB users had lower cumulative hazard than other AHM users or AHM non-users (P < 0.0001). In Cox PH analysis, ARB users showed a 20% lower adjusted hazard of developing ADRD compared to angiotensin-converting enzyme inhibitor (ACEI) users and a 29% and 18% reduced hazard when compared to non-ARB/ACEI AHM users and AHM non-users (all P < 0.0001). Consumption of BBB-crossing ARBs was linked to a lower hazard of ADRD development than non-BBB-crossing ARBs, undetermined ARBs, and non-consumption of AHMs by 11%, 25%, and 31% (all P < 0.0001). INTERPRETATION: This study suggests that ARBs are superior to ACEIs, non-ARB/ACEI AHMs, or non-use of AHMs in reducing the hazard of ADRD among patients with hypertension. Also, BBB-permeability in ARBs was associated with lower ADRD incidence. There is no cure for AD, ADRD, or vascular dementia; hence, these findings are significant in preventing those disorders in an inexpensive, convenient, and safe way. Limitations in claims data should be considered when interpreting our findings. A growing body of research has begun to link exposure to environmental contaminants, such as heavy metals, with a variety of negative health outcomes. In this paper, we sought to review the current research describing the impact of certain common contaminant metals on cardiovascular (CV) health. We reviewed ten metals: lead, barium, nickel, chromium, cadmium, arsenic, mercury, selenium, zinc, and copper. After a literature review, we briefly summarized the routes of environmental exposure, pathophysiological mechanisms, CV health impacts, and exposure prevention and/or mitigation strategies for each metal. The resulting article discloses a broad spectrum of pathological significance, from relatively benign substances with little to no described effects on CV health, such as chromium and selenium, to substances with a wide-ranging and relatively severe spectrum of CV pathologies, such as arsenic, cadmium, and lead. It is our hope that this article will provide clinicians with a practical overview of the impact of these common environmental contaminants on CV health as well as highlight areas that require further investigation to better understand how these metals impact the incidence and progression of CV diseases.

Conspicuous ornaments are often considered a result of evolution by sexual selection. According to the social selection hypothesis, such conspicuous traits may also evolve as badges of status associated with increased boldness or aggression toward conspecifics in conflicts about ecological resources. This study tested predictions from the social selection hypothesis to explain evolution of conspicuous red color of the pelvic spines of the three-spine stickleback (Gasterosteus aculeatus). Wild nonreproducing sticklebacks were presented to pairs of dummies which differed at their pelvic spines, having either (i) normal-sized gray or red pelvic spines or (ii) normal-sized gray or large red pelvic spines. The experimental tank was illuminated by white or green light, since green light impedes the sticklebacks' ability to detect red color. The dummies moved slowly around in circles at each end of the experimental tank. We quantified the parameters (i) which of the two dummies was visited first, (ii) time taken before the first visit to a dummy, (iii) distribution of the focal sticklebacks in the two zones close to each of the two dummies and in the neutral zone of the tank, (iv) close to which of the two dummies did the focal fish eat its first food-piece, and (v) time spent until the first piece of food was eaten. This was carried out for 22 females and 29 males sticklebacks. The results suggested no effect of the color or size of the dummies' pelvic spines, on none of the five behavioral parameters. Moreover, neither the color of the pelvic spines of the focal sticklebacks themselves (as opposed to redness of the dummies' spines) nor their body length was associated with behavior toward the dummies. Thus, this study did not support predictions from the social selection hypothesis to explain evolution of red pelvic spines in sticklebacks.

Apilactobacillus kunkeei is a fructophilic lactic acid bacterium found in fructose-rich environments such as flowers, fruits, fermented food, honey, and honeydew, as well as in the guts of fructose-feeding insects. We report here the draft genome sequences of three Apilactobacillus kunkeei strains isolated from the gut microbial community of three honeybees.

Background: Reassuring information is recommended in clinical guidelines for the treatment of low back pain (LBP), but has not been clearly defined. The Consultation-based Reassurance Questionnaire (CRQ) was developed as a tool for measuring to what extent reassurance is present in back pain consultations and may provide important information about the clinical encounter. Until now the CRQ has only been tested in general practice patients in the UK although many patients with LBP are seen outside of this setting. The objectives of this study were to translate the CRQ into Danish, test its feasibility in chiropractic practice, and determine if CRQ scores were associated with satisfaction with care and perceived pain control. Methods: On the day of the first visit for a LBP episode, patients received an electronic survey including the CRQ. Distributions and completeness of responses on the four subscales of the CRQ (data-gathering, relationship-building, generic reassurance, cognitive reassurance) were assessed, and internal consistency for each subscale calculated as Cronbach's alpha. Outcomes at 2 weeks were; satisfaction with care (5-point Likert scale dichotomised into yes/no) and ability to control pain (0-10). Associations of the CRQ with patient characteristics and outcomes were determined in mixed models to account for dependency of observations within clinics. Results: From 964 patients visiting between November 2016 and October 2017 with new episodes of LBP, 717 completed the CRQ with no more than 1% missing values on any single item. The internal consistency was acceptable for all subscales (0.67-0.86). Scores were generally high, and more so in patients visiting a chiropractor for the first time. All four subscales were positively associated with satisfaction (Odds ratios 1.08-1.23) and generic reassurance was weakly associated with pain control (ß = 0.07 [95% CI 0.03-0.11]). Conclusions: The CRQ was feasible for use in a Danish chiropractic setting and scores on all four reassurance subscales related positively to patients' satisfaction. Patients who had visited a chiropractor previously reported slightly lower levels of reassuring information, and it should be explored if this is in accordance with the patients' needs. The potential impact on patient outcomes needs investigation.

Microbiota of beehive products are very little known. We report here for the first time six metagenomes of royal jelly, pollen, and different types of honey from wild and cultivated lavender, chestnut, and fir honeydew. Four metagenomes of epiphytic and endophytic microbiota of lavender and rose flowers are also reported.

A redox-active compound has been purified from the barley powdery mildew fungus Blumeria ( Erysiphe ) graminis f. sp. hordei. A combination of spectrophotometry, MS and NMR has identified it as 3-hydroxykynurenine (3OHKyn). This compound, never previously detected in any fungus or pathogen, is best known for its role in vertebrate cataracts. It is found abundantly in developing and germinating spores and also in runner hyphae. Two roles for 3OHKyn are discussed: first, the presence of active oxygen species would enable 3OHKyn to cross-link the spore chemically with the plant. Secondly, it may be acting as an UV protectant and an antioxidant.
