RESUMEN

Elastic waves guided along bars of rectangular cross sections exhibit complex dispersion. This paper studies in-plane modes propagating at low frequencies in thin isotropic rectangular waveguides through experiments and numerical simulations. These modes result from the coupling at the edge between the first order shear horizontal mode SH0 of phase velocity equal to the shear velocity VT and the first order symmetrical Lamb mode S0 of phase velocity equal to the plate velocity VP. In the low frequency domain, the dispersion curves of these modes are close to those of Lamb modes propagating in plates of bulk wave velocities VP and VT. The dispersion curves of backward modes and the associated zero group velocity (ZGV) resonances are measured in a metal tape using noncontact laser ultrasonic techniques. Numerical calculations of in-plane modes in a soft ribbon of Poisson's ratio ν≈0.5 confirm that, due to very low shear velocity, backward waves and ZGV modes exist at frequencies that are hundreds of times lower than ZGV resonances in metal tapes of the same geometry. The results are compared to theoretical dispersion curves calculated using the method provided in Krushynska and Meleshko [J. Acoust. Soc. Am. 129, 1324-1335 (2011)].

BACKGROUND: Basophil activation contributes to inflammatory reactions, especially in allergy. It is controlled, both positively and negatively, by several mechanisms. High-affinity IgE receptors (FcεRI) generate a mixture of activation and inhibition signals when aggregated, the ratio of which depends on the concentration of allergen recognized by receptor-bound IgE. Low-affinity IgG receptors (FcγRIIA/B) generate inhibition signals when coengaged with FcεRI by allergen-antibody immune complexes. Commensal and probiotic bacteria, such as Lactobacillus paracasei, generate inhibition signals through still unclear mechanisms. OBJECTIVE: We sought to investigate whether mechanisms that control, both positively and negatively, basophil activation, which were unraveled and studied in basophils from healthy donors, are functional in allergic patients. METHODS: FcεRI and FcγRIIA/B expression, FcεRI-dependent activation, FcεRI-dependent inhibition, and FcγRIIB-dependent inhibition were examined in blood basophils incubated overnight with or without L paracasei and challenged under 10 experimental conditions. Basophils from healthy donors were compared with basophils from patients who consulted an allergology outpatient clinic over a period of 3 months with respiratory allergy, anaphylaxis antecedents, chronic urticaria, and/or atopic dermatitis. RESULTS: Patients' basophils expressed neither more FcεRI nor less FcγRIIB than basophils from healthy donors. They were neither hyperreactive to positive regulation nor hyporeactive to negative regulation, irrespective of the receptors or mechanisms involved and the allergic manifestations of the patients. CONCLUSION: Regulatory mechanisms that control basophil activation are fully functional in allergic patients. Intrinsic defects in these mechanisms do not explain allergic manifestations. Based on these mechanisms, immune checkpoint modifiers can be developed as novel therapeutic tools for allergy.

RESUMEN

A method to recover the elastic properties, thickness, or orientation of the principal symmetry axes of anisotropic plates is presented. This method relies on the measurements of multimode guided waves, which are launched and detected in arbitrary directions along the plate using a multi-element linear transducer array driven by a programmable electronic device. A model-based inverse problem solution is proposed to optimally recover the properties of interest. The main contribution consists in defining an objective function built from the dispersion equation, which allows accounting for higher-order modes without the need to pair each experimental data point to a specific guided mode. This avoids the numerical calculation of the dispersion curves and errors in the mode identification. Compared to standard root-finding algorithms, the computational gain of the procedure is estimated to be on the order of 200. The objective function is optimized using genetic algorithms, which allow identifying from a single out-of-symmetry axis measurement the full set of anisotropic elastic coefficients and either the plate thickness or the propagation direction. The efficiency of the method is demonstrated using data measured on materials with different symmetry classes. Excellent agreement is found between the reported estimates and reference values from the literature.

BACKGROUND: As a consequence of drug sequestration, increase in volume of distribution, or alteration of elimination, extracorporeal membrane oxygenation (ECMO) might lead to inadequate plasma concentrations of vital drugs. The aim of this experimental study was to develop an ex vivo model to better characterize the impact of ECMO procedure on beta-lactam antibiotics pharmacokinetics. METHODS: Plasma concentrations of cefotaxime, ceftazidime, cefepime, piperacillin, oxacillin, amoxicillin, and ceftriaxone were measured in an ex vivo ECMO circuit primed with whole human blood and compared with controls stored in glass tubes and polyvinyl chloride tubing. Serial blood samples were collected over 48 hours, and the concentrations of beta-lactam antibiotics were quantified using a validated high-performance liquid chromatography assay. The concentrations' decay rate over time was compared between the ECMO circuits and controls using nonlinear mixed-effect modeling. RESULTS: Cefotaxime concentrations decreased markedly: 86% of the initial concentration remained after 4 hours and only 21% after 48 hours (P < 0.05 for the comparison in rate of decrease with both glass and polyvinyl chloride controls). There was no difference in the rate of decrease between ECMO circuit and controls for the other beta-lactam antibiotics. The average drug recoveries from the ECMO circuits at 48 hours were as follows: ceftazidime, 73%; cefepime, 67%; piperacillin, 71%; oxacillin, 46%; and amoxicillin, 72%. Concentrations of ceftriaxone remained stable throughout the 48-hour study both in ECMO circuits and in controls. CONCLUSIONS: Significant losses of cefotaxime were observed, whereas ceftazidime, cefepime, piperacillin, oxacillin, and amoxicillin decrease was moderate and similar to that of the control group, and ceftriaxone concentrations remained unchanged. These results are reassuring for the use of beta-lactam antibiotics in critically ill patients treated with ECMO.

Optimization of Lamb modes induced by laser can be achieved by adjusting the spatial source distribution to the mode wavelength (λ). The excitability of Zero-Group Velocity (ZGV) resonances in isotropic plates is investigated both theoretically and experimentally for axially symmetric sources. Optimal parameters and amplitude gains are derived analytically for spot and annular sources of either Gaussian or rectangular energy profiles. For a Gaussian spot source, the optimal radius is found to be λZGV/π. Annular sources increase the amplitude by at least a factor of 3 compared to the optimal Gaussian source. Rectangular energy profiles provide higher gain than Gaussian ones. These predictions are confirmed by semi-analytical simulation of the thermoelastic generation of Lamb waves, including the effect of material attenuation. Experimentally, Gaussian ring sources of controlled width and radius are produced with an axicon-lens system. Measured optimal geometric parameters obtained for Gaussian and annular beams are in good agreement with theoretical predictions. A ZGV resonance amplification factor of 2.1 is obtained with the Gaussian ring. Such source should facilitate the inspection of highly attenuating plates made of low ablation threshold materials like composites.

The paper studies the interaction of Lamb waves with the free edge of a plate. The reflection coefficients of a Lamb mode at a plate free edge are calculated using a semi-analytical method, as a function of frequency and angle of incidence. The conversion between forward and backward Lamb modes is thoroughly investigated. It is shown that at the zero-group velocity (ZGV) frequency, the forward S1 Lamb mode fully converts into the backward S2b Lamb mode at normal incidence. Besides, this conversion is very efficient over most of the angular spectrum and remains dominant at frequencies just above the ZGV-point. This effect is observed experimentally on a Duralumin plate. First, the S1 Lamb mode is selectively generated using a transducer array, second the S2b mode is excited using a single circular transducer. The normal displacement field is probed with an interferometer. The free edge is shown to retro-focus the incident wave at different depths depending on the wave number mismatch between the forward and backward propagating modes. In the vicinity of the ZGV-point, wave numbers coincide and the wave is retro-reflected on the source. In this frequency range, the free edge acts as a perfect phase conjugating mirror.

The transient response of an elastic cylinder to a laser impact is studied. When the laser source is a line perpendicular to the cylinder axis, modes guided along the cylinder are generated. For a millimetric steel cylinder up to ten narrow resonances can be locally detected by a laser interferometer below 8 MHz. These resonances correspond to Zero-Group Velocity guided modes or to circumferential modes. The authors observe that the theory describing the propagation of elastic waves in an isotropic cylinder is not sufficient to precisely predict the resonance spectrum. In fact, the texture of such elongated structure manifests as elastic anisotropy. Thus, a transverse isotropic (TI) model is used to calculate the dispersion curves and compare them with the measured ones, obtained by moving the source along the cylinder. The five elastic constants of a TI cylinder are adjusted, leading to a good agreement between measured and theoretical dispersion curves. Then, all the resonance frequencies are satisfactorily identified.

Zero-group velocity (ZGV) Lamb modes are associated with sharp local acoustic resonances and allow, among other features, local measurement of Poisson's ratio. While the thermoelastic generation of Lamb waves in metal plates has been widely studied, the case of materials of low-optical absorption remains unexplored. In materials such as glasses, the generation of bulk elastic waves has been demonstrated to be sensitive to the refracted light distribution. In this paper, a detailed analysis of the effect of light refraction on the laser-based generation of ZGV Lamb modes is presented. Experiments are performed on a bare glass plate without the need for an additional layer for light absorption or reflection. Using an appropriate tilted volume source, it is shown that the laser-ultrasonic technique allows non-contact measurement of the Poisson's ratio.

Zero-Group Velocity (ZGV) Lamb waves are studied in a structure composed of two plates bonded by an adhesive layer. The dispersion curves are calculated for a Duralumin/epoxy/Duralumin sample, where the adhesion is modeled by a normal and a tangential spring at both interfaces. Several ZGV modes are identified and their frequency dependence on interfacial stiffnesses and on the bonding layer thickness is numerically studied. Then, experiments achieved with laser ultrasonic techniques are presented. Local resonances are measured using a superimposed source and probe. Knowing the thicknesses and elastic constants of the Duralumin and epoxy layers, the comparison between theoretical and experimental ZGV resonances leads to an evaluation of the interfacial stiffnesses. A good agreement with theoretical dispersion curves confirms the identification of the resonances and the parameter estimations. This non-contact technique is promising for the local evaluation of bonded structures.

RESUMEN

Thirty years ago, theorists showed that a properly designed combination of incident waves could be fully transmitted through (or reflected by) a disordered medium, based on the existence of propagation channels which are essentially either closed or open (bimodal law). In this Letter, we study elastic waves in a disordered waveguide and present direct experimental evidence of the bimodal law. Full transmission and reflection are achieved. The wave field is monitored by laser interferometry and highlights the interference effects that take place within the scattering medium.

BACKGROUND: Hereditary angioedema (HAE) is a rare and potentially life-threatening disease. New specific treatments are available. OBJECTIVE: To identify patients' features and patients' best therapeutic option. METHODS: A 1-year, multicenter, retrospective study was performed. The primary objective was to examine the clinical presentation of HAE. Secondary objectives included patient characteristics, management of HAE over 12 months, and health-related quality of life using the SF-36v2 questionnaire. RESULTS: One hundred ninety-three patients were included, and 69.4% were women. In the 12-month period, the mean number of HAE attacks was 7.6. Among the 568 reported attacks, localizations were the abdomen (57.1%), peripheral limbs (42.5%), upper airway (7.9%), and face (6.9%); 31.6% of attacks were severe and occurred statistically more often in women (P < .02). Compared with a population of allergic patients, all age- and sex-adjusted scores were significantly lower in patients with HAE (P < .05) except for the physical component summary. Health-related quality of life negatively correlated with the annual number of attacks and was markedly altered for patients having more than 5 attacks per year (P < .05 for all dimensions). CONCLUSION: HAE is a severe disease that places a heavy burden on quality of life.

Basophils express not only high-affinity IgE receptors, but also low-affinity IgG receptors. Which, among these receptors, are expressed by human basophils is poorly known. Low-affinity IgG receptors comprise CD32 (FcgammaRIIA, FcgammaRIIB, and FcgammaRIIC) and CD16 (FcgammaRIIIA and FcgammaRIIIB). FcgammaRIIA, FcgammaRIIC, and FcgammaRIIIA are activating receptors, FcgammaRIIB are inhibitory receptors, FcgammaRIIIB are GPI-anchored receptors whose function is poorly understood. Basophils were reported to express FcgammaRII, but not FcgammaRIII. We aimed at further identifying basophil IgG receptors. Basophils from normal donors and from patients suffering from an allergic skin disease (atopic dermatitis), allergic respiratory diseases (allergic rhinitis and asthma), or a nonallergic skin disease (chronic urticaria) were examined. We found that normal basophils contain FcgammaRIII transcripts and express FcgammaRIIIB, but not FcgammaRIIIA, which were detected on 24-81% basophils from normal donors and on 12-100% basophils from patients. Noticeably, the proportion of FcgammaRIIIB(+) basophils was significantly lower in atopic dermatitis patients than in other subjects. This decreased FcgammaRIII expression was not correlated with an activated phenotype of basophils in atopic dermatitis patients, although FcgammaRIIIB expression was down-regulated upon basophil activation by anti-IgE. Our results challenge the two dogmas 1) that basophils do not express FcgammaRIII and 2) that FcgammaRIIIB is exclusively expressed by neutrophils. They suggest that a proportion of basophils may be lost during enrichment procedures in which FcgammaRIII(+) cells are discarded by negative sorting using anti-CD16 Abs. They unravel an unexpected complexity of IgG receptors susceptible to modulate basophil activation. They identify a novel systemic alteration in atopic dermatitis.

We report on experimental and numerical implementations of devices based on the negative refraction of elastic guided waves, the so-called Lamb waves. Consisting in plates of varying thickness, these devices rely on the concept of complementary media, where a particular layout of negative index media can cloak an object with its anti-object or trap waves around a negative corner. The diffraction cancellation operated by negative refraction is investigated by means of laser ultrasound experiments. However, unlike original theoretical predictions, these intriguing wave phenomena remain, nevertheless, limited to the propagating component of the wave-field. To go beyond the diffraction limit, negative refraction is combined with the concept of metalens, a device converting the evanescent components of an object into propagating waves. The transport of an evanescent wave-field is then possible from an object plane to a far-field imaging plane. Twenty years after Pendry's initial proposal, this work thus paves the way towards an elastic superlens.

The propagation of waves in complex media can be harnessed either by taming the incident wave-field impinging on the medium or by forcing waves along desired paths through its careful design. These two alternative strategies have given rise to fascinating concepts such as time reversal or negative refraction. Here, we show how these two processes are intimately linked through the negative reflection phenomenon. A negative reflecting mirror converts a wave of positive phase velocity into its negative counterpart and vice versa. In this article, we experimentally demonstrate this phenomenon with elastic waves in a 2D billiard and in a disordered plate by means of laser interferometry. Despite the complexity of such configurations, the negatively reflected wave field focuses back towards the initial source location, thereby mimicking a phase conjugation operation while being a fully passive process. The super-focusing capability of negative reflection is also highlighted in a monochromatic regime. The negative reflection phenomenon is not restricted to guided elastic waves since it can occur in zero-gap systems such as photonic crystals, chiral metamaterials or graphene. Negative reflection can thus become a tool of choice for the control of waves in all fields of wave physics.

Zero-group-velocity (ZGV) waves have the peculiarity of being stationary, and thus locally confining energy. Although they are particularly useful in evaluation applications, they have not yet been tracked in two dimensions. Here we image gigahertz zero-group-velocity Lamb waves in the time domain by means of an ultrafast optical technique, revealing their stationary nature and their acoustic energy localization. The acoustic field is imaged to micron resolution on a nanoscale bilayer consisting of a silicon-nitride plate coated with a titanium film. Temporal and spatiotemporal Fourier transforms combined with a technique involving the intensity modulation of the optical pump and probe beams gives access to arbitrary acoustic frequencies, allowing ZGV modes to be isolated. The dispersion curves of the bilayer system are extracted together with the quality factor Q and lifetime of the first ZGV mode. Applications include the testing of bonded nanostructures.

The effect of microstructural elongation on ultrasonic backscattered fields was studied. Two methods for determining the elongation direction of macrozones in titanium alloys, using the anisotropic spatial coherence of the backscattered field, are presented. Both methods use a phased array attached on a rotative holder that records the array response matrix at several angles. Two titanium alloys were investigated: TA6V and Ti17. TA6V exhibited a strong macrozone elongation, whereas Ti17 macrozones were found equiaxial. The first method is based on the measurement of backscattered intensity in function of the probe angle relative to the macrozones elongation direction. An angular dependence of backscattered intensity is observed in presence of elongated scatterers, and their elongation direction is collinear with the probe direction corresponding to a minimal intensity. This variability is linked to both piezoelectric shape and the backscattered field spatial properties. The second method is based on the measurement of the relative proportion of single to multiple scattering in a diffusive media, using a simplified version of the single scattering filter developed in Aubry and Derode (2009). It allows the measurement of the level of multiple scattering: both titanium alloys exhibited strong multiple scattering. The elongation direction was determined as the direction of minimal multiple scattering. Furthermore, these results were confirmed by the measurement of the coherent backscattering cone on both samples.

PURPOSE: Acquired angioedema type 1 is characterized by a C1 inhibitor deficiency in patients with lymphoproliferative disorders, whereas acquired angioedema type 2 is characterized by anti-C1 inhibitor antibodies, and has not been thought to be associated with lymphoproliferative disease. We studied the clinical features, complement profiles, and associated diseases in 19 new patients with diagnosed acquired angioedema type 2. SUBJECTS AND METHODS: Plasma concentrations and functional activity of complement components were measured by conventional techniques. Functional C1 inhibitor activity was assessed by a chromogenic assay. Autoantibodies to C1 inhibitor were detected using an enzyme-linked immunosorbent assay. RESULTS: The 11 men and 8 women (median age, 60 years) presented with recurrent attacks of angioedema. All patients had detectable anti-C1 inhibitor antibodies in serum. A monoclonal gammopathy was detected in 12 patients (63%) at the time of diagnosis, 11 of whom had an immunoglobulin peak of the same heavy- and light-chain isotypes as the acquired anti-C1 inhibitor antibody. Three of these 12 patients developed a malignant lymphoproliferative disease. CONCLUSION: As with type 1 disease, a large proportion of patients with acquired angioedema type 2 have a lymphoproliferative disorder.
