Mesenchymal stem/stromal cells (MSCs) are spindle-like heterogeneous cell populations with advantageous bidirectional immunomodulatory and hematopoietic support effects. Vascular cellular adhesion molecule-1 (VCAM-1)+ MSCs have been reported to exhibit immunoregulatory and proangiogenic capacities. Here, we studied the effects of VCAM-1+ human umbilical cord (hUC)-MSCs on neuroprotection against cerebral infarction. Sprague-Dawley rats were subjected to middle cerebral artery occlusion (MCAO), and VCAM-1- and VCAM-1+ hUC-MSCs were intravenously injected into the rat 4 h post-MCAO surgery. Thereafter, modified neurological severity scores (mNSS) were determined, and the Morris water maze test, 2,3,5-triphenyltetrazolium chloride (TTC), hematoxylin and eosin (H&E), Nissl, TUNEL staining, and qRT-PCR were conducted. Following induction of oxygen-glucose deprivation/reoxygenation (OGD/R), SH-SY5Y cells were co-cultured with VCAM-1- and VCAM-1+ hUC-MSCs. CCK-8, flow cytometry, ELISA, and western blot analyses were performed in vitro. Compared with VCAM-1- hUC-MSCs, administration of VCAM-1+ hUC-MSCs revealed improved therapeutic efficacy against cerebral infarction in rats, as confirmed by lower mNSS scores and infarct volumes, as well as improved learning and memory capacities. In addition, VCAM-1+ hUC-MSCs exhibited improved efficacy against neurological defects in rats with cerebral infarction, accompanied by inhibition of the NLRP3-mediated inflammatory response. VCAM-1+ hUC-MSC co-culture improved the viability and diminished NLRP3-mediated inflammatory response in OGD/R-treated SH-SY5Y cells. Moreover, NLRP3 overexpression in SH-SY5Y cells prevented the beneficial effects of VCAM-1+ hUC-MSC co-culture. Overall, our findings demonstrated the relevance of VCAM-1+ hUC-MSC-based cytotherapy for preclinical neuroprotection against cerebral infarction.

The optical interference effect originating from the multiple reflections between the two-dimensional (2D) materials and the substrates has been used to dramatically enhance their Raman signal. However, this effect in the hybrid structures of colloidal quantum dots (QD) coupled to 2D materials is always overlooked. Here we theoretically prove that the photoluminescence (PL) intensities of the QD films in the QD-2D hybrid structures can be strongly enhanced and modulated by the optical interference effect between QD and 2D interfaces, breaking the inherent standpoint that PL intensities of the QD films are always prominently quenched in these hybrid structures. The theoretical predictions have been well confirmed by experimental measurements of PL properties of CdSe/ZnS and CdSeTe/ZnS QD on different 2D materials (such as WSe2, MoS2, and h-BN). PL intensities of these QD films have been periodically modulated from almost disappearing to strong enhancement (with the enhancement of about 6 times). The optical interference effect uncovered in this work enables a powerful method to manipulate the PL property of the QD films in the different QD-2D hybrid structures. These results can boost the optical performance of the QD-based electronic and optoelectronic devices in the hybrid QD-2D structures.

Fomesafen herbicide application has become major pollution in the growth and production of crops. Spraying fomesafen on the target crops may drift out to non-target crops. In northeast China, sugar beets are always planted adjacent to soybeans. Salicylic acid (SA) plays an important role in crop growth and alleviating abiotic stress, however, the role of SA in relieving fomesafen stress in sugar beet growth has rarely been investigated. Therefore, a pot study was conducted to elucidate the effects of different concentrations (0.025, 0.25, 0.5, 1, 5, and 10 mM) of SA on morphological parameters, photosynthetic performance, and antioxidant defense system in sugar beet seedlings under fomesafen (22.5 g a.i. ha-1) stress. The results showed that fomesafen stress inhibited the growth of sugar beet seedlings, and photosynthetic performance, while increased membrane lipid peroxidation and oxidative stress. However, exogenous SA alleviated the fomesafen stress and increased plant height, biomass, photosynthetic pigment contents, net photosynthetic rate (Pn), and photochemical efficiency of PSⅡ (Fv/Fm) in sugar beet leaves. Meanwhile, exogenous SA maintained the cell membrane integrity by reducing the content of malondialdehyde (MDA) and electrolyte permeability and regulating the activities of antioxidant enzymes including superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), and polyphenol (PPO). Therefore, it is concluded that exogenous SA ameliorated the adverse effects of fomesafen on the growth of sugar beet seedlings, with a pronounced effect at 1 mM SA. The present study results may have useful implications in managing other plants that are poisoned by herbicides.

GaTe nanoflakes have been receiving much research attention recently due to their applications in optoelectronic devices, such as anisotropic non-volatile memory, solar cells, and high-sensitivity photodetectors from the ultraviolet to the visible region. Further applications, however, have been impeded due to the limited understanding of their exciton dynamics. In this work we perform temperature- and power-dependent time-resolved photoluminescence (PL) spectra to comprehensively investigate the exciton dynamics of GaTe nanoflakes. Temperature-dependent PL measurements manifest that spectral profiles of GaTe nanoflakes change dramatically from cryogenic to room temperature, where the bound exciton and donor-to-acceptor pair transition normally disappear above 100 K, while the charged exciton survives to room temperature. The lifetimes of these excitons and their evolution vs temperature have been uncovered by time-resolved PL spectra. Further measurements reveal the entirely different power-dependent exciton behaviors of GaTe nanoflakes between room and cryogenic temperatures. The underlying mechanisms have been proposed to explore the sophisticated exciton dynamics within GaTe nanoflakes. Our results offer a more thorough understanding of the exciton dynamics of GaTe nanoflakes, enabling further progress in engineering GaTe-based applications, such as photodetectors, light-emitting diodes, and nanoelectronics.

BACKGROUND: Continuous cropping stress involves such factors as biological barriers, allelopathic autotoxicity, deterioration of soil physicochemical properties, and soil fertility imbalance and is regarded as a kind of comprehensive stress limiting soybean yield and quality. Genomic DNA methylation is an important regulatory mechanism for plants to resist various environmental stresses. Therefore, it is especially worthwhile to reveal genomic methylation characteristics under stress and clarify the relationship between DNA methylation status and continuous cropping stress adaptability in soybean. RESULTS: We generated a genome-wide map of cytosine methylation induced by this kind of comprehensive stress in a tolerant soybean variety (Kang Xian 2, KX2) and a sensitive variety (He Feng, HF55) using whole-genome bisulfite sequencing (WGBS) technology. The expression of DNA demethylase genes was detected using real-time quantitative PCR (qRT-PCR). The functions of differentially methylated genes (DMGs) involved in stress response in biochemical metabolism and genetic information transmission were further assessed based on Gene Ontology (GO) annotation and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis. The results showed that genomic DNA demethylation was closely related to continuous cropping comprehensive stress adaptability in soybean, which was further verified by the increasing expression of DNA demethylases ROS1 and DML. The demethylation of mCpG and mCpHpG (mCpApG preferred) contexts was more critical, which mainly occurred in gene-regulatory regions at the whole-chromosome scale. Moreover, this kind of stress adaptability may be related to various stress responders generated through strengthened glucose catabolism and amino acid and fatty acid anabolism, as well as fidelity transmission of genetic information. CONCLUSIONS: Genomic DNA demethylation was closely associated with continuous cropping comprehensive stress adaptability, highlighting the promising potential of screening continuous cropping-tolerant cultivars by DNA methylation index and further exploring the application of DNA demethylases in soybean breeding.

Plants synthesize and accumulate a series of stress-resistance proteins to protect normal physiological activities under adverse conditions. Chimonanthus praecox which blooms in freezing weather accumulates late embryogenesis abundant proteins (LEAs) in flowers, but C. praecox LEAs are little reported. Here, we report a group of five LEA genes of C. praecox (CpLEA5, KT727031). Prokaryotic-expressed CpLEA5 was employed in Escherichia coli to investigate bioactivities and membrane permeability at low-temperature. In comparison with the vacant strains, CpLEA5-containing strains survived in a 20% higher rate; and the degree of cell membrane damage in CpLEA5-containing strains was 55% of that of the vacant strains according to a conductivity test, revealing the low-temperature resistance of CpLEA5 in bacteria. CpLEA5 was also expressed in Pichia pastoris. Interestingly, besides low-temperature resistance, CpLEA5 conferred high resistance to salt and alkali in CpLEA5 overexpressing yeast. The CpLEA5 gene was transferred into Arabidopsis thaliana to also demonstrate CpLEA5 actions in plants. As expected, the transgenic lines were more resistant against low-temperature and drought while compared with the wild type. Taken together, CpLEA5-conferred resistances to several conditions in prokaryote and eukaryotes could have great value as a genetic technology to enhance osmotic stress and low-temperature tolerance.

High salt in soils is one of the abiotic stresses that significantly reduces crop yield, although saline lands are considered potential resources arable for agriculture. Currently, genetic engineering for enhancing salt tolerance is being tested as an efficient and viable strategy for crop improvement. We previously characterized a large subunit of the ribosomal protein RPL44, which is involved in osmotic stress in the extremely halophilic fungus Aspergillus glaucus. Here, we screened another ribosomal protein (AgRPS3aE) that also produced high-salt tolerance in yeast. Bioinformatics analysis indicated that AgRPS3aE encodes a 29.2 kDa small subunit of a ribosomal protein belonging to the RPS3Ae family in eukaryotes. To further confirm its protective function against salinity, we expressed AgRPS3aE in three heterologous systems, the filamentous fungus Magnaporthe oryzae and two model plants Arabidopsis and tobacco. Overexpression of AgRPS3aE in all tested transformants significantly alleviated stress symptoms compared with controls, suggesting that AgRPS3aE functions not only in fungi but also in plants. Considering that ribosomal proteins are housekeeping components in organisms from prokaryotes to eukaryotes, we propose that AgRPS3aE is one of the optimal genes for improving high-salt tolerance in crops.

Cold acclimation is an important process to increase freezing tolerance for over-winter survival in many organisms. The apoplastic area is very important in cold acclimation. Two-dimensional electrophoresis was used to identify apoplastic proteins involved in the cold acclimation process of the filamentous fungus Magnaporthe oryzae, and nine protein spots showed at least 1.5-fold increase during cold treatment. These proteins were further analyzed by matrix-assisted laser-desorption/ionization time-of-flight mass spectrometry. One of these proteins was identified to be an adenosine kinase (MoAK), an ortholog of the adenosine kinase from Saccharomyces cerevisiae. The MoAK gene showed significantly increased in transcription level. Microscopic analyses showed that an MoAK::GFP fusion protein was localized in the apoplastic region. The MoAk protein showed anti-freezing activity when expressed in yeast. These results indicated that cold acclimation is crucial for fungal freezing tolerance and MoAK played an important role in this process in M. oryzae.

Introduction: Rice blast disease caused by Magnaporthe oryzae has long been the main cause of rice (Oryza sativa L.) yield reduction worldwide. The quinone external inhibitor pyraclostrobin is widely used as a fungicide to effectively control the spread of pathogenic fungi, including M. oryzae. However, M. oryzae can develop resistance through multiple levels of mutation, such as target protein cytb mutation G143A/S, leading to a decrease in the effectiveness of the biocide after a period of application. Therefore, uncovering the possible mutational mechanisms from multiple perspectives will further provide feasible targets for drug development. Methods: In this work, we determined the gene expression changes in M. oryzae in response to pyraclostrobin stress and their relationship with DNA methylation by transcriptome and methylome. Results: The results showed that under pyraclostrobin treatment, endoplasmic reticulum (ER)-associated and ubiquitin-mediated proteolysis were enhanced, suggesting that more aberrant proteins may be generated that need to be cleared. DNA replication and repair processes were inhibited. Glutathione metabolism was enhanced, while lipid metabolism was impaired. The number of alternative splicing events increased. These changes may be related to the elevated methylation levels of cytosine and adenine in gene bodies. Both hypermethylation and hypomethylation of differentially methylated genes (DMGs) mainly occurred in exons and promoters. Some DMGs and differentially expressed genes (DEGs) were annotated to the same pathways by GO and KEGG, including protein processing in the ER, ubiquitin-mediated proteolysis, RNA transport and glutathione metabolism, suggesting that pyraclostrobin may affect gene expression by altering the methylation patterns of cytosine and adenine. Discussion: Our results revealed that 5mC and 6mA in the gene body are associated with gene expression and contribute to adversity adaptation in M. oryzae. This enriched the understanding for potential mechanism of quinone inhibitor resistance, which will facilitate the development of feasible strategies for maintaining the high efficacy of this kind of fungicide.

Cydonia Oblonga Mill. is widely distributed in Turkey, Uzbekistan and China and commonly used by the food industry to produce jam, jelly and candies. The aim of this study was to investigate the in vitro antidiabetic activity and anti-diabetic mechanism of Cydonia Oblonga Mill. fruit (COMF). The chemical compositions were further characterized in COMF by UPLC-Q-Orbitrap/MS and 65 components including 22 flavonoids, 16 organic acids, 11 polyphenols, 5 amino acids, 3 pentacyclic triterpenoids and 8 other compounds were identified. The antioxidant activity by DPPH scavenging method and α-glucosidase inhibitory activity were tested. Furthermore, we detected the effects of COMF extract on the proliferation activity of HUVECs, cell viability of HUVECs under H2O2-induced oxidative stress, and NO production. Then, molecular docking activity and α-glucosidase inhibitory activity of seven key flavonoid components selected by bioinformatics analysis and literature in the COMF were studied. Among them, quercetin showed potent inhibitory activity, kaempferol, isorhamnetin, luteolin and apigenin demonstrated moderate inhibitory activity, while rutin and epicatechin exhibited poor inhibitory activity. Subsequently, the effects of quercetin, kaempferol, isorhamnetin, leteolin and apigenin on the gene expression levels of AKT1, IL-6 and VEGFA were verified by real-time fluorescence quantification (RT-qPCR). Molecular biology result showed that different active ingredients can significantly recover the levels of AKT1, IL-6 and VEGFA in HUVECs injured by high glucose.

Mung bean is characterized by having a good edible and medicinal value, while its flowers and pods have low production. Being a tertiary amine, DCPTA [2-(3,4-dichlorophenoxy) triethylamine] substantially regulates the growth and development of crops, maintaining production. Yet it is still limited in terms of the regulation of DCPTA on growth and development, including the yield and sugar metabolism of mung bean. In this study, DCPTA was sprayed at the beginning of mung flowering through a two-season cultivation, to assess its effects on the yield, leaf area per plant, plant height, seed setting rate, photosynthesis, chlorophyll content, and endogenous protective enzymes. Experimental results illustrated that relative to the control (CK), the DCPTA application significantly (p < 0.05) improved the yield of Bailv 11 mung bean, which rose to 6.9% in 2020 and 7.8% in 2021, respectively. This effect positively corresponded to a significant (p<0.05) increase in the number of pods and grains per plant and pod setting rate, but a non-significant difference in 1,000-grain weight. DCPA application also increased the area and fresh weight of leaf, mung height, and its organ dry weight (i.e., leaf, branch, and stem). During plant growth over DCPTA application, the increased activities of SOD, POD, and CAT improved the net photosynthetic rate, stomatal conductance, and transpiration. In addition, transcriptome sequencing further demonstrated that DCPTA treatment significantly (p < 0.05) up-regulated the sucrose synthase, invertase, and fructose kinase in all organs (i.e., leaves, pod skins, and grains) of the plant. In particular, this effect was much greater in the sucrose synthesis (i.e., sucrose content) in leaves. Our study, therefore, concludes that DCPTA application promotes the yield of mung bean via likely enhancing its photosynthetic capacity and sucrose synthase, fructokinase, and beta-fructofuranosidase expression regulation.

The nano-opto-electro-mechanical systems (NOEMS) are a class of hybrid solid devices that hold promises in both classical and quantum manipulations of the interplay between one or more degrees of freedom in optical, electrical and mechanical modes. To date, studies of NOEMS using van der Waals (vdW) heterostructures are very limited, although vdW materials are known for emerging phenomena such as spin, valley, and topological physics. Here, we devise a universal method to easily and robustly fabricate vdW heterostructures into an architecture that hosts opto-electro-mechanical couplings in one single device. We demonstrated several functionalities, including nano-mechanical resonator, vacuum channel diodes, and ultrafast thermo-radiator, using monolithically sculpted graphene NOEMS as a platform. Optical readout of electric and magnetic field tuning of mechanical resonance in a CrOCl/graphene vdW NOEMS is further demonstrated. Our results suggest that the introduction of the vdW heterostructure into the NOEMS family will be of particular potential for the development of novel lab-on-a-chip systems.

As two coexisting abiotic stresses, salt stress and alkali stress have severely restricted the development of global agriculture. Clarifying the plant resistance mechanism and determining how to improve plant tolerance to salt stress and alkali stress have been popular research topics. At present, most related studies have focused mainly on salt stress, and salt-alkali mixed stress studies are relatively scarce. However, in nature, high concentrations of salt and high pH often occur simultaneously, and their synergistic effects can be more harmful to plant growth and development than the effects of either stress alone. Therefore, it is of great practical importance for the sustainable development of agriculture to study plant resistance mechanisms under saline-alkali mixed stress, screen new saline-alkali stress tolerance genes, and explore new plant salt-alkali tolerance strategies. Herein, we summarized how plants actively respond to saline-alkali stress through morphological adaptation, physiological adaptation and molecular regulation.

Monolayer transition metal dichalcogenides, manifesting strong spin-orbit coupling combined with broken inversion symmetry, lead to coupling of spin and valley degrees of freedom. These unique features make them highly interesting for potential spintronic and valleytronic applications. However, engineering spin-orbit coupling at room temperature as demanded after device fabrication is still a great challenge for their practical applications. Here we reversibly engineer the spin-orbit coupling of monolayer MoS2 by laser irradiation under controlled gas environments, where the spin-orbit splitting has been effectively regulated within 140 meV to 200 meV. Furthermore, the photoluminescence intensity of the B exciton can be reversibly manipulated over 2 orders of magnitude. We attribute the engineering of spin-orbit splitting to the reduction of binding energy combined with band renormalization, originating from the enhanced absorption coefficient of monolayer MoS2 under inert gases and subsequently the significantly boosted carrier concentrations. Reflectance contrast spectra during the engineering stages provide unambiguous proof to support our interpretation. Our approach offers a new avenue to actively control the spin-orbit splitting in transition metal dichalcogenide materials at room temperature and paves the way for designing innovative spintronic devices.

van der Waals (vdW) heterostructures of transition metal dichalcogenides (TMDCs) provide an excellent paradigm for next-generation electronic and optoelectronic applications. However, the reproducible fabrications of vdW heterostructure devices and the boosting of practical applications are severely hindered by their unstable performance, due to the lack of criteria to assess the interlayer coupling in heterostructures. Here we propose a physical model involving ultrafast electron transfer in the heterostructures and provide two criteria, η (the ratio of the transferred electrons to the total excited electrons) and ζ (the relative photoluminescence variation), to evaluate the interlayer coupling by considering the electron transfer in TMDC heterostructures and numerically simulating the corresponding rate equations. We have proved the effectiveness and robustness of two criteria by measuring the pump-probe photoluminescence intensity of monolayer WS2 in the WS2/WSe2 heterostructures. During thermal annealing of WS2/WSe2, ζ varies from negative to positive values and η changes between 0 and 4.5 × 10-3 as the coupling strength enhanced; both of them can well characterize the tuning of interlayer coupling. We also design a scheme to image the interlayer coupling by performing PL imaging at two time delays. Our scheme offers powerful criteria to assess the interlayer coupling in TMDC heterostructures, offering opportunities for the implementation of vdW heterostructures for broadband and high-performance electronic and optoelectronic applications.

RESUMEN

Monolayer transition metal dichalcogenides (TMDs) are direct gap semiconductors with promising applications in diverse optoelectronic devices. To improve devices' performance, recent investigations have been systematically focused on the tuning of their optical properties. However, an all-optical approach with the reversible feature is still a challenge. Here we demonstrate the tunability of the photoluminescence (PL) properties of monolayer WS2 via laser irradiation. The broad-range and continuous modulation of PL intensity, as well as the conversion between neutral and charged excitons have been readily and reversibly achieved by only switching the two laser power densities. We attribute the reversible manipulation to the laser-assisted adsorption and desorption of gas molecules, which will deplete or release free electrons from the surface of WS2 and thus modify its PL properties. This all-optical manipulation, with advantages of reversibility, quantitative control, and high spatial resolution, suggests promising applications of TMDs monolayers in optoelectronic and nanophotonic applications, such as erasable optical data storage, micropatterning, and display.
