RESUMEN

U6 snRNA, as an essential component of the catalytic core of the pre-mRNA processing spliceosome, is heavily modified post-transcriptionally, with 2'-O-methylation being most common. The role of these modifications in pre-mRNA splicing as well as their physiological function in mammals have remained largely unclear. Here we report that the La-related protein LARP7 functions as a critical cofactor for 2'-O-methylation of U6 in mouse male germ cells. Mechanistically, LARP7 promotes U6 loading onto box C/D snoRNP, facilitating U6 2'-O-methylation by box C/D snoRNP. Importantly, ablation of LARP7 in the male germline causes defective U6 2'-O-methylation, massive alterations in pre-mRNA splicing, and spermatogenic failure in mice, which can be rescued by ectopic expression of wild-type LARP7 but not an U6-loading-deficient mutant LARP7. Our data uncover a novel role of LARP7 in regulating U6 2'-O-methylation and demonstrate the functional requirement of such modification for splicing fidelity and spermatogenesis in mice.

Inspired by energy conversion and waste reuse, hybridized Ni-MOF derivative-CdS-DETA/g-C3N5, a type-II heterojunction photocatalyst, is synthesized by a hydrothermal method for simultaneous and highly efficient photocatalytic degradation and hydrogen evolution in dye wastewater. Without the addition of cocatalysts and sacrificial agents, the optimal MOF-CD(2)/CN5 (i.e. Ni-MOF derivative-CdS-DETA (20 wt.%)/g-C3N5) exhibit good bifunctional catalytic activity, with a H2 evolution rate of 2974.4 µmol g-1 h-1 during the degradation of rhodamine B (RhB), and a removal rate of 99.97% for RhB. In the process of H2-evolution-only, triethanolamine is used as a sacrificial agent, exhibiting a high H2 evolution rate (19663.1 µmol g-1 h-1) in the absence of a cocatalyst, and outperforming most similar related materials (such as MOF/g-C3N5, MOF-CdS, CdS/g-C3N5). With the help of type-II heterojunction, holes are scavenged for the oxidative degradation of RhB, and electrons are used in the decomposition of water for H2 evolution during illumination. This work opens a new path for photocatalysts with dual functions of simultaneous efficient degradation and hydrogen evolution.

A novel ternary photocatalyst Ni3S4@ZIS@C3N5 with type II and Z-type heterojunctions was synthesized for the first time by hydrothermal and electrostatic self-assembly methods, effectively avoiding the thermal decomposition of C3N5 during the synthesis of the complex. The best ternary catalyst Ni3S4@ZIS@C3N5 is capable of achieving an optimal hydrogen evolution rate of 9750 mmol g-1 h-1, which is approximately 10.89 times as high as that of C3N5, indicating that the complex effectively enhanced the photocatalytic properties of the monomer. The coexistence of two types of heterojunctions in the complex effectively enhances photocatalytic performance, in which the monomer ZIS constructs type II scheme with Ni3S4 and Z-type scheme with C3N5, respectively. The two heterojunctions complement each other and jointly promote the rapid electron transfer from Ni3S4 and C3N5 to the ZIS surface. In conclusion, the cooperation of the two heterojunctions efficiently facilitates the migration of photogenerated carriers, thus enhancing the photocatalytic hydrogen generation performance of Ni3S4@ZIS@C3N5.

We experimentally apply incoherent Fourier ptychography to enhance the resolution of recorded images by projecting known, uncorrelated, random patterns at high speed onto 3D moving and distant objects. We find that the resolution enhancement factor can be greater than 2, depending on the projection and camera optics.

In this study, simple-structured wavelength sensors were developed by depositing two back-to-back Au/MAPbI3/Au photodetectors on an MAPbI3 single crystal. This sensor could quantitatively distinguish wavelengths. Further device analysis showed that both photodetectors possess entirely disparate optoelectronic properties. Consequently, the as-developed wavelength sensor could accurately distinguish incident-light wavelengths ranging from 265 to 860 nm with a resolution of less than 1.5 nm based on the relation between the photocurrent ratios of both photodetectors and the incident light wavelengths. Notably, a high resolution and wide detection range are among the optimum reported values for such sensors and enable full-color imaging. Furthermore, technology computer-aided design (TCAD) simulations showed that a mechanism involved in distinguishing wavelengths is attributed to the wavelength-dependent photon generation rate in MAPbI3 single crystals. The high-performance MAPbI3 wavelength sensor can potentially drive the research progress of perovskites in wavelength recognition and full-color imaging.

Developing cost-effective and highly efficient photocathodes toward polysulfide redox reduction is highly desirable for advanced quantum dot (QD) photovoltaics. Herein, we demonstrate nitrogen doped carbon (N-C) shell-supported iron single atom catalysts (Fe-SACs) capable of catalyzing polysulfide reduction in QD photovoltaics for the first time. Specifically, Fe-SACs with FeN4 active sites feature a power conversion efficiency of 13.7% for ZnCuInSe-QD photovoltaics (AM1.5G, 100 mW/cm2), which is the highest value for ZnCuInSe QD-based photovoltaics, outperforming those of Cu-SACs and N-C catalysts. Compared with N-C, Fe-SACs exhibit suitable energy level matching with polysulfide redox couples, revealed by the Kelvin probe force microscope, which accelerates the charge transferring at the interfaces of catalyst/polysulfide redox couple. Density functional theory calculations demonstrate that the outstanding catalytic activity of Fe-SACs originates from the preferable adsorption of S42- on the FeN4 active sites and the high activation degree of the S-S bonds in S42- initiated by the FeN4 active sites.

We have developed a multi-spectral SWIR lidar system capable of measuring simultaneous spatial-spectral information for imaging and spectral discrimination through partial obscurations. We employ objects in the presence and absence of a series of obscurants to evaluate the capability of the system in classifying the objects of interest based on spectral and range information. We employ a principal component analysis-based algorithm in classifying the objects and quantifying the accuracy of detection under various obscured scenarios. The merits of multi-spectral lidar over hyperspectral imaging are highlighted for target identification in the presence of obscurants.

Optical multicasting, which involves delivering an input signal to multiple different channels simultaneously, is a key function to improve network performance. By exploiting individual spatial modes as independent channels, mode-division-multiplexing (MDM) can solve the capacity crunch of traditional standard single-mode fiber (SSMF) transmission system. In order to realize mode multicasting with high flexibility in future hybrid wavelength-division-multiplexing (WDM) and MDM networks, we propose a mode multicasting scheme without parasitic wavelength conversion, based on the inter-modal four-wave mixing (FWM) arising in the few-mode fiber (FMF). The operation mechanism including nonlinear phase shift for efficient mode multicasting is analytically identified. Then, based on the derived operation condition, we numerically investigate the impact of the dual-pump power and the FMF length on the performance of mode multicasting. By properly setting the pump wavelength and the dual-pump power, mode multicasting performance, in terms of mode multicasting efficiency, 3-dB bandwidth, and destination wavelength, can be tuned according to various application scenarios. After the performance optimization, mode multicasting of 25-Gbaud and 100-Gbaud 16-quadratic-amplitude modulation (16-QAM) signals is numerically demonstrated. The proposed reconfigurable mode multicasting is promising for future WDM-MDM networks.

In this article, it is shown experimentally that a planar laser-generated ultrasound source with a hard reflective backing will generate higher acoustic pressures than a comparable source with an acoustically matched backing when the stress confinement condition is not met. Furthermore, while the source with an acoustically matched backing will have a broader bandwidth when the laser pulse is short enough to ensure stress confinement, the bandwidths of both source types will converge as the laser pulse duration increases beyond stress confinement. The explanation of the results is supported by numerical simulations.

The COVID-19 pandemic created unprecedented challenges for people's health. Studies have demonstrated the positive impact of urban green spaces, particularly urban parks, on physical and mental health. However, few studies have evaluated social health, which is a component of human health, and more understanding of the relationship between urban parks and human health during the COVID-19 pandemic is required. This study examined the effects of urban parks on people's health using a canonical correlation model. Physical, mental, and social health were the dependent health variables, and five factors related to urban parks were the independent variables. This study investigated 22 urban parks inside the Forth Ring Road in Beijing, China using a questionnaire survey. The results demonstrated a positive association between urban parks and human health during the pandemic. Distance to the parks, park area, and park size were positively correlated with physical, mental, and social health. Furthermore, frequency and duration of visits to urban parks were positively associated with mental health and contact with neighbors. The health effects of urban park use varied with park types and locations' urbanization background. These findings can provide insights for health-oriented urban park planning and construction.

BACKGROUND: Although hormesis induced by heavy metals is a well-known phenomenon, the involved biological mechanisms are not fully understood. Cadmium (Cd) is a prevalent heavy metal in the environment. Exposure of Cd, via intake or consumption of Cd-contaminated air or food, poses a huge threat to human health. Chinese cabbage pakchoi (Brassica chinensis L.) is widely planted and consumed as a popular vegetable in China. Therefore, studying the response of Chinese cabbage pakchoi to Cd- stressed conditions is critical to assess whether cabbage can accumulate Cd and serve as an important Cd exposure pathway to human beings. In this study, we investigated the influence of Cd stress on growth, photosynthetic physiology, antioxidant enzyme activities, nutritional quality, anatomical structure, and canopy temperature in Chinese cabbage pakchoi. A partial least squares (PLS) model was used to quantify the relationship between physical and chemical indicators with Cd accumulation in cabbage, and identify the main controlling factors. RESULTS: Results showed that Cd stress significantly inhibited cabbage's growth and development. When Cd stress was increased, the phenotypic indicators were significantly reduced. Meanwhile, Cd stress significantly enhanced the oxidative stress response of cabbage, such as the activities of catalase (CAT), superoxide dismutase (SOD), peroxidase (POD), ascorbate peroxidase (APX), and the content of malondialdehyde (MDA) in leaves. Such a change tended to increase fenestrated tissues' thickness but decrease the thickness of leaf and spongy tissues. Moreover, Cd stress significantly increased soluble sugar, protein, and vitamin C contents in leaves as well as the temperature in the plant canopy. The PLS model analysis showed that the studied phenotypic and physicochemical indicators had good relationships with Cd accumulation in roots, shoots, and the whole plant of cabbage, with high coefficient of determination (R2) values of 0.891, 0.811, and 0.845, and low relative percent deviation (RPD) values of 3.052, 2.317, and 2.557, respectively. Furthermore, through analyzing each parameter's variable importance for projection (VIP) value, the SOD activity was identified as a key factor for indicating Cd accumulation in cabbage. Meanwhile, the effects of CAT on Cd accumulation in cabbage and the canopy mean temperature were also high. CONCLUSION: Cd stress has significant inhibitory effects and can cause damage cabbage's growth and development, and the SOD activity may serve as a key factor to indicate Cd uptake and accumulation in cabbage.

OBJECTIVE: To investigate the utility of 18F-fluorodeoxyglucose positron emission tomography/computed tomography (18F-FDG-PET/CT) in assessing disease activity in Takayasu arteritis (TA). METHODS: Ninety-one patients with TA were recruited from a Chinese cohort. Clinical data, acute-phase reactants and 18F-FDG-PET/CT findings were simultaneously recorded. The value of using 18F-FDG-PET/CT to identify active disease was evaluated, using ESR as a reference. Disease activity assessment models were constructed and concordance index (C-index), net reclassification index (NRI), and integrated discrimination index (IDI) were evaluated to compare the benefits of the new modes with ESR and the Kerr score. RESULTS: In total, 64 (70.3%) cases showed active disease. Higher levels of ESR and CRP, and lower IL-2 receptor (IL-2R) levels were observed in active cases. 18F-FDG-PET/CT parameters measured by determining the standard uptake value (SUV), including SUVmean, SUVratio1, SUVratio2, sum of SUVmean and sum of SUVmax, were significantly higher in active disease groups. The C-index threshold of ESR to indicate active disease was 0.78 (95% CI: 0.69, 0.88). The new activity assessment model combining ESR, sum of SUVmean and IL-2R showed significant improvement in C-index over the ESR method (0.96 vs 0.78, P < 0.01; NRI 1.63, P < 0.01; and IDI 0.48, P < 0.01). The new model also demonstrated modest superiority to the Kerr score assessment (0.96 vs 0.87, P = 0.03; NRI 1.19, P < 0.01; and IDI 0.33, P < 0.01). CONCLUSIONS: A novel 18F-FDG-PET/CT-based method that involves combining the sum of SUVmean with ESR score and IL-2R levels demonstrated superiority in identifying active TA compared with conventional methods.

OBJECTIVES: To identify valuable ultrasonography findings combined with clinical markers for predicting carotid progression of Takayasu's arteritis (TAK) on imaging during a 1-year follow-up period. METHODS: From May 2016 to June 2019, 77 Chinese TAK patients with carotid artery involvement were enrolled in the present study. The patients' clinical characteristics and serological test and carotid ultrasonography results were recorded at baseline and each visit. Carotid progression was evaluated by ultrasonography every 3 months during the 1-year follow-up. Baseline clinical characteristics and ultrasonography results for predicting progression on imaging were identified. RESULTS: Sixteen (20.8%) patients presented with carotid progression on imaging during the 1-year follow-up period. The patients in the progressive group were younger (23.4±3.7 vs. 32.3±9.8 years, p<0.01) than those in the non-progressive group. At baseline, the vessel wall was thicker in the progressive group than in the non-progressive group (2.4±0.8 vs. 1.9±0.5 mm, p=0.041). Furthermore, the proportion of patients with refractory disease (87.5% vs. 16.4%, p<0.01) was higher in the progressive group than in the non-progressive group. Patients with a thickened carotid wall (≥1.9 mm), refractory disease, and younger age (≤30 years) might be at a high risk of carotid progression on imaging (75%, AUC: 0.93, sensitivity: 75%, specificity: 93.4%). CONCLUSIONS: Younger patients with early vascular structural changes at baseline as well as refractory disease seemed more likely to show carotid progression on imaging.

We present a method for recovering the perspective image of a non-line-of-sight scene based on plenoptic observations of indirect photons scattered from a flat homogeneous surface. The plenoptic information is used to ascertain locations of non-line-of-sight features in a hidden scene. This latent knowledge is then used for the partial reconstruction of the scene by extracting the imagery perceived by the non-line-of-sight features from the scattered light field measured along the surface. We demonstrate the efficacy of our technique by applying it to synthetic and measured light field data to retrieve the locations of multiple knife-edge occlusions in a test scene along with their perceived imagery.

The scattered light distribution of surfaces in the long-wave infrared (λ∼8-12µm) is measured using a small set of thermal camera images. This method can extract scatter patterns considerably faster than standard laboratory bidirectional reflectance distribution function measurements and is appropriate for passive homogeneous surfaces. Specifically, six images are used in this study, each taken with respect to a thermal light source at an angle ranging from 10° to 60° to the normal of the surface. This data is deconvolved with the shape of the light source to estimate the scattering pattern. Both highly specular (black Masonite) and diffuse (painted drywall) surfaces are tested. Errors between the estimated scattering distribution and a directly measured one using a goniometer stage and quantum-cascade laser (QCL) are less than or equal to 3% except for extremely specular surfaces where viable QCL measurements cannot be made due to the increased relative contribution of speckle noise.

RESUMEN

A custom fibre laser designed as an excitation source for biomedical photoacoustic tomography has been developed. It is based on a custom-drawn large core diameter fibre (200 µm) that enables high pulse energies (∼10 mJ) to be achieved. The system can provide variable pulse durations (10 - 500 ns) and pulse repetition frequencies (100 Hz - 1 kHz), as well as arbitrary pulse bursts according to specific user defined sequences. The system is also compact and does not require external water cooling. This, along with the flexibility in the temporal characteristics of its output that it offers, will aid the translation of photoacoustic imaging to practical application in medicine and biology.

High concentration rare earth doped, large mode area (LMA) step-index fibers, which feature a very high cladding absorption per unit length at the pump wavelength, high efficiency, and excellent beam quality, are ideal for high power pulsed fiber lasers/amplifiers where large effective mode areas and short device lengths are crucial in order to reduce detrimental nonlinear effects associated with high peak power operation. In this Letter, we realize low numerical aperture (NA) high absorption fibers, simply by employing a germanium (Ge)-doped cladding rather than a pure silica cladding to offset the high refractive index associated with using a high concentration of ytterbium (Yb) in the core. This approach allows us to separate the two inter-linked fiber design parameters of pump absorption and NA in a step-index fiber. Using a conventional modified chemical vapor deposition process combined with solution doping, a low NA (0.04), LMA (475µm2) silica fiber is fabricated with a cladding absorption value of >20dB/m, which is the highest value among LMA step-index fibers with NA<0.06 so far reported to the best of our knowledge. The fabricated Yb-doped fiber was tested in a high-power picosecond amplifier system and enabled the generation of 190 ps laser pulses with a 101 µJ pulse energy and 0.5 MW peak power at an average power of 150 W.

We present a methodology for recovering the perspective imagery of a non-line-of-sight scene based on plenoptic observations of indirect photons scattered from a homogeneous surface. Our framework segregates the visual contents observed along the scattering surface into angular and spatial components. Given the reflectance characteristics of the scatterer, we show that the former can be deduced from scattering measurements employing diversity in angle at individual surface points, whereas the latter can be deduced from captured images of the scatterer based on prior knowledge of occlusions within the scene. We then combine the visual contents from both components into a plenoptic modality capable of imaging at higher resolutions than what is allowed by the angular information content and discriminating against extraneous signals in complex scenes that spatial information struggles to discern. We demonstrate the efficacy of this approach by reconstructing the imagery of test scenes from both synthetic and measured data.

OBJECTIVES: As a rare systemic autoinflammatory disease, adult-onset Still's disease (AOSD) has heterogeneous clinical manifestations, response to treatment and outcome. This study tried to assess the clinical characteristics, laboratory tests, and treatments of Chinese AOSD patients, and make a retrospective analysis. METHODS: We collected from 7 hospitals in China a total of 517 Chinese patients with AOSD who satisfied the Yamaguchi criteria. We retrospectively evaluated their clinical features, laboratory tests, treatments and compared them with published data from different studies. All the data in this study were from medical records and further statistic analyses. RESULTS: We evaluated a total of 517 AOSD patients, 72% female, average age of onset was 37.7; spiking fever, rash and arthralgia occurred in 472 (91.3%), 413 (79.9%), 378 (73.1%) cases, respectively. There were 439/513 (85.6%) cases with leukocytosis and 456/476 (95.8%) cases with raised serum ferritin. The highest frequently used medications and regimens for remission were glucocorticoids (498/517, 96.3%), methotrexate (273/517, 52.8%) and hydroxychloroquine (174/517, 33.7%). 84.4%. 357/423 of AOSD cases were able to achieve initial remission with different regimens, mostly including glucocorticoids, methotrexate or hydroxychloroquine. 47.2% of them (244/517) received 30

Methods: A total of 144,856 subjects aged 20 to 79 years were enrolled in this cross-sectional study. Serum uric acid level, renal function, hepatic function, and lipid profile were investigated. Results: The prevalence of hyperuricemia decreased with age in men but increased in women. In men, the correlation coefficient between the serum urate level and BMI declined steadily with age. Underweight was associated with a 53 to 68% and a 66% lower prevalence of hyperuricemia in men aged 20 to 69 years and in women aged 20 to 29 years, respectively. Overweight and obesity were correlated with a higher odds ratio (OR) (95% confidence interval [CI]) for hyperuricemia in both genders. In individuals with overweight or obesity, younger subjects had a higher OR (95% CI) for hyperuricemia than older subjects. Among subjects aged 20 to 59 years, as they gained weight, the OR (95% CI) for hyperuricemia increased faster in women than in men compared with their respective normal-weight controls. Conclusion: Underweight was associated with a lower prevalence of hyperuricemia in men aged ≤69 years. In individuals with overweight or obesity, younger subjects were more likely to develop hyperuricemia than older subjects. With active weight gain, the likelihood for developing hyperuricemia increased faster in women than in men compared with their respective normal-weight controls.
