RESUMEN

An artificial metalloenzyme based on the covalent grafting of a nonheme Fe(II) polyazadentate complex into bovine ß-lactoglobulin has been prepared and characterized by using various spectroscopic techniques. Attachment of the Fe(II) catalyst to the protein scaffold is shown to occur specifically at Cys121. In addition, spectrophotometric titration with cyanide ions based on the spin-state conversion of the initial high spin (S=2) Fe(II) complex into a low spin (S=0) one allows qualitative and quantitative characterization of the metal center's first coordination sphere. This biohybrid catalyst activates hydrogen peroxide to oxidize thioanisole into phenylmethylsulfoxide as the sole product with an enantiomeric excess of up to 20 %. Investigation of the reaction between the biohybrid system and H2 O2 reveals the generation of a high spin (S=5/2) Fe(III) (η(2) -O2 ) intermediate, which is proposed to be responsible for the catalytic sulfoxidation of the substrate.

RESUMEN

Voluntary drug intoxication is mainly due to drug overdose or the interaction of several drugs. Coma and its associated complications such as hypoventilation, aspiration pneumopathy, and heart rhythm disorders are the main hallmarks of drug intoxication. Conventional detoxification treatments, including gastric lavage or vomiting, administration of ipecac or activated charcoal (CH), and the use of antidotes, have proven to be inefficient and are generally associated with severe adverse effects. To overcome these limitations, titanate nanotubes (TiNTs) are proposed as an efficient emerging detoxifying agent because of their tubular shape and high adsorption capacity. In the present study, the detoxifying ability of TiNTs was evaluated on paracetamol (PR)-intoxicated rats. Results indicate that the loading ability of PR into TiNTs (70%) was significantly higher than that recorded for CH (38.6%). In simulated intestinal medium, TiNTs showed a controlled drug release of less than 10% after 72 h of incubation. In PR-intoxicated rats, TiNTs treatment resulted in a 64% decrease of PR after 4 h of poisoning versus 40% for CH. Concomitantly, TiNTs efficiently reduced PR absorption by 90% after 24 h of poisoning, attenuated the elevated levels of biochemical markers (i.e., alanine aminotransferase, aspartate aminotransferase, creatinine, and TNF-α) and mitigated oxidative stress by increasing the activity of superoxide dismutase and reducing the oxidized glutathione/total glutathione ratio, suggesting a histoprotective effect of TiNTs against paracetamol-induced toxicity in rats. In addition to their safety and high stability in the entire gastro-intestinal tract, biodistribution analysis revealed that TiNTs exhibited low intestinal absorption owing to their large cluster size of compact aggregate nanomaterials across the intestinal villi hindering the absorption of paracetamol. Collectively, these data provide a new and promising solution for in vivo detoxification. TiNTs are expected to have great potential for the treatment of voluntary and accidental intoxication in emergency care.

RESUMEN

Ultrasmall sub-10 nm nanoparticles of Prussian blue analogues incorporating GdIII ions at their periphery revealed longitudinal relaxivities above 40 mM-1 s-1 per GdIII regardless of the nature of the core and the polymer coating. Large T1-weighted contrast enhancements were achieved in addition to a highly efficient photothermal effect and in vivo photoacoustic imaging in tumors.

Gas chromatography-mass spectrometry has been used to determine the nature of organic materials used in mummification balms. A comparative analysis of samples taken from Egyptian mummies is developed. The results are given in two parts. First, it is shown that the chemical composition of the balm is practically independent of the part of the mummy from which it is taken. This study was done on a Ptolemaic mummy (circa 100 BC from the Guimet Museum in Lyon). Fats, beeswax, and diterpenic resins were the main components: they were found everywhere. Castor oil was also very often detected (in half of the samples). This particular fat is present in the balm inside the thorax but not in the skull. Moreover it is shown that a vegetable tannin was employed. Components indicative of vegetable tannin input (gallic acid and inositols) were found in seven samples out of eighteen, particularly close to the body and on the canopic pack of the heart. Secondly, some conclusions from a comparative study of the composition of balms from mummies of various social levels as well as of different Egyptian periods are reported. It is shown that beeswax was used as from very early times (XVIIIth dynasty). The mixture of beeswax, fats, and diterpenoid resins would appear to be more recent. The balms of three mummies dating from more recent Egyptian periods (XIXth to XXVth dynasty) were analysed. No evidence of a resin, gum-resin, or plant gum could be found. Some mummies would appear to have been embalmed with fats or beeswax. Finally, the entrails canopic pack said to belong to Ramses II undoubtedly shows an embalming process with a triterpenic resin of the mastic type. The adopted analytical methodology enabled us to achieve simultaneous detection of four components of the balm of the Ptolemaic mummy. Analysis of the other five mummies revealed far less complex chemical compositions for the balms. This may be an indication of different embalming processes, although we should bear in mind the question of organic matter preservation through the ages.
