RESUMEN

WHAT IS KNOWN AND OBJECTIVE: Continuous infusion of dobutamine plays an important role in the management of patients with end-stage heart failure. Home infusion of dobutamine using a continuous ambulatory delivery device (CADD) facilitates the management of patients in their home, avoiding complications associated with long-term hospitalization. However, the stability of dobutamine in CADD is currently unknown. Therefore, this study investigated the physicochemical stability of dobutamine in CADDs at three different temperatures over various time points. METHODS: Six CADDs (three containing dobutamine 10 mg/mL in 0.9% sodium chloride and three containing dobutamine 10 mg/mL in 5% glucose) were prepared and stored at 4°C for 7 days, followed by 12 hours at 35°C and then for another 12 hours at 25°C. An aliquot (n = 3) was withdrawn aseptically at 0, 24, 48, 72, 96, 120, 144 and 168 hours when stored at 4°C, and at 0, 6 and 12 hours when stored at the other two temperatures. Each sample was analysed for dobutamine concentration using a stability-indicating high-performance liquid chromatography. All the samples were also evaluated for change in pH, colour and for particle content. RESULTS AND DISCUSSION: No evidence of particle formation, colour or pH change was observed throughout the study period. Dobutamine, when admixed with 0.9% sodium chloride or 5% glucose, was found to be chemically stable for at least 168 hours at 4°C and for another 12 hours at 35°C and for another 12 hours at 25°C. WHAT IS NEW AND CONCLUSIONS: Our findings will allow health professionals to provide a weekly supply of dobutamine-containing CADDs to patients for home infusions. Continuous infusion over a 24-hour period using one CADD per day will also decrease the number of exchanges required and thus reduce the risk of catheter-related bloodstream infections.

RESUMEN

Croton antisyphiliticus Mart. is a medicinal plant native to Cerrado vegetation in Brazil, and it is popularly used to treat urogenital tract infections. The objective of the present study was to assess the genetic variability of natural C. antisyphiliticus populations using AFLP molecular markers. Accessions were collected in the states of Minas Gerais, São Paulo, and Goiás. The genotyping of individuals was performed using a LI-COR® DNA Analyzer 4300. The variability within populations was found to be greater than the variability between them. The F(ST) value was 0.3830, which indicated that the populations were highly structured. A higher percentage of polymorphic loci (92.16%) and greater genetic diversity were found in the population accessions from Pratinha-MG. Gene flow was considered restricted (N(m) = 1.18), and there was no correlation between genetic and geographic distances. The populations of C. antisyphiliticus exhibited an island-model structure, which demonstrates the vulnerability of the species.

RESUMEN

OBJECTIVE: Methyl-2-(4-chloro- phenyl)-5-benzoxazoleacetate (MCBA), a synthetic benzoxazole derivative with established antipsoriatic efficacy, was investigated for potential antinociceptive effects. This study employs various nociceptive assays in mice to elucidate MCBA's antinociceptive mechanisms. MATERIALS AND METHODS: MCBA's antinociceptive potential was tested against various nociception models induced by formalin, glutamate, capsaicin, a transient receptor potential vanilloid 1 (TRPV1) receptor agonist, and phorbol 12-myristate 13-acetate, a protein kinase C (PKC) activator. It was then assessed using the hot plate test and examined within the acetic acid-induced writhing test. During the acetic acid-induced writhing test, MCBA was pre-challenged against selective receptor antagonists such as naloxone, caffeine, atropine, yohimbine, ondansetron, and haloperidol. It was also pre-challenged with ATP-sensitive potassium channel inhibitor (glibenclamide) to further elucidate its antinociceptive mechanism. RESULTS: The results showed that oral administration of MCBA led to a dose-dependent and significant inhibition (p < 0.05) of nociceptive effects across all evaluated models at doses of 60, 120, and 240 mg/kg. Moreover, the efficacy of MCBA's antinociceptive potential was significantly counteracted (p < 0.0001) by specific antagonists: (i) directed at adenosinergic, alpha-2 adrenergic, and cholinergic receptors using caffeine, yohimbine, and atropine, respectively; and (ii) targeting ATP-sensitive potassium channels, employing glibenclamide. Antagonists aimed at opioidergic and serotoninergic receptors (naloxone and ondansetron, respectively) had poor utility in inhibiting antinociceptive activity. Conversely, the dopaminergic receptor antagonist haloperidol potentiated locomotor abnormalities associated with MCBA treatment. CONCLUSIONS: MCBA-induced antinociception involves modulation of glutamatergic-, TRVP1 receptors- and PKC-signaling pathways. It impacts adenosinergic, alpha-2 adrenergic, and cholinergic receptors and opens ATP-sensitive potassium channels.

RESUMEN

Samples of olivine (Fo(0)Fa(100), Fo(60)Fa(40), Fo(80)Fa(20), and Fo(100)Fa(0)) and of spinel (Fo(50)Fa(50), Fo(2)Fa(100), where Fo is forsterite and Fa is fayalite) were subjected to pressures up to 250 kilobars in a diamond anvil press and were heated in situ up to ~ 1700 degrees C by an infrared beam from a continuous-wave YAG (yttrium-aluminum-garnet) laser. The brightness temperature was determined from the intensity of incandescence of the sample by means of an optical pyrometer. X-ray diffraction patterns of the samples, obtained after quenching and unloading, show conclusively that these compositions disproportionate to (Mg, Fe)O and SiO(s) (stishovite) under these conditions.

RESUMEN

The new diamond-like B-C phase was obtained from the graphite-like BC phase in a laser-heated diamond anvil cell at high temperature 2230+/-140 K and high pressure 45 GPa. Raman spectra of the new phase measured at ambient conditions revealed a peak at 1315 cm(-1), which was attributed to longitudinal-optical (LO) mode. The X-Y Raman mapping was used to investigate spatial distribution of the diamond-like phases and was shown to be a powerful tool in studying the sp(2)-to-sp(3) phase transformations occurring in the diamond cell under high temperature and high pressure.

RESUMEN

The objective of this study was to verify the occurrence of Dioscorea trifida in Brazil and to obtain information concerning its distribution, management and diversity. Farmers from 21 communities were interviewed in the states of São Paulo, Santa Catarina and Mato Grosso. During the visits, semi-structured interviews were conducted to collect socio-economic, management and diversity data for this crop. Fifty-one collected accessions, plus two accessions obtained at local markets of Amazonas, were characterized using 12 morphological traits. Most the interviewed farmers were men (75%) with a mean age of 59.5 years. Just a few young people and labor force were available for agricultural activities, with an average of only three individuals per farm. Most farmers (56%) grew only one variety of D. trifida, although 44% had more than one variety in their fields, which aims to provide greater assurance at harvest. Many popular names were observed for D. trifida, and cará roxo (purple yam) was the name most used by farmers (43.4%). Characters referring to the tuber, such as skin and flesh color, were most relevant for the distinction of the accessions. The results of this study may collaborate to develop strategies for conservation, both ex situ and in situ, within the view of on farm conservation.

RESUMEN

The Advanced Sleep Phase Syndrome (ASPS) is a sleep disorder characterized by an early sleep onset and early awakening without any disturbance of the sleep structure. The management of this disease requires clinical and laboratory investigations in an attempt to confirm the phase advance of body core temperature and melatonin rhythm. The use of light therapy, possibly associated with chronotherapy or melatonin intake has been proposed. The evolution is variable. Seven subjects, aged 15 to 72 were diagnosed in our sleep disorders unit by mean of sleep log, actigraphy, sleep and temperature recording. The sleep onset and sleep offset times were approximately the same according to sleep log, actigraphy and night polysomnography. The nadir of body core temperature was at 01:38 +/- 01:03. Two familial cases were identified of which 1 was investigated in constant routine condition with hourly blood sampling. An advanced phase of melatonin and cortisol was evidenced. The disease temporarily improved in 3 cases with light therapy and in one case with the association of light therapy and chronotherapy. These data show the difficulties of the management and the treatment of this rarely diagnosed disease.

RESUMEN

Widely spread in the tropics, yams were introduced into Brazil during the colonial period and are currently grown throughout the country. Despite its importance as a pharmacological and food source, there is a lack of studies describing how and where this tuber is grown in Brazil. The aim of this study was to provide an overview of the cultivation and distribution of Dioscorea alata in different Brazilian regions. A total of 63 farmers were visited in different municipalities and communities in four regions in the country: South, Southeast, Northeast and Midwest. Semi-structured interviews were conducted to collect socio-economic, use, management and diversity data for this crop. The majority of interviewers were men, married, with children, using retirement benefits and agriculture as income and family labour as the main support in the yam cultivation. A wide distribution of this species was found, with the occurrence of D. alata in the four sampled regions. A variety of vernacular names for this species was collected, differing according to the region where it is cultivated. Most farmers cultivate yams in fields, however an increased usage of home gardens for the cultivation of this tuber was found. Also, most farmers cultivate yams in association with other crops in areas of different sizes and slash and burn practices, although mostly disappearing, are still being used by many farmers. The results of this study provide more concrete data on the distribution and diversity of this important crop.

RESUMEN

ABSTRACTIn the current work we performed a review of the Araceae family species traditionally used to treat malaria and its symptoms. The aim is to reveal the large number of antimalarial Araceae species used worldwide and their great unexplored potential as sources of antimalarial natural products. The SciFinder Scholar, Scielo, PubMed, ScienceDirect and Google books search engines were consulted. Forty-three records of 36 species and 23 genera of Araceae used for malaria and symptoms treatment were found. The neotropical genera Philodendron Schott and Anthurium Schott were the best represented for the use in the treatment of malaria, fevers, liver problems and headaches. Leaves and tubers were the most used parts and decoction was the most common preparation method. The extracts of Araceae species inhibit the in vitro growth of the human malaria parasite, the Plasmodium falciparum Welch, and significant median inhibitory concentrations (IC50) for extracts of guaimbê-sulcado (Rhaphidophora decursiva (Roxb.) Schott), aninga (Montrichardia linifera (Arruda) Schott), Culcasia lancifolia N.E. Br. and forest anchomanes (Anchomanes difformis (Blume) Engl.) have been reported demonstrating the antimalarial and cytotoxicity potential of the extracts and sub-fractions. In the only report about the antimalarial components of this family, the neolignan polysyphorin and the benzoperoxide rhaphidecurperoxin presented strong in vitro inhibition of the D6 and W2 strains of Plasmodiumfalciparum (IC50 = 368-540 ng/mL). No live study about antimalarial activity in animal models has been conducted on a species of Araceae. More bioguided chemical composition studies about the in vitro and also thein vivo antimalarial activity of the Araceae are needed in order to enhance the knowledge about the antimalarial potential of this family.

Schott), aninga (Montrichardia linifera (Arruda) Schott), Culcasia lancifoliaN.E.

The aim of this study was to analyze the genetic structure of populations of Pothomorphe umbellata (L.) Miq. based on RAPD molecular polymorphisms. Analysis included four natural populations from São Paulo State (Jacareí, Jundiaí, Piquete, Ubatuba) and one population from Paraná State (Adrianópolis). Twenty-five polymorphic loci (96.15%) were identified. There were high levels of genetic diversity within populations (Hs = 0.2220). Of the total genetic variability, 65.33% is within populations and 34.67% among populations (G ST = 0.3467). Results suggest that these populations have high levels of genetic variability, which can be strongly impacted by human action.

This study aimed to verify whether the harvest time of the "Brazilian erva-cidreira" [Lippia alba (Mill.) N. E. Br.], limonene-carvone chemotype, influences leaf mass production and essential oil yield and composition. Five different harvest times were evaluated at 145 days after transplanting: 8:00, 10:00, 12:00, 14:00 and 16:00, with five replicates, distributed in randomized blocks. The experiment was carried out at Lageado Experimental Farm of the School of Agronomical Sciences FCA-UNESP/Botucatu, Horticulture Sector, Department of Plant Production. Harvest was conducted from 15 cm above the soil surface and essential oil was obtained through steam distillation in a Clevenger-type apparatus. Both the agronomical factors and the yield obtained through steam distillation were considered. The essential oil was analyzed in a gas chromatographer coupled to a mass spectrometer (GC/MS). Results were subjected to analysis of variance (F Test) and means were compared according to Tukey's test. There was no statistical difference for leaf mass production, essential oil content and essential oil yield in fresh and dry matter. However, considering the major essential oil compounds, carvone and limonene, the best yield was obtained at 10:00 for carvone in fresh (2050 L ha-1) and dry matter (2068 L ha-1), and at 16:00 for limonene in fresh (1068 L ha-1) and dry matter (1060 L ha-1).

With the release of the list of medicinal plants by Agência Nacional de Vigilância Sanitária (ANVISA), according to RDC Nº10, March, 9th , 2010, the use of these plants is replaced by the official seal from the government regulator, in consequence, have greatly increased their demand. The production of these materials then acquires a great importance, as there will be a need to produce these plants. Aiming to assess the status of agronomic research on these species, particularly the species which occur in Atlantic Forest, a survey was made of publications from the scientific names in the CAB Abstract electronic database, ranging from 1990 to 2011 . The survey showed that the number of publications varies from 2 to 1129, and the species with the largest number of articles are those already cultivated as food. Of the 66 species listed, 36 are exotic, 24 are native of the Atlantic Forest and 6 are to other biomes. Among the Atlantic Forest species, were excluded the ruderal, fruit, and tree, due to most studies in Agronomy are related to the management, control or fruit production, and not to the cultivation with the medicinal viewpoint. The only arboreus specie excluded was Maytenus ilicifolia. Thus, we selected 16 species, which had its publications divided into three areas: Agronomy; Phytochemistry and Biological Assays. This research identified that 32% of published articles are agronomic, area that has less content than the area of biological activity, which has 40% of publications, and the area of phytochemistry have 20% of publications. These results show that researchers are aware of the importance of agronomic research with medicinal plants, but it is necessary to perform work of domestication of wild species and crop science with the less-studied species, to improve the culture, the conservation of plant genetic resources and the environment.

Stoves] seguido do Panamá e Costa Rica.

Brazil is among the leading exporters of ipecac [Psychotria ipecacuanha (Brot.) Stoves], followed by Panama and Costa Rica. The roots of Brazilian ipecac have high pharmacological value due to their levels of emetine and cephalin. This study aimed to describe how families of "poaieiros" maintain the cultural memory of Psychotria ipecacuanha (Brot.) Stoves. Information was collected in the city of Cáceres, Mato Grosso State, Brazil, through structured interviews and participating observation involving 20 men and 10 women aged from 45 to 86 years. The cited forms of use were in animal nutrition, as insecticide, acaricide, emetic, against diarrhea, relief from headache, and against malaria, bronchitis and stomach ache. The root is the most used part and its preparation form is tincture or mixed with tobacco, wine or sugarcane. Few interviewees transmitted to their children the knowledge of P. ipecacuanha. The cultural memory of P. ipecacuanha is due to experience, extraction and marketing of the plant, as well as to hearing conversations between parents and friends. The loss of knowledge associated with ipecac is caused by rural exodus, destruction of its habitat due to deforestation and agricultural occupation. The extinction of this species in the region contributes to cultural erosion.

The effect of planting density and age of Phyllanthus amarus- genotype CPQBA-14, on the content and productivity of phyllanthin were evaluated. The experimental design was randomized blocks in factorial scheme 4 x 6, with five replicates, consisting of 4 treatments of planting density (400.000 pl ha-1; 200.000 pl ha-1; 100.000 pl ha-1 and 66667.68 pl ha-1) with six ages of plants (30, 45, 60, 75, 90 and 105 days after transplanting (DAT)). Data were subjected to analysis of variance and regression (P>0.005). For phyllanthin content independent and significant effects of harvest and planting density were observed. The phyllanthin contents increased with plant age, peaking at 105 DAT, with a content of 11.52 g kg-1. The estimated population density that provided the high phyllanthin contents (8.66 g kg-1) was at 299.860 pl ha-1. The productivity of phyllanthin showed significant interaction between planting density and plants ages. It was observed during the plant growth, the treatments with 200 and 400.000 pl ha -1 had the highest yield of phyllanthin (39.3 and 37.8 kg ha-1) with 95.17 and 97.12 DAT, respectively. We conclude that the proper spacing for best spatial arrangement of the cultivation is 299.860 pl ha-1 for the yield of 3974.19 kg (dry weight leaf) x 0.00866 kg (phyllanthin contents) = 34.416 kg ha-1 of phyllanthin with harvest scheduled at 97 DAT.

Pfaffia glomerata occurs in several states of Brazil and its neighboring countries in the south region at riverbanks and gallery forests. It is a hydrophyte and heliophyte species. The roots of the genus Pfaffia are used in Brazilian folk medicine especially as tonic, aphrodisiac and to control diabetes. The aim of this work was to establish an in vitro germplasm bank for Pfaffia glomerata. The experiment was carried out in completely randomized design with six treatments: 1) DM + 2 percent sucrose + 4 percent sorbitol; 2) DM/2 + 2 percent sucrose + 4 percent sorbitol; 3) DM + 2 percent sucrose + 4 percent sorbitol + 2 mg L-1 calcium pantothenate; 4) DM/2 + 2 percent sucrose + 4 percent sorbitol + 2 mg L-1 calcium pantothenate; 5) DM + 2 percent sucrose + 3 percent mannitol + 2 mg L-1 calcium pantothenate; 6) DM/2 + 2 percent sucrose + 3 percent mannitol + 2 mg L-1 calcium pantothenate. Results were subjected to analysis of variance and Scott Knott test for mean grouping. Treatments 1, 3 and 4 had a significantly larger number of nodal segments per stem, compared to Treatments 2, 5 and 6. Treatment 2 was the most appropriate for the in vitro conservation of this species since it led to the lowest growth (3.1±1.9 cm height), high survival rate, 100 percent explants with sprouting, and the largest number of sprouts per explant after six months of culture. All seedlings produced root and showed no formation of calluses or hyperhydricidity under the evaluated treatments. Acclimatized plants showed 100 percent survival in the ex vitro environment. Maintaining P. glomerata accessions in an in vitro germplasm bank is viable both economically and for conservation.
