Although infections resulting from cosmetic surgery performed outside the United States have been regularly reported, deaths have rarely been identified. During 2009-2022, 93 U.S. citizens died after receiving cosmetic surgery in the Dominican Republic. The number of deaths increased from a mean of 4.1 per year during 2009-2018 to a mean of 13.0 during 2019-2022 with a peak in of 17 in 2020. A subset of post-cosmetic surgery deaths occurring during peak years was investigated, and most deaths were found to be the result of embolic events (fat emboli or venous thromboembolism) for which a high proportion of the patients who died had risk factors, including obesity and having multiple procedures performed during the same operation. These risk factors might have been mitigated or prevented with improved surgical protocols and postoperative medical care, including prophylactic measures against venous thromboembolism. U.S. citizens interested in receiving elective cosmetic surgery outside the United States should consult with their health care professionals regarding their risk for adverse outcomes. Public health authorities can support provider education on the importance of preoperative patient evaluation and the potential danger of performing multiple cosmetic procedures in one operation.

The metathesis reaction has been an important tool in both organic and inorganic synthetic chemistry. More specifically in polymer chemistry, ring opening metathesis polymerization (ROMP), via the formation of an active metal-carbene species (MîCHR), has been widely used. The elucidation of the mechanism for ROMP opened the way for the development of well-defined catalysts, suited to local conditions. In the present study, we employed density functional theory (DFT) to investigate three reaction pathways for the formation of a species capable of activating ROMP. The active species is formed from the [RuCl2(PPh3)2(pip)] complex in the presence of norbornadiene (NBD) and the carbene source ethyl diazoacetate (EDA). Formation of a hexacoordinated intermediate [RuCl2(PPh3)2(pip)(NBD)] is favored in the first step, with NBD doubly coordinated to the [RuCl2(PPh3)2(pip)] moiety. Analysis of donation (X â Ru) and back-donation (Ru â X) processes in the [RuCl2(PPh3)2(pip)(NBD)] complex shows that piperidine behaves as a σ donor, while NBD behaves as a π donor and the PPh3 groups act as π acceptors. The intensity of the orbital component is predominant in relation to the steric component in the complex. Thus, we propose that the reaction occurs through the formation of a hexacoordinated complex, followed by the dissociation of a PPh3 group, thus forming a complex where NBD is doubly coordinated to the metal center. Coordination of EDA leads finally to the catalyst capable of forming the metallocyclobutane intermediate required for the ROMP reaction.

BACKGROUND: Trans-abdominal pre-peritoneal (TAPP) hernia repair is a complex procedure that presents several challenges. Even though, due to the high prevalence of inguinal hernia, TAPP technique is increasing in frequency and robotic Abdominal Wall Surgery (rAWS) is emerging as a valuable tool in this regard. Although inguinal TAPP procedure principles have been published and simulation is needed, the availability of validated models remains scarce. METHODS: A new low-cost model was developed to simulate inguinal rTAPP repair. For validity assessment, a new TAPP-specific fidelity questionnaire and assessment scale were developed to compare the performance of novices and experts in the simulated procedure. The models used were assessed at 60 min for execution and quality score. RESULTS: Twenty-five residents and specialists from all over the country participated in this study. Execution, quality, and global performance was higher in the seniors group compared to juniors (8.91 vs 6.36, p = 0.02; 8.09 vs 5.14, p < .001; and 17 vs. 11,5, p < .001, respectively). Overall fidelity was assessed as being very high [4.41 (3.5-5.0), α = .918] as well as face [4.31 (3.0-5.0), α = .867] and content validity [4.44 (3.2-5.0), α = .803]. Participants strongly agreed that the model is adequate to be used with the DaVinci® Robot [4.52 (3.5-5.0), α = .758]. CONCLUSION: This study shows face, content, and construct validity of the model for inguinal TAPP simulation, including for robotic surgery. Therefore, the model can be a valuable tool for learning, understanding, practicing, and mastering the TAPP technique prior to participating in the operating room.

The influence of pulp carryover on the efficiency of the xylanase (X) treatment of industrial unbleached and oxygen-delignified eucalypt kraft pulps (A1 and A2 pulps, with kappa number (KN) values of 16 and 10, respectively), collected at the same pulp mill, was studied regarding the consumption of bleaching chemicals and pulp bleachability. Another non-oxygen-delignified eucalyptus kraft pulp of KN 13 was received after the extended cooking from a different pulp mill (pulp B). The assays were performed with both lab-washed (carryover-free) and unwashed (carryover-rich) pulps. Both lab-washed and unwashed pulps with carryover were subjected to X treatment, the former being demonstrating considerably higher ClO2 savings than the pulps containing carryover. The savings of bleaching reagents were higher when the X stage was applied to the A1 pulp than to the A2 pulp. This advantage of A1 pulp, however, was not confirmed when using unwashed pulps. In contrast, the gains obtained from the X treatment of unwashed pulp A2 were practically as high as those observed for the lab-washed A2 pulp. Furthermore, a similar effect in X stage was recorded for unwashed pulps having close KN: oxygen-delignified A2 pulp and non-oxygen-delignified B pulp. The results suggest that pulp carryover and initial pH were the key factors relating to the effectiveness of X treatment. The application of X treatment to the A2 unwashed pulp (after the oxygen stage) not only saved 20% of the ClO2 and 10% of the sodium hydroxide, but also improved the brightness stability of the bleached pulp without affecting its papermaking properties. Presently, the prevailing approaches to assessing hinge joint damage predominantly rely on predefined damage indicators or updating finite element models (FEMs). However, these methods possess certain limitations. The damage indicator method requires high-quality monitoring data and demonstrates variable sensitivities of distinct indicators to damage. On the other hand, the FEM approach mandates a convoluted FEM update procedure. Hinge joint damage represents a major kind of defect in prefabricated assembled multi-girder bridges (AMGBs). Therefore, effective damage detection methods are imperative to identify the damage state of hinge joints. To this end, a stiffness-based method for the performance evaluation of hinge joints of AMGBs is proposed in this paper. The proposed method estimates hinge joint stiffness by solving the characteristic equations of the multi-beam system. In addition, this study introduces a method for determining baseline joint stiffness using design data and FEM. Subsequently, a comprehensive evaluation framework for hinge joints is formulated, coupling a finite element model with the baseline stiffness, thereby introducing a damage indicator rooted in stiffness ratios. To verify the effectiveness of the proposed method, strain and displacement correlations are analyzed using actual bridge monitoring data, and articulation joint stiffness is identified. The results underscore the capability of the proposed method to accurately pinpoint the location and extent of hinge joint damage.

PURPOSE: To evaluate, through in vitro studies, the bond strength of vitreous and hybrid ceramics with self-etching surface treatment compared to conventional treatment. METHODS: This systematic review followed the preferred reporting items for systematic reviews and meta-analyses (PRISMA) and was registered on the open science framework (OSF) platform for in vitro studies. A population, intervention, control, and outcome (PICO) question was formulated: "Does the surface conditioning of glass and hybrid ceramics with self-etching silane present a bond strength similar to that of conventional bonding?". A literature search was carried out in the PubMed, Embase, Web of Science, Cochrane Library, and ProQuest databases until September 2023. The Joanna Briggs Institute (JBI) critical appraisal guidelines for quasi-experimental studies were used for risk assessment of bias. The meta-analysis was based on the inverse variance (IV) method (p < 0.05). RESULTS: A total of 29 in vitro studies published between 2017 and 2022 were included in this systematic review, totaling 1889 ceramic samples. The meta-analysis indicated a significant decrease in the bond strength of HF 4%-5% with silane compared to self-etching (p < 0.05; MD: 0.34; 95% CI: 0.13-0.35; I2 = 3%, p = 0.42), while it indicated that there was no significant difference between self-etching compared to 9%-10% HF with silane (p = 0.92; MD: 0.02; 95% CI: -0.32 to 0.36; I2 = 14%, p = 0.32). CONCLUSION: Self-etching primer presents bond strength that is superior to or similar to conventional surface treatment on glass and hybrid ceramics.

The Nerium oleander extract PBI 05204 (PBI) and its cardiac glycoside constituent oleandrin have direct anti-viral properties. Their effect on the immune system, however, is largely unknown. We used an in vitro model of human peripheral blood mononuclear cells to document effects under three different culture conditions: normal, challenged with the viral mimetic polyinosinic:polycytidylic acid Poly I:C, and inflamed by lipopolysaccharide (LPS). Cells were evaluated for immune activation marks CD69, CD25, and CD107a, and culture supernatants were tested for cytokines. Both PBI and oleandrin directly activated Natural Killer (NK) cells and monocytes and triggered increased production of cytokines. Under viral mimetic challenge, PBI and oleandrin enhanced the Poly I:C-mediated immune activation of monocytes and NK cells and enhanced production of IFN-γ. Under inflammatory conditions, many cytokines were controlled at similar levels as in cultures treated with PBI and oleandrin without inflammation. PBI triggered higher levels of some cytokines than oleandrin. Both products increased T cell cytotoxic attack on malignant target cells, strongest by PBI. The results show that PBI and oleandrin directly activate innate immune cells, enhance anti-viral immune responses through NK cell activation and IFN-γ levels, and modulate immune responses under inflamed conditions. The potential clinical impact of these activities is discussed.

Corner-truncated cubic ß-Ag1.94Cu0.06MoO4 microcrystals were synthesized using the hydrothermal method. These were investigated by X-ray diffraction, confirming obtention of the spinel structure Fd3Ì m. Through Raman spectroscopy are confirmed all modes for the point group of Oh7. The Egap shows a decrease of the band gap from 3.20 to 3.07 eV, with reduction of the conduction band occurring from -0.20 eV (ß-Ag2MoO4) to -0.13 eV (ß-Ag1.94Cu0.06MoO4), suggesting a p-type behavior for the Cu2+ ion. The field-emission scanning electron microscopy images confirm the morphological changes for ß-Ag2MoO4, where potato-like microcrystals were found. Meanwhile, corner-truncated cubic microcrystals for ß-Ag1.94Cu0.06MoO4. The photoluminescence (PL) spectrum confirms the increase in the PL emission for ß-Ag1.94Cu0.06MoO4, with suppression of the deep defects occurring in the structure caused by oxygen and silver atoms. In contrast, the green region is intensified because of distortions of the Ag-O and Mo-O bonds. Therefore, the ß-Ag1.94Cu0.06MoO4 solid solution has PL emission with CCT (4510 K) and CIE coordinates (x = 0.372 and y = 0.433), which could be interesting properties for applications as light-emitting diodes.

Biomaterials used in tissue regeneration processes represent a promising option for the versatility of its physical and chemical characteristics, allowing for assisting or speeding up the repair process stages. This research has characterized a polyurethane produced from castor oil monoacylglyceride (Ricinus communis L) and tested its effect on reconstructing bone defects in rat calvaria, comparing it with commercial castor oil polyurethane. The characterizations of the synthesized polyurethane have been performed by spectroscopy in the infrared region with Fourier transform (FTIR); thermogravimetric analysis (TG/DTG); X-ray diffraction (XRD) and Scanning Electron Microscopy (SEM). For the in vivo test, 24 animals have been used, divided into 3 groups: untreated group (UG); control group treated with Poliquil® castor polyurethane (PCP) and another group treated with castor polyurethane from the Federal University of Piauí - UFPI (CPU). Sixteen weeks after surgery, samples of the defects were collected for histological and histomorphometric analysis. FTIR analysis has shown the formation of monoacylglyceride and polyurethane. TG and DTG have indicated thermal stability of around 125 °C. XRD has determined the semi-crystallinity of the material. The polyurethane SEM has shown a smooth morphology with areas of recesses. Histological and histomorphometric analyzes have indicated that neither CPU nor PCP induced a significant inflammatory process, and CPU has shown, statistically, better performance in bone formation. The data obtained shows that CPU can be used in the future for bone reconstruction in the medical field.

Molybdates are biocidal materials that can be useful in coating surfaces that are susceptible to contamination and the spread of microorganisms. The aim of this work was to investigate the effects of copper doping of hydrated cobalt molybdate, synthesized by the co-precipitation method, on its antibacterial activity and to elucidate the structural and morphological changes caused by the dopant in the material. The synthesized materials were characterized by PXRD, Fourier Transformed Infrared (FTIR), thermogravimetric analysis/differential scanning calorimetry (TG/DSC), and SEM-Energy Dispersive Spectroscopy (SEM-EDS). The antibacterial response of the materials was verified using the Minimum Inhibitory Concentration (MIC) employing the broth microdilution method. The size of the CoMoO4·1.03H2O microparticles gradually increased as the percentage of copper increased, decreasing the energy that is needed to promote the transition from the hydrated to the beta phase and changing the color of material. CoMoO4·1.03H2O obtained better bactericidal performance against the tested strains of Staphylococcus aureus (gram-positive) than Escherichia coli (gram-negative). However, an interesting point was that the use of copper as a doping agent for hydrated cobalt molybdate caused an increase of MIC value in the presence of E. coli and S. aureus strains. The study demonstrates the need for caution in the use of copper as a doping material in biocidal matrices, such as cobalt molybdate.

Acetylcholine (ACh) spillover from motor endplates occurs after neuronal firing bursts being potentiated by cholinesterase inhibitors (e.g., neostigmine). Nicotinic α7 receptors (α7nAChR) on perisynaptic Schwann cells (PSCs) can control ACh spillover by unknown mechanisms. We hypothesized that adenosine might be the gliotransmitter underlying PSCs-nerve terminal communication. Rat isolated hemidiaphragm preparations were used to measure (1) the outflow of [3 H]ACh, (2) real-time transmitter exocytosis by video-microscopy with the FM4-64 fluorescent dye, and (3) skeletal muscle contractions during high-frequency (50 Hz) nerve stimulation bursts in the presence of a selective α7nAChR agonist, PNU 282987, or upon inhibition of cholinesterase activity with neostigmine. To confirm our prediction that α7nAChR-mediated effects require direct activation of PSCs, we used fluorescence video-microscopy in the real-time mode to measure PNU 282987-induced [Ca2+ ]i transients from Fluo-4 NW loaded PSCs in non-stimulated preparations. The α7nAChR agonist, PNU 282987, decreased nerve-evoked diaphragm tetanic contractions. PNU 282987-induced inhibition was mimicked by neostigmine and results from the reduction of ACh exocytosis measured as decreases in [3 H]ACh release and FM4-64 fluorescent dye unloading. Methyllycaconitine blockage of α7nAChR and the fluoroacetate gliotoxin both prevented inhibition of nerve-evoked ACh release and PSCs [Ca2+ ]i transients triggered by PNU 282987 and neostigmine. Adenosine deamination, inhibition of the ENT1 nucleoside outflow, and blockage of A1 receptors prevented PNU 282987-induced inhibition of transmitter release. Data suggest that α7nAChR controls tetanic-induced ACh spillover from the neuromuscular synapse by promoting adenosine outflow from PSCs via ENT1 transporters and retrograde activation of presynaptic A1 inhibitory receptors.

In this work, we report the room-temperature synthesis of pure calcium tungstate (CaWO4) and copper-doped calcium tungstate solid solution (Ca0.99Cu0.01WO4) by using a sonochemistry method. These materials were structurally characterized by X-ray diffraction (XRD) and Raman spectroscopy. The obtained XRD patterns were submitted to a Rietveld refinement showing, in both materials, a tetragonal phase with space group and point group of I41/a and C4h6, respectively. Microscopy images of both materials, obtained by field emission scanning electron microscopy, showed spherical agglomerated structures composed by spherical nanoparticles, while calcium and tungstate elements were identified by energy-dispersive X-ray spectroscopy for pure calcium tungstate and copper, calcium, and tungstate for Ca0.99Cu0.01WO4 solid solution. The decrease of optical band gap (Egap) from 4.0 eV (CaWO4) to 3.45 eV (Ca0.99Cu0.01WO4) confirmed the substitution of calcium atoms for copper atoms in the clusters [CaO8]. Maximum photoluminescence (PL) emission was shifted from 522 nm in the pure CaWO4 to 475 nm in the Ca0.99Cu0.01WO4 solid solution. Consequently, there was an increase of PL emissions intensity in the blue and green regions of the visible spectrum, due to electronic transitions between the orbitals O 2p, Cu 3d, and W 5d.

This study was carried out to evaluate the effect of natural ventilation and intermittent pumping events in hydrogen sulfide and methane dynamics, in terms of system operation and risk of gas exposure. Work was conducted in a full scale gravity sewer downstream of pumping stations, in Portugal. Different ventilation rates and locations were assessed, as well as H2S removal rates and potential exposure risk, through the opening of distinct manhole covers. Increased ventilation, resulting from opening of one manhole cover, saw a 38% increase in average pipe air velocity peaks, doubling the estimated rate of air turnovers per day, accompanied by an increase of nearly 20% in H2S average removal rate. Simultaneous opening of two manhole covers induced similar airflow rates through the vent stack, but different rates throughout the pipe. H2S removal rates were also found to differ, according to location of open manholes, but also initial H2S headspace concentration. Under more unfavourable conditions, natural ventilation did not suffice in attaining recommended safety concentrations, regardless of number and location of open manhole covers. H2S concentrations above defined thresholds were verified for all studied setups. Headspace oxygen concentrations below an 18.5% asphyxiation threshold also occasionally occurred, even at manholes immediately downstream of ventilation point.

BACKGROUND/AIMS: In this study, we evaluated the functional impact of facilitatory presynaptic adenosine A2A and muscarinic M1 receptors in the recovery of neuromuscular tetanic depression caused by the blockage of high-affinity choline transporter (HChT) by hemicholinium-3 (HC-3), a condition that mimics a myasthenia-like condition. METHODS: Rat diaphragm preparations were indirectly stimulated via the phrenic nerve trunk with 50-Hz frequency trains, each consisting of 500-750 supramaximal intensity pulses. The tension at the beginning (A) and at the end (B) of the tetanus was recorded and the ratio (R) B/A calculated. RESULTS: Activation of A2A and M1 receptors with CGS21680 (CGS; 2 nmol/L) and McN-A-343c (McN; 3 µmol/L) increased R values. Similar facilitatory effects were obtained with forskolin (FSK; 3 µmol/L) and phorbol 12-myristate 13-acetate (PMA; 10 µmol/L), which activate adenylate cyclase and protein kinase C respectively. HC-3 (4 µmol/L) decreased transmitter exocytosis measured by real-time videomicroscopy with the FM4-64 fluorescent dye and prevented the facilitation of neuromuscular transmission caused by CGS, McN, and FSK, with a minor effect on PMA. The acetylcholinesterase inhibitor, neostigmine (NEO; 0.5 µmol/L), also decreased transmitter exocytosis. The paradoxical neuromuscular tetanic fade caused by NEO (0.5 µmol/L) was also prevented by HC-3 (4 µmol/L) and might result from the rundown of the positive feedback mechanism operated by neuronal nicotinic receptors (blocked by hexamethonium, 120 µmol/L). CONCLUSION: Data suggest that the recovery of tetanic neuromuscular facilitation by adenosine A2A and M1 receptors is highly dependent on HChT activity and may be weakened in myasthenic patients when HChT is inoperative.

The insulin receptor substrate (IRS) proteins serve as essential signaling intermediates for the activation of PI3K by both the insulin-like growth factor 1 receptor (IGF-1R) and its close family member, the insulin receptor (IR). Although IRS-1 and IRS-2 share significant homology, they regulate distinct cellular responses downstream of these receptors and play divergent roles in breast cancer. To investigate the mechanism by which signaling through IRS-1 and IRS-2 results in differential outcomes, we assessed the involvement of the microtubule cytoskeleton in IRS-dependent signaling. Treatment with drugs that either stabilize or disrupt microtubules reveal that an intact microtubule cytoskeleton contributes to IRS-2- but not IRS-1-mediated activation of AKT by IGF-1. Proximal IGF-1R signaling events, including IRS tyrosine phosphorylation and recruitment of PI3K, are not inhibited by microtubule disruption, indicating that IRS-2 requires the microtubule cytoskeleton at the level of downstream effector activation. IRS-2 colocalization with tubulin is enhanced upon Taxol-mediated microtubule stabilization, which, together with the signaling data, suggests that the microtubule cytoskeleton may facilitate access of IRS-2 to downstream effectors such as AKT. Of clinical relevance is that our data reveal that expression of IRS-2 sensitizes breast carcinoma cells to apoptosis in response to treatment with microtubule-disrupting drugs, identifying IRS-2 as a potential biomarker for the response of breast cancer patients to Vinca alkaloid drug treatment.

Mass transfer of a range of volatile substances was studied under highly turbulent conditions. The applied setup mimicked drop structures, where the release of volatile organic carbons likely occurs at a high rate. The experiments covered several substances in a range of resistances from residing entirely in the liquid film to being fully in the gas film. The two-film theory yielded a good prediction of the whole measured range. This allowed the experimental validation of a method where two reference substances are applied, to determine the transfer of any other substance - independently of where its resistance to mass transfer resides. One finding was that the range of dimensionless Henry's constants, where both films contributed by more than 5%, was 0.0027 to 1.05, which is over five times higher than the accepted rule of thumb (0.0005-0.18). Another finding was that the ratio between the liquid and the gas film mass transfer coefficients of the reference substances was similar for the two drop configurations studied. If this holds true over a wider range of configurations, such a ratio constitutes a valuable shortcut to the current practice of ignoring gas film resistance in the estimation of mass transfer rates.

Production and build-up of sulfide in wastewater systems, especially downstream of rising mains, may lead to severe odour nuisance, toxic environments and high risk of corrosion. Due to increased population migration towards cities and lower area availability for treatment facilities, rising mains for the conveyance of wastewater sludge are becoming more frequent, and research on sulfide build-up in such cases is needed. In this paper the findings of the work carried out in a full scale wastewater sludge rising main, operated during different seasons and under distinct conditions are presented (comprising both the start-up and normal operation stages of the facility). Results showed a sulfide build-up rate of 3.24 g S-2 m-2 d-1 in the summer and of 2.30 g S-2 m-2 d-1 during the winter. The ratio of sulfate reduction to sulfide production (SO4-2/S-2) was of roughly 3 to 1, as expected. Furthermore, obtained results allowed adjusting a second order polynomial empirical equation for the forecasting of sulfide build-up within the sludge rising main. The obtained equation for sludge significantly differs from existing equations obtained for wastewater. Moreover, this work also allowed obtaining new insight into the positive influence of biofilm and hydraulic retention time in the biological sulfide generation, as well as into its variation along the length of the rising main.

For engineering purposes it is especially useful to be able to predict and control sewer corrosion rates and odor impacts as well as to design effective measures aiming to reduce effects related to hydrogen sulfide formation and release. Doing so, it is important to use modeling tools that are capable of assessing variations of dissolved oxygen, dissolved sulfide and hydrogen sulfide gas concentrations for a wide range of environmental scenarios. Two such models were assessed: AEROSEPT, an empirical formulation, and WATS, a conceptual and more complex approach. The models were applied to evaluate the effects of transitions between pressure mains and gravity sewers in the air-liquid mass transfer of hydrogen sulfide at the Ericeira sewer system in Portugal. This network is known to have odor and corrosion problems, especially during summer. Despite the unavoidable uncertainties due to the unsteady flow rate and the quantification of air velocity and turbulence, the simulation results obtained with both models have been shown to adequately predict the overall behavior of the system.

BACKGROUND AND PURPOSE: Crotoxin (CTX), a heterodimeric phospholipase A2 (PLA2) neurotoxin from Crotalus durissus terrificus snake venom, promotes irreversible blockade of neuromuscular transmission. Indirect electrophysiological evidence suggests that CTX exerts a primary inhibitory action on transmitter exocytosis, yet contribution of a postsynaptic action of the toxin resulting from nicotinic receptor desensitization cannot be excluded. Here, we examined the blocking effect of CTX on nerve-evoked transmitter release measured directly using radioisotope neurochemistry and video microscopy with the FM4-64 fluorescent dye. EXPERIMENTAL APPROACH: Experiments were conducted using mice phrenic-diaphragm preparations. Real-time fluorescence video microscopy and liquid scintillation spectrometry techniques were used to detect transmitter exocytosis and nerve-evoked [3H]-acetylcholine ([3H]ACh) release, respectively. Nerve-evoked myographic recordings were also carried out for comparison purposes. KEY RESULTS: Both CTX (5µg/mL) and its basic PLA2 subunit (CB, 20µg/mL) had biphasic effects on nerve-evoked transmitter exocytosis characterized by a transient initial facilitation followed by a sustained decay. CTX and CB reduced nerve-evoked [3H]ACh release by 60% and 69%, respectively, but only the heterodimer, CTX, decreased the amplitude of nerve-evoked muscle twitches. CONCLUSION AND IMPLICATIONS: Data show that CTX exerts a presynaptic inhibitory action on ACh release that is highly dependent on its intrinsic PLA2 activity. Given the high safety margin of the neuromuscular transmission, one may argue that the presynaptic block caused by the toxin is not enough to produce muscle paralysis unless a concurrent postsynaptic inhibitory action is also exerted by the CTX heterodimer.

BACKGROUND: Leptospirosis is a re-emerging bacterial zoonosis caused by spirochetes of the genus Leptospira. Severe disease has been reported in dogs in Europe despite vaccination with bivalent Leptospira vaccines. Recently, a tetravalent canine Leptospira vaccine (Nobivac® L4) was licenced in Europe. The goal of this study was to investigate clinical signs, microscopic agglutination test (MAT) titres, haematology, blood biochemistry, cardiac (c) Troponin I levels and echocardiography before and after vaccination with this tetravalent vaccine. Forty-eight healthy dogs were prospectively enrolled and vaccinated twice, 3-4 weeks apart (T0 and T1). Before vaccination (T0) and 16-31 days after the second vaccination (T2), MAT (n = 48), haematology (n = 48), blood biochemistry (n = 36) and cTroponin I measurements (n = 29) were performed, and MAT was repeated 347-413 days after the second vaccination (T3, n = 44). Echocardiography was performed before the first and second vaccination (T0 and T1, n = 24). RESULTS: Mild and transient clinical signs within 5 days following the first and second vaccination occurred in 23% and 10% of the dogs, respectively. Before the first vaccination (T0), all dogs showed negative MAT titres for the tested serovars except for Canicola (50% with titres 100-400). At T2, positive MAT titres to the serovars Canicola (100%), Australis (89%), Grippotyphosa (86%), Bratislava (60%), Autumnalis (58%), Copenhageni (42%), Pomona (12%), Pyrogenes (8%) and Icterohaemorrhagiae (2%) were found. Median to high titres (≥ 400) were most common to the serovar Canicola (92%) and less common to the serovars Australis (41%), Grippotyphosa (21%), Bratislava (12%), Autumnalis (4%), Pyrogenes (4%) and Pomona (2%). At T3, positive MAT titres (titre range: 100-400) were found in 2-18% of the dogs to serovars of the vaccine serogroups and in 2-18% of the dogs to the non-vaccine serovars Pomona, Autumnalis, Pyrogenes and Ballum. Haematology, blood biochemistry, cTroponin I levels and echocardiography results did not change significantly following vaccination. CONCLUSIONS: Clinical signs following vaccination with Nobivac® L4 were transient and mild in all cases. Seroconversion differed considerably among individual dogs and among the vaccine serogroups.
