RESUMEN

The number of local and traditional fruit cultivars in Andalusia (Southern Spain) has decreased dramatically since the 1970s when new commercial cultivars from breeding programs were introduced, replacing old varieties, and thus decreasing genetic diversity. The present study was included in a genetic resources project with the objective of identifying and preserving traditional fruit tree cultivars in Southern Spain. The goal of this study was to begin the characterization of 29 apple accessions (Malus x domestica Borkh) belonging to 13 traditional cultivar denominations. For molecular characterization studies, 12 simple sequence repeat markers previously developed for apple species were used. Morphological characterization was performed using 33 fruit traits. A total of 115 alleles were amplified for the 12 loci, ranging from 7 (CH01h01, CH01h10, and GD 12) to 13 alleles per locus (CH02c11). Forty-one alleles were unique to specific genotypes. The locus with the highest number of detected unique alleles was CH01f03b with 6 alleles. Expected heterozygosity ranged from 0.74 for CH01h10 to 0.88 for CH02c11, with an average of 0.82. Observed heterozygosity varied from 0.45 for CH01h01 to 1.0 for CH02d08, with an average of 0.86. Three homonyms were found for accessions belonging to varieties 'Maguillo', 'Pero Minguela', and 'Castellana'. The most discriminant morphological characters studied revealed no homonyms or synonyms among cultivar denominations, although they are useful for describing varietal characteristics that have not been previously defined.

The objective of the current study was to evaluate the effect of a subtilisin protease, without or with inclusion of carbohydrases, on digestibility and retention of energy and protein, as well as the solubilization and disappearance of non-starch polysaccharides (NSP) from corn-soybean meal based diets fed to broiler chickens. Two hundred eighty-eight Ross 308 male broiler chickens were used for the experiment. On d 14, the birds were weighed and allocated to 6 treatments and 8 replicates per treatment with 6 birds per replicate. Treatments were: 1) corn-soybean meal based control diet; 2) control diet plus supplemental protease at 5,000 (P5000) protease units (PU)/kg); 3) control plus 10,000 PU/kg protease (P10000); or control plus an enzyme combination containing xylanase, amylase, and protease (XAP) added to achieve protease activity of: 4) 2,500 PU/kg (XAP2500); 5) 5,000 PU/kg (XAP5000); or 6) 10,000 PU/kg (XAP10000). The enzymes in XAP were combined at fixed ratios of 10:1:25 of xylanase:amylase:protease. Data were analyzed by ANOVA and specific orthogonal contrasts between treatments were performed. Addition of xylanase and amylase increased (P < 0.05) the ileal digestibility of protein by 4.2% and 2.1% at XAP5000 and XAP10000, respectively (relative to P5000 and P10000, respectively), exhibiting a plateau after the XAP5000 dose. Increment (P < 0.05) in AME due to protease was evident, particularly in P10000. At the ileal level, XAP reduced (P < 0.05) the flow of insoluble xylose and arabinose, which indicates an increase in the solubilization of arabinoxylan polymers in the small intestine. Protease on its own reduced (P < 0.05) the flow of insoluble arabinose but did not affect the flow of insoluble xylose. XAP reduced (P < 0.05) the pre-cecal flow of insoluble and total glucose and galactose. It was concluded that whereas protease by itself improved nutrient utilization and increased solubilization of NSP components, at the lower dose, a combination of xylanase, amylase, and protease produced effects greater than those of protease alone.

The effects of high dose of microbial phytase and an emulsifier on the performance, apparent metabolisable energy (AME) and nitrogen (N) retention in broilers fed on diets containing different fat sources were examined in a 5-week trial. Two fat sources (soy oil and tallow), two inclusion levels of E. coli phytase (500 or 1000 phytase units (FTU)/kg diet) and two inclusion levels of lysolecithin emulsifier (0 or 3.5 g/kg of diet) were evaluated in a 2 × 2 × 2 factorial arrangement of treatments. Throughout the 5-week trial, soy oil supplementation improved weight gain and feed per gain compared with tallow, but had no effect on feed intake. The high dose of phytase increased the weight gain and feed intake and lowered the feed per gain during d 1-21, but had no effect on performance parameters over the whole trial period. An effect of emulsifier was observed for feed intake during d 1-21 and over the whole trial period. Addition of emulsifier increased feed intake compared with diets without emulsifier. During weeks 1, 2, 3 and 5, birds fed on soy oil-based diets had higher nitrogen-corrected AME (AMEN) compared with those fed on tallow-based diets. During weeks 2, 3 and 5, the effect of phytase was significant for AMEN, with the high dose increasing the AMEN. During week 2, AMEN was increased with emulsifier addition. During weeks 1, 2, 3 and 5, birds fed on soy oil-based diets had higher fat retention compared with those fed on tallow-based diets. The high dose of phytase improved the retention of fat during week 5 and the addition of emulsifier resulted in higher fat retention during week 1. During weeks 2, 3 and 5, an interaction between fat source × phytase × emulsifier was observed for N retention. In soy oil-based diets, emulsifier plus 1000 FTU/kg phytase increased N retention compared with other groups, while in tallow-based diets, emulsifier addition increased N retention in diets with 500 FTU/kg, but not in 1000 FTU/kg diet. Overall, the present data suggest that the dietary fat source influenced performance, AMEN and fat retention in broiler chickens. There is opportunity to improve bird performance during d 1-21, AMEN and fat retention with higher doses of microbial phytase. Addition of the emulsifier increased the AMEN during week 2 and tract retention of fat during week 1, but this effect was not translated into improvements in performance.

Efficacy of supplemental xylanase on growth performance, nutrient utilization, and digesta characteristics in broiler chickens fed corn- or wheat-based diets was investigated. In experiment 1, 192 male broilers (8 birds/pen; n = 6) were fed 4 diets (corn or wheat without or with 1,250 xylanase units/kg) in 2 phases (starter, d 0-21 and grower, d 22-42). There was no interaction (P > 0.05) between diet and xylanase on performance (d 0-42). Wheat diets resulted (P < 0.01) in better performance than corn diets, whereas xylanase-fed birds had improved (P < 0.01) BW gain (2,457 vs. 2,275 g) and feed per gain (1.677 vs. 1.762) relative to birds not fed xylanase. In experiment 2, TiO2 (0.3%) was added in starter diets used in experiment 1, allocated to 13-d-old broiler chicks (n = 6) housed in cages (6 birds/cage) and fed up to d 21. Excreta samples were obtained from d 17 to 20 and birds were euthanized on d 21 for digesta. Corn diets had a greater concentration (10.7 vs. 9.8%) of insoluble nonstarch polysaccharides (NSP) than wheat diets, which in turn had more than twice the concentration of soluble NSP. There was an interaction (P < 0.03) between diet type and xylanase on jejunal digesta viscosity but not (P > 0.10) on apparent ileal digestibilities of nutrients, cecal volatile fatty acids, and AMEn. In this context, diet type influenced (P < 0.05) cecal volatile fatty acids and retention of nutrients and fiber but did not affect (P = 0.45) AMEn. In contrast, xylanase-fed birds showed higher (P < 0.05) ceca digesta acetic acid, apparent ileal digestibilities of nutrients, and retention of components. As a result, birds fed xylanase had higher AMEn (3,059 vs. 2,995 kcal/kg; P < 0.01) compared with birds not fed xylanase. Although wheat diets had superior growth performance, the AMEn was similar in both diets. Xylanase improved growth performance and AMEn independent of diet type, suggesting hydrolysis of both soluble and insoluble NSP.

The ileal energy contribution of protein, starch, and fat in response to 2 exogenous enzyme combinations was studied in 2 digestibility assays with 21- (experiment 1; 432 birds) and 42-d-old (experiment 2; 288 birds) Ross 308 broiler chickens. A 2 × 2 × 3 factorial arrangement of treatments with 2 base grains (corn or wheat), without or with high fiber ingredients (corn distillers dried grains with solubles and canola meal), and 3 enzyme treatments was implemented. Enzyme treatments, fed from 12 to 21 d or 32 to 42 d, were 1) without enzymes, 2) with xylanase from Trichoderma ressei (2,000 U/kg) and amylase from Bacillus licheniformis (200 U/kg; XA), or 3) with XA plus protease from Bacillus subtilis (4,000 U/kg; XAP). All diets contained Escherichia coli phytase (500 FTU/kg). Apparent ileal digestibility (AID) of protein, starch, and fat, as well as the apparent ileal digestible energy, were determined using titanium dioxide as inert marker. A generalized mixed model was used to test main effects and 2-way interactions at P < 0.05. An enzyme × grain interaction was detected for AID of starch at 21 and 42 d, and AID of fat at 21 d, with greater effects of enzymes in wheat-based compared with corn-based diets, but significant increments due to enzymes compared with controls in both diet types. Apparent ileal digestibility of fat at 42 d increased with enzyme supplementation compared with the control treatments. The XA and XAP treatments gradually (P < 0.05) increased AID of protein at 21 d, but only XAP increased AID of protein compared with the control at 42 d. Compared with the controls, XA increased AID energy by 52 or 87 kcal, and XAP by 104 or 152 kcal/kg of DM at 21 or 42 d, respectively. The caloric contribution of starch, fat, and protein were affected differentially by base grain and the presence of fibrous ingredients at 21 and 42 d of age.

Methionine (Met) supplementation is common practice in broilers to support nutrition, yet there are gaps in the understanding of its role in systemic physiology. Furthermore, several different Met sources are available that may have different physiological effects. This study evaluated the mode of action of Met deficiency (no Met-supplementation) and supplementation (0.25% DL- or L-Met, 0.41% liquid methionine hydroxy analog-free acid (MHA-FA)), and of Met source (DL-, L- or MHA-FA) in broiler chickens, via host transcriptomics. Biological pathway activation modeling was performed to predict the likely phenotypic effects of differentially expressed genes (DEGs) in tissue samples from the jejunum, liver and breast obtained at 10, 21 and 34/35 d of age from three experiments in a combined analysis. Animal performance data showed that Met deficiency reduced BW, daily BW gain, daily feed intake, and breast yield, and increased feed conversion ratio in all experiments (P < 0.05). Effects of Met deficiency on gene expression were least evident in the jejunum and most evident in the liver and breast, as evidenced by the number of DEG and activated pathways. Activated pathways suggested Met deficiency was associated with inhibited protein turnover, gut barrier integrity, and adaptive immunity functions in the jejunum, that predicted reduced breast yield. There was an interaction with age; in Met-deficient birds, there were 333 DEGs in the jejunum of starter vs finisher birds suggesting young birds were more sensitive to Met deficiency than older birds. In the liver, Met deficiency activated pathways associated with lipid turnover, amino acid metabolism, oxidative stress, and the immune system, whereas in breast, it activated pathways involved in metabolic regulation, hemostasis, the neuronal system, and oxidative stress, again predicting a negative impact on breast yield. In the starter phase, supplementation with DL-Met compared to MHA-FA inhibited gamma-aminobutyric acid activity and oxidative stress in breast tissue. When data from all tissues were integrated, increased expression of a liver gene (ENSGALG00000042797) was found to be correlated with the expression of several genes that best explained variation due to the Met deficiency in jejunum and breast muscle. Some of these genes were involved in anti-oxidant systems. Overall, the findings indicate that impaired growth performance due to Met deficiency results from an array of tissue-specific molecular mechanisms in which oxidative stress plays a key systemic role. Young birds are more sensitive to Met-deficiency and DL-Met was a preferential source of Met than L- or MHA-FA during the starter phase.

1. The effects of microbial phytase on the performance and nutrient utilisation in broilers fed on phosphorus-adequate starter diets were examined in this study. The effect of phytase on the apparent ileal digestibility of fatty acids was of particular interest. Two grain types (maize and wheat) and two inclusion concentrations of a phytase enzyme from Escherichia coli expressed in Schizosaccaromyces pombe (0 or 500 phytase units (FTU)/kg of feed) were evaluated in a 2 × 2 factorial arrangement of treatments. 2. Supplemental phytase improved the weight gain and feed per gain, but had no effect on the feed intake of young broilers receiving phosphorus-adequate diets. 3. Phytase supplementation improved the apparent ileal digestibility of nitrogen and phosphorus in both diet types. Phytase supplementation tended to improve the apparent ileal digestible energy in wheat-based diets, but had no effect on the apparent metabolisable energy in both diet types. 4. Supplementation of phytase increased the apparent ileal digestibility of fat, with similar effects for the different fatty acids measured. Increments on ileal fat digestibility due to phytase were not dependent on the type of diet. 5. Dietary supplementation of microbial phytase enhanced not only the digestibility of phosphorus, but also that of nitrogen and fat, exhibiting increased ileal digestibility for all fatty acids in P-adequate maize- and wheat-based diets.

The presence of fruits provokes significant modifications in plant water relations and leaf gas exchange. The underlying processes driving these modifications are still uncertain and likely depend on the water deficit level. Our objective was to explain and track the modification of leaf-water relations by the presence of fruits and water deficit. With this aim, net photosynthesis rate (AN), stomatal conductance (gs), leaf osmotic potential (Ψπ), leaf soluble sugars and daily changes in a variable related to leaf turgor (leaf patch pressure) were measured in olive trees with and without fruits at the same time, under well-watered (WW) and water stress (WS) conditions. Leaf gas exchange was increased by the presence of fruits, this effect being observed mainly in WW trees, likely because under severe water stress, the dominant process is the response of the plant to the water stress and the presence of fruits has less impact on the leaf gas exchange. Ψπ was also higher for WW trees with fruits than for WW trees without fruits. Moreover, leaves from trees without fruits presented higher concentrations of soluble sugars and starch than leaves from trees with fruits for both WW and WS, these differences matching those found in Ψπ. Thus, the sugar accumulation would have had a dual effect because on one hand, it decreased Ψπ, and on the other hand, it would have downregulated AN, and finally gs in WW trees. Interestingly, the modification of Ψπ by the presence of fruits affected turgor in WW trees, the change in which can be identified with leaf turgor sensors. We conclude that plant water relationships and leaf gas exchange are modified by the presence of fruits through their effect on the export of sugars from leaves to fruits. The possibility of automatically identifying the onset of sugar demand by the fruit through the use of sensors, in addition to the water stress produced by soil water deficit and atmosphere drought, could be of great help for fruit orchard management in the future.

The effects of mild coccidial vaccine (5× dose) challenge and fiber levels on ileal endogenous amino acid (EAA) losses was determined in 26-d-old broiler chickens using the regression method. Birds were fed semipurified diets containing 3 levels of casein (40, 80, or 120 g/kg of diet) as the only source of dietary protein at 2 levels of fiber (purified cellulose at 25 or 75 g/kg of diet). One half of the experimental birds were gavaged with coccidial vaccine on d 20. The 3 levels of casein were used to determine EAA losses for each replicate cage of birds. The design of the study was a randomized complete block employing a factorial arrangement of treatments with 2 levels of coccidial challenge (control or unchallenged, and challenged) and 2 levels of fiber, with 6 replicate cages per treatment and 8 birds per cage. Birds were killed on d 26 and contents from the entire ileum were flushed with distilled water and stored at -20°C until processed. Ileal EAA losses were determined from the ordinate intercept, at zero amino acid (AA) intake, of the regression of ileal digesta AA concentration in milligrams per kilogram of DM intake against dietary AA intake in milligrams per kilogram of DM. The effect of fiber level on EAA losses, expressed in milligrams per kilogram of DM intake, were higher (P < 0.05) for most of the AA in birds fed 25 g of fiber/kg of diet. Coccidial vaccine challenge decreased (P < 0.05) EAA losses for 8 of the 18 AA. A significant interaction between fiber level and coccidial vaccine challenge was obtained for all AA except His and Lys. Coccidial vaccine challenge resulted in increased (P < 0.05) NaPi-IIb expression. The results suggest that ileal EAA loss is higher in challenged birds fed the diet containing 25 g of fiber/kg of diet. Changes in EAA losses as a result of dietary fiber level and coccidial infection are important in amino acid nutrition of the broiler chicken.

This study investigated the effect of maternal energy efficiency on broiler chicken growth and energy efficiency from 7 to 40 d of age. Residual feed intake (RFI) and residual maintenance ME requirement (RME) were used to measure energetic efficiency. Residual feed intake was defined as the difference between observed and predicted ME intake, and RME(m) as the difference between observed and predicted maintenance ME requirements. A total of 144 Ross-708 broiler breeder pullets were placed in individual laying cages at 16 wk of age. Hens with the greatest RFI (n = 32) and lowest RFI (n = 32) values from 20 to 56 wk of age were selected (maternal RFI; RFI(mat)). Selected hens were retrospectively assigned to a high- or low-RME(m) category (maternal RME(m); RME(mmat)). At 59 wk, eggs were collected for 8 d and pedigree hatched. A total of 338 broilers grouped by dam and sex were raised in 128 cages where feed intake, BW, and temperature were recorded from 7 to 40 d to calculate broiler feed conversion ratios, RFI, and RME(m). The design was a 2 × 2 × 2 factorial with 2 levels of RFI(mat), 2 levels of RME(mmat), and 2 sexes. Neither the RFI(mat) nor RME(mmat) category affected broiler offpring BW or total conversion ratio. The high-RFI(mat) × low-RME(mmat) broilers had decreased growth to 40 d. Low-RFI(mat) × low-RME(mmat) broilers had a lower RME(m) (-5.93 kcal of ME/kg(0.60) per day) and RFI (-0.86 kcal of ME/d) than high-RFI(mat) × low-RME(mmat) broilers (RME(m) = 1.70 kcal of ME/kg(0.60) per day; RFI = 0.38 kcal of ME/d). Overall, hens with low maintenance requirements (low RME(m)) produced more efficient broilers when other efficiency related traits, represented in a lower RFI, were present. Exclusion of high-RFI × low-RME(m) hens from selection programs may improve energy efficiency at the broiler level. The RME(m) methodology is a viable alternative to evaluate energy efficiency in broilers because it avoids confounding environmental effects and allows measurement standardization.

This study evaluated the effect of feed allocation and energetic efficiency on technical and economic efficiency of broiler breeder hens using the data envelope analysis methodology and quantified the effect of variables affecting technical efficiency. A total of 288 Ross 708 pullets were placed in individual cages at 16 wk of age and assigned to 1 of 4 feed allocation groups. Three of them had feed allocated on a group basis with divergent BW targets: standard, high (standard x 1.1), and low (standard x 0.9). The fourth group had feed allocated on an individual bird basis following the standard BW target. Birds were classified in 3 energetic efficiency categories: low, average, and high, based on estimated maintenance requirements. Technical efficiency considered saleable chicks as output and cumulative ME intake and time as inputs. Economic efficiency of feed allocation treatments was analyzed under different cost scenarios. Birds with low feed allocation exhibited a lower technical efficiency (69.4%) than standard (72.1%), which reflected a reduced egg production rate. Feed allocation of the high treatment could have been reduced by 10% with the same chick production as the standard treatment. The low treatment exhibited reduced economic efficiency at greater capital costs, whereas high had reduced economic efficiency at greater feed costs. The average energetic efficiency hens had a lower technical efficiency in the low compared with the standard feed allocation. A 1% increment in estimated maintenance requirement changed technical efficiency by -0.23%, whereas a 1% increment in ME intake had a -0.47% effect. The negative relationship between technical efficiency and ME intake was counterbalanced by a positive correlation of ME intake and egg production. The negative relationship of technical efficiency and maintenance requirements was synergized by a negative correlation of hen maintenance and egg production. Economic efficiency methodologies are effective tools to assess the economic effect of selection and flock management programs because biological, allocative, and economic factors can be independently analyzed.

Understanding factors affecting ME availability for productive processes is an important step in optimal feed formulation. This study compared a modelling methodology with the comparative slaughter technique (CST) to estimate energy partitioning to heat production and energy retention (RE) and to investigate differences in heat dissipation. At hatch, 50 broilers were randomly allocated in one of 4 pens equipped with a precision feeding station. From day 14 to day 45, they were either fed with a low-ME (3,111 kcal/kg ME) or a high-ME (3,383 kcal/kg ME) diet. At day 19, birds were assigned to pair-feeding in groups of 6 with lead birds eating ad libitum (100%) and follow birds eating at either 50, 60, 70, 80, or 90% of the paired lead's cumulative feed intake. Heat production and RE were estimated by CST and with a nonlinear mixed model explaining daily ME intake (MEI) as a function of metabolic BW and average daily gain (ADG). The energy partitioning model predicted MEI = (145.10 + u) BW0.83 + 1.09 × BW-0.18 × ADG1.19 + Îµ. The model underestimated heat production by 13.4% and overestimated RE by 22.8% compared with the CST. The model was not able to distinguish between net energy for gain values of the diets (1,448 ± 18.5 kcal/kg vs. 1,493 ± 18.0 kcal/kg for the low-ME and high-ME diet, respectively), whereas the CST found a 148 kcal/kg difference between the low-ME and high-ME diets (1,101 ± 22.5 kcal/kg vs. 1,249 ± 22.0 kcal/kg, respectively). The estimates of the net energy for gain values of the 2 diets decreased with increasing feed restriction. The heat increment of feeding did not differ between birds fed with the low- or high-ME diet (26% of MEI). Additional measurements on heat dissipation, physical activity, and immune status indicated that the energetic content of the diet and feed restriction affect some parameters (shank temperature, feeding station visits) but not others (leukocyte counts, heterophil to lymphocyte ratio, and immune cell function).

This trial characterized residual feed intake (RFI) and residual maintenance requirement (RME(m)) as measures of energetic efficiency in broiler breeder hens. The RFI was defined as the difference between observed and expected ME intake and RME(m) as the difference between observed and expected maintenance requirements. A total of 600 Ross 708 1-d-old pullets were placed in floor pens. At 16 wk, 144 hens were caged and randomly assigned to 1 of 2 feed allocation treatments (72 birds each). The control treatment had feed allocated on a group basis (GRP) following the standard BW target. A second treatment had feed allocated on an individual-bird basis (IND) and followed the same BW target as GRP. Sexual maturity age, egg and chick production, and several feed conversion ratios were correlated to standardized efficiency indices of RFI (SRFI) and RME(m) (SRME(m)) in each treatment. Greater SRFI and SRME(m) values described a greater energetic efficiency. Residual feed intake was more variable in IND than GRP hens (P < 0.001). The variability of RME(m) did not differ between treatments (P = 0.14). The SRFI was positively correlated to egg production in the GRP hens (r = 0.31), but negatively correlated in IND hens (r = -0.40) and was correlated to feed conversion per chick only in the GRP-based feed allocation (r = -0.44). The SRME(m) correlated strongly to egg production (r = 0.64), chick production (r = 0.64), and feed conversion per chick (r = -0.59) in both feed allocation treatments. Feed intake confounded the RFI calculation, which limits the value of RFI as a selection criterion in meat-producing animals. The independence of RME(m) from feed intake is desirable for energetic efficiency assessment in selection programs because consistent values can be obtained across different management schemes. Hens with lower maintenance requirements (greater RME(m) efficiency) partitioned more energy toward reproduction than did high-maintenance hens. The RME(m) methodology provided an unbiased estimate of energetic efficiency by adjusting the maintenance requirement for the effect of dietary thermogenesis.

This study assessed egg traits, chick traits, growth, yield, and meat quality characteristics of the offspring from broiler breeders classified by 2 measurements of energetic efficiency: residual feed intake (RFI), defined as the difference between observed and expected ME intake, and residual maintenance requirement (RME(m)), defined as the residual of the relationship between hen maintenance requirement and feed intake. A group of 72 pullets were placed in laying cages from 16 to 60 wk of age. Individual hen-based feed allocation was provided following a standard BW target. At 41wk, eggs from 8 d of production were collected and pedigree hatched. Chicks were assigned to 1 of 3 maternal RFI (RFI(mat)) categories: low, average, and high. A total of 366 chicks were placed in 36 floor pens, 6 per sex x RFI(mat) interaction, and raised to 38 d. At the end of the breeder experiment (60 wk), broilers were retrospectively assigned to a low or high maternal RME(m) (RME(mmat)) category. Low RFI(mat) broilers had greater 38-d BW than average and high RFI(mat) broilers. That was achieved through a greater BW gain and feed intake of low RFI(mat) broilers from 21 to 28 d. It was found that RFI(mat) had no effect on feed conversion, yield, or meat quality characteristics. Low RME(m) hens produced heavier eggs (62.3 g) and chicks (42.5 g) than high RME(m) hens (60.0 g; 41.0 g), but RME(mmat) did not affect broiler 38-d BW. High RME(mmat) broilers had greater breast yield (29.5%) and lower breast shear force (4.7 kg of force/g) than low RME(mmat) broilers (28.5%; 5.6 kg of force/g). The low RFI(mat) x high RME(mmat) broilers had the greatest growth to 38 d. It was found that RFI(mat) was inversely related to broiler growth, particularly when RME(mmat) was high. Although low maintenance requirements may be desirable for egg and chick production, hens with a high maintenance requirement produced broilers with greater breast yield and tenderness. Minimizing maintenance requirements may not be compatible with maximizing broiler performance and meat yield.

A study was performed to assess the effect of reducing BW variability on sexual maturation and reproductive performance of broiler breeder hens. A total of 208 Ross 708 1-d-old pullets were randomly assigned at hatch to 1 of 2 feed allocation treatments starting at 16 wk of age when all birds were placed in individual cages. A control treatment had feed allocated on a group basis (GRP) and followed the recommended BW target. A second treatment had feed allocated on an individual bird basis (IND). By design, the BW of IND pullets converged at 20 wk. This design dictated that IND birds had a lower BW variability and a higher feed intake variability than GRP birds. Pullets were retrospectively classified into 3 initial (16 wk) BW categories: low, average, or high, using the mean+/-0.5 SD as threshold. After their first egg, 64 birds were killed and dissected for determination of fleshing, fatness, and reproductive morphology. Egg production traits were analyzed to 60 wk, when the remaining birds were killed and dissected. Reducing BW variability did not reduce variability of age, follicle numbers, and ovary and oviduct weight at sexual maturity. The IND feed allocation accelerated the onset of production of low BW birds, which increased total egg production (177 eggs) and average sequence length (3.9 d) with respect to GRP x low birds (163 eggs; 3.0 d). However, INDxlow hens produced more eggs<52 g than GRPxlow hens (22 versus 8 eggs). The IND treatment increased variability of ovary weight, large yellow follicle number, and large yellow follicle weight at 60 wk. Reducing BW variability increased variation in ovarian morphology at the end of production, which suggests that optimal BW for reproduction varied among birds. Correcting BW from 16 wk to reach the BW target primarily affected low initial BW pullets, which entered lay sooner and produced more eggs, although 91% of the additional eggs weighed<52 g.

This study developed mathematical models to overcome limitations of linear models of energy partitioning in hens. The fit of 1 linear and 2 nonlinear models of ME intake (MEI) were compared, using empirical data of 288 caged broiler breeder hens from 20 to 60 wk of age. Pullets were individually caged at 16 wk and assigned to 1 of 4 feed allocation groups. Three groups had feed allocated on a group basis with divergent target BW: standard (STD), HIGH (STD x 1.1), and LOW (STD x 0.9). The fourth group had individual-based feed allocation (IND) and followed the STD BW target. The linear model expressed MEI as a function of BW(0.75), ADG, egg mass (EM), and temperature. Nonlinear mixed models employed a normally distributed term associated with hen metabolic BW, and exponential terms of ADG and EM, or Cobb-Douglas form interactions between terms. Fit was evaluated with the Bayesian information criterion and systematic bias was analyzed through linear regressions of observed versus expected values. The linear model estimated energy partitioned to maintenance and retention in the range of reported values in the literature. However, this model had the poorest fit (R(2) = 0.64) and exhibited a slope bias of 0.91 (i.e., MEI was overestimated at low values and underestimated at high values). The first nonlinear mixed model indicated that MEI partitioned to ADG was a factor of ADG(1.15), whereas the ME partitioned to EM was a factor of EM(1.12). This model had improved fit (R(2) = 0.71) relative to the linear model. The second nonlinear mixed model indicated that the energy requirement for ADG increased by 0.60% and the EM energy requirement decreased by 2.07% for each 1% increment in BW. This model further improved fit (R(2) = 0.75). Nonlinear mixed models reduced estimation bias by accounting for individual variation in maintenance energy expenditure. These nonlinear mixed models may be used to analyze energy partitioning in animals, to develop prediction equations of MEI, to evaluate individual efficiency for maintenance, and to assess diets regarding the slope of bias in coefficients of maintenance energy requirements.

A total of 90 male Ross 308 broiler chicks were used in a digestibility and performance bioassay to explore the effect of reduction in dietary protein and digestible amino acids and inclusion of an exogenous mono-component protease on amino acid digestibility, net energy, jejunal gene expression, and bird performance. Four dietary treatments were created by the supplementation, or not, of 2 control diets with a mono-component exogenous protease. The control diets were corn/wheat/soybean meal-based and were formulated to be either nutritionally adequate or reduced in protein and amino acids (around 3%). The 2 control diets were supplemented with xylanase and phytase (2000 FYT). Treatments were therefore arranged as a 2 × 2 factorial design. The reduction in diet nutrient density had no significant effect on various experimental outcomes (including bird performance, amino acid digestibility, and net energy [NE]) that were measured with the exception of a reduction in the expression of aminopeptidase N and glucose transporter 2. However, the addition of exogenous protease resulted in an increase in weight gain and a reduction in feed conversion ratio (around 4%; P < 0.05) and an increase in the digestibility of several amino acids (P < 0.05) and starch (P = 0.06). Protease addition also resulted in an increase in both apparent metabolizable energy (AME) (+73 kcal/kg; P < 0.05) and NE (+107 kcal/kg; P < 0.05). The addition of exogenous protease to the diet also increased the jejunal expression of genes responsible for peptide transport (PepT2; P < 0.01) and starch digestion (sucrase isomaltase; P = 0.06). These results confirm the efficacy of exogenous protease in broiler diets that contain both xylanase and phytase and suggest substantial beneficial effects that extend beyond protein and amino acid nutrition. The effect of exogenous protease on energy partitioning, starch digestibility and the efficiency of nitrogen cycling is an area for further study.

The efficacy of two exogenous enzyme combinations and a multi-strain Bacillus probiotic (DFM) on the growth performance, nutrient digestibility, disappearance of non-starch polysaccharides (NSP) and gut microbial composition was investigated in broilers. One-day old Ross 308 chicks were assigned to 36 pens with 22 birds/pen and 6 pens/treatment (Experiment 1) or 36 cages with 8 birds/cage and 6 cages/treatment (Experiment 2). Treatment additives were added to nutritionally complete corn/soy based starter (d 1 to 21) and finisher (d 22 to 42) diets. Treatments included 1) a control diet containing 500 FTU/kg phytase (CTL), 2) CTL + xylanase (2,000 U/kg) and amylase (200 U/kg; XA), 3) CTL+XA + protease (4000 U/g; XAP), 4) CTL+DFM (150,000 cfu/g of 3 strains of Bacillus spp), 5) CTL+DFM+XA, and 6) CTL+DFM+XAP. Supplementation with DFM increased BW, BWG, and FI compared with the CTL (P < 0.05); XAP, but not XA, resulted in increased final BW, BWG and FI compared to the control (P < 0.05). XA and XAP improved apparent ileal digestibility (AID) of starch and fat on d 22 to 42 with XAP improving AMEn (by ∼82 kcal) compared with CTL birds (P < 0.01). DFM+XAP improved apparent ileal digestible energy (AIDE), AID of fat and starch on d 22 to 42, and additionally had a greater than additive effect on AIDE and AMEn. Supplementation with DFM+XAP reduced the ileal and total tract flow of insoluble arabinose and additionally total tract flow of soluble and insoluble xylose and total galactose (P < 0.05); similar effects of XA+DFM were not seen or were lower in magnitude, suggesting that the protease component plays an important role in increasing the availability of NSP for hydrolysis. Supplementation with DFM alone did not affect gut bacterial populations, but XA and XAP reduced numbers of Campylobacter species (by > 2.5 log cfu/g; P < 0.001) and Bacteroides (P < 0.02) in the cecum compared with CTL birds.

Two trials (a 42-d performance and a 21-d cohort digestibility) were conducted to evaluate the performance and nutrient digestibility of broilers fed corn diets supplemented with exogenous xylanase, amylase, and protease as single or combined activities. A nutritionally adequate, positive control (PC) diet was formulated. The negative control (NC) diet was formulated to be lower in metabolizable energy (∼86 kcal/kg diet) and digestible amino acids (1 to 2%) compared to PC. The other 4 treatments were based on the NC and they were either supplemented with xylanase (X), amylase (A), protease (P), or a combination of X, A, and P (XAP; to provide 2,000 U of X, 200 U of A, and 4,000 U of P/kg diet). All diets were marginal in AvP and Ca and contained a background of phytase (1,000 FTU/kg). In each trial, male broiler (Ross 308) chicks were allocated to the 5 treatments (10 replicates of 20 birds/pen and 9 replicates of 8 birds/cage for the performance and digestibility trials, respectively). In the digestibility trial, ileal digesta was collected on d21 for the determination of nutrient utilization. Data were subjected to one-way ANOVA and means were separated by Tukey's HSD test. Only the XAP improved (P < 0.05) AMEn compared to NC. X, A or XAP improved (P < 0.05) N digestibility and apparent ileal digestible energy (AIDE). Both P and XAP improved N retention. The relative improvement in energy digestibility due to enzyme supplementation was greater at the ileal level than that measured in the excreta. The measured changes on AIDE due to supplemental enzymes were much higher than the sum of calculated contributions from starch, fat, and protein. Supplementation of all enzymes reduced (P < 0.05) ileal flow of soluble rhamnose and mannose relative to NC. In the performance trial, both X and XAP improved (P < 0.05) weight gain (WG) and only XAP improved (P < 0.05) FCR compared to NC during the starter phase (1-21d). Over the entire period (1-42d), WG and FI were not influenced (P > 0.05) by dietary treatments. Both X and XAP had lower (P < 0.05) FCR compared to NC (1.540 and 1.509 vs 1.567, respectively). However, birds fed diet supplemented with XAP had an improved (P < 0.05) FCR compared to birds fed single activities and had similar (P > 0.05) FCR compared to PC. In conclusion, these results suggest a synergistic effect between X, A and P on broiler performance and nutrient digestibility. In the current study, AIDE measurements appeared to overestimate the enzyme response. Calculation of the energy contribution by supplemental enzymes using the improvements in the digestibility of the undigested fraction of starch, fat and protein may be a more accurate measurement for the enzyme response than the absolute response in AIDE.
