BACKGROUND: Mammalian intestinal epithelium constantly undergoes rapid self-renewal and regeneration sustained by intestinal stem cells (ISCs) within crypts. Inducible nitric oxide synthase (iNOS) is an important regulator in tissue homeostasis and inflammation. However, the functions of iNOS on ISCs have not been clarified. Here, we aimed to investigate the expression pattern of inducible nitric oxide synthase (iNOS) within crypts and explore its function in the homeostatic maintenance of the ISC niche. METHODS: Expression of iNOS was determined by tissue staining and qPCR. iNOS-/- and Lgr5 transgenic mice were used to explore the influence of iNOS ablation on ISC proliferation and differentiation. Enteroids were cultured to study the effect of iNOS on ISCs in vitro. Ileum samples from wild-type and iNOS-/- mice were collected for RNA-Seq to explore the molecular mechanisms by which iNOS regulates ISCs. RESULTS: iNOS was physiologically expressed in Paneth cells. Knockout of iNOS led to apparent morphological changes in the intestine, including a decrease in the small intestine length and in the heights of both villi and crypts. Knockout of iNOS decreased the number of Ki67+ or BrdU+ proliferative cells in crypts. Loss of iNOS increased the number of Olfm4+ ISCs but inhibited the differentiation and migration of Lgr5+ ISCs in vivo. iNOS depletion also inhibited enteroid formation and the budding efficiency of crypts in vitro. Moreover, iNOS deficiency altered gluconeogenesis and the adaptive immune response in the ileum transcriptome. CONCLUSION: Paneth cell-derived iNOS is required to maintain a healthy ISC niche, and Knockout of iNOS hinders ISC function in mice. Therefore, iNOS represents a potential target for the development of new drugs and other therapeutic interventions for intestinal disorders.

Diffraction-free electromagnetic beam propagates in free space without change in its two-dimensional transverse profile. Elongating diffraction-free length can benefit the practical application of this beam. Here, we demonstrate that a THz diffraction-free beam with meter-scale length can be achieved by using only one optical element. By circumscribing the line-shape of spherical harmonic function on a traditional axicon, such optical element is designed, and then can be fabricated by 3D-printing technique. Simulated, experimental, and theoretical results all show that the diffraction-free length of generated beam is over 1000 mm. Further analysis based on Fourier optics theory indicates that the spatial frequency of this beam has a comb distribution, which plays a key role during the beam generation process. Moreover, such distribution also demonstrates the beam generated by our invented optical element is not the Bessel beam, but a new diffraction-free beam. It is believed that this meter-scale THz diffraction-free beam can be useful in a non-contact and non-destructive THz imaging system for large objects.

BACKGROUND: Rice is the staple food of many people around the world. However, most rice varieties, especially widely grown indica varieties and hybrids, are sensitive to cold stress. In order to provide a basis for the utilization of a common wild rice (CWR, Oryza rufipogon Griff.) named 'Chaling' CWR in cold-tolerant rice breeding and deepen the understanding of rice cold tolerance, the cold tolerance of ratoon 'Chaling' CWR was studied under the stress of the natural low temperature in winter in Changsha, Hunan province, China, especially under the stress of abnormal natural low temperature in Changsha in 2008, taking other ratoon CWR accessions and ratoon cultivated rice phenotypes as control. RESULTS: The results showed that ratoon 'Chaling' CWR can safely overwinter under the natural conditions in Changsha (28° 22' N), Hunan province, China, which is a further and colder northern place than its habitat, even if it suffers a long-term low temperature stress with ice and snow. In 2008, an extremely cold winter appeared in Changsha, i.e., the average daily mean temperature of 22 consecutive days from January 13 to February 3 was - 1.0 °C, and the extreme low temperature was - 4.7 °C. After subjected to this long-term cold stress, the overwinter survival rate of ratoon 'Chaling' CWR was 100%, equals to that of ratoon 'Dongxiang' CWR which is northernmost distribution in the word among wild rice populations, higher than those of ratoon 'Fusui' CWR, ratoon 'Jiangyong' CWR, and ratoon 'Liujiang' CWR (63.55-83.5%) as well as those of ratoon 'Hainan' CWR, ratoon 'Hepu' CWR, and all the ratoon cultivated rice phenotypes including 3 japonica ones, 3 javanica ones, and 5 indica ones (0.0%). CONCLUSIONS: The results indicate that ratoon 'Chaling' CWR possesses strong cold tolerance and certain freezing tolerance.

Intestinal stem cells (ISCs) are pivotal for the maintenance and regeneration of the intestinal epithelium. Berberine (BBR) exhibits diverse biological activities, but it remains unclear whether BBR can modulate ISCs' function. Therefore, we investigated the effects of BBR on ISCs in healthy and radiation-injured mice and explored the potential underlying mechanisms involved. The results showed that BBR significantly increased the length of the small intestines, the height of the villi, and the depth and density of the crypts, promoted the proliferation of cryptal epithelial cells and increased the number of OLFM4+ ISCs and goblet cells. Crypts from the BBR-treated mice were more capable of growing into enteroids than those from untreated mice. BBR alleviated WAI-induced intestinal injury. BBR suppressed the apoptosis of crypt epithelial cells, increased the quantity of goblet cells, and increased the quantity of OLFM4+ ISCs and tdTomato+ progenies of ISCs after 8 Gy WAI-induced injury. Mechanistically, BBR treatment caused a significant increase in the quantity of p-S6, p-STAT3 and p-ERK1/2 positive cryptal epithelial cells under physiological conditions and after WAI-induced injury. In conclusion, BBR is capable of enhancing the function of ISCs either physiologically or after radiation-induced injury, indicating that BBR has potential value in the treatment of radiation-induced intestinal injury.

The construction of an n-p heterojunction through the self-assembly of a dyad based on tetraphenylporphyrin (TPP) and 1,4,5,8-naphthalenedimide (NDI) (1) is described. Proton transfer from the lysine head group of 1 to the porphyrin ring occurs concomitantly with self-assembly into 1D nanorods in CHCl3. TEM and AFM studies showed that the nanorods are formed by the lateral and vertical fusion of multilameller vesicles into networks and hollow ribbons, respectively. These intermediate structures transitioned to nanorods over the course of 4-6 days. Time-resolved spectroscopy revealed that photoinduced charge separation occurs with rate constants that depend on the nature of the aggregation.

Mitochondria are critical for cellular energetic metabolism, intracellular signaling orchestration and programmed death regulation. Therefore, mitochondrial dysfunction is associated with various pathogeneses. The maintenance of mitochondrial homeostasis and functional recovery after injury are coordinated by mitochondrial biogenesis, dynamics and autophagy, which are collectively referred to as mitochondrial quality control. There is increasing evidence that mitochondria are important targets for melatonin to exert protective effects under pathological conditions. Melatonin, an evolutionarily conserved tryptophan metabolite, can be synthesized, transported and metabolized in mitochondria. In this review, we summarize the important role of melatonin in the damaged mitochondria elimination and mitochondrial energy supply recovery by regulating mitochondrial quality control, which may provide new strategies for clinical treatment of mitochondria-related diseases.

7075 Aluminum alloy (AA7075) samples undergone four aging sequences were examined using a scanning electron microscope (SEM) and a transmitted electron microscope (TEM). The measurements results validate the correlation between stress corrosion cracking (SCC) resistance and the size and inter-distance of the grain boundary precipitates (GBPs). To evaluate the size and inter-distance of GBPs, we demonstrate in this study a highly efficient SEM imaging technique that can unfold grain boundary in a two-dimensional view. Compared to TEM, imaging with backscattered electrons in SEM (SEM-BSE) is more advantageous for GBPs presentation and measurements. The major reason is that about 900 times more sampling area can be imaged with SEM from the same specimen for TEM observation, thus enabling frequent appearances of GBPs at normal top view perspective, a planar view best for GBPs quantitative analysis but not well-documented. The acceleration tension of SEM for imaging was optimized at 10 kV with an information depth of around 330 nm. RESEARCH HIGHLIGHTS: Scanning electron microscope (SEM) imaging using backscattered electrons is efficient for AA7075 grain boundary precipitate imaging. The precipitate size and inter-distance can be more accurately measured with the perspective of normal top view under SEM than transmitted electron microscope.

The self-assembly of bolaamphiphile 1 into nanotubes containing a nanostructured electron donor/acceptor heterojunction is reported. In 10% MeOH/H(2)O, the tetraphenylporphyrin (TPP) and 1,4,5,8-naphthalenetetracarboxylic acid diimide chromophores engage in strong J-type π-π interactions within monolayer rings that further stack into the nanotube assemblies. In 10% MeOH/H(2)O at pH 1 or 11 or in pure MeOH, assembly is driven exclusively by the TPP ring, leading to the formation of nonspecific, unstructured aggregates. Steady-state, time-resolved fluorescence and femtosecond transient absorption spectroscopy revealed a strong dependence of the fluorescence decay and electron-transfer/charge-recombination time constants on the nature of the assemblies. These studies highlight the importance of local nanostructure in determining the photophysical properties of optoelectronic materials.

Sol-gels are found in many different scientific fields and have very broad applications. They are often prepared by the hydrolysis and condensation of alkoxysilanes such as trimethoxysilanes, which are commonly used as precursors in the preparation of silsequioxanes via the sol-gel process. The reaction rates of such reactions are influenced by a wide range of experimental factors such as temperature, pH, catalyst, etc. In this study, we combined multiple in situ spectroscopic techniques to monitor the hydrolysis and partial condensation reactions of methyltrimethoxysilane and phenyltrimethoxysilane. A rich set of kinetics information on intermediate species of the hydrolysis reactions were obtained and used for kinetics modeling. Raman and nuclear magnetic resonance (NMR) spectroscopy provided the most information about hydrolysis and NMR provided the most information about condensation. A quantitative method based on Raman spectra to quantify the various transient intermediate hydrolysis products was developed using NMR as the primary method, which can be deployed in the field where it is impractical to carry out NMR measurements.
