Human papillomavirus (HPV) infection can lead to HPV-related cancer in men, including the anus, penile, and oropharyngeal cancers and precancerous lesions. This study retrospectively investigated HPV prevalence and genotype distribution in Liaocheng men between 2016 and 2022. The total HPV positive rate was 64.87% (2388/3681, 95% confidence interval [CI]: 63.32%-66.40%), where high risk (HR)-HPV and low risk (LR)-HPV accounted for 42.49% (1564/3681, 95% CI: 40.90%-44.09%) and 69.71% (2566/3681, 95% CI: 68.20%-71.17%), respectively. The mixed HPV infection rate of two and more genotypes was 35.72%. The infection rate of HR-HPV increased with the number of positive cases annually from 2016 (16.91%) to 2022 (46.59%). The most common HR-HPV genotypes were HPV16 (11.60%), HPV52 (6.95%), and HPV59 (6.28%), whereas the least common HR-HPV was HPV26. The most common LR-HPV genotypes were HPV6 (56.99%), HPV11 (23.79%), and HPV43 (6.37%). The 9 v HPV vaccine preventable for LR-HPV and HR-HPV accounted for 80.78% and 30.40%, respectively, in this study. Most HPV-positive patients aged 1-86 were in the 30-39 age group. This study confirmed that HPV prevalence in Liaocheng men was common and diverse. HPV16, HPV52, and HPV59 are widely distributed in Liaocheng men, and the male HR-HPV infection rate remained high in this region. Regarding public health and cancer prevention, it is recommended and effective to include the HPV vaccination in the national vaccination program for men.

PURPOSE: Hypertensive disorder complicating pregnancy (HDCP) is a serious clinical disorder syndrome during pregnancy. This study aims at finding novel targets for HDCP therapy. METHODS: HDCP-related mRNAs were firstly screened out and subjected to gene enrichment analysis. We chose protein kinase AMP-activated catalytic subunit alpha 2 (PRKAA2) as the research object. Thirty-nine HDCP patients at 32 to 40 weeks of gestation were selected as the HDCP group, and 39 normal controls who received cesarean section delivery at 37-42 weeks of pregnancy were enrolled in this study. Chorionic villi samples were collected within 30 min of delivery. The apoptosis of isolated placental trophoblasts was monitored to investigate the regulatory role of PRKAA2. RESULTS: PRKAA2 expression was further proven to be enhanced in the placental tissues of HDCP patients compared with that of normal puerpera. Subsequently, the results of flow cytometry analysis and western blot indicated that PRKAA2 overexpression accelerated primary placental cell apoptosis, while its knockdown attenuated cell apoptosis. Mechanistically, we determined that the level of PRKAA2 succinylation was elevated in the placental tissue of HDCP patients. Through in vitro succinylation assay and mutagenesis, we confirmed that sirtuin 5 (SIRT5) interacts with PRKAA2 at K69 and K260 to induce PRKAA2 desuccinylation. SIRT5 regulated primary HDCP cell apoptosis through PRKAA2. Finally, the animal study revealed that PRKAA2 elevates the systolic blood pressure of HDCP rat model. CONCLUSION: Our findings indicated that SIRT5-mediated PRKAA2 succinylation modulates placental cell apoptosis in HDCP, suggesting that PRKAA2 is a potential therapeutic target for HDCP treatment.

Background: The nursing work in the operating room is heavy, intensive, and irregular, and the quality of nursing work can directly affect the surgical effect and patient prognosis. Therefore, nursing management in the operating room should be strengthened to protect patients' life safety effectively. Objective: To assess the effectiveness of applying the 6S management model in operating room nursing. Design: This was a retrospective study. Setting: This study was conducted at the Department of Anesthesia Surgery, Nanfang Hospital, Southern Medical University. Participants: The research included 100 operating room nurses on duty between January 2020 and December 2022. Intervention: From January 2020 to June 2021, the hospital conducted routine training programs for nurses in the operating room. From July 2021 to December 2022, the hospital adopted the 6S management model for overseeing nursing work in the operating room. Primary Outcome Measures: (1) nursing quality score (2) nursing staff safety awareness (3) nursing disputes and complaints (4) incidence of adverse reactions (5) patient satisfaction with the quality of nursing care. Results: Following the adoption of the 6S management model, there was a noticeable improvement in the nursing quality scores, the nursing staff's awareness of safety, and the satisfaction levels of patients with the quality of care provided by operation nurses (P < .05). Additionally, the incidence of nursing disputes, complaints, and adverse events among patients decreased significantly compared to before the implementation of 6S (P = .01). Conclusion: Implementing 6S management with a focus on the work of operation room nurses enhanced the competence of the nursing staff and improved management effectiveness, ultimately leading to increased patient satisfaction.

The road passenger transportation enterprise is a complex system, requiring a clear understanding of their active safety situation (ASS), trends, and influencing factors. This facilitates transportation authorities to promptly receive signals and take effective measures. Through exploratory factor analysis and confirmatory factor analysis, we delved into potential factors for evaluating ASS and extracted an ASS index. To predict obtaining a higher ASS information rate, we compared multiple time series models, including GRU (gated recurrent unit), LSTM (long short-term memory), ARIMA, Prophet, Conv_LSTM, and TCN (temporal convolutional network). This paper proposed the WDA-DBN (water drop algorithm-Deep Belief Network) model and employed DEEPSHAP to identify factors with higher ASS information content. TCN and GRU performed well in the prediction. Compared to the other models, WDA-DBN exhibited the best performance in terms of MSE and MAE. Overall, deep learning models outperform econometric models in terms of information processing. The total time spent processing alarms positively influences ASS, while variables such as fatigue driving occurrences, abnormal driving occurrences, and nighttime driving alarm occurrences have a negative impact on ASS.

Here we show that it is possible to control magnon-magnon entanglement in a hybrid magnon-atom-cavity system based on atomic coherence. In a four-level V-type atomic system, two strong fields are applied to drive two dipole-allowed transitions and two microwave cavity modes are coupled with two dipole forbidden transitions as well as two magnon modes simultaneously. It is found that the stable magnon-magnon entanglement, one-way steering and two-way EPR steering can be generated and controlled by atomic coherence according to the following two points: (i) the coherent coupling between magnon and atoms is established via exchange of virtual photons; (ii) the dissipation of magnon mode is dominant over amplification since one of the atomic states mediated one-channel interaction always keeps empty. The coherent control of magnon-magnon correlations provides an effective approach to modify macroscopic quantum effects using the laser-driven atomic systems.

Depression is a high-incidence mental illness that seriously affects human health. AQP4 has been reported to be closely associated with depression, while the underlying mechanism is still unclear. This work aimed to investigate the functional role of AQP4 in depression. Depression mouse model was constructed by administration of chronic social defeat stress (CSDS). We found that AQP4 was highly expressed in the hippocampal tissues of CSDS mice. AQP4 knockdown alleviated depression and enhanced the expression of NR2B and PSD95 in CSDS mice. Moreover, primary hippocampal neurons were treated with N-methyl-d-aspartate (NMDA) to induce neuron injury. AQP4 overexpression repressed cell viability and promoted apoptosis of NMDA-treated primary hippocampal neurons. AQP4 up-regulation repressed the expression of NR2B (surface), and enhanced the expression of NR2B (intracellular), P-NR2B, CaMK II and CK2 in the NMDA-treated primary hippocampal neurons. The influence conferred by AQP4 up-regulation was abolished by KN-93 (CaMK II inhibitor) or TBB (CK2 inhibitor) treatment. Rapamycin treatment enhanced the expression of NR2B (surface), and repressed the expression of AQP4, NR2B (intracellular) and P-NR2B in the primary hippocampal neurons by activating autophagy. The activated autophagy alleviated depression in CSDS mice by repressing AQP4 expression. In conclusion, our data demonstrated that autophagy ameliorated depression by repressing AQP4 expression in mice, and AQP4 knockdown promoted membrane trafficking of NR2B and inhibited phosphorylation of NR2B via CaMK II/CK2 pathway. Thus, our work suggests that AQP4 may be a promising molecular target for the development of antidepressant drugs.

Uranium is well-known to have serious adverse effects on the ecological environment and human health. Bioremediation stands out among many remediation methods owing to its being economically feasible and environmentally friendly. This study reported a great promising strategy for eliminating uranium by Stenotrophomonas sp. CICC 23833 in the aquatic environment. The bacterium demonstrated excellent uranium adsorption capacity (qmax = 392.9 mg/g) because of the synergistic effect of surface adsorption and intracellular accumulation. Further analysis revealed that hydroxyl, carboxyl, phosphate groups and proteins of microorganisms were essential in uranium adsorption. Intracellular accumulation was closely related to cellular activity, and the efficiency of uranium processing by the permeabilized bacterial cells was significantly improved. In response to uranium stress, the bacterium was found to release multiple ions in conjunction with uranium adsorption, which facilitates the maintenance of bacterial life activities and the conversion of uranyl to precipitates. These above results indicated that Stenotrophomonas sp. Had great potential application value for the remediation of uranium.

Evidence suggests that CXC motif chemokines are involved in neuronal injury and inflammatory processes. Bioinformatics analysis by using data from the Gene Expression Omnibus (GEO) database was performed and identified CXC motif chemokine ligands (CXCLs) as associated with diabetic peripheral neuropathy (DPN). The present study focused on CXC motif chemokine ligand 2 (CXCL2), and the role and potential mechanisms of CXCL2 in DPN were investigated. The DPN rat model was generated by streptozotocin (STZ) injection in vivo, and high-glucose (HG)-stimulated Schwann cell RSC96 was considered a cell model of DPN in vitro. Neuropathic symptoms of DPN were explored by neurological tests and histological examinations. DPN rats showed a decreased level of motor nerve conduction velocity (MNCV) along with typical histological changes. CXCL2 expression was significantly increased in STZ-induced DPN rat sciatic nerve and HG-induced RSC96 cells. Functionally, CXCL2 knockdown inhibited cell apoptosis and inflammation activation under diabetic conditions in vitro and in vivo. CXCL2 knockdown increased cell viability in HG-treated RSC96 cells and reduced apoptosis concerning the decreased expression of cleaved Caspase 3/9. In addition, CXCL2 knockdown protected against NOD-like receptor protein 3 (NLRP3) inflammasome activation and reduced levels of pro-inflammatory cytokines, interleukin (IL)-1ß and IL-18. The repressive effects of CXCL2 knockdown on inflammasome activation under HG conditions were significantly abolished by treatment of the NLRP3 activator nigericin. In conclusion, these results indicated that CXCL2 knockdown exhibited amelioration of hyperglycemia-induced DPN by inhibiting cell apoptosis and NLRP3 inflammasome activation, suggesting that targeting CXCL2 might be a potential strategy for DPN treatment.

Objective: To investigate the job burnout situation in different seniority staff in a public hospital disinfection supply center. Methods: The hospital Disinfection supply center staff in 4 grade 3 A public hospitals received a questionnaire From January 1st to 7th, 2023 including Maslach Burnout Inventory-General Survey, MBI-GS, and other programs. 158 subjects were included in this study. They were divided into three groups according to working years: A short seniority group of 68 cases (≤5 years), middle seniority: 49 cases (5 ~15 years), and long seniority: 41 cases (≥15 years). The correlation between seniority and job burnout was explored by comparing the job burnout scores of three groups. MBI-GS is divided into three parts: the sense of achievement, work attitude, and emotional failure. The full score of achievement is 36 (reverse scored). Higher scores indicate lower achievement from work and severe burnout. Besides, the gender rate, educational background, and job title, as well as the scores in achievability, job attitude, emotional exhaustion, and job burnout were compared among the three groups. Results: By comparing with the short seniority group, the proportion of men in the long seniority group (41.4%) and the middle seniority group (44.8%) were higher. Compared with the long seniority group (39%), the highly educated rate of the short seniority group (92.6%) and middle seniority group (77.5%) was higher. Compared with the long seniority group (63.5%), the proportion of low job titles including primary nurse and no job title in the short seniority group (95.6%) and middle seniority group (77.6%) were higher. The achievability of the short seniority group (19.4 ± 4.2) was significantly lower than those of the middle (11.4 ± 3.6) and long seniority group (9.4 ± 4.0). Compared with the short (10.2 ± 2.3) and long subgroups group (13.5 ± 3.1), the job attitude of the middle seniority group (9.7 ± 3.3) was poorer. Compared with the short (9.7 ± 3.3) and long seniority group (12.3 ± 4.2), the emotional control ability and thus emotional exhaustion of the middle seniority group (14.2 ± 3.8) was poorer. In contrast with the long-seniority group, job burnout situations in the short and middle-seniority groups were more serious. Conclusion: There was a significant difference in the job burnout of different seniority staff in public hospital disinfection supply centers, with lower achievability for short seniority, emotional exhaustion, and poorer job attitude for middle seniority staff.

Tyrosinase (TYR) plays a pivotal role in the biosynthesis of melanin, and its activity level holds critical implications for vitiligo, melanoma cancer, and food nutritional value. The sensitive determination of TYR activity is of great significance for both fundamental research and clinical investigations. In this work, we successfully synthesized silicon-doped carbon quantum dots (Si-CQDs) through a one-pot hydrothermal method with trans-aconitic acid as carbon source and N-[3-(trimethoxysilyl)propyl]ethylenediamine as the dopant, exhibiting remarkable fluorescence quantum yield (QY) and photostability. Correspondingly, Si-CQDs were used as a probe to construct a sensitive, rapid, and user-friendly fluorescence method for TYR detection. The method relied on the oxidation of isoprenaline (ISO) by TYR, where Si-CQDs were employed as a highly efficient probe. The testing mechanism was the internal filtering effect (IFE) observed between Si-CQDs and the oxidative system of ISO and TYR. Under the optimized conditions, the fluorescence strategy exhibited a detection range of 0.05-2.0 U/mL for TYR with a limit of detection (LOD) of 0.041 U/mL. Furthermore, we successfully demonstrated the accurate determination of TYR levels in human serum, showcasing the promising potential of this method in various practical scenarios.

BACKGROUND: Vascular dementia (VAD) is the second most common type of dementia lacking effective treatments. Pentoxifylline (PTX), a nonselective phosphodiesterase inhibitor, displays protective effects in multiple cerebral diseases. In this study, we aimed to investigate the therapeutic effects and potential mechanisms of PTX in VAD. METHODS: Bilateral common carotid artery stenosis (BCAS) mouse model was established to mimic VAD. Mouse behavior was tested by open field test, novel object recognition test, Y-maze and Morris water maze (MWM) tests. Histological staining, magnetic resonance imaging (MRI) and electron microscopy were used to define white matter integrity. The impact of PTX on microglia phagocytosis, peroxisome proliferator-activated receptors-γ (PPAR-γ) activation and Mer receptor tyrosine kinase (Mertk) expression was assessed by immunofluorescence, western blotting and flow cytometry with the application of microglia-specific Mertk knockout mice, Mertk inhibitor and PPAR-γ inhibitor. RESULTS: Here, we found that PTX treatment alleviated cognitive impairment in novel object recognition test, Y-maze and Morris water maze tests. Furthermore, PTX alleviated white matter injury in corpus callosum (CC) and internal capsule (IC) areas as shown by histological staining and MRI analysis. PTX-treatment group presented thicker myelin sheath than vehicle group by electron microscopy. Mechanistically, PTX facilitated microglial phagocytosis of myelin debris by up-regulating the expression of Mertk in BCAS model and primary cultured microglia. Importantly, microglia-specific Mertk knockout blocked the therapeutic effects of PTX in BCAS model. Moreover, Mertk expression was regulated by the nuclear translocation of PPAR-γ. Through modulating PPAR-γ, PTX enhanced Mertk expression. CONCLUSIONS: Collectively, our results demonstrated that PTX showed therapeutic potentials in VAD and alleviated ischemic white matter injury via modulating Mertk-mediated myelin clearance in microglia.

Phonon nonlinearities play an important role in hybrid quantum networks and on-chip quantum devices. We investigate the phonon statistics of a mechanical oscillator in hybrid systems composed of an atom and one or two standard optomechanical cavities. An efficiently enhanced atom-phonon interaction can be derived via a tripartite atom-photon-phonon interaction, where the atom-photon coupling depends on the mechanical displacement without practically changing a cavity frequency. This novel mechanism of optomechanical interactions, as predicted recently by Cotrufo et al. [Phys. Rev. Lett.118, 133603 (2017)10.1103/PhysRevLett.118.133603], is fundamentally different from standard ones. In the enhanced atom-phonon coupling, the strong phonon nonlinearity at a single-excitation level is obtained in the originally weak-coupling regime, which leads to the appearance of phonon blockade. Moreover, the optimal parameter regimes are presented both for the cases of one and two cavities. We compared phonon-number correlation functions of different orders for mechanical steady states generated in the one-cavity hybrid system, revealing the occurrence of phonon-induced tunneling and different types of phonon blockade. Our approach offers an alternative method to generate and control a single phonon in the quantum regime and could have potential applications in single-phonon quantum technologies.

Soluble fms-like tyrosine kinase-1 (sFlt-1), a circulating antiangiogenic protein, is involved in the pathogenesis of atherosclerosis (AS), and the underlying mechanism is still unclear. Here, we attempted to investigate the mechanism of action of sFlt-1 in AS. Human umbilical vein endothelial cells (HUVECs) were treated with oxidized low density lipoprotein (ox-LDL) to induce cell injury. ox-LDL treatment increased LC3-II/LC3-I ratio, Beclin-1 expression and GFP-LC3 puncta in HUVECs, suggesting that ox-LDL may induce autophagic flux impairment in HUVECs. ox-LDL-treated HUVECs displayed a decrease of sFlt-1 levels. Moreover, ox-LDL treatment reduced cell proliferation and elevated apoptosis in HUVECs, which was abrogated by sFlt-1 overexpression. Up-regulation of sFlt-1 repressed the activity of PI3K/AKT/mTOR signaling pathway and enhanced autophagy in HUVECs following ox-LDL treatment. Additionally, sFlt-1 overexpression-mediated increase of autophagy in ox-LDL-treated HUVECs was abolished by 3-methyladenine (autophagy inhibitor). 3-methyladenine abrogated the impact of sFlt-1 overexpression on proliferation and apoptosis in ox-LDL-treated HUVECs. This work confirmed that overexpression of sFlt-1 activated autophagy by repressing PI3K/Akt/mTOR signaling pathway, and thus alleviated ox-LDL-induced injury of HUVECs. Therefore, this study suggests that sFlt-1 may be a potential target for AS treatment.

General CuCo2O4electrodes suffer a very low reversible capacity and poor cycling stability because of easily fading phenomena and volume change during cycling. To optimize the electrode, a facile method is conducted to fabricate a novel electrode of Cu@CuCo2O4@polypyrrole nanoflowers. Due to larger specific surface area and more electrochemical reactive areas of CuCo2O4@polypyrrole nanoflowers, the pseudocapacitance of thein situgrown CuCo2O4@polypyrrole (912 F g-1at 2 A g-1) is much higher than the pristine CuCo2O4(618 F g-1at 2 A g-1). Remarkably, the CuCo2O4@polypyrrole (cathode) and active carbon (anode) are used to assemble an asymmetric supercapacitor, which exhibits a relatively high energy density of 90 Wh kg-1at a power density of 2519 W kg-1and 35 Wh kg-1at a high-power density of 9109 W kg-1, and excellent cycling stability (about 90.4% capacitance retention over 10 000 cycles). The prominent performance of CuCo2O4@polypyrrole makes it as a potential electrode for supercapacitor.

RESUMEN

This paper studies a general framework for high-order tensor SVD. We propose a new computationally efficient algorithm, tensor-train orthogonal iteration (TTOI), that aims to estimate the low tensor-train rank structure from the noisy high-order tensor observation. The proposed TTOI consists of initialization via TT-SVD [1] and new iterative backward/forward updates. We develop the general upper bound on estimation error for TTOI with the support of several new representation lemmas on tensor matricizations. By developing a matching information-theoretic lower bound, we also prove that TTOI achieves the minimax optimality under the spiked tensor model. The merits of the proposed TTOI are illustrated through applications to estimation and dimension reduction of high-order Markov processes, numerical studies, and a real data example on New York City taxi travel records. BACKGROUND The incidence of intra-abdominal hypertension (IAH) and abdominal compartment syndrome (ACS) in intensive care units is high. Dynamic monitoring of intra-abdominal pressure (IAP) is important to treat patients with these conditions. The World Society of Abdominal Compartment Syndrome revised IAP measurement and treatment guidelines in 2013. IAP is measured by instilling ≤25 mL of sterile saline into the bladder, but there is no requirement for the saline to be at a specific temperature. Many doctors presume that using cold saline will trigger bladder muscle spasms, resulting in measurement error. In the present study, we investigated the effect of body-temperature saline on IAP measurements. MATERIAL AND METHODS A single-center study was conducted in 12 patients with IAH over a 2-year period. IAP was measured via the bladder with instillation of sterile saline at temperatures of 35°C, 25°C, and 15°C. We analyzed the data using R software, version 4.1.0. Paired t tests were used for comparisons between groups. A Spearman rank correlation analysis was performed to compare groups. Analysis results were plotted using the R packages ggplot2, ggpubr, and BlandAltmanLeh. P<0.05 was considered statistically significant. RESULTS There was a significant difference in IAP measurement between the 15°C and 35°C groups (t=-2.55, P=0.027). There was no significant difference between the 25°C and 35°C groups (t=0.73, P=0.48). Bland-Altman analysis showed that IAP was consistent in the 25°C and 35°C groups. CONCLUSIONS Although it is preferable to measure IAP with saline at body temperature (35°C), a temperature >25°C is associated with accurate results. Using saline at <15°C should be avoided.

BACKGROUND: The reproducibility of intravoxel incoherent motion (IVIM)-based radiomics studies in humans has not been reported. PURPOSE: To determine the inter- and intra-observer variability on the reproducibility of IVIM-based radiomics features in cervical cancer (CC). MATERIAL AND METHODS: The IVIM images of 25 patients with CC were retrospectively collected. Based on the high-resolution T2-weighted images, the regions of interest (ROIs) were independently delineated twice in diffusion-weighted images at a b value of 1000 s/mm2 (interval time was one month) by two radiologists. This was done at the largest transversal cross-sections of the tumors. The ROI was subsequently used in apparent diffusion coefficient (ADC), true diffusion coefficient (D), pseudo-diffusion coefficient (D*), and perfusion fraction (f) maps derived from IVIM images. In total, 105 radiomics features were then finally extracted from the IVIM-derived maps. The inter- and intra-observer reproducibility of IVIM-derived features was then evaluated using the intraclass correlation coefficient. RESULTS: Inter- and intra-observer variability affected the reproducibility of radiomics features. D* map had 100% and 95% reproducible features, ADC map had 89% and 93%, D map had 97% and 86%, while f map had 54% and 62% reproducible features with good to excellent reliability in the intra-observer analysis. Similarly, D* map had 90% and 94%, ADC map had 85% and 70%, D map had 81% and 78%, while f map had 41% and 93% reproducible features with good to excellent reliability in the inter-observer analysis. CONCLUSION: Inter- and intra-observer variability can affect radiomics analysis. Cognizant to this, multicenter studies should pay more attention to intra- and inter-observer variability.

Mesenchymal stem/stromal cells (MSCs) play an important role in bone tissue engineering because MSCs possess multilineage potential of differentiation to mesenchymal tissues. Semaphorin 3A (Sema3A) and hypoxia-inducible factor-1α (HIF1α) are proved as important regulatory factors for osteogenesis and angiogenesis. The aim of this study was to investigate the effects of Sema3A and HIF1α co-overexpression on the osteogenesis and angiogenesis in induced pluripotent stem cell-derived mesenchymal stem cells (iPSC-MSCs). Importantly, we assessed the potential osteogenic effectiveness of Sema3A and HIF1α co-overexpressed iPSC-MSCs seeded on hydroxyapatite (HA) scaffold in a mouse calvarial defect model. The overexpression for Sema3A, HIF1α, or Sema3A-HIF1α fusion in iPSC-MSCs was performed by separately infecting with conducted lentiviral vector. We determined the cell proliferation, the expressions of osteogenic, and endothelial markers of iPSC-MSCs cultured in osteogenic or endothelial induction medium in vitro. A mouse model calvarial defect was created and implanted with the Empty implant, HA scaffold alone, HA scaffold combined with iPSC-MSCs that infected with negative control or Sema3A-HIF1α fusion for 8 weeks in vivo. The results showed that Sema3A and HIF1α co-overexpression reversed the reduced cell proliferation that reduced by Sema3A overexpression alone. Importantly, the co-overexpression significantly increased the expressions of osteogenic and angiogenic related-genes compared with negative control after induction. Moreover, the Sema3A-HIF1α co-overexpressed iPSC-MSCs seeded on HA scaffold boosted the new bone and collagen fiber formation and facilitated repair of calvarial defect in a mouse model, which might have the potential application for bone defect reconstruction.

BACKGROUND: The majority of patients with chronic fatigue have a risk of comorbidity with depression. Pinocembrin (PB) is a kind of flavonoid molecule isolated from honey and propolis and has antimicrobial, anti-inflammatory, antioxidant, and anticancer function. The purpose of the current study was to determine the possible function of PB on treatment of depression. METHODS: A chronic unpredictable mild stress (CUMS) mouse model was established to mimic the depressive-like behaviors in vivo. The depressive-like behaviors of CUMS mice were measured by sucrose preference test (SPT), open field test (OFT), forced swim test (FST) and tail suspension test (TST). The concentration of reactive oxygen species (ROS), malondialdehyde (MDA) and the activity or superoxide dismutase (SOD) were detected by commercial kit. The inflammatory factor including interleukin (IL)-1ß, tumor necrosis factor (TNF)-α, IL-10 and transforming growth factor (TGF)-ß were examined. RESULTS: We found that PB alleviated the decreasing of sucrose preference and body weight. CUMS mice significantly increased the immobility time but decreased latency to abandon in FST, increased the immobility time in TST, and reduced crossing score and rearing score in OFT, whereas these changes were reversed by PB treatment. More importantly, PB decreased the concentration of ROS and MDA, but increased the SOD activity, suggesting that it could protected against oxidative stress in CUMS mice. Interestingly, PB inhibited cell apoptosis and regulated inflammatory factors expression in hippocampus of CUMS mice. Moreover, PB activated Nrf2/HO-1 signal pathway but inhibited the phosphorylation of NF-kB. CONCLUSIONS: In conclusion, PB mitigated CUMS-induced depressive-like behaviors through ameliorating neuroinflammation and apoptosis. TRIAL REGISTRATION: Not Applicable.

Microglia polarization is associated with the pathogenesis of depression. A previous study shows that long non-coding RNA uc.80- is down-regulated in the hippocampus of depressed rats. Thus, this article aims to investigate the role of uc.80- in microglia polarization in depression. We first established depression model rats by chronic unpredictable mild stress (CUMS) regiment. We found that hippocampus of depressed rats exhibited an increase of M1 microglias and a decrease of M2 microglias. uc.80- was down-regulated in hippocampus of depressed rats. Furthermore, the detection of behaviouristics of depressed rats showed that uc.80- overexpression alleviated depression of rats. In addition, uc.80- overexpression promoted M2 polarization of microglias in vivo and in vitro. uc.80- overexpression led to a decrease in apoptosis of hippocampal neurons in vivo and in vitro. In conclusion, our study confirms that lncRNA uc.80- overexpression ameliorates depression in rats by promoting M2 polarization of microglias. Thus, our work suggests that uc.80- may be a target gene for depression treatment.
