OBJECTIVE: To explore the trajectory of psychological resilience and its impact factors of adolescents in earthquake-hit areas by latent variable growth curve model (LGM) and to find out protective factors for psychological resilience. METHODS: Longitudinal design was administrated. All grade one students in two junior high schools in the heavily and slightly earthquake-hit areas respectively were investigated by follow-up study in one year. Repeated measures analysis of variance and latent variable growth curve model were used for statistical analysis. RESULTS: The three-time measurements of psychological resilience of adolescents in the heavily earthquake-hit areas were significantly lower than corresponding measurements of the slightly earthquake-hit areas all along. There was an ascending trend of adolescent's psychological resilience and objective support over one year (P < 0.05), while a descending trend of self-esteem (P < 0.05). The directly protective factors of resilience were self-esteem and subjective support and indirectly protective factors were objective support and utilization of social support. The psychological resilience of boys was higher than that of girl, but the change trend of psychological resilience was no difference between boys and girls. CONCLUSION: Psychological resilience of adolescents had dynamic characteristics. Psychological resilience of adolescents would be damaged in disasters but it could be recovered through enhance internal and external protective factors.

Porcine reproductive and respiratory syndrome virus (PRRSV) is one of the most economically important pathogens to the swine industry worldwide over the past three decades. No approved effective antiviral drug is available to control this virus. The antiviral effects of allicin (diallyl thiosulfinate) on many human and animal viruses have been documented. However, the antiviral effect of allicin on PRRSV infection remains unknown. In this study, we found that allicin exhibited an inhibitory effect on HP-PRRSV and NADC30-like PRRSV in a dose-dependent manner by interfering with viral entry, replication, and assembly. Furthermore, allicin alleviated the expression of pro-inflammatory cytokines (IFN-ß, IL-6, and TNFα) induced by PRRSV infection. The pro-inflammatory signaling pathways, TNF signaling pathway and MAPK signaling pathway, up-regulated by PRRSV infection were restored by allicin treatment. Taken together, these results demonstrate that allicin has antiviral activity against PRRSV and ameliorates inflammatory responses induced by PRRSV infection, suggesting that allicin is a promising drug candidate for anti-PRRSV therapy in vivo.

RESUMEN

The reverse genetics system is a valuable tool in the virological study of RNA viruses. With the availability of reverse genetics, the porcine reproductive and respiratory syndrome virus (PRRSV) has been utilized as a viral vector for the expression of foreign genes of interest. Here, we constructed a full-length cDNA clone of a highly pathogenic PRRSV (HP-PRRSV) TA-12 strain. Using this cDNA clone, we generated a reporter virus expressing a gaussia luciferase (Gluc) via an additional subgenomic RNA between ORF7 and 3'UTR. This reporter virus exhibited similar growth kinetics to the wild-type (WT) virus and remained genetically stable for at least ten passages in MARC-145 cells. In cells infected with this reporter virus, the correlation between the expression levels of Gluc in culture media and the virus titers suggested that Gluc is a good indicator of the reporter virus infection. With this reporter virus, we further established the Gluc readout-based assays for antiviral drug screening and serum neutralizing antibody detection that exhibited comparable performance to the classical assays. Taken together, we established a reverse genetics system of HP-PRRSV and generated a novel reporter virus that could serve as a valuable tool for antiviral drug screening and serum neutralizing antibody detection.

Porcine epidemic diarrhea virus (PEDV), a swine enteric coronavirus causing acute diarrhea in piglets, is one of the major threatens to the pork industry globally. Reverse genetics is a valuable tool for the virological study and vaccine development for coronaviruses. Due to the large size and unstable problem in Escherichia coli of coronavirus genome, construction and manipulation of reverse genetics system for coronaviruses remain laborious and time-consuming. In this study, a reverse genetics system of the genotype II PEDV strain HM was generated using the transformation-associated recombination (TAR) technology in yeast within 1 week. The rescued virus (rPEDV) exhibited similar growth properties to the wild-type virus in vitro. With this PEDV infectious cDNA clone, CRISPR/Cas9 technology and homologous recombination were combined to generate a recombinant virus rPEDV-EGFP in which the ORF3 gene was swapped with an EGFP gene. The reporter virus displayed similar growth properties to the parental virus rPEDV and remained stable during serial passage in vitro. Of note, the strategies of construction and manipulation of PEDV infectious cDNA clone are extremely simple and efficient, which could be applied for other RNA viruses and DNA viruses.

Japanese encephalitis virus (JEV) belongs to a zoonotic flavivirus and is the main cause of viral encephalitis in humans throughout Asia. During the past two decades, both genotypes I (G) and III (GIII) JEVs co-existed in many Asian countries, and the prevalent strains have shifted from GIII to GI. Since all licensed JE vaccines are derived from GIII strains and no specific treatment is available, the development of novel vaccines and specific antiviral drugs against both genotypes is urgently required. To solve the unstable issue of the JEV infectious cDNA clone and establish a reliable antiviral screening assay, we established the yeast artificial chromosome (YAC)-based reverse genetics systems for GI and GIII JEV strains through transformation-associated recombination (TAR) technology in yeast. The YAC-based infectious clones of GI and GIII JEV exhibited high genetic stability both in yeast and E. coli. Using these reverse genetics systems, recombinant EGFP-reporter viruses were generated and remained stable for at least 10 passages in vitro. An image-based antiviral assay for JEV was developed with the EGFP-reporter viruses, and two drugs were identified to have a broad-spectrum inhibitory effect on GI and GIII JEV replication, which provide potential new therapeutic for the treatment of multiple genotypes JEV infection.

In recent years, African swine fever (ASF) has caused a devastating blow to the swine industry globally. Since no effective vaccine is available, strict biosafety measures and rapid diagnosis are the most effective strategies for ASF control. ASFV p30 is one of the most antigenic viral proteins that have been widely used in the field for serological diagnosis of ASF infection. In this study, we developed a luciferase immunoprecipitation system (LIPS) assay for the detection of ASFV antibodies in pig serum using Gaussia luciferase (GLuc)-tagged p30 as a diagnostic antigen. The optimal GLuc-p30 input of 107 luminance units (LU) and optimal serum dilution factor of 1/100 were set to achieve the highest P/N ratio. Based on 87 ASFV-positive and negative pig sera, the cutoff value of the S/N ratio could be set between 2.298 and 30.59 to achieve 100% sensitivity and 100% specificity. Moreover, the diagnostic sensitivity of this LIPS is comparable to that of a commercial enzyme-linked immunosorbent assay (ELISA) and the specificity of LIPS is even superior to the tested ELISA. In conclusion, we have established a LIPS assay for ASFV antibody detection, which could be a potential method for ASFV diagnosis in laboratories and farms.

Air pollution adversely affects skin, leading to skin inflammation and premature skin aging. Plant derived antioxidant compounds have been considered to be promising in discovery of effective agents for the protection of skin from the damage by air pollutants. Our previous studies demonstrated that Averrhoa carambola fruit (known as star fruit) is rich in flavonoid C-glycosides with unique structures and potent antioxidant activity. Thus, the star fruit extract (SFE) and main flavonoid C-glycoside components, carambolasides I, J, and P (1-3), carambolaflavone B (4), and isovitexin 2″-O-α-L-rhamnoside (5), were investigated for the activity against air pollutant stress in human epidermis. As a result, SFE and compounds 1-5 exhibited significant inhibitory activity against protein carbonylation in oxidative-stressed stratum corneum with the best activity being shown by compound 3. SFE and compounds 2-5 were also active against engine exhaust-induced protein carbonylation in stratum corneum. When further evaluated, SFE and compound 3 significantly inhibited gene expression of the key inflammation mediators IL-1α and COX-2 in PM-stressed keratinocytes. The results indicated that SFE and the flavonoid C-glycosides are potentially effective against air pollutant-induced skin inflammation and premature aging.

Tripartite motif 35 (TRIM35) protein is a ubiquitin E3 ligase that mediates interferon-beta (IFN-ß) production via regulating ubiquitination of multiple adaptor proteins in innate immune signaling pathways. Here, we cloned the porcine TRIM35 (porTRIM35) gene and analyzed its involvement in IFN-ß expression as well as the antiviral response against Japanese encephalitis virus (JEV). The full-length porTRIM35 gene encoded a 493-amino acid protein and exhibited 79.6%-89.5% sequence similarity with its orthologues in humans, mice, monkeys and rabbits. porTRIM35 possessed typical structural features of TRIMs, including a RING domain, a B-box domain, a coiled-coil domain and a PRY/SPYR domain. Exogenous overexpression of porTRIM35 significantly up-regulated the mRNA expression level of IFN-ß in swine testicular (ST) cell in response to poly(I:C) stimulation, whereas knockdown endogenous expression of porTRIM35 lead to a decrease in the expression level of IFN-ß. Mechanically, porTRIM35 directly interacted with porcine TNF-receptor associated factor 3 (TRAF3) and catalyzed its Lys63-linked polyubiquitination, thereby leading to the up-regulation of IFN-ß production. Meanwhile, we demonstrated that the RING and PRY/SPRY domains were essential for the E3 ligase activity of porTRIM35. In response to JEV infection, the endogenous expression of porTRIM35 was markedly inhibited at the mRNA level, while exogenous expression of porTRIM35 significantly elevated the expression of IFN-ß induced by JEV infection and reduced viral titers in ST cells, suggesting that porTRIM35 is a negative regulator for JEV replication. These data demonstrate the importance of porTRIM35 in IFN-ß expression as well as the antiviral response against JEV replication.

OBJECTIVE: A pulsed amperometric detector-high performance anion chromatography method for the determination of L-carnitine in health food was established. METHODS: After samples were dissolved and dispersed with ether, L-carnitine in samples was extracted with 0. 1 mmol/L HC1. Using 200 mmol/L NaOH as the mobile phase, amino PAC PA10 Amino acid-separation column as separation column and pulse ampere detector with An as working electrode throughout the experiment. RESULTS: The linear was good in the range of 0.08 - 1.00 mg/ml. The intra-day RSDs of peak area and retention time for L-carnitine standard solution were 3.97% and 0.62% and the inter-day RSDs were 5.66% and 0.69%, respectively. The average recoveries were 95.6% - 113.5% and the RSD of sample analysis was 7.34%. The results of the proposed method for the analysis of L-carnitine in health foods were satisfactory. CONCLUSION: The proposed method is simple, accurate and reproducible. One analysis can be completed within 15 min. The method is adaptive for the analysis of L-carnitine in health food.

In this paper, we reported an integrated method to generate ultra-wideband (UWB) pulses of different orders based on a reconfigurable silicon micro-ring resonator-coupled Mach-Zehnder interferometer. Under proper operating conditions, the device can produce Fano resonances with a peak-to-valley extinction ratio of above 20 dB. UWB monocycle and doublet signals with picosecond pulse widths are produced when the microring resonator is modulated by square and Gaussian electrical pulses, respectively. With our Fano resonance modulator on silicon photonics, it is promising to foresee versatile on-chip microwave signal generation.

OBJECTIVE: To identify the prevalence of posttraumatic stress disorder (PTSD) and its determinants among adolescents more than 4 years after the 2008 Wenchuan earthquake. METHOD: Adolescents (1,125 total) from 2 junior high schools in areas affected by the catastrophic earthquake were followed up for 3 years. The self-rating PTSD scale based on the Manual of Mental Disorders, 4th Edition (DSM-IV) and the Chinese Classification and Diagnostic Criteria of Mental Disorders, 2nd Edition, Revised (CCMD-2-R) was collected at 53 months, and determinant data were collected repeatedly. Logistic regression was used for a determinants analysis. RESULTS: The prevalence of overall PTSD was 23.4% among the sample. The risk factors for PTSD were older age (OR=1.52, 95% CI: 1.20~1.92), and death or injury of a family member in the earthquake (OR=1.61, 95% CI: 1.09~2.37). Adolescents who had moderate-to-severe common mental health problems were more likely to have PTSD symptoms, with ORs from 3.98 to 17.67 (All P<0.05). Self-esteem remained a protective factor for PTSD regardless of age, whereas positive coping was a protective factor for PTSD when adolescents were older. CONCLUSION: PTSD symptoms among adolescent survivors of a catastrophic earthquake seemed to persist over time. Long-term interventions are needed to alleviate PTSD symptoms among adolescent survivors. (Disaster Med Public Health Preparedness. 2019;13:414-418).

BACKGROUND: Habitat fragmentation, water resources and biological characteristics are important factors that shape the genetic structure and geographical distribution of desert plants. Analysis of the relationships between these factors and population genetic variation should help to determine the evolutionary potential and conservation strategies for genetic resources for desert plant populations. As a traditional Chinese herb, Glycyrrhiza inflata B. (Fabaceae) is restricted to the fragmented desert habitat in China and has undergone a dramatic decline due to long-term over-excavation. Determining the genetic structure of the G. inflata population and identifying a core collection could help with the development of strategies to conserve this species. RESULTS: We investigated the genetic variation of 25 G. inflata populations based on microsatellite markers. A high level of population genetic divergence (FST = 0.257), population bottlenecks, reduced gene flow and moderate genetic variation (HE = 0.383) were detected. The genetic distances between the populations significantly correlated with the geographical distances, and this suggests that habitat fragmentation has driven a special genetic structure of G. inflata in China through isolation by distance. STRUCTURE analysis showed that G. inflata populations were structured into three clusters and that the populations belonged to multiple water systems, which suggests that water resources were related to the genetic structure of G. inflata. In addition, the biological characteristics of the perennial species G. inflata, such as its long-lived seeds, asexual reproduction, and oasis ecology, may be related to its resistance to habitat fragmentation. A core collection of G. inflata, that included 57 accessions was further identified, which captured the main allelic diversity of G. inflata. CONCLUSIONS: Recent habitat fragmentation has accelerated genetic divergence. The population genetic structure of G. inflata has been shaped by habitat fragmentation, water resources and biological characteristics. This genetic information and core collection will facilitate the conservation of wild germplasm and breeding of this Chinese medicinal plant.

In this article, we present poly (methyl methacrylate)-b-polystyrene (PMMA-b-PS) with different block ratios and high molecular weight, which was synthesized by environmentally friendly seeded emulsion polymerization with 1,1-diphenylethylene (DPE) as a chain transfer agent. Polymerization kinetics in the first and second stage was investigated. Stable latex and homogeneous latex particles were obtained with the characterization of laser light scattering (LLS) and transmission electron microscopy (TEM). SEC and (1)H NMR revealed the successful preparation of block copolymers with high molecular weight and two different block ratios. The morphology of microphase separation of block copolymer thin films was investigated by AFM, and long-range order lamellar morphology was observed after vapor-annealing. The block copolymer with block ratio of almost 1:1 and higher molecular weight than that of previous PMMA-b-PS was sulfonated with acetyl sulfate, and the sulfonation was confirmed by FTIR, (1)H NMR, and TGA. Then, the sulfonated PMMA-b-PS was casted as membranes. The electrochemical impedance spectroscopy displayed that membranes possessed favorable proton conductivity and fine dimensional stability, and they could be candidates as proton exchange membranes.

One-dimensional (1D) magnetic Fe(3)O(4)/P(GMA-DVB) peapod-like nanochains have been successfully synthesized by magnetic-field-induced precipitation polymerization using Fe(3)O(4) as building blocks and P(GMA-DVB) as linker. The Fe(3)O(4) microspheres without surface modification can be arranged with the direction of the external magnetic field in a line via the dipolar interaction between Fe(3)O(4) microspheres and linked permanently via P(GMA-DVB) coating during precipitation polymerization. The length of peapod-like nanochains can be controlled by magnetic field intensity, and the thickness of polymer shell can be tuned by the amount of monomers. Magnetic measurement revealed that these 1D peapod-like nanochains showed highly magnetic sensitivity. In the presence of magnetic field, 1D magnetic Fe(3)O(4)/P(GMA-DVB) peapod-like nanochains can be oriented and aligned along the direction of external magnetic field.
