RESUMO

The Laser Communications Relay Demonstration is NASA's multi-year demonstration of laser communication from the Earth to a geosynchronous satellite. The mission currently has two optical ground stations (OGSs), with one in California (OGS1) and one in Hawaii (OGS2). Each ground terminal optical system consists of a high-order adaptive optics (AO) system, a laser transmit system, and a camera for target acquisition. The OGS1 AO system is responsible for compensating for the downlink beam for atmospheric turbulence and coupling it into the modem's single mode fiber. The mission requires a coupling efficiency of 50%, which necessitates a high-order AO system. To achieve this performance, the AO system uses two deformable mirrors with one mirror correcting for low-spatial-frequency aberrations with large amplitude and a second deformable mirror correcting for high-spatial-frequency aberrations with small amplitude. Turbulence is sensed with a Shack-Hartmann wavefront sensor. To meet its performance requirements in the most stressing conditions, the system can operate at frame rates of 20 kHz. This high frame rate is enabled by the design of the real-time control system. We present an overview of both the hardware and software design of the system, and describe the control system and methods of reducing non-common path aberrations. Finally, we show measured system performance.

Glucocorticoid-induced glaucoma is a secondary open-angle glaucoma. About 40% of the general population may develop elevated intraocular pressure on prolonged glucocorticoid treatment secondary to damages in the trabecular meshwork (TM), a tissue that regulates intraocular pressure. Therefore, identifying the key molecules responsible for glucocorticoid-induced ocular hypertension is crucial. In this study, Dickkopf-related protein 1 (Dkk1), a canonical Wnt signaling inhibitor, was found to be elevated in the aqueous humor and TM of glaucoma patients. At the signaling level, Dkk1 enhanced glucocorticoid receptor (GR) signaling, whereas Dkk1 knockdown or Wnt signaling activators decreased GR signaling in human TM cells as indicated by luciferase assays. Similarly, activation of the GR signaling inhibited Wnt signaling. At the protein level, glucocorticoid-induced extracellular matrix was inhibited by Wnt activation using Wnt activators or Dkk1 knockdown in primary human TM cells. In contrast, inhibition of canonical Wnt signaling by ß-catenin knockdown increased glucocorticoid-induced extracellular matrix proteins. At the physiological level, adenovirus-mediated Wnt3a expression decreased glucocorticoid-induced ocular hypertension in mouse eyes. In summary, Wnt and GR signaling inhibit each other in the TM, and canonical Wnt signaling activators may prevent the adverse effect of glucocorticoids in the eye.

Glaucoma is a leading cause of blindness worldwide. The loss of vision in glaucoma patients is due to optic nerve damage. The most important risk factor of glaucoma is elevated intraocular pressure (IOP) which is due to glaucomatous changes in the trabecular meshwork. Animal models, especially mouse models for ocular hypertension (OHT), are important for studying glaucoma. Published studies showed that 2.5 × 107 PFU adenoviral vectors expressing the biologically active form of human TGFß2 elevate IOP in female C57BL/6J mice when they are intravitreally delivered. In this study, we found that 2.5 × 107 PFU adenoviral TGFß2 vector did not elevate IOP in 3- or 5-month old male C57BL/6J mice. In contrast, 5 × 107 PFU of the same viral vectors elevated IOP in both 3- and 5-month old male C57BL/6J mice. Also, 5-month old mice showed earlier OHT and higher IOP compared to 3-month old mice. In summary, our data showed that age and sex play roles in adenoviral vector-mediated TGFß2-induced OHT in C57BL/6J mice.

Elevated levels of TGFß2 in the aqueous humor is associated with the pathological changes in the trabecular meshwork (TM). These changes lead to ocular hypertension (OHT), the most important risk factor for the development and progression of primary open angle glaucoma (POAG), a leading cause of blindness worldwide. Therefore, TGFß2 is frequently used to develop OHT models including in perfusion cultured eyes and in mouse eyes. Adenovirus-mediated overexpression of human mutant TGFß2 has demonstrated great success in increasing intraocular pressure (IOP) in mouse eyes. However, adenoviruses have limited capacity for a foreign gene, induce transient expression, and may cause ocular inflammation. Here, we explored the potential of using lentiviral vectors carrying the mutant human TGFß2C226S/C228S (ΔhTGFß2C226S/C228S) gene expression cassette for the induction of OHT in C57BL/6J mice. Lentiviral vectors using CMV or EF1α promoter to drive the expression of ΔhTGFß2C226S/C228S were injected into one of the mouse eyes and the fellow eye was injected with the same vector but expressing GFP/mCherry as controls. Both intravitreal and intracameral injection routes were tested in male and female mice. We did not observe significant IOP changes using either promoter or injection route at the dose of 8 × 105 PFU/eye. Immunostaining showed normal anterior chamber angle structures and a slight increase in TGFß2 expression in the TM of the eyes receiving intracameral viral injection but not in those receiving intravitreal viral injection. At the dose of 2 × 106 PFU/eye, intracameral injection of the lentiviral vector with the CMV-ΔhTGFß2C226S/C228S cassette induced significant IOP elevation and increased the expression of TGFß2 and fibronectin isoform EDA in the TM. Our data suggest that lentiviral doses are important for establishing the TGFß2-induced OHT model in the C57BL/6J strain.

Cross-linked actin networks (CLANs) in trabecular meshwork (TM) cells may contribute to increased IOP by altering TM cell function and stiffness. However, there is a lack of direct evidence. Here, we developed transformed TM cells that form spontaneous fluorescently labelled CLANs. The stable cells were constructed by transducing transformed glaucomatous TM (GTM3) cells with the pLenti-LifeAct-EGFP-BlastR lentiviral vector and selection with blasticidin. The stiffness of the GTM3-LifeAct-GFP cells were studied using atomic force microscopy. Elastic moduli of CLANs in primary human TM cells treated with/without dexamethasone/TGFß2 were also measured to validate findings in GTM3-LifeAct-GFP cells. Live-cell imaging was performed on GTM3-LifeAct-GFP cells treated with 1 µM latrunculin B or pHrodo bioparticles to determine actin stability and phagocytosis, respectively. The GTM3-LifeAct-GFP cells formed spontaneous CLANs without the induction of TGFß2 or dexamethasone. The CLAN containing cells showed elevated cell stiffness, resistance to latrunculin B-induced actin depolymerization, as well as compromised phagocytosis, compared to the cells without CLANs. Primary human TM cells with dexamethasone or TGFß2-induced CLANs were also stiffer and less phagocytic. The GTM3-LifeAct-GFP cells are a novel tool for studying the mechanobiology and pathology of CLANs in the TM. Initial characterization of these cells showed that CLANs contribute to at least some glaucomatous phenotypes of TM cells.

The human anterior segment perfusion culture model is a valuable tool for studying the trabecular meshwork (TM) and aqueous humor outflow in glaucoma. The traditional model relies on whole eye globes resulting in high cost and limited availability. Here, we developed a glue-based method which enabled us to use human corneal rims for perfusion culture experiments. Human corneal rim perfusion culture plates were 3D printed. Human corneal rims containing intact TM were attached and sealed to the plate using low viscosity and high viscosity glues, respectively. The human corneal rims were perfused using the constant flow mode, and the pressure changes were recorded using a computerized system. Outflow facility, TM stiffness, and TM morphology were evaluated. When perfused at rates from 1.2 to 3.6 µl/min, the outflow facility was 0.359 ± 0.216 µl/min/mmHg among 10 human corneal rims. The stiffness of the TM in naïve human corneal rim was similar to that of perfusion cultured human corneal rim. Also, the stiffness of TM of corneal rims perfused with dexamethasone was significantly higher than the control. Human corneal rims with glue contamination in the TM could be differentiated by high baseline intraocular pressure as well as high TM stiffness. Histology studies showed that the TM tissues perfused with plain medium appeared normal. We believed that our glued-based method is a useful tool and low-cost alternative to the traditional anterior segment perfusion culture model.

Noninvasive in vivo imaging of the mouse retina is essential for eye research. However, imaging the mouse fundus is challenging due to its small size and requires specialized equipment, maintenance, and training. These issues hinder the routine evaluation of the mouse retina. In this study, we developed a noncontact imaging system consisting of a smartphone, a 90D condensing lens, a homemade light diaphragm, a tripod, and a Bluetooth remote. With minimal training, examiners were able to capture fundus images from the mouse retina. We also found that fundus images captured using our system from wild type mice, mice with laser-induced retinal injury, and a mouse model of retinitis pigmentosa showed a quality similar to those captured using a commercial fundus camera. These images enabled us to identify normal structures and pathological changes in the mouse retina. Additionally, fluorescein angiography was possible with the smartphone system. We believe that the smartphone imaging system is low cost, simple, accessible, easy to operate, and suitable for the routine screening and examination of the mouse eye.

BACKGROUND: The adenoma detection rate (ADR) is of primary importance to the quality of screening colonoscopy. An online survey was conducted to assess knowledge and practices on ADR. METHODS: Paid questionnaire distributed by email. Eligible respondents were board certified gastroenterologists who perform >80 colonoscopies per month with 3 to 35 years practice after fellowship. RESULTS: There were 250 respondents. In total, 39% were unaware that ADR applies only to screening colonoscopies and 76% incorrectly answered that ADR includes sessile-serrated polyps/adenomas. A total of 51% of respondents incorrectly believe the threshold is set at 25% because 25% is a national ADR average. Many also believe the threshold depends on the patient population (current evidence suggests adjusting ADR for factors other than age and sex is unnecessary). 75% ranked ADR as highly important. 80% reported tracking ADR. A busy practice was the most common reason for not tracking ADR. Caps, chromoendoscopy, and good bowel preparation were viewed as valuable for improving ADR (this is true except for caps). High-definition colonoscopes and education were considered less valuable, despite evidence indicating that both are associated with improved ADR. In total, 57% reported not sharing ADR information with their patients, and 59% reported that no patients in the last 6 months asked for their ADR. CONCLUSIONS: The importance of ADR as a quality measure is well understood, but there are misconceptions among gastroenterologists regarding the definition and measurement of ADR, and which methods are proven to increase ADR. Patients are having very little impact on ADR measurement.

OBJECTIVES: This study aimed to estimate rates of cancer or opportunistic infection in patients with psoriatic arthritis (PsA) compared with patients without PsA. METHODS: Using the Clinical Practice Research Datalink, we conducted a cohort study of patients with a PsA diagnosis and patients without such diagnosis, matched on age, sex, general practice, and calendar time, to assess the incidence of cancers (solid, hematologic, and nonmelanoma skin cancer) and opportunistic infections. We estimated incidence rates (IRs) and IR ratios (IRRs) with 95% confidence intervals (CIs) for each outcome and stratified results in the PsA cohort by receipt of systemic PsA drugs. RESULTS: The rate of hematologic cancer was slightly higher in the PsA cohort compared with the non-PsA cohort (IRR, 1.52; 95% CI, 1.10-2.10), whereas the rates of solid cancer and of nonmelanoma skin cancer were similar between the PsA and non-PsA cohorts (IRR, 1.01; 95% CI, 0.90-1.13; and IRR, 0.97; 95% CI, 0.82-1.14, respectively). Incidence rates were higher for PsA patients who received prescriptions for PsA drugs compared with those who did not. The IRs for infection were higher in the PsA compared with the non-PsA cohort (IRR, 1.39; 95% CI, 1.31-1.47) and were significantly higher in patients who received prescriptions (IRR, 1.71; 95% CI, 1.52-1.91). CONCLUSIONS: The rates of solid and nonmelanoma skin cancers were similar in patients with PsA compared with patients without PsA, but the rates of hematologic cancer and opportunistic infections were higher in patients with PsA. In patients with PsA, rates of all outcomes were higher among those who received prescriptions for systemic PsA therapy.

OBJECTIVE: To estimate rates of suicidal behaviors and treated depression in patients with psoriatic arthritis (PsA) in comparison to non-PsA patients. METHODS: Using the Clinical Practice Research Datalink, we conducted a cohort study of patients with PsA compared to non-PsA patients. Patients with codes for suicidal behaviors (ideation, attempts, and suicide) and treated depression (diagnosis plus anti-depressant prescription) recorded during follow-up were identified as cases. We estimated incidence rates (IRs) and incidence rate ratios (IRRs) with 95% confidence intervals (CIs) for each outcome and stratified results in the PsA cohort by receipt of systemic PsA drugs. RESULTS: The rates of suicide ideation, attempt, and suicide were similar for PsA and non-PsA patients [IRR = 0.99 (95%CI: 0.67-1.47), IRR = 1.07 (95%CI: 0.86-1.34), and 0.34 (95%CI: 0.05-2.48), respectively] and rates of suicidal behaviors were slightly higher among PsA patients who received PsA drugs compared to those who did not. PsA patients had slightly higher rate of treated depression compared to non-PsA patients [IRR = 1.38 (95%CI: 1.27-1.49)] and were significantly higher in PsA patients who received drugs [IRR = 1.59 (95%CI: 1.35-1.86)]. CONCLUSIONS: Rate of depression was higher in patients with PsA compared to non-PsA patients. The rate of suicidal behaviors was similar between the two cohorts.

BACKGROUND: Few studies report estimates of cardiovascular disease (CVD) or major adverse cardiovascular events (MACE) in patients with psoriatic arthritis (PsA). OBJECTIVE: To estimate rates of incident CVD and MACE in patients with PsA compared to patients without PsA. METHODS: Using the Clinical Practice Research Datalink, we conducted 2 cohort studies of patients with PsA compared to patients without PsA or psoriasis matched on age, sex, general practice, and calendar time: 1 study of CVD and 1 study of MACE. In each study, we excluded patients who had a study outcome before cohort entry. Cases were patients with a first-time diagnosis of CVD or MACE recorded during follow-up. We estimated incidence rates (IRs) and incidence rate ratios (IRRs) with 95% confidence intervals (95% CI) and stratified results in the PsA cohort by exposure to systemic PsA treatments. RESULTS: The IR of CVD was higher in the patients with PsA compared to those without PsA (12.8/1000 person-years [PYs] [95% CI, 11.9-13.7] and 9.6/1000 PYs [95% CI, 9.3-9.0]; IRR, 1.33 [95% CI, 1.23-1.44]). The IR of MACE was slightly higher in the PsA compared to the non-PsA cohort (4.6/1000 PYs [95% CI, 4.1-5.1] and 3.5/1000 PYs [95% CI, 3.4-3.7]; IRR, 1.30 [95% CI, 1.15-1.47]). Among the patients with PsA, IRs were higher for those who received PsA treatments for both outcomes but did not differ significantly by type of treatment received. CONCLUSIONS: The rates of CVD and MACE were slightly higher in the patients with PsA compared to the patients without PsA. Among the patients with PsA, rates of both outcomes were higher among those who received prescriptions for systemic PsA treatments.

We demonstrate that balanced optical-microwave phase detectors (BOMPD) are capable of optical-RF synchronization with sub-femtosecond residual timing jitter for large-scale timing distribution systems. RF-to-optical synchronization is achieved with a long-term stability of < 1 fs RMS and < 7 fs pk-pk drift for over 10 hours and short-term stability of < 2 fs RMS jitter integrated from 1 Hz to 200 kHz as well as optical-to-RF synchronization with 0.5 fs RMS jitter integrated from 1 Hz to 20 kHz. Moreover, we achieve a -161 dBc/Hz noise floor that integrates well into the sub-fs regime and measure a nominal 50-dB AM-PM suppression ratio with potential improvement via DC offset adjustment.

Long-term stable timing distribution over a 3.5-km polarization maintaining (PM) fiber link using balanced optical cross-correlators (BOC) for optical-to-optical synchronization is demonstrated. Remote laser synchronization over 40 hours showed a residual timing jitter and drift of 2.5 fs for the whole locking period and only 1.1 fs integrated from 100 µHz to 1 MHz. This result corresponds to the lowest jitter and drift achieved to date for a multi-km fiber link and remote timing synchronization operating continuously over multiple days.

The overall objective of this study was to construct a 3D neuroanatomical map of the saphenous nerve based on cartesian coordinate data to define its course in 3D space relative to bony and soft tissue landmarks. Ten lower limb embalmed specimens were meticulously dissected, digitized, laser scanned, and modelled in 3D. The course of the main branches, number of collateral branches, and relationship of saphenous nerve to the great saphenous vein were defined and quantified using the high-fidelity 3D models. In 60% of specimens, the saphenous nerve was found to have three branches in the leg, infrapatellar, anterior, and posterior. In 40% of specimens, the posterior branch was absent. Three landmarks were found to consistently localize the anterior branch: the medial border of tibia at the level of the tibial tuberosity, the medial border of tibia at the level of the mid-point of leg, and the mid-point of the anterior border of the medial malleolus. The posterior branch, when present, had variable branching patterns but did not extend as far distally as the medial malleolus in any specimen. Anatomically, the anterior and posterior branches at the level of the tibial tuberosity could be most advantageous for nerve stimulation due to their close proximity to the bifurcation of the saphenous nerve where the branches are larger and more readily localizable than distally. Additionally, the tibial tuberosity is a prominent landmark that can be easily identified in most individuals and could be used to localize the anterior and posterior branch using ultrasound or other imaging modalities. These findings will enable implementation of highly realistic computational models that can be used to simulate saphenous nerve stimulation using percutaneous and implanted devices.

Long-term stable, sub-femtosecond timing distribution over a 1.2-km polarization-maintaining (PM) fiber-optic link using balanced optical cross-correlators for link stabilization is demonstrated. Novel dispersion-compensating PM fiber was developed to construct a dispersion-slope-compensated PM link, which eliminated slow timing drifts and jumps previously induced by polarization mode dispersion in standard single-mode fiber. Numerical simulations of nonlinear pulse propagation in the fiber link confirmed potential sub-100-as timing stability for pulse energies below 70 pJ. Link operation for 16 days showed ~0.6 fs RMS timing drift and during a 3-day interval only ~0.13 fs drift, which corresponds to a stability level of 10(-21).

With the impact of COVID-19, many university students may not be able to learn English in the physical classroom in a traditional way. Students' English learning effectiveness and outcome were threatened when English learning was forced to turn online. Thus, a variety of technological media and platforms to improve their learning outcomes are in need. Mobile learning (M-learning) that involves interacting with other devices through mobile devices and wireless networks can also be a solution to improve students' online English learning effectiveness. In order to explore the learning behaviors and attitudes of university students when learning English with M-learning, this study integrated technology acceptance model and Stimulus Organism Response model including the concepts of perceived convenience, curiosity and self-efficacy in addition to the original technology acceptance model to verify university students' usage cognition and attitude toward English M-learning. This study disseminated surveys to 10 targeted universities/colleges and collected 1432 valid surveys. This study implemented Smart-PLS 4.0 to examine structural model and verify the hypotheses. Results indicated that perceived convenience have positive impact on perceived ease of use, perceived usefulness and attitude toward using; there is a significant and positive relationship among perceived ease of use, perceived usefulness, attitude toward using and intention to using; curiosity and self-efficacy have positive impact on intention to using. Based on the findings, this study further provides abundant theoretical insights and practical significance on language learning.

After the COVID-19 outbreak, this study examines the influence of modifications in China's Sustainable Growth Goals (SDGs) and economic development goals on Chinese enterprises' energy conservation and emissions reduction behavior. Meanwhile, the COVID-19 epidemic has erupted, displacing the flimsy traditional techniques. As a result, the post-COVID-19 pandemic emphasizes the need for a long-term sustainable development method compatible with the local and regional environmental systems. The main objective of this study is used as a roadmap to steer the post-COVID-19 pandemic on a sustainable green path by emphasizing sustainable energy strategies to connect in SDG-related efforts. The investigation in this paper begins with examining significant impacts in the energy industry and their impact on progress toward sustainability. The empirical findings that the CO2 emissions reduction objectives in long-term development plans had a considerable impact on energy saving and emissions reduction, lowering energy consumption intensity by 3.33% and carbon emission intensity by 4.23% between 2010 and 2019. Besides, the results and long and short run techniques are built to describe the Sustainable Development Goals interface, with the result revealing that Sustainable Development Goals enhance the green economic recovery performance. Furthermore, this study recommends that the key natural resources and green economic recovery policies to overcome the climate change impacts by COVID-19 pandemic.

Accurate conversion of wideband multi-GHz analog signals into the digital domain has long been a target of analog-to-digital converter (ADC) developers, driven by applications in radar systems, software radio, medical imaging, and communication systems. Aperture jitter has been a major bottleneck on the way towards higher speeds and better accuracy. Photonic ADCs, which perform sampling using ultra-stable optical pulse trains generated by mode-locked lasers, have been investigated for many years as a promising approach to overcome the jitter problem and bring ADC performance to new levels. This work demonstrates that the photonic approach can deliver on its promise by digitizing a 41 GHz signal with 7.0 effective bits using a photonic ADC built from discrete components. This accuracy corresponds to a timing jitter of 15 fs - a 4-5 times improvement over the performance of the best electronic ADCs which exist today. On the way towards an integrated photonic ADC, a silicon photonic chip with core photonic components was fabricated and used to digitize a 10 GHz signal with 3.5 effective bits. In these experiments, two wavelength channels were implemented, providing the overall sampling rate of 2.1 GSa/s. To show that photonic ADCs with larger channel counts are possible, a dual 20-channel silicon filter bank has been demonstrated.

Quarantine policies introduced in the context of COVID-19 are affecting business operations and slowing down the flow rate of the overall economy. Different degrees and types of threats occur in both the living environment and the working environment during the epidemic prevention, which causes many additional uncertainties. The impact on employees is the identity threat from environment and organizations. This is different from the related research on the identification and impact of the threat before the occurrence of COVID-19. However, in the post-pandemic period, companies continue to strengthen important factors that can increase innovation and recovery, including the role of employee knowledge sharing. The organizational inequity and lack of organizational justice bring about the threat of internal identification in organizations. In order to ensure their own interests in organizations, employees may think twice when sharing knowledge. Therefore, this study explores the relationship among employees' identity threat, social capital and knowledge sharing behavior from the perspective of organizational behavior. In this study, a sample of high-tech employees was conducted, and a total of 434 questionnaires were obtained. The research results show that employees' perception of identity threat has a negative impact on knowledge sharing behavior and positively affects social capital; employees' social capital positively affects their knowledge sharing behaviors; and organizational reward system moderates the relationship between identity threat and knowledge sharing behavior. Based on the comprehensive research findings, this research proposes corresponding theoretical and practical implications.

The use of multiple media kiosks (MMKs) is witnessing an increasingly strong uptrend in 24-h chain convenient stores in developed countries. However, as the functions of MMKs intensifies and increases, how to retain consumers has been a topic that raises concerns of managers and researchers. In this study, we adopt the integrated technology acceptance model, which combines technology readiness and acceptance model with interactivity that serves as the moderating factor, with the purpose of discussing the relationships among all these variables and their impacts on behavior intention to use. Through the cross-sectional survey and purposive sampling, a total of 623 copies of questionnaire from Taiwan were collected in this study. Smart-PLS for PLS-SEM was applied in the structural model to conduct a verification of the hypotheses and comparative analysis in this study. The results show that all hypotheses were significantly supported; technology readiness has a positive impact on perceived ease to use and perceived usefulness; and interactivity also positively moderates the relationship among perceived ease to use, perceived usefulness and attitude toward using. Our results will offer more insights and advice concerning theories of service technology.
