RESUMO

The neurotransmitter dopamine (DA) is a key regulatory component of executive functioning and dysfunction in dopaminergic circuity has been shown to result in impaired working memory. Studies have identified multiple common genetic variants suggested to functionally impact the DA system and behaviorally alter working memory performance. Here, we aimed to develop a predictive model of affective working memory and to examine whether specific combinations of polymorphisms differently influence later encoding processes in affective working memory. Specifically, we examined the effects of the dopamine D2 and D1 receptors and Catechol-O-methyltransferase (COMT), on affective working memory in 155 older adults. Our model identified genotype variants, and scores on the Mini-Mental State exam and Geriatric depression scales as significant influencers in the predictive model whereas behavioral results showed specific patterns of performance linked to valence and string length but not to specific genetic variants. That is, all participants remembered a more positive words compared to negative and neutral words when remembering short strings of 3 or 4 words whereas performance on long strings, 5 or 6 words, revealed a more general affective enhancement independent of genotype. These findings are some of the first to investigate the effects dopaminergic enzyme and receptor interactions on affective working memory.

The female reproductive system represents a sensitive target of the harmful effects of cigarette smoke, with folliculogenesis as one of the ovarian processes most affected by this exposure. The aim of this study was to analyze the impact of tobacco smoking on expression of oxidative stress-related genes in cumulus cells (CCs) from smoking and non-smoking women undergoing IVF techniques. Real time PCR technology was used to analyze the gene expression profile of 88 oxidative stress genes enclosed in a 96-well plate array. Statistical significance was assessed by one-way ANOVA. The biological functions and networks/pathways of modulated genes were evidenced by ingenuity pathway analysis software. Promoter methylation analysis was performed by pyrosequencing. Our results showed a down-regulation of 24 genes and an up-regulation of 2 genes (IL6 and SOD2, respectively) involved in defense against oxidative damage, cell cycle regulation, as well as inflammation in CCs from smoking women. IL-6 lower promoter methylation was found in CCs of the smokers group. In conclusion, the disclosed overall downregulation suggests an oxidant-antioxidant imbalance in CCs triggered by cigarette smoking exposure. This evidence adds a piece to the puzzle of the molecular basis of female reproduction and could help underlay the importance of antioxidant treatments for smoking women undergoing IVF protocols.

The COVID-19 pandemic has shaken not only the global economy but every development field, including all levels of the education sector and in every place in the world. The wide spread of this pandemic disaster has undoubtedly changed the education landscape worldwide. Online teaching and learning become the primary instruction method and the global world of schools, colleges and universities were forced to adapt this model. The first concern about online learning is whether this method is effective compared to traditional face-to-face lessons. In this paper, we carried out a quantitative analysis to explore variations in university students' feedback on learning experience in the context of this new challenging situation caused by the COVID-19 pandemic. By adopting an IRT modeling, we compared the appreciation of some aspects of 41 courses taught at the University of Chieti-Pescara (Italy) during the educational emergency with that of the previous year. Overall, from the results of this study, it is arisen that students have given positive feedbacks on their learning experiences and their effectiveness.

Faced with novel coronavirus outbreak, the most hard-hit countries adopted a lockdown strategy to contrast the spread of virus. Many studies have already documented that the COVID-19 control actions have resulted in improved air quality locally and around the world. Following these lines of research, we focus on air quality changes in the urban territory of Chieti-Pescara (Central Italy), identified as an area of criticality in terms of air pollution. Concentrations of NO 2 , PM 10 , PM 2.5 and benzene are used to evaluate air pollution changes in this Region. Data were measured by several monitoring stations over two specific periods: from 1st February to 10 th March 2020 (before lockdown period) and from 11st March 2020 to 18 th April 2020 (during lockdown period). The impact of lockdown on air quality is assessed through functional data analysis. Our work makes an important contribution to the analysis of variance for functional data (FANOVA). Specifically, a novel approach based on multivariate functional principal component analysis is introduced to tackle the multivariate FANOVA problem for independent measures, which is reduced to test multivariate homogeneity on the vectors of the most explicative principal components scores. Results of the present study suggest that the level of each pollutant changed during the confinement. Additionally, the differences in the mean functions of all pollutants according to the location and type of monitoring stations (background vs traffic), are ascribable to the PM 10 and benzene concentrations for pre-lockdown and during-lockdown tenure, respectively. FANOVA has proven to be beneficial to monitoring the evolution of air quality in both periods of time. This can help environmental protection agencies in drawing a more holistic picture of air quality status in the area of interest.

Cigarette smoking among women of reproductive age is known to take a toll on systemic health and fertility potential by severely impacting ovarian tissues and cells, such as granulosa and cumulus cells (CCs). The purpose of this study was to determine the potential damage caused by tobacco smoke at a molecular level in the CCs of females who had undergone in vitro fertilization. The level of intracellular damage was determined by estimating the average telomere length (TL) and mitochondrial DNA copy number (mtDNA-CN), as well as the expression profile of telomere maintenance genes TERF1, TERF2, POT1 and microRNAs miR-155, miR-23a and miR-185. Western blotting analysis was performed to detect consequent protein levels of TERF1, TERF2 and POT1. Our results evidenced significantly lower relative TL and mtDNA-CN and a down-regulation pattern for all three described genes and corresponding proteins in the CCs of smokers compared with controls (p < 0.05). No significant differences were found in the miRNAs' modulation. Combined, our data add another piece to the puzzle of the complex regulatory molecular networks controlling the general effects of tobacco smoke in CCs. This pilot study extends the until now modest number of studies simultaneously investigating the mtDNA-CN and TL pathways in the human CCs of smoking women.

COVID-19 pandemic has hit people's health, economy, and society worldwide. Great confidence in returning to normality has been placed in the vaccination campaign. The knowledge of individual immune profiles and the time required to achieve immunological protection is crucial to choose the best vaccination strategy. We compared anti-S1 antibody levels produced over time by BNT162b2 and AZD1222 vaccines and evaluated the induction of antigen-specific T-cells. A total of 2569 anti-SARS-CoV-2 IgG determination on dried blood spot samples were carried out, firstly in a cohort of 1181 individuals at random time-points, and subsequently, in an independent cohort of 88 vaccinated subjects, up to the seventeenth week from the first dose administration. Spike-specific T-cells were analysed in seronegative subjects between the two doses. AZD1222 induced lower anti-S1 IgG levels as compared to BNT162b2. Moreover, 40% of AZD1222 vaccinated subjects and 3% of BNT162b2 individuals resulted in seronegative during all the time-points, between the two doses. All these subjects developed antigen-specific T cells, already after the first dose. These results suggest that this test represents an excellent tool for a wide sero-surveillance. Both vaccines induce a favourable immune profile guaranteeing efficacy against severe adverse effects of SARS-CoV-2 infection, already after the first dose administration.

Many studies have found that memory for affective material is better than memory for neutral information and memory for positive material compared to negative material is better in older adults. Behavioral, neurophysiological as well as single polymorphism differences have been advanced to account for these effects. Here, we aimed to examine whether the combination of two polymorphisms (ADRA2B and CB1) in older adults influences active maintenance and manipulation of emotional information in aging working memory. We examined genotype data from 207 older adults (56 double deletion carriers, 116 single deletion carriers and 35 no deletion carriers) who performed a verbal operation span-like task with positive, negative and neutral words. We found that subjects carrying both ADRA2B and CB1 variants generally remembered a higher number of words. In addition, double carriers showed positivity effects while single carriers showed more general emotional enhancement effects, especially as strings lengthened. These findings are amongst the first to suggest a haplotype account of positivity effects in older adults' memory.

The storage of surplus oocytes by cryopreservation (OC) is a widely used tool in assisted reproductive technology, but there is a great debate about the effects of cryopreservation on oocyte competence. It is known that OC may affect meiotic spindles but remains unclear if OC may increase the risk of aneuploidy. The aim of this study was to test the effects of OC and women aging on the expression of cytokinesis-related genes playing an important role in the segregation of chromosomes (DCTN1, DCTN2, DCTN3, DCTN6 and PLK1). Results highlighted that OC do not modify the expression of the selected genes, whereas women aging modulate the expression of all transcripts, confirming that aging is the crucial factor affecting meiosis and aneuploidy risk. A new role for Dynactin and PLK1 was shed in light, providing information on the ageing process in the oocyte which may be associated to reduced fertility.

Albeit the dominant source of radon in indoor environments is the geology of the territory, many studies have demonstrated that indoor radon concentrations also depend on dwelling-specific characteristics. Following a stepwise analysis, in this study we propose a combined approach to delineate radon prone areas. We first investigate the impact of various building covariates on indoor radon concentrations. To achieve a more complete picture of this association, we exploit the flexible formulation of a Bayesian spatial quantile regression, which is also equipped with parameters that controls the spatial dependence across data. The quantitative knowledge of the influence of each significant building-specific factor on the measured radon levels is employed to predict the radon concentrations that would have been found if the sampled buildings had possessed standard characteristics. Those normalised radon measures should reflect the geogenic radon potential of the underlying ground, which is a quantity directly related to the geological environment. The second stage of the analysis is aimed at identifying radon prone areas, and to this end, we adopt a Bayesian model for spatial cluster detection using as reference unit the building with standard characteristics. The case study is based on a data set of more than 2000 indoor radon measures, available for the Abruzzo region (Central Italy) and collected by the Agency of Environmental Protection of Abruzzo, during several indoor radon monitoring surveys.
