RESUMO

The presence of numerous chemical contaminants from industrial, agricultural, and pharmaceutical sources in water supplies poses a potential risk to human and ecological health. Current chemical analyses suffer from limitations, including chemical coverage and high cost, and broad-coverage in vitro assays such as transcriptomics may further improve water quality monitoring by assessing a large range of possible effects. Here, we used high-throughput transcriptomics to assess the activity induced by field-derived water extracts in MCF7 breast carcinoma cells. Wastewater and surface water extracts induced the largest changes in expression among cell proliferation-related genes and neurological, estrogenic, and antibiotic pathways, whereas drinking and reclaimed water extracts that underwent advanced treatment showed substantially reduced bioactivity on both gene and pathway levels. Importantly, reclaimed water extracts induced fewer changes in gene expression than laboratory blanks, which reinforces previous conclusions based on targeted assays and improves confidence in bioassay-based monitoring of water quality.

RESUMO

Antimicrobial resistance (AMR) is a grand societal challenge with important dimensions in the water environment that contribute to its evolution and spread. Environmental monitoring could provide vital information for mitigating the spread of AMR; this includes assessing antibiotic resistance genes (ARGs) circulating among human populations, identifying key hotspots for evolution and dissemination of resistance, informing epidemiological and human health risk assessment models, and quantifying removal efficiencies by domestic wastewater infrastructure. However, standardized methods for monitoring AMR in the water environment will be vital to producing the comparable data sets needed to address such questions. Here we sought to establish scientific consensus on a framework for such standardization, evaluating the state of the science and practice of AMR monitoring of wastewater, recycled water, and surface water, through a literature review, survey, and workshop leveraging the expertise of academic, governmental, consulting, and water utility professionals.

Reduced availability of agricultural water has spurred increased interest in using recycled irrigation water for U.S. food crop production. However, there are significant knowledge gaps concerning the microbiological quality of these water sources. To address these gaps, we used 16S rRNA gene and metagenomic sequencing to characterize taxonomic and functional variations (e.g., antimicrobial resistance) in bacterial communities across diverse recycled and surface water irrigation sources. We collected 1 L water samples (n = 410) between 2016 and 2018 from the Mid-Atlantic (12 sites) and Southwest (10 sites) U.S. Samples were filtered, and DNA was extracted. The V3-V4 regions of the 16S rRNA gene were then PCR amplified and sequenced. Metagenomic sequencing was also performed to characterize antibiotic, metal, and biocide resistance genes. Bacterial alpha and beta diversities were significantly different (p < 0.001) across water types and seasons. Pathogenic bacteria, such as Salmonella enterica, Staphylococcus aureus, and Aeromonas hydrophilia were observed across sample types. The most common antibiotic resistance genes identified coded against macrolides/lincosamides/streptogramins, aminoglycosides, rifampin and elfamycins, and their read counts fluctuated across seasons. We also observed multi-metal and multi-biocide resistance across all water types. To our knowledge, this is the most comprehensive longitudinal study to date of U.S. recycled water and surface water used for irrigation. Our findings improve understanding of the potential differences in the risk of exposure to bacterial pathogens and antibiotic resistance genes originating from diverse irrigation water sources across seasons and U.S. regions.

There is a growing consensus that recycled water, as an alternative and renewable water source, can serve as a vital water supply to alleviate water scarcity problem and in support of water resilience. Accordingly, recycled water infrastructure investment has seen a significant growth in recent years in many regions of the world. However, previous studies found the perceptions of public, the main end user, toward using recycled water for potable or non-potable purposes remain negatively stereotyped. The negative stereotypes led to public rejections to the construction and operation of recycled water infrastructure. Traditionally, public perceptions of recycled water uses are captured through self-reporting interview or survey techniques. To gain a more accurate measurement of the implicit public stereotypes toward recycled water uses, this study employed an event-related potential (ERPs) technique to collect neurophysiological responses with participants and presented a few research findings. Firstly, the negative stereotypes of recycled water still exist. Secondly, the degree of human contact impacts the negative stereotypes of participants toward recycled water uses more significantly on the supply side (referring to the whole supply chain of recycled water) rather than on the demand side (referring to the potential consumers of recycled water) Third, knowledge level significantly impacts the negative stereotypes of participants toward recycled water uses that have close human contact, at both supply and demand sides, and shows a more significant impact on the supply side. The findings of study contributed to the literature through creatively dividing the negative stereotypes of recycled water into the "supply-side" and the "demand-side" ones, and meanwhile have managerial implication for policymaking and scheme implementation in the area.

Treating and reusing greywater for agricultural irrigation is becoming increasingly prevalent in water-scarce regions such as the Middle East. However, the potential for antibiotic-resistant bacteria to be introduced into food systems or the environment via greywater reuse is a potential area of concern. It is known that off-grid treated greywater often has elevated levels of bacteria, however, little is known regarding the prevalence of antibiotic-resistant bacteria in this water source. To address this knowledge gap, samples (n = 61) of off-grid, household greywater (influent), treated greywater effluent, and irrigation pond water were collected between October 2017 and June 2018 from four farms in the West Bank, Palestinian Territories. Samples were tested for pH, turbidity, dissolved oxygen, electrical conductivity, and oxidation reduction potential. Standard membrane filtration was used to enumerate presumptive Escherichia coli, and isolates (n = 88) were purified, confirmed using 16S rRNA sequencing, and subjected to antimicrobial susceptibility testing using microbroth dilution. The majority of influent (76.5%) and effluent (70.6%) samples had detectable presumptive E. coli. Interestingly, the majority of the isolates were confirmed as Klebsiella sp. (n = 37), followed by E. coli (n = 32), and the remainder were classified as other Enterobacteriaceae (n = 19). A higher percentage of effluent isolates were fully susceptible to all tested antibiotics when compared to influent isolates (28.6% vs 18.6%). Resistance was most commonly observed against ampicillin (69.3% of all isolates), trimethoprim-sulfamethoxazole (11.4%), tetracycline (9.1%), and cefazolin (7.9%), and 7.9% of isolates were observed to be multidrug-resistant. While most water quality parameters were within Israeli and Palestinian wastewater reuse requirements, E. coli levels in effluent violated available standards. These findings suggest that, despite observed decreases in bacteria and an overall decrease in isolates expressing antibiotic resistance from influent to effluent, off-grid greywater treatment systems are still a potential source of both susceptible and antibiotic-resistant bacteria in the agricultural environment.

Reliable clean drinking water is becoming increasingly scarce. One potential additional source of drinking water is recycled water. However, public acceptance of potable recycled drinking water is low. One likely factor involved in the acceptance of recycled drinking water is objective knowledge about recycled water. In three studies (N = 229, 590, and 200), we developed a 34-item measure of objective knowledge of recycled drinking water. The objective knowledge measure was often a strong and unique predictor of intentions to accept and use recycled drinking water compared to other prominent factors including disgust and subjective knowledge of recycled water. Measuring knowledge of recycled drinking water holds the potential to estimate how and whether educational interventions aimed at increasing acceptance of recycled drinking water programs work.

AIMS: To investigate the presence of noroviruses in recycled water and sewage sludge obtained from a wastewater treatment plant in Bangkok Metropolitan Region, Thailand. METHODS AND RESULTS: Twenty-seven recycled water and twenty-three sewage sludge samples were tested for the presence of norovirus genogroup (G)I and GII using RT-nested PCR. Molecular characterization of noroviruses was undertaken by DNA sequencing and phylogenetic analysis. The level of the RNA genome of the noroviruses was determined using quantitative real-time RT-PCR. Noroviruses were detected in 44·4% of recycled water samples and 73·9% of sewage sludge samples. Norovirus GI.2 and GII.4 were identified in recycled water samples at levels of 2·19 × 101 and 3·26 × 104 RNA copies per litre. Six different genotypes of GI (GI.1, GI.2, GI.5a, GI.5b, GI.6b and GI.7) and GII.17 were identified in sewage sludge samples at levels ranging from 1·99 × 101 -1·43 × 105 RNA copies per gram wet weight. Four recombinant norovirus strains were detected in sewage sludge samples, namely GII.P16-GII.2, GII.P16-GII.4, GII.P16-GII.13 and GII.P21-GII.13. CONCLUSIONS: These findings provide evidence that noroviruses may be spread to the community and environment via the use of recycled water for plant areas, and sewage sludge for land application. SIGNIFICANCE AND IMPACT OF THE STUDY: This is the first study demonstrating recombinant norovirus strains in sewage sludge samples. The presence of noroviruses in recycled water and sewage sludge contributes to a health risk of environmental exposure.

Reclaimed water is often presented as a cost-effective, reliable, and safe solution to increasingly common water shortages in the United States and across the globe, but studies have shown that consumers tend to object to the use of this water. Broad adoption of this technology will require consumer acceptance or at least tolerance of it, and studies have suggested that better branding could minimize consumers' concerns. In this study, we first test twenty-one potential branding names for reclaimed water using survey responses to identify the top-six most favored names. We then determine whether an opportunity for consumers to try reclaimed water can change their preferences. The results suggest that the common names for this water, such as Recycled, Reclaimed, Nontraditional, Treated Wastewater, and Reused, are the least appealing, as they all scored at the bottom. In contrast, names that invoke desirable characteristics of the water-Pure, Eco-Friendly, and Advanced Purified, were viewed significantly more favorable than the others. Having an opportunity to taste reclaimed water treated to a potable standard seems to clarify consumers' preferences by increasing the differences in favorability between the names. Based on these results, it appears that while there are a couple of appealing names, the most consistently preferred is Pure Water.

RESUMO

Climate change and population growth are contributing to a growing global freshwater crisis that is exacerbating agricultural water scarcity and compromising food security and public health. In light of these challenges, the increased reliance on nontraditional irrigation water sources, such as reclaimed or recycled water, is emerging as a potentially viable strategy to address water and food insecurity worldwide. This editorial provides an overview of the global need for agricultural water reuse and outlines the environmental and public health impacts associated with this practice. Policy implications, including an emphasis on "One Water" approaches, are discussed. Finally, the editorial leads off a Special Issue that includes a collection of articles reporting on the initial research and extension/outreach findings of CONSERVE: A Center of Excellence at the Nexus of Sustainable Water Reuse, Food and Health, funded by the U.S. Department of Agriculture, National Institute of Food and Agriculture. Taken together, this compilation of articles addresses the overarching theme that transdisciplinary teams are key with regard to moving the science of agricultural water reuse forward to achieve water and food security and advance public health in a changing climate.

The use of nontraditional water sources, including reclaimed or recycled water, has become a desirable option to meet increasing demands in water stressed regions. In the Southwest United States, utilization of alternative water sources is becoming increasingly common, including use for landscape irrigation, environmental enhancement, cooling and power generation, potable reuse, and as a source water for agricultural irrigation. While much research has gone into identifying public perception towards water reuse schemes, little attention has been given to understanding grower attitudes, perceptions, and knowledge on the use of nontraditional water, including reclaimed water, in agriculture and how that may influence grower acceptance and production practices. This unique study utilized a needs assessment survey of growers (n = 521) within the Southwest region of the United States to gain an understanding of industry attitudes and needs regarding nontraditional water in agriculture. Results indicate that the majority of survey respondents were concerned with water availability (67.49%) yet less than half (48.30%) thought using a nontraditional water source in agriculture was 'very important'. Interestingly, respondents rated irrigation of 'food crops' third (42.20%) among agricultural activities for which they would be willing to use nontraditional water sources, behind irrigation of forage crops (61.60%) and dust control (61.60%). The importance of the use of nontraditional water sources in agriculture was influenced mostly by farm size (p = 0.007) and primary water source (p = 0.016), and the level of education was significant in respondent's level of concern over water availability (p = 0.021). Information on the quality of nontraditional water sources, showing that it is as good or better than respondents current sources, was found to shift rejection and uncertainty towards acceptance by 16.04%. The results of this study provide insight into perceived risks, willingness to use, drivers and constraints to grower adoption, and preferred methods of education regarding water reuse in agriculture. These findings can be used by water managers and planners to aid in the adoption of nontraditional waters, including reclaimed or recycled water, in agriculture thus extending water resources, securing food supplies, and protecting public health.

Questions related to the safety of alternative water sources, such as recycled water or reclaimed water (including grey water, produced water, return flows, and recycled wastewater), for produce production have been largely un-explored at the detail warranted for protection of public health. Additionally, recent outbreaks of Escherichia coli (E. coli) in fresh produce, in which agricultural water was suspected as the source, coupled with heightened media coverage, have elevated fruit and vegetable safety into the forefront of public attention. Exacerbating these concerns, new Federal regulations released by the U.S. Food and Drug Administration (FDA) as part of implementation of the FDA Food Safety Modernization Act (FSMA), require testing of agricultural water quality for generic E. coli. Here, we present a review of water quality criteria - including surface water, groundwater recreational water, and water reuse - in an attempt to better understand implications of new FDA regulations on irrigated produce. In addition, a Quantitative Microbial Risk Assessment (QMRA) was conducted to estimate risks from pathogen contamination of food crops eaten fresh under the context of FDA regulations to provide perspective on current water reuse regulations across the country. Results indicate that irrigation water containing 126 CFU/100 mL of E. coli correspond to a risk of GI illness (diarrhea) of 9 cases in 100,000,000 persons (a 0.000009% risk) for subsurface irrigation, 1.1 cases in 100,000 persons (a 0.0011% risk) for furrow irrigation, and 1.1 cases in 1000 persons (a 0.11% risk) for sprinkler irrigation of lettuce. In comparison to metrics in states that currently regulate the use of recycled water for irrigation of food crops eaten fresh, the FDA FSMA water quality metrics are less stringent and therefore the use of recycled water presents a reduced risk to consumers than the FDA regulations. These findings, while limited to a one-time exposure event of lettuce irrigated with water meeting FSMA water quality regulations, highlight the need for additional assessments to determine if the scientific-basis of the regulation is protective of public health.

The water-energy nexus (WEN) is dynamic and complicated in megacities, most of which are challenged by water scarcity and the mandate to reduce energy consumption. A salient feature of water and energy services in megacities is that they are supported by a web of regional infrastructure, extending far beyond the geographic boundaries of the cities, resulting in a strong dependence on resources imported from outside. Understanding the WEN of megacities has implications not only for more efficient resource utilization but also for synergistic regional development and corporation. This study provides a quantitative assessment of the WEN of Beijing within and beyond its geographic boundaries. Results show that water for local internal energy production and transformation accounts for 220 million m3/year, or 5.6% of its total freshwater use in 2016, and the energy for local water abstraction, supply, and treatment is 3.06 billion kWh, accounting for 1.1% of its total energy consumption for the same year. The external water for "imported" energy is 290 million m3/year, 1.3 times of Beijing's freshwater use for internal energy. This means that more water for energy is consumed outside Beijing than that within Beijing. The energy for external water is negligible because the bulk of the water transfer into Beijing relies on gravity and because the energy for construction of the transfer infrastructure is not included. Analysis of the WEN revealed the contradiction between the two independently conceived policies of Beijing: to meet the "three-red-line" target in the water sector, recycled water and transferred water use will rise, making it more difficult to meet the carbon emission control targets. Therefore, adopting low energy intensity, nature-based water recycling is a wise policy choice.

This study investigated people's preferences for different water sources and factors that predict such preferences using a blind taste test. Water preferences of 143 participants for one name-brand bottled water, one groundwater-sourced tap water, and one indirect potable reuse (IDR) water were assessed. For predictors of water preference, we measured each participant's PTC taste sensitivity and assessed two personality traits (Neuroticism, Openness to Experience). We also explored participants' descriptions of each water source. Results indicate a preference for water treated with Reverse Osmosis (RO) (bottled and IDR water) over groundwater-sourced water, which had higher pH levels and lower concentrations of Ca and HCO3-. PTC taste sensitivity did not predict preferences, while Openness to Experience and Neuroticism predicted preference for IDR water. Positive relations between Openness to Experience and preferences for bottled and IDR water were moderated by gender and were stronger among females. Participants described water primarily by its taste and texture. Findings suggest that (1) tap water treated by RO is equally preferable to some bottled water, (2) personality traits may affect water preferences, and (3) prior findings of gender differences in preferences for bottled water may reflect personality characteristics. Efforts to increase acceptance for sustainable water alternatives, such as IDR, may be more successful by assuring consumers about taste and addressing personality traits that encourage or inhibit use.

Greywater considers being a highly reclaimable water source particularly important for water-stressed nations. In this work, heterogeneous photocatalysis using artificial and solar illumination has been applied for the mineralization of simulated light greywater (effluents from dishwashers and kitchen sinks were excluded from the study). The effects on the process' efficiency of TiO2 P25 catalyst's concentration, initial concentration of H2O2 and Fe3+, pH of the solution, as well as the type of radiation, were evaluated in a bench-scale Pyrex reactor and a pilot-scale slurry fountain photoreactor. The treatment efficiency has been followed through the evolution of the organic matter content expresses as dissolved organic carbon (DOC). Best results were obtained with the photo-Fenton-assisted TiO2 photocatalytic process with 72% DOC removal after 210 min of bench scale treatment, while under the same photocatalytic conditions in the pilot reactor the DOC removal reached almost 64%. Moreover, the decrease in toxicity, phytotoxicity and biodegradability of the simulated wastewater has been observed after solar-induced photocatalytic treatment.

This paper has considered risk management, financial evaluation and funding in seven Australian wastewater and stormwater reuse projects. From the investigated case studies it can be seen that responsible parties have generally been well equipped to identify potential risks. In relation to financial evaluation methods some serious discrepancies, such as time periods for analysis, and how stormwater benefits are valued, have been identified. Most of the projects have required external, often National Government, funding to proceed. As National funding is likely to become less common in the future, future reuse projects may need to be funded internally by the water industry. In order to enable this the authors propose that the industry requires (1) a standard project evaluation process, and (2) an infrastructure funders' forum (or committee) with representation from both utilities and regulators, in order to compare and prioritise future reuse projects against each other.

Quantitative microbial risk assessment was used to assess the risk of norovirus gastroenteritis associated with consumption of raw vegetables irrigated with highly treated municipal wastewater, using Melbourne, Australia as an example. In the absence of local norovirus concentrations, three methods were developed: (1) published concentrations of norovirus in raw sewage, (2) an epidemiological method using Melbourne prevalence of norovirus, and (3) an adjustment of method 1 to account for prevalence of norovirus. The methods produced highly variable results with estimates of norovirus concentrations in raw sewage ranging from 10(4) per milliliter to 10(7) per milliliter and treated effluent from 1 × 10(-3) per milliliter to 3 per milliliter (95th percentiles). Annual disease burden was very low using method 1, from 4 to 5 log10 disability adjusted life years (DALYs) below the 10(-6) threshold (0.005-0.1 illnesses per year). Results of method 2 were higher, with some scenarios exceeding the threshold by up to 2 log10 DALYs (up to 95,000 illnesses per year). Method 3, thought to be most representative of Melbourne conditions, predicted annual disease burdens >2 log10 DALYs lower than the threshold (∼ 4 additional cases per year). Sensitivity analyses demonstrated that input parameters used to estimate norovirus concentration accounted for much of the model output variability. This model, while constrained by a lack of knowledge of sewage concentrations, used the best available information and sound logic. Results suggest that current wastewater reuse behaviors in Melbourne are unlikely to cause norovirus risks in excess of the annual DALY health target.

Faced with a severe drought, the residents of the regional city of Toowoomba, in South East Queensland, Australia were asked to consider a potable wastewater reuse scheme to supplement drinking water supplies. As public risk perceptions and trust have been shown to be key factors in acceptance of potable reuse projects, this research developed and tested a social-psychological model of trust, risk perceptions and acceptance. Participants (N = 380) were surveyed a few weeks before a referendum was held in which residents voted against the controversial scheme. Analysis using structural equation modelling showed that the more community members perceived that the water authority used fair procedures (e.g., consulting with the community and providing accurate information), the greater their sense of shared identity with the water authority. Shared social identity in turn influenced trust via increased source credibility, that is, perceptions that the water authority is competent and has the community's interest at heart. The findings also support past research showing that higher levels of trust in the water authority were associated with lower perceptions of risk, which in turn were associated with higher levels of acceptance, and vice versa. The findings have a practical application for improving public acceptance of potable recycled water schemes.

Growing concern over the presence of per- and polyfluoroalkyl substances (PFAS) in agricultural compartments (e.g., soil, water, plants, soil fauna) has led to an increased interest in scalable and economically feasible remediation technologies. Biochar is the product of pyrolyzing organic materials (crop waste, wood waste, manures, grasses) and has been used as a low-cost adsorbent to remove contaminants including PFAS. This review frames biochar as a strategy for mitigating the detrimental impacts of PFAS in agricultural systems and discusses the benefits of this strategy within the framework of the needs and challenges of contaminant remediation in agriculture. To gauge the optimal physicochemical characteristics of biochar in terms of PFAS adsorption, principal component analysis using >100 data points from the available literature was performed. The main biochar-based PFAS treatment strategies (water filtration, soil application, mixing with biosolids) were also reviewed to highlight the benefits and complications of each. Life cycle analyses on the use of biochar for contaminant removal were summarized, and data from selected studies were used to calculate (for the first time) the global warming potential and net energy demand of various agriculturally important biochar classes (crop wastes, wood wastes, manures) in relation to their PFAS adsorption performance. This review serves to identify key gaps in our knowledge of (i) PFAS adsorption by biochars in agricultural remediation applications and (ii) environmental costs/benefits of biochars in relation to their adsorptive properties toward PFAS. The concepts introduced in this review may assist in developing large-scale biochar-based PFAS remediation strategies to help protect the agricultural food production environment.

Recycled water contributes to sustainable development by providing a unified approach to wastewater treatment and rational water supply. However, the environmental benefit-sharing feature of recycled water may encourage some residents to show "free-rider" behavior. In this study, the evolutionary game model was applied to a game system consisting of two types of people, environmental preference and risk aversion. We analyzed internal and external factors and conducted a numerical simulation to seek both players' behavior evolutionary and stabilization trends. The study found that interventions are indispensable during recycled water use's initial and intermediate stages. Moreover, interventions should be tailored to different stages. In the initial stage, the benefits of co-use and the recycled water scientific information should be advertised. In the intermediate and mature stages, it is necessary to increase the trust coefficient or knowledge base of decided people who belong to early adopters of the recycled water diffusion system.

Introduction: The development of advanced sewage technologies empowers the industry to produce high-quality recycled water, which greatly influences human's life and health. Thus, this study investigates the mechanism of individuals' adoption of recycled water from the technology adoption perspective. Methods: Employing the mixed method of structural equation modeling and artificial neural network analysis, we examined a research model developed from the extended Unified Theory of Acceptance and Use of Technology (UTAUT2) framework. To examine the research model, this study employs a leading web-survey company (Sojump) to collect 308 valid samples from the residents in mainland China. Results: The structural equation modeling results verified the associations between the six predictors (performance expectancy, effort expectancy, social influence, facilitating conditions, environmental motivation, and price value), individuals' cognitive and emotional attitudes, and acceptance intention. The artificial neural network analysis validates and complements the structural equation modeling results by unveiling the importance rank of the significant determinants of the acceptance decisions. Discussion: The study provides theoretical implications for recycled water research and useful insights for practitioners and policymakers to reduce the environmental hazards of water scarcity.
