ABSTRACT

The cardiotoxic effects of synthetic cathinones remain largely unknown. In this study, we present two cases, a case series and a scoping review, to explore synthetic cathinone associated cardiotoxicity. Case 1 involved a 28-year-old male with non-ST-elevation myocardial infarction after ingesting a substance containing 4-methylmethcathinone (4-MMC), 3-methylmethcathinon (3-MMC), and methcathinone. Case 2 involved a 49-year-old male with ventricular fibrillation after 4-methylmethcathinone ingestion, who was diagnosed with severe three-vessel disease. A retrospective analysis was performed on self-reported synthetic cathinone poisonings reported to the Dutch Poisons Information Centre from 2012 to 2022. A total of 222 mono-intoxications with cardiotoxicity were included, mostly involving 3-methylmethcathinon (63%). Often tachycardia, hypertension, palpitations, and chest pain were reported. A comprehensive literature search was performed on PubMed to identify the studies reporting cardiac arrest, myocardial infarction, cardiac inflammation, cardiomyopathy, and life-threatening arrhythmias following synthetic cathinone use. A total of 30 articles reporting 40 cases were included. The reported complications included cardiac arrest (n = 28), ventricular tachycardia (n = 4), supraventricular tachycardia (n = 1), ST-elevation myocardial infarction (n = 2), non-ST-elevation myocardial infarction (n = 2), cardiomyopathy (n = 1), and myocarditis (n = 2). A total of ten different associated synthetic cathinones were identified. Cardiac arrest, myocardial infarction, and ventricular arrhythmias have been reported following the use of synthetic cathinones, underscoring the importance of obtaining a detailed recreational drug use history from patients presenting with syncope, chest pain, or palpitations.

ABSTRACT

A new method, based on shotgun spectral matching of peptide tandem mass spectra, was successfully applied to the identification of different food species. The method was demonstrated to work on raw as well as processed samples from 16 mammalian and 10 bird species by counting spectral matches to spectral libraries in a reference database with one spectral library per species. A phylogenetic tree could also be constructed directly from the spectra. Nearly all samples could be correctly identified at the species level, and 100% at the genus level. The method does not use any genomic information and unlike targeted methods, no prior knowledge of genetic variation within a genus or species is necessary.

ABSTRACT

Scutellaria agrestis é utilizada por comunidades ribeirinhas do Amazonas principalmente para o tratamento de otites por via tópica utilizando-se o extrato bruto obtido por maceração. O presente trabalho visou investigar preliminarmente o perfil fitoquímico, a segurança toxicológica e as ações analgésica, anti-inflamatória e antiedematogência do extrato aquoso das folhas de S. agrestis. Foram coletados 80 indivíduos da espécie no horto medicinal da Universidade Nilton Lins, Manaus, Brasil. O perfil fitoquímico foi obtido por meio de prospecção da droga vegetal para heterosídeos cianogênicos, terpenos, compostos fenólicos e alcaloides. A toxicologia foi avaliada pelo teste de toxicidade aguda. As atividades analgésicas/ anti-inflamatórias foram analisadas por meio dos testes de formalina em camundongos e a atividade antiedematogência, pelo teste de edema de pata em ratos. Os metabólitos detectados foram fenóis (taninos hidrolisáveis, cumarinas e várias classes de flavonoides) e terpenos (esteroides livres, saponinas). Não foi possível estabelecer DL50, haja visto que o extrato não provocou a morte de nenhum animal durante o teste de toxicidade aguda, provavelmente devido à ausência de heterosídeos cianogênicos na sua composição. Apesar de não provocar morte, considerou-se que o extrato apresenta uma discreta toxicidade, uma vez que foi observada a ocorrência de espasmos na primeira hora de observação dos animais. O extrato apresentou ainda efeito analgésico e anti-inflamatório significativo nas doses de 30, 100 e 300 mg/kg pelo teste da formalina, sendo o resultado na maior dose equivalente ao obtido com a droga padrão (fentanil). No entanto, não observamos efeito antiedematogênico nas doses testadas durante as 5 horas de registro do edema de pata. Os resultados obtidos nesta pesquisa conferem base científica preliminar quanto à segurança e ao efeito analgésico e antiinflamatório da droga vegetal, o que indica que tal espécie é promissora e expressamente recomendada para maiores estudos farmacológicos in vitro e in vivo.


The Scutellaria agrestis is used by Amazonas riverine communities, especially for otitis externa topical treatment, by using the crude extract obtained by maceration. This study aimed to investigate the preliminary phytochemical profile, the safety/toxicity and the analgesic, anti-inflammatory and antiedematogenic activities of the aqueous extract of the S. agrestis leaves. Eighty individuals were collected at the Nilton Lins University medicinal garden, Manaus, Brazil. The phytochemical profile was obtained through a plant drug survey for cyanogenic heterosides, terpenes, alkaloids and phenolic compounds. The extract safety was evaluated by acute toxicity test. Analgesic and anti-inflammatory activities were accessed using formalin test in mice and the antiedematogenic activity, using paw edema test in mice. We detected phenolic (hydrolysable tannins, coumarins and several classes of flavonoids) and terpenoid (free steroids, saponins) metabolites. We could not establish LD50 because no animals died during the acute toxicity test, probably because of the absence of cyanogenic glycosides on the composition of the extract. However, we found that the extract is slightly toxic as animal spasms were observed in the first hour of the test. The extract showed significant analgesic and anti-inflammatory activity on the formalin test (30, 100 and 300 mg/kg p.o.), and the highest dose result was equivalent to the standard drug (Fentanyl). However, no significant antiedematogenic effect was observed during the paw edema test. The results obtained in this study provide preliminary scientific basis about the safety and analgesic/anti-inflammatory actions of the aqueous extract of S. agrestis, which indicates that this species is a promising option for further in vitro and in vivo pharmacological studies.

ABSTRACT

Several cationic antimicrobial oligo-acyl-lysyl (OAK) peptide mimetics can form cochleate structures, that is, elongated multilayered cylindrical structures, with lipid mixtures mimicking the composition of bacterial cytoplasmic membranes. These cochleate structures do not require divalent cations for their assembly. In the present work, we use light microscopy to screen for cochleate formation in several OAK-lipid systems and freeze-fracture electron microscopy to assess their morphological features and size. We identify several factors that facilitate a structural change in these assemblies. Dehydration of the membrane interface and a high melting temperature are features of the lipids that enhance cochleate formation in OAK-based lipid systems. In addition, we observed that there is a specific length of the hydrocarbon linker in the OAK of 8-9 carbon atoms that provides optimal formation of these structures. The biophysical properties established in this study will allow for a better understanding of their role and suitability for biological studies.

Studies on population dynamics and vertical distribution in the soil of Scaptocoris carvalhoi Becker were carried out in a pasture area in Paraúna, State of Goias, Brazil, from November 2004 to October 2005. One meter-deep holes were excavated in the soil, except on the season of lower precipitation, when 2-meter holes were excavated (June, August and September 2005). On each evaluation, the specimens were collected and grouped according to the depth they were found in the soil profile (0-20, 21-40, 41-60, 61-80 and 81-100 cm). Nymphs occurred during the entire length of the study and were more abundant than adults. Adult population augmented with the increase of rainfall. Nymphs and adults were located in the top layer of the soil during the rainy season and were found deeper in the soil during droughts. However, adults were less tolerant to hydric stress and burrowed deeper into the soil at the beginning of the dry season, while nymphs burrowed deeper only at the end of the season. Results of this study are in accordance to other studies on various species of Scaptocoris, in other regions of Brazil.

ABSTRACT

Os estudos de levantamento populacional e distribuição vertical de Scaptocoris carvalhoi Becker foram realizados em área de pastagem de Paraúna, GO, entre os meses de novembro de 2004 e outubro de 2005. Em cada avaliação foram realizadas escavações de 1 m de profundidade, coletando os indivíduos e agrupando-os de acordo com a profundidade em que se localizavam no perfil do solo (0-20, 21-40, 41-60, 61-80 e 81-100 cm). Quando os insetos foram encontrados em quantidades baixas até 1 m, as escavações foram realizadas até 2 m de profundidade (junho, agosto e setembro de 2005). As ninfas foram predominantes durante todo o período. O número de adultos foi dependente das chuvas, constatando-se maior incidência com o incremento da precipitação. Ninfas e adultos mantiveram-se nas camadas superficiais do solo no período chuvoso e aprofundaram-se nos períodos de deficiência hídrica. No entanto, os adultos demonstraram menor tolerância ao estresse hídrico e aprofundaram-se logo no início da época seca, enquanto as ninfas o fizeram somente no final desse período. Os resultados obtidos nesse trabalho são semelhantes aos verificados para outras espécies de Scaptocoris, em outras regiões.
