RESUMEN

The Chinese mitten crab (Eriocheir sinensis) is an important commercial species in China. E. sinensis is typically farmed in rice-crab symbiosis, as an important ecological farming model. However, E. sinensis is often exposed to a high ammonia environment due to the application of nitrogen fertilizers essential for rice growth. We investigated the molecular mechanisms in the gills of E. sinensis exposed to high ammonia at transcriptional and histological levels. We randomly assigned E. sinensis to two groups (control group, CG; ammonia stress group, AG), and gill samples were excised from the CG and AG groups for histopathological and transcriptome analyses. The histopathological evaluation revealed that ammonia stress damaged the gills of E. sinensis. The transcriptome analysis showed that some essential genes, including Xanthine dehydrogenase (XDH), Ubiquitin C-terminal hydrolase-L3 (UCHL3), O-linked N-acetylglucosamine transferase (OGT), Cathepsin B (CTSB), and Ubiquitin-conjugating enzyme E2 W (UBE2W) changed significantly during ammonia exposure. These genes are related to ammonia detoxification, the immune response, and apoptosis. This study demonstrated the molecular response mechanism of E. sinensis gills to ammonia stress at the transcriptional and histological levels. This study provides insight for further study on the molecular mechanism of ammonia stress in crustaceans and supplies technical support for rice crab symbiosis.

OBJECTIVE: To evaluate the dynamics of killing of oral multispecies biofilms grown on dental restorative materials by commercially available mouthrinses and a d-enantiomeric peptide. METHODS: Four composite resins (3 M Supreme, 3 M Supreme flow, Kerr Sonicfill, and Shofu Beautifil II) and one glass ionomer (GC Fuji II) were used as restorative materials. Plaque biofilms were grown on the surfaces of restorative material discs for 1 week. The surface roughness and biofilm attachment were assessed by atomic force microscopy and scanning electron microscopy. One-week-old biofilms grown anaerobically at 37 °C were exposed to each of five solutions for one minute (twice daily for seven days): Listerine Total care and Paroex Gum mouthrinses, 0.12% chlorhexidine, 0.001% d-enantiomeric peptide DJK-5, and sterile water. The dynamic variation of the biovolume of the biofilms and the percentage of dead bacteria were monitored and analyzed using confocal laser scanning microscopy. RESULTS: All restorative materials had similar surface roughness with intact biofilm attachment. The percentage of dead bacteria and biovolume of biofilms treated by each oral rinse solution remained constant between days 1 and 7, with no statistically significant difference. DJK-5 showed the highest percentage of dead bacteria (up to 75.7%; cf. ∼20-40% for other mouthrinses) of all solutions tested within 7 days. CONCLUSIONS: DJK-5 outperformed conventional mouthrinses in killing bacteria in oral multispecies biofilms grown on dental restorative materials. CLINICAL SIGNIFICANCE: The antimicrobial peptide DJK-5 is effective against oral biofilms and serves as a promising candidate for the development of future mouthrinses to improve long-term oral hygiene.

Aging poses a major risk factor for cardiovascular diseases, the leading cause of death in the aged population. However, the cell type-specific changes underlying cardiac aging are far from being clear. Here, we performed single-nucleus RNA-sequencing analysis of left ventricles from young and aged cynomolgus monkeys to define cell composition changes and transcriptomic alterations across different cell types associated with age. We found that aged cardiomyocytes underwent a dramatic loss in cell numbers and profound fluctuations in transcriptional profiles. Via transcription regulatory network analysis, we identified FOXP1, a core transcription factor in organ development, as a key downregulated factor in aged cardiomyocytes, concomitant with the dysregulation of FOXP1 target genes associated with heart function and cardiac diseases. Consistently, the deficiency of FOXP1 led to hypertrophic and senescent phenotypes in human embryonic stem cell-derived cardiomyocytes. Altogether, our findings depict the cellular and molecular landscape of ventricular aging at the single-cell resolution, and identify drivers for primate cardiac aging and potential targets for intervention against cardiac aging and associated diseases.

This study aimed to compare the effect of a bioceramic sealer (iRoot SP) and a resin-based sealer (AH Plus) on the outcome of root canal treatment in a 2-year follow-up. Seventy-six teeth with irreversibly or necrotic pulp were recruited. After instrumentation and disinfection, the root canals were obturated using warm vertical compaction with iRoot SP (n = 43) or AH Plus (n = 33). Patients were followed up by clinical and radiographic examination at 6 12 and 24 months with recall rates of 84.2%, 65.8% and 48.7%, respectively. During each recall session, the success rates were 80%, 85.2% and 85% in the iRoot SP group and 82.8%, 91.3% and 88.2% in the AH Plus group. The success rates of the two groups did not differ significantly (p > 0.05). The bioceramic sealer resulted in a similar clinical performance and success rate to the resin-based sealer in endodontic treatment during a 2-year follow-up.

Agriculture is the primary source of human survival, which provides the most basic living and survival conditions for human beings. As living standards continue to improve, people are also paying more attention to the quality and safety of agricultural products. Therefore, the detection of agricultural product quality is very necessary. In the past decades, the spectroscopy technique has been widely used because of its excellent results in agricultural quality detection. However, traditional spectral inspection methods cannot accurately describe the internal information of agricultural products. With the continuous research and development of optical properties, it has been found that the internal quality of an object can be better reflected by separating the properties of light, such as its absorption and scattering properties. In recent years, spatially resolved spectroscopy has been increasingly used in the field of agricultural product inspection due to its simple compositional structure, low-value cost, ease of operation, efficient detection speed, and outstanding ability to obtain information about agricultural products at different depths. It can also separate optical properties based on the transmission equation of optics, which allows for more accurate detection of the internal quality of agricultural products. This review focuses on the principles of spatially resolved spectroscopy, detection equipment, analytical methods, and specific applications in agricultural quality detection. Additionally, the optical properties methods and direct analysis methods of spatially resolved spectroscopy analysis methods are also reported in this paper.

OBJECTIVES: To determine the effect of the location of the canal curvature on the fatigue resistance of WaveOne (WO), WaveOne Gold (WOG), Reciproc (Rec), and Reciproc Blue (RecB) files, and to examine the phase transformation behaviors of the reciprocating file systems. MATERIAL AND METHODS: The instruments were subjected to fatigue testing in five artificial canals with a curvature of 60° angle and a 3-mm radius. The location of the curvature was unique for each canal. Each file was inserted 16 mm into the canal and operated until fracture occurred. The time to fracture was recorded and the length of the fragment was measured. Differential scanning calorimetry was used to characterize the thermal behavior of the files. The number of cycles to failure was analyzed using two-way analysis of variance and the post hoc Tukey test. The Kruskal-Wallis test was used to compare the mean fragment lengths between groups. RESULTS: The instruments had significantly lower fatigue resistance in canals with curvatures in the middle and coronal canals compared with those with apical curvatures (p < 0.05). At all tested curvature locations, RecB had superior fatigue resistance compared with WO and Rec (p < 0.05). There were no significant differences between WOG and Rec in canals with curvatures in the middle and coronal canals. The DSC thermograms for RecB exhibit a single exothermic peak during cooling but double endothermic peaks during heating indicating that a two-step phase transformation from martensite to R-phase to austenite takes place. CONCLUSIONS: The reciprocating instruments experience decreased cyclic fatigue resistance when operated in canals with coronal- and middle-third curvatures when compared with curvatures in the apical-third. Instrumenting coronally positioned curvatures with reciprocating files needs to be performed with caution. CLINICAL RELEVANCE: The location of the root canal's curvature influences the fracture resistance of rotary files that are used with reciprocating movements. Therefore, caution needs to be exercised when using reciprocating instruments in canals with coronal or middle curvatures.

INTRODUCTION: The purpose of this study was to evaluate the effect of 5 different curvature locations on the fatigue resistance of thermomechanically treated nickel-titanium (NiTi) files and superelastic NiTi files at body temperature and to document the corresponding phase transformations. METHODS: EndoSequence (ES; Brasseler USA, Savannah, GA), EndoSequence CM (ESCM, Brasseler USA), K3 (SybronEndo, Orange, CA), K3XF (SybronEndo), and Vortex Blue (VB; Dentsply Tulsa Dental Specialties, Tulsa, OK) NiTi files (size 25/.04) were subjected to fatigue tests inside customized artificial canals containing a curvature of 60° and a 3-mm radius. There were 5 different canals based on the location of initial curvature; these included groups in which the distance between the canal orifice and the location of the curvature (DOC) was 5, 6, 8, 10, and 11 mm. The model canal was immersed in water at 37°C ± 1°C. The number of cycles to failure (Nf) was recorded, and the fracture surface of the fragments was examined by scanning electron microscopy. Two unused files of each brand were examined by differential scanning calorimetry. RESULTS: The Nf was highest in the 10-mm and 11-mm DOC groups and lowest with the 5-mm and 6-mm DOC groups (P < .05). ESCM files had the highest fatigue resistance followed by the VB, K3XF, K3, and ES files (P < .05). ESCM files had the highest Nf (P < .05), and ES and K3 files had the lowest Nf (P < .05) depending on the curvature location. Two endothermic peaks were observed on the heating curve of the heat-treated files (ESCM, K3XF, and VB). CONCLUSIONS: The location of the canal curvature had a significant effect on the fatigue resistance of both heat-treated and superelastic NiTi files. The fatigue life of files in the coronal curve was quite short.

MOTIVATION: High-throughput sequencing discovers many naturally occurring disulfide-rich peptides or cystine-rich peptides (CRPs) with diversified bioactivities. However, their structure information, which is very important to peptide drug discovery, is still very limited. RESULTS: We have developed a CRP-specific structure prediction method called Cystine-Rich peptide Structure Prediction (CRiSP), based on a customized template database with cystine-specific sequence alignment and three machine-learning predictors. The modeling accuracy is significantly better than several popular general-purpose structure modeling methods, and our CRiSP can provide useful model quality estimations. BACKGROUND An accurate and valid caries prevention policy is absent in Zhejiang because of insufficient data. Therefore, the aim of this study was to investigate oral health status and related risk factors in 12- to 14-year-old students in Zhejiang, China. MATERIAL AND METHODS Using multi-stage, stratified, random sampling, we recruited a total of 4860 students aged 12 to 14 years old from 6 regions in Zhejiang in this cross-sectional study. Dental caries was measured using the Decayed, Missing and Filled Teeth (DMFT) index and the Significant Caries Index (SiC). Information concerning family background and relevant behaviors was collected in a structured questionnaire. Logistic regression analysis was used to study risk factors related to dental caries. RESULTS The overall prevalence of dental caries was 44% and the mean DMFT and SiC scores were 1.14 and 3.11, respectively. Female students had a higher level of dental caries than male students (P<0.01). The annual increase in caries prevalence was 3% with increasing age, and the DMFT score was 0.15. The results of logistic regression analysis showed that female sex, older age, snacks consumption once or more per day, fair or poor self-assessment of dental health, toothache experience, and dental visits were the most significant risk factors for dental caries, with odds ratios ranging from 1.24 to 2.25 (P<0.01). CONCLUSIONS The prevalence of dental caries in 12- to 14-year-old students in Zhejiang was low, with a tendency to increase compared with previous oral surveys. Female sex, older age, increased sugar intake, poor oral health self-assessment, and bad dental experience were the most important factors increasing dental caries risks.
