RESUMEN

We studied the ability of the polyphenolic complex from Maackia amurensis, the active substance of Maksar, to inhibit the cytopathogenic effect induced by the SARS-CoV-2 and to reduce the concentration of viral RNA in infected Vero E6 cells. Polyphenolic complex showed significant anti-SARS-CoV-2 activity and effectively inhibited viral replication by direct action on viral particles and the early stage of viral infection.

RESUMEN

The quinoid pigment echinochrome A isolated from the sea urchin Scaphechinus mirabilis, the product of its oxidation dehydroechinochrome, and structurally similar antiviral agent oxolin were tested for their ability to inhibit plaque formation induced by herpes simplex virus type 1 (HSV-1) in Vero cells. The tested compounds showed significant anti-HSV-1 activity, mainly due to their direct effect on viral particles and on virus attachment to cells. The antiviral efficacy of the test compounds increased in the following order: oxolin→echinochrome A→dehydroechinochrome.

RESUMEN

Two patients with locally advanced gastric cancer are reported. Both patients underwent colonoscopy in preoperative period. Preoperative examination revealed synchronous colorectal cancer. Preoperative colonoscopy in patients with gastric cancer ensured timely diagnosis of synchronous colorectal cancer and adequate minimally invasive treatment with favorable results.

We studied the effect of echinochrome A on the primary culture of pulmonary fibroblasts under normal conditions and under oxidative stress. Exposure to echinochrome A (100 µM, 6 h) did not affect the production of superoxide radical by fibroblasts, area of their nuclei, and number of nucleoli, but reduced the total area of nucleolar organizer regions against the background of stable proliferative activity of the culture. Two-hour oxidative stress induced by hydrogen peroxide solution (60 µM) increased the generation of superoxide radical, decreased DNA-synthetic activity of fibroblasts, area of fibroblast nuclei, and total area of nucleolar organizer regions. Preliminary administration of echinochrome A significantly attenuated the damaging effect of oxidative stress: the intensity of production of superoxide radicals decreased, DNA-synthetic activity and nucleus area of fibroblasts partially recovered; normalization of the total area of nucleoli was accompanied by an increase in their number.

RESUMEN

Small bowel tumor causes gastrointestinal bleeding in 1-4% of cases. Gastrointestinal bleeding from metastases of renal cell carcinoma is a rare and little-known manifestation of this disease. We report a rare clinical case of a solitary metastasis of clear cell renal cell carcinoma into small bowel in 5 years after nephrectomy. The first symptom was intestinal bleeding. This example emphasizes the need for more thorough examination of patients with symptoms of latent and anamnestic blood loss.

Aim to determine the efficacy of drug aminodihydrophthalazinedione sodium (Galavit) for prevention of progression of the coronavirus infection pulmonary complications: acceleration of regression of pulmonary infiltrates and resolution of COVID-induced pneumonia. 22 patients with medium and severe COVID-induced pneumonia were included in the study. The study included 8 men and 14 women, the average age was 62.17.4 years. Patients with more than one adverse prognostic factor made 82%. Average volume of pulmonary tissue affection (computer tomography CT-2, 2550% of lung volume) was registered in 13 (59.1%) patients, significant volume (CT-3, 5075% of lung volume), in 9 (40.9%) patients. All patients had progressive respiratory failure manifestations due to hypoxemia and related diseases. Aminodihydrophthalazinedione sodium was administered for 714 days from the beginning of disease, at the end of the course of standard complex therapy, in case of preservation of signs of intoxication, negative dynamics according to computer tomography data. Administration of aminodihydrophthalazinedione sodium had a positive effect on the dynamics of clinical scores. The progression of respiratory failure was halted and there was an increase in SpO2 values. According to the control computer tomography data the stabilization of the pulmonary parenchyma affection degree was noted, as well as reduction of the size of the compacted areas in the pulmonary tissue and formation of the picture of organising pneumonia that contributed to reduction of respiratory failure grade. The use of aminodihydrophthalazinedione sodium in complex therapy of COVID-induced pneumonia has a modulating effect on the immune system, prevents the progression of pulmonary tissue affection, promotes regression of infiltration foci, preventing the development of excessive pneumofibrosis and the progression of respiratory failure.

KEY MESSAGE: Overexpression of both native and mutant forms of AtCPK1 in Rubia cordifolia cells increased anthraquinone production and transcript abundance of the RcIPPI, RcOSBL, RcOSBS , and RcICS genes to different extents. Calcium-dependent protein kinases (CDPKs) are involved in various cell processes and are regulated by a calcium signal system. CDPKs also function in plant defense against stress factors such as pathogens, temperature, and salinity. In this study, we compared the effect of heterologous expression of two forms of the Arabidopsis AtCPK1 gene, native and constitutively active (Ca(2+)-independent), on anthraquinone production in transgenic Rubia cordifolia cells. Significant qualitative and quantitative differences were found in the content of anthraquinone derivatives in control and AtCPK1-transgenic calli. Expression of the AtCPK1 gene increased anthraquinone production by 3 and 12 times for native and constitutively active forms, respectively, compared with control cells. In addition, we identified and quantified the expression of genes encoding key enzymes of the anthraquinone biosynthesis pathway, including isochorismate synthase (ICS), o-succinylbenzoate synthase (OSBS), o-succinylbenzoate ligase (OSBL), and isopentenyl diphosphate isomerase (IPPi). In all AtCPK1-transgenic cell lines, expression of ICS, OSBS, OSBL, and IPPi increased considerably at 14-15 days of subculture and decreased at the end of cultivation (30 days). The results suggest that both native and constitutively active AtCPK1 forms induced anthraquinone accumulation at the logarithmic growth stage via enhancement of expression of genes involved in the metabolism of anthraquinones or their regulatory mechanisms.

Histochrome is the medicinal form of echinochrome (2,3,5,6,8-pentahydroxy-7-ethyl-1,4-naphthoquinone). The drug pharmacodynamics and its relationship with metabolism, which was revealed in previous investigations, predetermined the aim of this work: to study excretion of echinochrome and its possible metabolites in urine of rats. Histochrome was introduced to Wistar rats (n = 10) subcutaneously in a dose of 10 mg/kg. Naphthoquinone derivatives were extracted from the acidified urine by ethyl acetate and analyzed by high performance liquid chromatography with UV detection (HPLC/UV) and mass-spectrometric detection (HPLC/MS). It was established that histochrome is completely metabolized in an organism and excreted by kidneys in the form of 3-methoxy-2,5,6,8-tetrahydroxy-7-ethyl-1,4-naphthoquinone and 2-metoxy-3,5,6,8-tetrahydroxy-7-ethyl-1,4-naphthoquinone.

RESUMEN

Hepatoprotective properties of ethanol extract from the roots of Maackia amurensis Ruper et Maxim have been studied on the model of toxic hepatitis induced by carbon tetrachloride damage. It is established that the extract contains daidzein, 7-O-gentobiosides of isoflavonoids genistein, formononetin, pseudobabtige-nin, and 5-O-methylgenistein, and 3-O-gentobiosides of pterocarpans (6aR, 11aR)-maakiain and (6aR, 11aR)-medicarpin. The administration of extract facilitates the restoration of antioxidant protection enzymes activity and reduced glutathione level, decreases the formation of toxic peroxidation products, produces normalizing impact on liver phospholipid pattern, and improves the erythrocyte tolerance to hemolytic agents. The action of isoflavonoids from Maackia amurensis in restoration of metabolic pathways of the liver and removal of toxic stress was more effective as compared to that of the reference hepatoprotector legalon.

RESUMEN

Ovariectomy in rats induces the activation of lipid peroxidation processes, reduces total lipid content, and decreases the accumulation of lysophospholipids in erythrocyte membranes. The administration of Maackia amurensis extract (200 mg/kg, p.o., during 14 days) reduces the level of lipid peroxidation, prevents a decrease in the total lipid content, normalizes the proportion of lipids and proteins, and eliminates the accumulation of lysophospholipids in erythrocyte membranes in ovariectomized rats.

Analysis of intraocular fluid and the vitreous body in 7 rabbits (14 eyes) by high performance liquid chromatography reveals histochrom metabolic products and, possibly, histrochrom itself during intravenous and subconjunctival injections and retrobulbar administration of histochrom through the irrigation system on models of experimental rabbit hyphema and hemophthalmia, which suggests that histochrom is able to penetrate across the blood-ocular barrier.

RESUMEN

Changes in the diameter of brain vessels and intensity of collateral blood flow typical of chronic ischemia were detected by magnetic resonance imaging in senescence-accelerated OXYS rats demonstrating reduced (compared to Wistar rats) research and exploratory activity. Histochrome (antioxidant drug) produced positive effects on cerebral vessels in OXYS rats by stimulating collateral blood flow and acting as a vasodilator agent. Analysis of correlations showed that these effects of histochrome were closely related to its capacity to activate research and exploratory activity and reduce anxiety of OXYS rats in the open field test.

RESUMEN

The changes developing in the perifocal area of hematoma and perspectives of antioxidant and chelate therapy were studied on the model of experimental hemorrhagic stroke and in clinical conditions. Microcirculatory, ischemic and inflammation disturbances with a certain time sequence were found in the perifocal areas. These changes, along with hypostasis and oxidative stress, form the pathobiochemical cascade of changes in hemorrhagic stroke and are potential therapeutic targets. Administering of an antioxidant and chelate drug histochrome reduces the intensity of changes in the perifocal area in the experimental conditions. In clinical conditions, it accelerates the dynamics of brain and meningeal symptoms regression and improves the blood rheological properties.

Presented are the results of the use of histochrome in animals with experimental hemorrhagic stroke. An influence of the compound on cerebral edema and the dynamics of hemoglobulin converting as well as a role of MRT in diagnostic of experimental hemorrhagic stroke in different periods of the disease are discussed.

The carrying out tests of water-extracts of carline thistle (plant pink, Caryophyllaceae) in Kazakhstan (extract 1) and gathered in the Primorye region (extract 2). The method of spectral and thin layer chromatography silicagel plates contain phenolys components. It is found out that both extracts contain the same bonds with the max, uptake of long waves lambda = 282 hm and lambda = 260 hm. Besides, extract 2 has significant quantity of substances with the max, uptake lambda = 272 hm and substance with the max, uptake lambda = 330 hm. When determining with the help of standard tap the chemical nature of the substances, which have antioxidant characteristics, we settled out that antioxidant of soapwort possibly can be coffee acid and nonpolar flavonoid of unstated nature.

RESUMEN

The efficacy of a new bioantioxidant histochrome is studied in 92 patients (98 eyes). The drug was injected subconjunctivally or parabulbarly in a dose of 0.5 ml 0.02% solution daily, 7-10 injections per course. The clinical effect of histochrome is determined by the localization, duration, and extent of the process. The drug was the most effective in the treatment of intraocular hemorrhages which occurred no more than 7 days before. Electroretinography demonstrated pronounced retinoprotective properties of the drug which improved the electrophysiological parameters in degenerative processes in the retina and optic nerve. Hence, histochrome is characterized by hemoresorption, retinoprotective, and antioxidant properties and is recommended for the treatment of ocular diseases involving metabolic disorders in the retina, vascular membrane, and cornea.

The effect of the antioxidant histochrom on the size of the myocardial infarction and activity of the cardioselective enzyme creatine phosphokinase was studied in 45 patients with acute myocardial infarction during thrombolytic therapy. Changes in blood plasma histochrom concentration were studied in 7 patients with myocardial infarction after a single intravenous infusion of 100 mg of the drug. The pharmacokinetic parameters of the drug are shown. The use of histochrom in thrombolytic treatment of acute myocardial infarction lead to limitation of the necrotic zone and increase of myocardial contractility, and results in a more favorable course of the processes of left-ventricular remodeling.

RESUMEN

Lipid peroxidation and blood rheology were assessed in the course of treatment with antioxidant histochrome and placebo of 66 unstable angina pectoris (UAP) patients. Histochrome has a high antioxidant activity and effectively inhibits accumulation of lipoperoxides in plasma and red cells of UAP patients, promotes stabilization of blood cell function and structures, reduces red cell and platelet aggregation.
